Characterization of catecholaminergic and serotonergic pathways in the trachea of the chicken and turkey.

The avian trachea is a critical component of respiration and vocalization, as well as pathogen defense and immunity. Tracheal neuroimmune pathways involving serotonin and catecholamines are well established in mammals. Although earlier work identified an endocrine system in the avian tracheal epithelium involved in dopamine metabolism, little remains known regarding the neurophysiological systems of the trachea in poultry species. As such, the present study sought to characterize the pathways involved in the synthesis, transport, and breakdown of indolamines and catecholamines in the trachea o

DNA-copper hybrid nanoflowers as efficient laccase mimics for colorimetric detection of phenolic compounds in paper microfluidic devices.

Laccases are important multicopper oxidases that are involved in many biotechnological processes; however, they suffer from poor stability as well as high cost for production/purification. Herein, we found that DNA-copper hybrid nanoflowers, prepared via simple self-assembly of DNA and copper ions, exhibit an intrinsic laccase-mimicking activity, which is significantly higher than that of control materials formed in the absence of DNA. Upon testing all four nucleobases, we found that hybrid nanoflowers composed of guanine-rich ssDNA and copper phosphate (GNFs) showed the highest catalytic acti

Modification of the association between early adversity and obsessive-compulsive disorder by polymorphisms in the MAOA, MAOB and COMT genes.

The monoamine oxidases (MAOA/B) and catechol-O-methyltransferase (COMT) enzymes break down regulatory components within serotonin and dopamine pathways, and polymorphisms within these genes are candidates for OCD susceptibility. Childhood trauma has been linked OCD psychopathology, but little attention has been paid to the interactions between genes and environment in OCD aetiology. This pilot study investigated gene-by-environment interactions between childhood trauma and polymorphisms in the MAOA, MAOB and COMT genes in OCD. Ten polymorphisms (MAOA: 3 variants, MAOB: 4 variants, COMT: 3 vari

Is prolactin playing a role in the regulation of catecholamine synthesis and release from male rat adrenal medulla?

Previous evidence allows one to suspect that prolactin (PRL) may be a physiological regulator of catecholamine (CA) synthesis and release in the adrenal gland of rodents. To explore this possibility, we studied the in vivo and in vitro metabolism and release of noradrenaline (NA) and adrenaline (A) in the adrenal gland of male rats. The study was carried out with animals exhibiting a moderate increase in plasma PRL levels induced by grafting of additional pituitaries or a severe hyperprolactinemia produced by diethylstilbestrol (DES)-induced pituitary hyperplasia. The latter animals exhibited

Catechol-O-methyltransferase activity in cultured human skin fibroblasts from controls and patients with dystonia musculorum deformans.

Fibroblasts provide a source of living cells that can be obtained easily from humans and used to evaluate inherited differences in the activities of enzymes important in neurotransmitter and drug metabolism. Here, we describe biochemical characteristics of catechol-O-methyltransferase (COMT, EC 2.1.1.6) activity in homogenates of cultured human skin fibroblasts. Many properties of the enzyme, including apparent affinity for dihydroxybenzoic acid and S-adenosyl methionine, optimal pH and (Mg++), and inhibition by Ca++, are similar to those reported in lysates of human erythrocytes. Culture and

A Review of the Recent Advances in the Pharmacological Management of Parkinson's Disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons in the substantia nigra, leading to motor and nonmotor symptoms. The pathophysiology of PD involves a complex interplay of genetic, environmental, and biochemical factors, including the accumulation of alpha-synuclein protein aggregates, mitochondrial dysfunction, oxidative stress, and neuroinflammation. Current pharmacological treatments primarily focus on alleviating symptoms, with levodopa (LD) being the gold standard for motor symptom management. Additionally, DOPA decarbo

Pharmacological properties of extracts and prenylated isoflavonoids from the fruits of Osage orange (Maclura pomifera (Raf.) C.K.Schneid.).

Osage orange trees (Maclura pomifera (Raf.) C.K.Schneid.) are distributed worldwide, particularly in south-east states of the USA. They produce large quantities of strong yellow fruits, bigger than oranges, but these fruits are inedible, with an acid milky juice which is little consumed by birds and insects. Extracts prepared from Osage orange fruits (hedge apple) have revealed a range of pharmacological properties of interest in human and veterinary medicine. In addition, Osage orange extracts can be used in agriculture and aquaculture, and as dyeing agent for the textile industry. Extracts c

Inhibition of catecholamine degradation ameliorates while chemical sympathectomy aggravates the severity of acute Friend retrovirus infection in mice.

Several lines of evidence indicate that the sympathetic nervous system (SNS) might be involved in the pathogenesis and progression of retroviral infections. However, experimental data are scarce and findings inconsistent. Here, we investigated the role of the SNS during acute infection with Friend virus (FV), a pathogenic murine retrovirus that causes polyclonal proliferation of erythroid precursor cells and splenomegaly in adult mice. Experimental animals were infected with FV complex, and viral load, spleen weight, and splenic noradrenaline (NA) concentration was analyzed until 25 days post

Molecular anatomy of tyrosinase and its related proteins: beyond the histidine-bound metal catalytic center.

The structure of tyrosinase (Tyr) is reviewed from a double point of view. On the one hand, by comparison of all Tyr found throughout nature, from prokaryotic organisms to mammals and on the other, by comparison with the tyrosinase related proteins (Tyrps) that appeared late in evolution, and are only found in higher animals. Their structures are reviewed as a whole rather than focused on the histidine (His)-bound metal active site, which is the part of the molecule common to all these proteins. The availability of crystallographic data of hemocyanins and recently of sweet potato catechol oxid

[The effect of droxidopa on the monoamine metabolsim in the human brain].

