Four thermostatic steps: A novel CRISPR-Cas12-based system for the rapid at-home detection of respiratory pathogens.

The outbreak of coronavirus disease 2019 (COVID-19) in 2019 has severely damaged the world's economy and public health and made people pay more attention to respiratory infectious diseases. However, traditional quantitative real-time polymerase chain reaction (qRT-PCR) nucleic acid detection kits require RNA extraction, reverse transcription, and amplification, as well as the support of large-scale equipment to enrich and purify nucleic acids and precise temperature control. Therefore, novel, fast, convenient, sensitive and specific detection methods are urgently being developed and moving to

Simplified Inhouse Nanoweb Membrane Enrichment Coupled Viral Whole Genome Shotgun Metagenomics Approach for Waste Water Surveillance.

Wastewater/sewage represents a highly complex environmental matrix and harbors various viruses including viruses of epidemic and pandemic potential like Influenza A and, SARS-CoV-2. The limitations of available methods for sample concentration are cost, efficiency and time. NGS- metagenomics offer sample to virus genomic characterization for even non-cultivable viruses at highest resolution. In this study, we have collected samples from sewage treatment plants located in Gwalior region of Central India (December 2023 to March 2025). In this study a novel in house nanoweb membrane-based sample

The Natural Product Eugenol Is an Inhibitor of the Ebola Virus In Vitro.

PURPOSE: Since the 2014 Ebola virus (EBOV) outbreak in West Africa there has been considerable effort towards developing drugs to treat Ebola virus disease and yet to date there is no FDA approved treatment. This is important as at the time of writing this manuscript there is an ongoing outbreak in the Democratic Republic of the Congo which has killed over 1000. METHODS: We have evaluated a small number of natural products, some of which had shown antiviral activity against other pathogens. This is exemplified with eugenol, which is found in high concentrations in multiple essential oils, and

Synergistic control of viral persistence via wettability and ion release on antiviral coatings.

Antiviral coating design remains largely empirical due to the lack of quantitative principles linking interfacial properties to viral persistence under realistic conditions. Here, we establish a survival time-based evaluation framework that captures the drying dynamics of 2-μL virus-containing microdroplets and enables quantitative assessment of antiviral coating performance using viral survival time (duration of infectivity) and survival time reduction rate. Using this framework, we identify surface virus density, defined as infectious virus per unit distribution area, as a physical determina

Virucidal Efficacy of Organic Acids and Plant Essential Oils.

Currently, the predominant commercially available disinfectants and sanitizers are formulated with active ingredients including alcohol, halogenated compounds (e.g., sodium hypochlorite), surfactants, oxidizing agents (eg., hydrogen peroxide), and quaternary ammonium compounds. Natural products represent an alternative since they are often effective microbicides and are generally recognized as safe (GRAS) due to their long history of use. The purpose of this study was to evaluate the efficacy of several organic acids and plant essential oils against three bacteriophages (MS2, ΦX174, PR772) wit

Feline Herpesvirus 1 and Mycoplasma spp. Conventional PCR Assay Results From Conjunctival Samples From Cats in Shelters With Suspected Acute Ocular Infections.

Signs of ocular infections like discharge and conjunctivitis occur commonly in cats in shelters and feline herpesvirus 1 (FHV-1), Chlamydia felis, Mycoplasma spp, and feline calicivirus (FCV) are thought to be the most common causes. While molecular assays are available to amplify nucleic acids of each of these agents as single tests or in panels, additional information is needed concerning whether the assay results can be used to predict response to treatment. The objectives of this study were to report results for conventional polymerase chain reaction (PCR) assays that amplify nucleic acids

Inactivation of pathogenic viruses by plant-derived tannins: strong effects of extracts from persimmon (Diospyros kaki) on a broad range of viruses.

Tannins, plant-derived polyphenols and other related compounds, have been utilized for a long time in many fields such as the food industry and manufacturing. In this study, we investigated the anti-viral effects of tannins on 12 different viruses including both enveloped viruses (influenza virus H3N2, H5N3, herpes simplex virus-1, vesicular stomatitis virus, Sendai virus and Newcastle disease virus) and non-enveloped viruses (poliovirus, coxsachievirus, adenovirus, rotavirus, feline calicivirus and mouse norovirus). We found that extracts from persimmon (Diospyros kaki), which contains ca. 22

Epidemiological investigation of feline chronic gingivostomatitis and its relationship with oral microbiota in Xi'an, China.

