Isoginkgetin protects chondrocytes and inhibits osteoarthritis through NF-κB and P21 signaling pathway.

OBJECTIVE: Osteoarthritis (OA) is the most prevalent chronic articular disease in adults. The degree of cartilage degradation and matrix depletion in OA have been substantially connected with chondrocyte inflammatory response. Consequently, pharmacological anti-inflammatory agents provide OA patients a new therapeutic option. Isoginkgetin (IGK), a bioactive bioflavonoid derived from the medicinal herb Ginkgo Biloba, defends against obesity-induced heart diastolic dysfunction and harmful remodeling. Whether IGK has a regulatory effect on OA remains unknown. This study investigated whether IGK c

Theaflavin-3,3'-digallate represses osteoclastogenesis and prevents wear debris-induced osteolysis via suppression of ERK pathway.

UNLABELLED: Peri-implant osteolysis (PIO) and the following aseptic loosening is the leading cause of implant failure. Emerging evidence suggests that receptor activator of nuclear factor kappa-B ligand (RANKL)-induced osteoclast formation and osteoclastic bone resorption are responsible for particle-stimulated PIO. Here, we explored the effect of theaflavin-3,3'-digallate (TF3) on titanium particle-induced osteolysis in vivo and in vitro. Twenty-eight male C57BL/6 mice were randomly separated into four groups: sham control (sham), titanium particles only (titanium), titanium particles with lo

Procyanidins-crosslinked heart valve matrix: anticalcification effect.

Calcification is one of the key factors for short durability of glutaraldehyde-crosslinked bioprosthetic heart valves. We have shown previously that procyanidins (PC)-crosslinked heart valve matrix has low cytotoxicity, perfect mechanical properties, and ideal stability. The aim of this study is to investigate the anticalcification effect of procyanidins and its crosslinked heart valves. Porcine aortic heart valve leaflets were decellularized and crosslinked with PC solution. The inhibition of calcification on PC-crosslinked heart valves was evaluated by soaking valves in simulated body fluid.

Tea enhances insulin activity.

The most widely known health benefits of tea relate to the polyphenols as the principal active ingredients in protection against oxidative damage and in antibacterial, antiviral, anticarcinogenic, and antimutagenic activities, but polyphenols in tea may also increase insulin activity. The objective of this study was to determine the insulin-enhancing properties of tea and its components. Tea, as normally consumed, was shown to increase insulin activity >15-fold in vitro in an epididymal fat cell assay. Black, green, and oolong teas but not herbal teas, which are not teas in the traditional sen

Procyanidin B2 alleviates triphenyl phosphate-induced colon injury through Tmigd1/NF-κB/NLRP3 signal axis.

Procyanidin B2 (PCB2), a natural dietary polyphenol with anti-inflammatory and antioxidant properties, demonstrates potential in mitigating chemical-induced toxicity. This study aimed to investigate the mechanism of triphenyl phosphate (TPHP)-induced colon injury and the protective role of PCB2. In a mouse model, TPHP exposure caused colon shortening, barrier damage, inflammation, and oxidative stress, all of which were dose-dependently reversed by PCB2. RNA-seq analysis identified the NF-κB signaling pathway as a key pathway and highlighted Tmigd1 as a core differential gene. TPHP downregulat

Theaflavin suppresses necroptosis by attenuating RIPK1-RIPK3-MLKL signaling and mitigates cisplatin-induced kidney injury in mice.

Necroptosis is a lytic form of regulated cell death (RCD) that is dependent on receptor-interacting protein kinase 3 (RIPK3) and mixed lineage kinase domain like pseudokinase (MLKL). This form of RCD has been implicated in various inflammatory diseases and organ injuries including cisplatin-induced acute kidney injury (AKI), thus representing a therapeutic target for such diseases. Theaflavin is an ingredient of black tea that exhibits beneficial effects on human health and has been shown to regulate pyroptosis, but its effects on necroptosis and cisplatin-induced AKI remain unclear.