Droxidopa (L-threo-3,4-dihydroxyphenylserine) is an artificial amino acid, which is used to supplement noradrenaline (NA) in neurodegenerative disorders. Droxidopa is decarboxylated into NA by aromatic L-amino acid decarboxylase in the brain, but its effects on other monoamine neurotransmitters, such as dopamine (DA) and serotonin (5-HT) have not been systematically examined. The monoamine metabolism has been suggested to interact with each other in the brain, and by analysis of the cerebrospinal fluid, L-DOPA, a precursor amino acid used for supplement of DA, was found to inhibit serotonin sy

D-DOPA Is a Potent, Orally Bioavailable, Allosteric Inhibitor of Glutamate Carboxypeptidase II.

Glutamate carboxypeptidase-II (GCPII) is a zinc-dependent metalloenzyme implicated in numerous neurological disorders. The pharmacophoric requirements of active-site GCPII inhibitors makes them highly charged, manifesting poor pharmacokinetic (PK) properties. Herein, we describe the discovery and characterization of catechol-based inhibitors including L-DOPA, D-DOPA, and caffeic acid, with sub-micromolar potencies. Of these, D-DOPA emerged as the most promising compound, with good metabolic stability, and excellent PK properties. Orally administered D-DOPA yielded high plasma exposures (AUCpla

Interactive effects of DAOA (G72) and catechol-O-methyltransferase on neurophysiology in prefrontal cortex.

BACKGROUND: Accumulating evidence indicates that genetic polymorphisms of D-amino acid oxidase activator (DAOA) (M24; rs1421292; T-allele) and catechol-O-methyltransferase (COMT) (Val¹⁵⁸Met; rs4680) likely enhance susceptibility to schizophrenia. Previously, clinical association between DAOA M24 (T-allele) and a functionally inefficient 3-marker COMT haplotype (that included COMT Val¹⁵⁸Met) uncovered epistatic effects on risk for schizophrenia. Therefore, we projected that healthy control subjects with risk genotypes for both DAOA M24 (T/T) and COMT Val¹⁵⁸Met (Val/Val) would produce prefrontal

The Gut Microbiota in Parkinson's Disease: Mechanistic Insights into Microbial-Host Interactions.

Parkinson's disease (PD) is a multifactorial neurodegenerative disorder characterized by progressive motor and non-motor manifestations, including early gastrointestinal dysfunction. Growing evidence implicates the gut microbiota as an active modulator of host immune tone and neurodegenerative vulnerability, extending beyond descriptive taxonomic associations toward functional and metabolic mechanisms. PD-associated dysbiosis is consistently characterized by altered microbial functional capacity, including reduced short-chain fatty acid (SCFA) production, enrichment of pro-inflammatory metabol

Rapid colorimetric detection of trace-level catechol and hydroquinone using a robust, highly active δ-MnO2 nanoflower nanozyme synthesized under mild conditions.

The colorimetric discrimination and detection of catechol (CC) and hydroquinone (HQ) remain a challenge due to their highly similar physicochemical properties. Herein, pure-phase δ-MnO2 nanoflower nanozyme (δ-MnO2-NF) was synthesized at a mild temperature of 80°C. The as-obtained material exhibits dual laccase- and oxidase-like activities, favorable substrate affinity and accelerated maximum reaction velocity, with its specific activity more than 11.77 times higher than previously reported nanozymes. Additionally, it features robust stability across broad ranges of pH (2-10), temperature (10-8

C-type lectin 9 participates in the immune response, development and reproduction of Tribolium castaneum.

C-type lectins (CTLs), as a large family of pattern recognition receptors (PRRs), have been reported to be involved in bacterial infection, but the role of CTLs in development has been poorly understood in insects. The orthologues of Tribolium castaneum CTL9 (TcCTL9) have been identified among insects, but its functions were currently unclear. Therefore, we performed functional analysis of TcCTL9 in this study. Our results indicated that TcCTL9 could bind to bacteria through lipopolysaccharide and peptidoglycan, and agglutinate Gram-positive and Gram-positive bacteria in a Ca2+-dependent manne

Process optimization of polyphenol oxidase immobilization: Isotherm, kinetic, thermodynamic and removal of phenolic compounds.

Chitosan/montmorillonite (CTS/MMT) and chitosan‑gold nanoparticles/montmorillonite (CTS-Au/MMT) composites were prepared, characterized through Fourier transformed infrared (FT-IR), X-ray powder diffraction (XRD), and scanning electron microscopy (SEM), and utilized as support for immobilization of polyphenol oxidase (PPO). PPO was immobilized on CTS/MMT (IPPO) and CTS-Au/MMT (IPPO-Au) by physical adsorption, respectively. In order to achieve simultaneous maximization of immobilization efficiency and enzyme activity, the immobilization process parameters were optimized by Taguchi-Grey relation

Evaluation of the efficacy and safety of adjuvant treatment to levodopa therapy in Parkinson s disease patients with motor complications.

BACKGROUND: One of the complications of long-term treatment of Parkinson's disease (PD) with levodopa is the development of motor complications. Generally, when motor complications develop, clinicians add in an additional drug (to the levodopa regimen) from one of three other classes of anti-Parkinsonian treatments (dopamine agonists, catechol-O-methyl transferase inhibitors (COMTIs) or monoamine oxidase type B inhibitors (MAOBIs)). However, despite trials having shown that these drugs are beneficial compared to placebo, it remains unclear as to the best way to treat patients experiencing moto
