Caffeic acid phenethyl ester attenuates liver fibrosis via inhibition of TGF-β1/Smad3 pathway and induction of autophagy pathway.

Caffeic acid phenethyl ester (CAPE) has been reported to possess the hepatoprotective effect. This study was to investigate the mechanism underlying CAPE against liver fibrosis in a liver fibrosis model induced by toxic carbon tetrachloride (CCl4) in male Sprague-Dawley rats and in vitro in CAPE (5 μM, 10 μM, 15 μM) treated hepatic stellate cells (HSC-T6). We found that CAPE treatment remarkably attenuated CCl4-induced liver fibrosis by blocking the activation of HSCs as determined by the expression alternation of transforming growth factor (TGF)-β1, phosphorylated Smad3 (p-Smad3), collage I,

Effect of caffeic acid phenethyl ester on oxidant and anti-oxidant status of liver and serum in a rat model with acute methanol intoxication.

BACKGROUND: Methanol toxicity is one of the major public health problems because it can cause severe morbidity and mortality. Methanol intoxication causes changes in the balance between the production of free radicals and antioxidant capacity. AIMS: We aimed to investigate the effects of caffeic acid phenethyl ester (CAPE) on the total oxidant status, total antioxidant status (TAS), and oxidative stress index (OSI) parameters of the liver and the serum in a rat model of acute methanol intoxication. MATERIALS AND METHODS: Rats were treated with intraperitoneal (i.p.) Methotrexate (MTX) for 7

Dietary caffeic acid mitigates Streptococcus agalactiae pathogenesis in Asian seabass (Lates calcarifer): antibacterial efficacy, immunomodulatory gene regulation, and hepatoprotection.

Caffeic acid (CA) has recently gained attention as a natural compound with antimicrobial and immunomodulatory potential relevant to aquaculture health management. However, its functional efficacy, optimal dietary inclusion level, and protective capacity against Streptococcus agalactiae infection in Asian seabass (Lates calcarifer) remain insufficiently characterized. This study evaluated the in vitro antibacterial activity of CA, assessed its short-term dietary safety (n = 150 fish), examined growth and immune responses (n = 400 fish) during a 4-week feeding trial, and determined its protectiv

Combined Treatment of Cisplatin With Caffeic Acid Phenethyl Ester Demonstrates Additive Growth Inhibition on Non-small Cell Lung Cancer Cells.

BACKGROUND/AIM: Non-small cell lung cancer (NSCLC) accounts for approximately 85% of all lung cancer cases. While cisplatin remains standard treatment for NSCLC, development of drug resistance causes failure of treatment. Caffeic acid phenethyl ester (CAPE) has been reported to cause synergistic inhibitory effects with cisplatin in other cancer types. In this study, we investigated whether combination of cisplatin with CAPE enhances the suppression of NSCLC cell proliferation and survival. MATERIALS AND METHODS: Proliferation assays, MTT assays, and flow cytometry were performed to examine th

Dual dynamic covalent hydrogel enabling pH-responsive release of carboxymethyl chitosan and caffeic acid for synergistic antibacterial activity and wound healing.

Hydrogels have shown broad application prospects in the treatment of complex wounds due to their good biocompatibility and adhesion. In this study, we developed the CMCS-FPBA-CA hydrogel by incorporating a dual dynamic covalent cross-linking system based on Schiff base and borate ester bonds. This design confers pH-responsive behavior to the acidic wound microenvironment. Under acidic conditions, the cleavage of Schiff base and boronate ester bonds enables the release of carboxymethyl chitosan (CMCS) and caffeic acid (CA), which act synergistically to inhibit bacterial growth and accelerate wo

A multimodal caffeic acid-derived alkyl-amide antidiabetic agent: targeting α-glucosidase, α-amylase, oxidative stress, and protein glycation.

Historically, natural products have been a primary source of new drugs and lead compounds. However, their direct application as therapeutic agents remains limited. Given the multifactorial nature of diabetes, therapeutic agents with multi-targeting properties offer a promising solution for effective management. Herein, we developed a library of seven lipophilic derivatives of caffeic acid (CA) and evaluated their multimodal antidiabetic activities. Among these, the most potent compound, CA14, showed an IC50 of 1.94 μM, representing a 113-fold improvement over acarbose (IC50 219.70 μM). Further

Caffeic acid mitigates restenosis post-angioplasty via oxidative stress and inflammation reduction and mitochondrial protection in vascular smooth muscle cells.

