Assessment of causality and impairment following unilateral hypoglossal nerve paralysis: A case report.

Isolated hypoglossal nerve injury is an infrequent occurrence in clinical and forensic traumatology practice. Its etiology includes trauma, malignancy, vascular events, autoimmune diseases, and complications of surgical procedures. Clinical manifestations resulting from nerve damage may present early or be delayed. We present the case of a 44-year-old woman who sustained a fracture of the third cervical vertebra following a traffic accident. An anterior approach was employed for instrumentation using an anterior plate spanning two cervical segments. The patient developed dysphagia and swallowi

Exploring the Causality Between Mood Swings and Neurological Diseases: A Mendelian Randomization Study.

BACKGROUND: Associations between genetically predicted mood swings and neurological diseases have been suggested. However, the causal relationships between these factors remain undefined. METHOD: We used a Mendelian randomization (MR) study to examine the causal relationship of genetically predicted mood swings with the risk of neurological diseases, including stroke, Alzheimer's disease (AD), multiple sclerosis (MS), epilepsy (EP), and headache. The single-nucleotide polymorphisms (SNPs) that exhibited genetic associations with mood swings were utilized as instrumental variables (IV) in the

Mendelian randomization study showed no causality between metformin treatment and polycystic ovary syndrome.

BACKGROUND: Despite previous clinical studies providing some evidence of an association between metformin treatment and polycystic ovary syndrome(PCOS), these findings remain controversial. To investigate whether the association reflect causality, a two-sample Mendelian randomization (MR) method was conducted. METHODS: Data from genome-wide association studies were analyzed, with the exposure factor being metformin and the outcome variable being PCOS. The inverse variance weighted(IVW) was used as the primary method for MR analysis. In addition, MR-Egger, weighted median, heterogeneity tests,

Localization of arrhythmogenic sites in post-ischemic ventricular tachycardia using Network Granger Causality.

Detecting abnormal electrical wave propagation in Sinus Rhythm (SR) in patient with Ventricular Tachycardia (VT) can enhance treatment strategies. However, traditional methods, like activation maps, often produce biased results due to the difficulty of calculating what is called Local Activation Time (LAT). This paper introduces a method based on Network Granger Causality (NGC) to assess VT mechanics. Instead of computing LATs, NGC generalizes the binary Granger causality framework and allows to leverage the EGM morphologies, identifying temporal causality links within a network of electrogram

Investigating Disruptions in Information Flow due to Sickle Cell Disease Using Granger Causality.

Sickle cell disease (SCD) is an inherited blood disorder caused by a mutation in the beta-globin gene, resulting in chronic complications, including cognitive decline-particularly in executive functions. Neuroimaging studies have identified structural and functional abnormalities associated with SCD; however, the directionality of information flow between brain networks and how disruptions in these interactions contribute to cognitive deficits remains poorly understood. This study employed Granger causality (GC) analysis to investigate effective connectivity and information flow between brain

Assessing causality between mitochondrial-associated proteins with musculoskeletal diseases: A Mendelian randomization study.

Musculoskeletal diseases are the leading cause of disability-adjusted life years. Mitochondria, often referred to as the "powerhouses" of cells, are believed to play a role in regulating cellular metabolism and differentiation, potentially influencing the occurrence and progression of musculoskeletal diseases. However, the exact causal relationships remain to be defined. This study aimed to investigate the causal relationships between mitochondrial biological functions and musculoskeletal diseases (including osteoarthritis (OA), osteoporosis, rheumatoid arthritis (RA), and ankylosing spondylit

Genetic Insights into the Association and Causality Between Blood Metabolites and Alzheimer's Disease.

BACKGROUND: Alzheimer's disease (AD) is an increasing public health concern with the aging of the global population. Understanding the genetic correlation and potential causal relationships between blood metabolites and AD may provide important insights into the metabolic dysregulation underlying this neurodegenerative disorder. OBJECTIVE: The aim of this study was to investigate the causal relationship between blood metabolites and AD using Mendelian randomization (MR) analysis. METHODS: Association data were obtained from three large-scale genome-wide association studies of 486 blood metab

Pleiotropy and genetically inferred causality linking multisite chronic pain to substance use disorders.

Individuals suffering from chronic pain develop substance use disorders (SUDs) more often than others. Understanding the shared genetic influences underlying the comorbidity between chronic pain and SUDs will lead to a greater understanding of their biology. Genome-wide association statistics were obtained from the UK Biobank for multisite chronic pain (MCP, Neffective = 387,649) and from the Million Veteran Program and the Psychiatric Genomics Consortium meta-analyses for alcohol use disorder (AUD, Neffective = 296,974), cannabis use disorder (CanUD, Neffective = 161,053), opioid use disorder

A Model for Causality of Pitching-related Overuse Injuries in Women's Fastpitch Softball.

Fastpitch softball is a popular women's sport, and athletes, particularly pitchers, are at high risk for overuse injury. Softball-related injury rates are low; however, the prevalence of overuse injuries is high. Injuries at the high school and collegiate levels occur early in the season, and approximately 50% of shoulder and elbow injuries in softball are attributable to overuse. Survey research showed 77% of high school pitchers pitch with pain, and 73% of collegiate pitchers reported an overuse injury in the previous season. Modifiable and non-modifiable intrinsic and extrinsic risk factors

Certolizumab-Induced Liver Injury in Ankylosing Spondylitis: A Case Report and Causality Assessment.

