BNCT (Boron Neutron Capture Therapy) is a binary radiotherapeutic modality in which high LET (Linear Energy Transfer) particles are generated from 10B(n,α)7Li reaction, ideally within boron-loaded tumour cells, so the therapeutic outcome depends critically on the pharmacokinetics and biodistribution of boron carriers. In this review, boron-containing agents for BNCT, with a focus on ADMET (absorption, distribution, metabolism, excretion and toxicity) and model-informed design, were examined. Low-MW (low-molecular-weight) compounds, peptide conjugates, polymeric and nanostructured platforms and

The exploration of innovative molecular scaffolds offers an exciting opportunity to venture into a previously unexplored chemical space. Boron clusters, such as carboranes, exhibit three-dimensional (3D) cage structures for exopolyhedral derivatization along multiple directions. This makes carboranes hold great potential as bioisosteric mimetics of benzene. However, the lack of efficient synthetic strategies for the regioselective functionalization of up to ten B-H sites has considerably hampered the broad application of carboranes as benzene bioisosteres. Herein, we report a novel reaction pa

The Li(p,n) and Be(p,n) reactions are widely used as accelerator-based neutron sources for boron neutron capture therapy (BNCT). Because the energy of the neutrons produced by these reactions is high for BNCT, a moderator is required to reduce the energy to the epithermal energy region. The neutron yields and energy spectra derived from the Li(p,n) and Be(p,n) reactions vary with the incident proton energy; a high proton energy results in a high neutron yield. However, a high proton energy can also increase the neutron energy, and in this case, a high neutron energy requires a thick moderator,

Clusters of main group elements, such as phosphorus, arsenic, germanium, and tin - called Zintl clusters - have been known for more than a century. However, their application in main group catalysis is largely unknown. Here, we tether boranes to a seven-atom phosphorus cluster ({C8H14}BCH2CH2SiMe2)3P7 (2) and we demonstrate Lewis acid catalysis as proof-of-principle that boron chemistry can be mapped onto clusters using this method. Catalyst 2 was employed to mediate key organic transformations, including the hydroboration of carbodiimides, isocyanates, ketones, alkenes, alkynes, and nitriles.

The development of tumor-targeting agents is of great significance for cancer therapy. Despite the promising potential of Boron Neutron Capture Therapy (BNCT), efficient and selective accumulation of boron-10 agents in cancer cells remains challenging. Conjugation of carborane with tumor-targeting ligands provides an alternative strategy for selective accumulation in the vicinity of cancer cells. In this study, we designed and synthesized aptamer-carborane derivatives, named AS1411-CB conjugates, via covalent linkage between a dodecahydro-arachno-bis(acetonitrile)decaborane and the DNA aptamer

PURPOSE: Meningiomas and schwannomas associated with neurofibromatosis 2 (NF2) are difficult to control by microsurgery and stereotactic radiotherapy alone. Boron neutron capture therapy (BNCT) is a chemically targeted form of radiotherapy requiring increased concentration of boron-10 in tumour tissue. PET with the boron carrier 4-borono-2-[(18)F]fluoro-L-phenylalanine ([(18)F]FBPA) allows investigation of whether 4-borono-L-phenylalanine (BPA) concentrates in NF2 tumours, which would make BNCT feasible.
METHODS: We studied dynamic uptake of [(18)F]FBPA in intracranial meningiomas (n=4) and s

This study systematically investigated the radiobiological effects of an accelerator-based boron neutron capture therapy (AB-BNCT) system on SAS cells, a human tongue squamous cell carcinoma (TSCC) cell line. In vitro experimental results showed that L-10B-BPA incubation within 12 h had no significant effect on cell viability. At 24 and 48 h post-irradiation, the 26N20 group exhibited 11 % and 31.91 % reductions in cell viability, accompanied by 82.15 % and 68.34 % decreases in proliferation rates, respectively, compared to controls. In the 26N10 group, 48-h post-irradiation analysis revealed

4-Borono-L-phenylalanine (BPA) is a key 10B carrier used in boron neutron capture therapy (BNCT), while its PET tracer analogue, 4-borono-2-18F-fluoro-L-phenylalanine (18F-BPA), enables non-invasive visualization of tumour boron uptake. Since BNCT efficacy depends on precise tumour boron accumulation, we evaluated whether 18F-BPA mirrors BPA's transport and biodistribution. In vitro, BPA exhibited a highly consistent uptake profile with its non-radioactive fluorinated analogue, 2-19F-4-borono-L-phenylalanine (19F-BPA), across nine cancer cell lines (r = 0.9455, P < 0.001) and tri-iodothyronine

We studied the effects of a single administration of two types of boron particles coated with silane-PEG and produced either by laser fragmentation of boron-10 micropowder or by laser ablation of a solid target of enriched boron on body weight, the weight and histological structure of organs (kidney, liver, spleen, heart, lungs, and brain) and on blood parameters in immunodeficient SCID mice. The particles were injected subcutaneously and intravenously at doses of 10, 30, and 100 mg/kg. We observed a dose-dependent effect of boron particles on the lung and spleen weights, with larger particles

Reaction-enhanced radiotherapy (RER) aims to improve the therapeutic ratio of particle therapy by exploiting isotope-specific nuclear reactions that generate localized high-linear energy transfer secondary radiation. Absorbed dose remains the central metric for radiotherapy planning and evaluation; however, RER introduces additional verification requirements related to nuclear-reaction data, mixed-field transport, isotope delivery, and measurable secondary-field observables. This review frames RER as a verification-oriented physics problem built on three interdependent pillars: covariance-awar

