Spatial and Temporal Analysis of Plasmodium knowlesi Infection in Peninsular Malaysia, 2011 to 2018.

The life-threatening zoonotic malaria cases caused by Plasmodium knowlesi in Malaysia has recently been reported to be the highest among all malaria cases; however, previous studies have mainly focused on the transmission of P. knowlesi in Malaysian Borneo (East Malaysia). This study aimed to describe the transmission patterns of P. knowlesi infection in Peninsular Malaysia (West Malaysia). The spatial distribution of P. knowlesi was mapped across Peninsular Malaysia using Geographic Information System techniques. Local indicators of spatial associations were used to evaluate spatial patterns

Plasmodium knowlesi - an emerging pathogen.

Ten years have passed since the publication of a large focus of Plasmodium knowlesi infections in the human population. The discovery was made during a molecular investigation of atypical P. malariae cases in the Kapit Health Division, Sarawak, Malaysian Borneo. Patients were more symptomatic with higher parasite counts than expected in P. malariae infections. The investigation found only P. knowlesi DNA present in patient blood samples. Morphological similarity had allowed P. knowlesi to masquerade as P. malariae during routine diagnostic microscopy for malaria. P. knowlesi, a malaria parasit

Inter-Population Genetic Diversity and Clustering of Merozoite Surface Protein-1 (pkmsp-1) of Plasmodium knowlesi Isolates from Malaysia and Thailand.

The genetic diversity of pkmsp-1 of Malaysian Plasmodium knowlesi isolates was studied recently. However, the study only included three relatively older strains from Peninsular Malaysia and focused mainly on the conserved blocks of this gene. In this study, the full-length pkmsp-1 sequence of recent P. knowlesi isolates from Peninsular Malaysia was characterized, along with Malaysian Borneo and Thailand pkmsp-1 sequences that were retrieved from GenBank. Genomic DNA of P. knowlesi was extracted from human blood specimens and the pkmsp-1 gene was PCR-amplified, cloned, and sequenced. The sequen

Seasonal and Spatial Dynamics of the Primary Vector of Plasmodium knowlesi within a Major Transmission Focus in Sabah, Malaysia.

BACKGROUND: The simian malaria parasite Plasmodium knowlesi is emerging as a public health problem in Southeast Asia, particularly in Malaysian Borneo where it now accounts for the greatest burden of malaria cases and deaths. Control is hindered by limited understanding of the ecology of potential vector species. METHODOLOGY/PRINCIPAL FINDINGS: We conducted a one year longitudinal study of P. knowlesi vectors in three sites within an endemic area of Sabah, Malaysia. All mosquitoes were captured using human landing catch. Anopheles mosquitoes were dissected to determine, oocyst, sporozoites an

Early hominin arrival in Southeast Asia triggered the evolution of major human malaria vectors.

Some species of the Leucosphyrus Group of Anopheles mosquitoes in Southeast Asia are highly anthropophilic and efficient vectors of human malaria parasites, while others primarily feed on non-human primates (NHP) and transmit NHP malaria parasites. The evolutionary history of this group, particularly the origin of anthropophily, was studied using phylogenomic analysis of 2,657 high-confidence nuclear single-copy orthologous genes and 13 mitochondrial protein coding genes from 40 individuals of 11 species. Molecular dating and ancestral state reconstruction revealed that monkey-feeding is ances

Fish tapeworms (Cestoda) in the molecular era: achievements, gaps and prospects.

The tapeworms of fishes (Chondrichthyes and Actinopterygii) account one-third (1670 from around 5000) of the total tapeworm (Platyhelminthes: Cestoda) species diversity. In total 1186 species from 9 orders occur as adults in elasmobranchs (sharks, rays and chimaeras), and 484 species from 8 orders mature in ray-finned fishes (referred to here as teleosts). Teleost tapeworms are dominated by freshwater species (78%), but only 3% of elasmobranch tapeworms are known from freshwater rays of South America and Asia (Borneo). In the last 2 decades, vast progress has been made in understanding species

Two Genetically Distinct Plasmodium knowlesi Duffy Binding Protein Alpha Region II (PkDBPαII) Haplotypes Demonstrate Higher Binding Level to Fy(a+b+) Erythrocytes than Fy(a+b--) Erythrocytes.

Invasion of human erythrocytes by merozoites of Plasmodium knowlesi involves interaction between the P. knowlesi Duffy binding protein alpha region II (PkDBPαII) and Duffy antigen receptor for chemokines (DARCs) on the erythrocytes. Information is scarce on the binding level of PkDBPαII to different Duffy antigens, Fya and Fyb. This study aims to measure the binding level of two genetically distinct PkDBPαII haplotypes to Fy(a+b-) and Fy(a+b+) human erythrocytes using erythrocyte-binding assay. The binding level of PkDBPαII of Peninsular Malaysian and Malaysian Borneon haplotypes to erythrocyt

Cryptic Diversity and Demographic Expansion of Plasmodium knowlesi Malaria Vectors in Malaysia.