Certolizumab-induced liver injury is exceptionally rare, with only a few cases reported in the literature. We present the case of a 34-year-old man with axial ankylosing spondylitis (AS) who developed a drug-induced liver injury following treatment with certolizumab. Despite the initial ineffectiveness of non-steroidal anti-inflammatory drugs and an inadequate response to infliximab, the patient achieved remission of AS symptoms with certolizumab. However, he subsequently developed elevated liver enzymes indicative of hepatocellular injury. Investigations excluded viral hepatitis and autoimmun

Causality between ankylosing spondylitis and urolithiasis in European Populations: A Mendelian randomization analysis.

BACKGROUND: Ankylosing spondylitis (AS) is an inflammatory condition affecting the spine. Observational studies have suggested a heightened risk of urolithiasis in AS patients. However, due to the inherent limitations of observational research, the causal relationship between the two remains to be determined. OBJECTIVE: Utilizing the Mendelian randomization analysis approach, this study sought to explore the causal link between AS and urolithiasis. METHODS: Data from genome-wide association studies were employed for analysis. Mendelian randomization analyses were conducted using the IVW, MR-

An optimized instrument variable selection approach to improve causality estimation in association studies.

Mendelian randomization (MR) is an emerging tool for inferring causality in genetic epidemiology. MR studies suffer bias from weak genetic instrument variables (IVs) and horizontal pleiotropy. We introduce a robust integrative framework strictly adhering with STROBE-MR guidelines to improve causality inference through MR studies. We implemented novel t-statistics-based criteria to improve the reliability of selected IVs followed by various MR methods. Further, we include sensitivity analyses to remove horizontal-pleiotropy bias. For functional validation, we perform enrichment analysis of iden

Incorporating the Molecular Mimicry of Environmental Antigens into the Causality of Autoimmune Hepatitis.

Molecular mimicry between foreign and self-antigens has been implicated as a cause of autoimmune hepatitis in experimental models and cross-reacting antibodies in patients. This review describes the experimental and clinical evidence for molecular mimicry as a cause of autoimmune hepatitis, indicates the limitations and uncertainties of this premise, and encourages investigations that assess diverse environmental antigens as sources of disease-relevant molecular mimics. Pertinent articles were identified in PubMed using multiple search phrases. Several pathogens have linear or conformational e

Allergic Reactions to Current Available COVID-19 Vaccinations: Pathophysiology, Causality, and Therapeutic Considerations.

Vaccines constitute the most effective medications in public health as they control and prevent the spread of infectious diseases and reduce mortality. Similar to other medications, allergic reactions can occur during vaccination. While most reactions are neither frequent nor serious, anaphylactic reactions are potentially life-threatening allergic reactions that are encountered rarely, but can cause serious complications. The allergic responses caused by vaccines can stem from activation of mast cells via Fcε receptor-1 type I reaction, mediated by the interaction between immunoglobulin E (Ig

Type 2 Diabetes Mellitus and Amyotrophic Lateral Sclerosis: Genetic Overlap, Causality, and Mediation.

CONTEXT: Understanding phenotypic connection between type II diabetes (T2D) mellitus and amyotrophic lateral sclerosis (ALS) can offer valuable sight into shared disease etiology and have important implication in drug repositioning and therapeutic intervention. OBJECTIVE: This work aims to disentangle the nature of the inverse relationship between T2D mellitus and ALS. METHODS: Depending on summary statistics of T2D (n = 898 130) and ALS (n = 80 610), we estimated the genetic correlation between them and prioritized pleiotropic genes through a multiple-tissue expression quantitative trait lo

High prevalence of smoking in patients with adrenal incidentalomas: causality or case selection?

OBJECTIVE: Autonomous cortisol secretion and possible autonomous cortisol secretion (ACS/pACS) are associated to an increase of cardiovascular risk factors such as hypertension, diabetes mellitus and dyslipidaemia. To our knowledge, the prevalence of smoking, another well-established risk factor for cardiovascular disease, has not been studied in detail in people with ACS/pACS or adrenal incidentalomas. METHODS: Patients with adrenal incidentalomas were examined with the 1-mg overnight dexamethasone suppression test (cortisolONDST). Information about current smoking was collected from the pat

The Association Between Vascular Inflammation and Depressive Disorder. Causality, Biomarkers and Targeted Treatment.

Diabetes, obesity, atherosclerosis, and myocardial infarction are frequently co-morbid with major depressive disorder. In the current review, it is argued that vascular inflammation is a factor that is common to all disorders and that an endothelial dysfunction of the blood-brain barrier could be involved in the induction of depression symptoms. Biomarkers for vascular inflammation include a high plasma level of C-reactive protein, soluble cell-adhesion molecules, von Willebrand factor, aldosterone, and proinflammatory cytokines like interleukin-6 or tumor necrosis factor α.