Angioplasty is the primary treatment for lower extremity arteriosclerosis obliterans, but the incidence of post-operative restenosis remains high. This condition is characterized by abnormal dedifferentiation of vascular smooth muscle cells (VSMCs), leading to neointimal formation. Caffeic acid (CA) is known for its antioxidant and anti-atherosclerotic properties. This study aims to investigate the effect of CA on neointimal formation after injury and its underlying mechanisms. In vitro experiments showed that CA (100 μM) significantly inhibited PDGF-induced proliferation, migration, and dedif

Caffeic acid phenethyl ester restores mitochondrial homeostasis against peritoneal fibrosis induced by peritoneal dialysis through the AMPK/SIRT1 pathway.

Increasing evidence suggests that peritoneal fibrosis induced by peritoneal dialysis (PD) is linked to oxidative stress. However, there are currently no effective interventions for peritoneal fibrosis. In the present study, we explored whether adding caffeic acid phenethyl ester (CAPE) to peritoneal dialysis fluid (PDF) improved peritoneal fibrosis caused by PD and explored the molecular mechanism. We established a peritoneal fibrosis model in Sprague-Dawley rats through intraperitoneal injection of PDF and lipopolysaccharide (LPS). Rats in the PD group showed increased peritoneal thickness, s

Caffeic acid recovers ischemia-induced synaptic dysfunction without direct effects on excitatory synaptic transmission and plasticity in mouse hippocampal slices.

Caffeic acid is a polyphenolic compound present in a vast array of dietary components. We previously showed that caffeic acid reduces the burden of brain ischemia joining evidence by others that it can attenuate different brain diseases. However, it is unknown if caffeic acid affects information processing in neuronal networks. Thus, we now used electrophysiological recordings in mouse hippocampal slices to test if caffeic acid directly affected synaptic transmission, plasticity and dysfunction caused by oxygen-glucose deprivation (OGD), an in vitro ischemia model. Caffeic acid (1-10 μM) was d

Caffeic Acid Phenethyl Ester Induces Vascular Endothelial Growth Factor Production and Inhibits CXCL10 Production in Human Dental Pulp Cells.

The survival rate of root non-vital teeth is lower than that of vital teeth. Therefore, to preserve the dental pulp is very important. The vascular endothelial growth factor (VEGF) is the most potent angiogenic factor involved in the vitality of dental pulp including reparative dentin formation. Caffeic acid phenethyl ester (CAPE) is a physiologically active substance of propolis and has some bioactivities such as anti-inflammatory effects. However, there are no reports on the effects of CAPE on dental pulp inflammation. In this study, we investigated the effects of CAPE on VEGF and inflammato

Caffeic acid and resveratrol ameliorate cellular damage in cell and Drosophila models of spinocerebellar ataxia type 3 through upregulation of Nrf2 pathway.

Polyglutamine (polyQ)-expanded mutant ataxin-3 protein, which is prone to misfolding and aggregation, leads to cerebellar neurotoxicity in spinocerebellar ataxia type 3 (SCA3), an inherited PolyQ neurodegenerative disease. Although the exact mechanism is unknown, the pathogenic effects of mutant ataxin-3 are associated with dysregulation of transcription, protein degradation, mitochondrial function, apoptosis, and antioxidant potency. In the present study we explored the protective role and possible mechanism of caffeic acid (CA) and resveratrol (Res) in cells and Drosophila expressing mutant

Caffeic Acid phenethyl ester protects blood-brain barrier integrity and reduces contusion volume in rodent models of traumatic brain injury.

A number of studies have established a deleterious role for inflammatory molecules and reactive oxygen species (ROS) in the pathology of traumatic brain injury (TBI). Caffeic acid phenethyl ester (CAPE) has been shown to exert both antioxidant and anti-inflammatory effects. The primary objective of the present study was to examine if CAPE could be used to reduce some of the pathological consequences of TBI using rodent models. Male Sprague-Dawley rats and C57BL/6 mice were subjected to controlled cortical impact (CCI) injury. Blood-brain barrier (BBB) integrity was assessed by examining claudi

Caffeic acid phenethyl ester (CAPE) attenuates cerebral vasospasm after experimental subarachnoidal haemorrhage by increasing brain nitric oxide levels.

BACKGROUND: Cerebral vasospasm, a medical complication of aneurysmal subarachnoid hemorrhage (SAH), is associated with high morbidity and mortality rates, even after the aneurysm has been secured surgically or endovascularly. Evidence accumulated during the last decade suggest that scavenging a vasodilator, nitric oxide (NO), by superoxide anions (O(2)(-)), and activating a strong vasoconstructor, protein kinase C (PKC), are the two most important mechanisms in the pathogenesis of vasospasm. Our aim in this study was to determine whether caffeic acid phenethyl ester (CAPE), a non-toxic oxygen