Although Malaysia is considered free of human malaria, there has been a growing number of Plasmodium knowlesi cases. This alarming trend highlighted the need for our understanding of this parasite and its associated vectors, especially considering the role of genetic diversity in the adaptation and evolution among vectors in endemic areas, which is currently a significant knowledge gap in their fundamental biology. Thus, this study aimed to investigate the genetic diversity of Anopheles balabacensis, Anopheles cracens, Anopheles introlatus, and Anopheles latens-the vectors for P. knowlesi mala

Severe malaria - a case of fatal Plasmodium knowlesi infection with post-mortem findings: a case report.

BACKGROUND: Zoonotic malaria caused by Plasmodium knowlesi is an important, but newly recognized, human pathogen. For the first time, post-mortem findings from a fatal case of knowlesi malaria are reported here. CASE PRESENTATION: A formerly healthy 40 year-old male became symptomatic 10 days after spending time in the jungle of North Borneo. Four days later, he presented to hospital in a state of collapse and died within two hours. He was hyponatraemic and had elevated blood urea, potassium, lactate dehydrogenase and amino transferase values; he was also thrombocytopenic and eosinophilic. De

G6PD variants but not Filipino beta-thalassemia are associated with reduced risk of Plasmodium knowlesi infection in Sabah, Malaysia.

BACKGROUND: The emergence of Plasmodium knowlesi as the predominant cause of malaria in Malaysian Borneo raises important questions about protective genetic factors in endemic populations. This study investigates two common erythrocyte polymorphisms, glucose-6-phosphate dehydrogenase (G6PD) deficiency and Filipino β-thalassemia, for association with P. knowlesi infection risk. METHODS: A retrospective case-control study compared 106 PCR-confirmed P. knowlesi cases with 89 controls from Sabah, Malaysia. Genotyping used multiplex PCR (8 major Southeast Asian G6PD variants) and gap-PCR (Filipino

In silico profiling of the plasmodium knowlesi 32 kDa antigen: Diversity, epitope prediction, and structural modeling.

The emergence of Plasmodium knowlesi malaria in Southeast Asia, particularly Malaysia, necessitates effective interventions. However, high polymorphism often limits the utility of key merozoite surface proteins (MSPs) as viable vaccine candidates. Pk32, a predicted glycosylphosphatidylinositol (GPI)-anchored protein in P. knowlesi, shares homology with the relatively conserved Plasmodium vivax Pv32, suggesting it may be a promising vaccine candidate. We analyzed the genetic diversity, polymorphism, and signatures of natural selection and 3-D structure using 39 full-length Pk32 sequences, prima

Ethnobotany-guided exploration of plant-derived photosensitizer candidates for photodynamic therapy.

The integration of photosensitization with Indigenous knowledge offers a promising framework for exploring more sustainable therapeutic approaches. This review explores how the ethnobotanical knowledge of the Dayak communities of Borneo (Indonesia), particularly the traditional use of brightly coloured medicinal plants for dermatological treatments, may provide a biologically informed framework for prioritizing plant-derived photosensitizer candidates. The biosynthetic pathways and photochemical potential of key metabolites associated with selected Dayak medicinal taxa (Curcuma longa, Etlinger

Neuroanatomy education through the years: Revisiting the teaching slides used by Sir Wilfrid Edward Le Gros Clark.

Sir Wilfrid Edward Le Gros Clark (1895-1971) was a pioneering British anatomist, physician, and evolutionary biologist whose legacy spans the foundational development of twentieth century neuroanatomy, primate biology, and medical education. From his formative experiences as a young medical officer in the First World War to his later academic ascendancy at the University of Oxford, Le Gros Clark exemplified a rare fusion of clinical insight, anatomical precision, and philosophical curiosity. His tenure in Borneo, where he studied primates in their natural habitats, enriched his evolutionary pe

Two new species of Phoreiobothrium Linton, 1889 (Cestoda: Onchoproteocephalidea) off southern Iran, completing the puzzle of Phoreiobothrium faunas in Rhizoprionodon acutus species complex.

It has been shown that the milk shark, Rhizoprionodon acutus (Rüppell), is probably a complex of four narrowly distributed cryptic species. To confirm this hypothesis, the oioxenous species of the onchoproteocephalid genus Phoreiobothrium Linton, 1889 was recently used to recognize each shark species of this species complex so that P. nadiae Caira and Jensen, 2015, P. swaki Caira and Jensen, 2015, and P. jahki Caira and Jensen, 2015 were described respectively from Rhizoprionodon cf. acutus 1 off Senegal, R. cf. acutus 2 off northern Australia, and R. cf. acutus 3 off Borneo.