The synthesis of the first conjugates of acridine with cobalt bis(dicarbollide) are reported. A novel 9-azido derivative of acridine was prepared through the reaction of 9-methoxyacridine with N3CH2CH2NH2, and its solid-state molecular structure was determined via single-crystal X-ray diffraction. The azidoacridine was used in a copper (I)-catalyzed azide-alkyne cycloaddition reaction with cobalt bis(dicarbollide)-based terminal alkynes to give the target 1,2,3-triazoles. DNA interaction studies via absorbance spectroscopy showed the weak binding of the obtained conjugates with DNA. The antipr

INTRODUCTION: To plan the optimal BNCT for patients with malignant cerebral glioma, estimation of the ratio of boron concentration in tumor tissue against that in the surrounding normal brain (T/N ratio of boron) is important. We report a positron emission tomography (PET) imaging method to estimate T/N ratio of tissue boron concentration based on pharmacokinetic analysis of amino acid probes.
METHODS: Twelve patients with cerebral malignant glioma underwent 60 min dynamic PET scanning of brain after bolus injection of (18)F-borono-phenyl-alanine (FBPA) with timed arterial blood sampling. Usi

Boron neutron capture therapy (BNCT) is a binary cancer treatment strategy that relies on the selective delivery of a 10B-containing drug followed by thermal neutron irradiation, generating localized high-LET particles that destroy tumor cells. L-BPA is the most recent and currently the only marketed boron carrier; however, its poor tumor retention and low solubility significantly limit therapeutic efficacy. Thus, developing improved L-BPA derivatives, as well as tools for efficient monitoring of L-BPA biodistribution, is critically important. Herein, we report a turn-on fluorescent probe, WF3

BACKGROUND: Boron neutron capture therapy (BNCT) is a radiotherapeutic approach that can destroy cancer cells while sparing the surrounding normal cells. Currently, boronophenylalanine (BPA) is the most common boron delivery agent used in BNCT for treating recurrent cancers of the head and neck, gliomas, and melanomas. On the other hand, valproic acid (VPA) is one of the representative class I histone deacetylase inhibitors (HDACi), which is a promising sensitizer for cancer therapies. In this study, we aimed to verify whether VPA could induce an enhanced effect in destroying melanoma cells in

Boron has emerged as a versatile element in medicinal chemistry because its empty p-orbital enables reversible covalent interactions with biologically important nucleophiles while supporting diverse, tunable molecular architectures. Following approval of bortezomib in 2003, boron-containing therapeutics have expanded beyond proteasome inhibition to infectious disease, inflammation, thrombosis, metabolic and cardiovascular disease, cancer immunotherapy, and boron neutron capture therapy (BNCT). This chapter critically surveys boron-containing candidates that have entered clinical trials, organi

Cancer remains a major health challenge, with the effectiveness of chemotherapy often limited by its lack of specificity and systemic toxicity. Nanotechnology, particularly in targeted drug delivery, has emerged as a key innovation to address these limitations. This study introduces lipoic acid-boronophenylalanine (LA-BPA) derivatives that incorporate short-chain polyethylene glycol (PEG) as a spacer. These derivatives distinctively self-assemble into vesicles under specific pH conditions, exhibiting a pH-dependent reversible assembly characteristic. Notably, these vesicles target cancer cells

OBJECTIVE: The progress from Boron Neutron Capture Therapy (BNCT) development urged us to explore new targeted and selective boron carriers. Firstly, we reported the successful synthesis of CCB-2 which exerts a cytotoxic effect against triple negative breast cancer (TNBC) cells. We introduced the new modification of CCB-2 with sugar and alcohol sugars to enhance its solubility in hoping to increase cellular uptake.
METHODS: CCB-2 fructose complex (CCB-2-F), CCB-2 sorbitol complex (CCB-2-Sor), and CCB-2 xylitol complex (CCB-2-Xy) were obtained with small size within nano-specific particle. All

We have established methods of targeting a sufficient number of 10B atoms on human melanoma cells to allow selective destruction of the cancer cells by thermal neutron irradiation. Thermal neutron capture therapy (NCT)1-3 requires the presence of at least 10(9) 10B atoms on each target cell for specific killing of that cell without injuring normal tissues. In order to accumulate an adequate number of 10B atoms on target cells, we first created an effective compound containing 12 atoms of 10B per molecule (10B12-chlorpromazine) and 10B-dopa analogue (10B1-paraboronophenylalanine). In the presen

Boron neutron capture therapy (BNCT) is an encouraging therapeutic modality for cancer treatment. Prostate-specific membrane antigen (PSMA) is a cell membrane protein that is abundantly overexpressed in prostate cancer and can be targeted with radioligand therapies to stimulate clinical responses in patients. In principle, a spatially targeted neutron beam together with specifically targeted PSMA ligands could enable prostate cancer-targeted BNCT. Thus, we developed and tested PSMA-targeted poly(lactide-co-glycolide)-block-poly(ethylene glycol) (PLGA-b-PEG) nanoparticles (NPs) loaded with carb

Pathobiologic characteristics of cerebral and cutaneous melanoma may cause an increase in mortality resulting from brain metastases in advanced melanoma patients, in addition to anatomic lesions and biological effects caused by the tumor location. We established intracranial and subcutaneous melanoma models using cultured malignant cells derived from amelanotic melanoma. The median survival times in a mouse model with intracranial tumors was 20 days, but a mouse model with subcutaneous tumors did not show cachexia until they were killed 28 days after inoculation with tumor cells.