Feline chronic gingivostomatitis (FCGS) is an ulcerative and/or proliferative disease that typically affects the palatoglossal folds. Because of its unknown pathogenesis and long disease course, it is difficult to treat and has a high recurrence rate. Most of the bacteria in the oral microbiota exist in the mouth symbiotically and maintain a dynamic balance, and when the balance is disrupted, they may cause disease. Disturbance of the oral microbiota may play an important role in the development of FCGS. In this study, the medical records of 3109 cats in three general pet hospitals in Xi 'an w

Efficacy of commonly used disinfectants for inactivation of human noroviruses and their surrogates.

Human noroviruses (HuNoVs) are the most common cause of acute viral gastroenteritis worldwide and are a leading cause of foodborne disease. Their environmental persistence and purported resistance to disinfection undoubtedly contribute to their success as foodborne disease agents. The purpose of this study was to compare the efficacy of three commonly used disinfectant active ingredients against representative HuNoV strains and cultivable surrogates. Ethanol (50, 70, and 90%), sodium hypochlorite (5, 75, 250, 500, and 1,000 ppm), and a quaternary ammonium compound blend (at 0.1×, 1.0×, and 10×

Electrolyzed Saline Prevents Virus Transmission in Dental Procedures: An In Vitro Study.

In dentistry, disinfection with antimicrobials is employed under different conditions and at different time points. During the COVID-19 pandemic, the use of disinfectant dental sprays was proposed, among other measures, to help prevent the transmission of infections during dental procedures that require highly effective antiseptics at particularly short contact times. The study aimed to evaluate the efficacy of electrolyzed saline (EOS) compared with other antiseptics in terms of the spread of enveloped and nonenveloped viruses by ultrasonic scaler (USS)-generated dental spray. Suspension test

InP-Based Quantum Dots as Photosensitizers in Photodynamic Antimicrobial Materials.

Ligand-functionalized InP-based quantum dots (QDs) have been developed as an innovative class of nontoxic photosensitizer suitable for antimicrobial applications, aimed at reducing or preventing pathogen transmission from one host to another via high contact surfaces. A hot injection method followed by functionalization via ligand exchange with 9-anthracene carboxylic acid (ACA) yielded the desired core/shell InP/ZnSe/ZnS QDs. Transmission electron microscopy (TEM) revealed these QDs to be uniform in size (∼3.2 nm), with light absorption across the entire visible spectrum (λmax ∼550 nm). Under

First report of molecular epidemiology and phylogenetic characteristics of feline herpesvirus (FHV-1) from naturally infected cats in Kunshan, China.

BACKGROUND: Feline herpesvirus type 1 (FHV-1) is a life threatening highly contagious virus in cats and typically causes upper respiratory tract infections as well as conjunctival and corneal ulcers. Genetic variability could alter the severity of diseases and clinical signs. Despite regular vaccine practices against FHV-1 in China, new FHV-1 cases still commonly occur. The genetic and phylogenetic characteristics of FHV-1 in Kunshan city of China has not been studied yet. Therefore, this study was planned to investigate the prevalence, molecular characteristics of circulating strains, and phy

Potent Protease Inhibitors of Highly Pathogenic Lagoviruses: Rabbit Hemorrhagic Disease Virus and European Brown Hare Syndrome Virus.

Rabbit hemorrhagic disease (RHD) and European brown hare syndrome (EBHS) are highly contagious diseases caused by lagoviruses in the Caliciviridae family. These infectious diseases are associated with high mortality and a serious threat to domesticated and wild rabbits and hares, including endangered species such as riparian brush rabbits (Sylvilagus bachmani riparius). In the United States (U.S.), only isolated cases of RHD had been reported until Spring 2020. However, RHD caused by GI.2/rabbit hemorrhagic disease virus (RHDV)2/b was unexpectedly reported in April 2020 in New Mexico and has s
