Nitroxide Polymer Encapsulating Triptolide for Safe and Effective Small Cell Lung Cancer Therapy by Targeting Energy Metabolism and Pyroptosis.

Small cell lung cancer (SCLC) is highly aggressive neuroendocrine tumor with a paucity of effective therapeutic option. Triptolide (TP), a natural diterpenoid triepoxide, exhibits potent antitumor activities, yet its efficacy and underlying mechanisms in SCLC remain largely undefined. Here, we demonstrate that TP effectively inhibits cell viability across all molecular subtypes of SCLC, with greater potency than in nonsmall cell lung cancer. Mechanistically, TP transcriptionally represses hexokinase 1 and 2 (HK1/2), thereby disrupting the glycolysis-TCA-OXPHOS axis and resulting in ATP depleti

Poly(d,l-lactide-co-glycolide) Nanoparticles Encapsulating Doxorubicin for Improved Treatment in Cholangiocarcinoma and Drug-Resistant Cells.

Cholangiocarcinoma (CCA), a malignancy of the bile duct epithelium, represents a significant public health issue in the Greater Mekong Subregion, including Thailand. Its aggressive characteristics and late-stage diagnosis lead to poor prognosis and elevated mortality rates. Chemotherapy faces limitations, including the requirement for high and frequent dosages, low cellular uptake, and side effects. To address these challenges, poly(d,l-lactide-co-glycolide) (PLGA) nanoparticles (NPs) encapsulating doxorubicin (DOX), a chemotherapeutic drug, were developed via a modified nanoprecipitation tech

Evaluating the Effect of Exosome-Encapsulated miR-4289 on Menstrual Blood-Derived Mesenchymal Stem Cells from Endometriosis Patients.

Endometriosis is a benign yet chronic gynecological disorder characterized by dysregulation of processes such as inflammation, angiogenesis, migration, apoptosis, and proliferation. Menstrual blood-derived endometrial stem cells play a crucial role in the retrograde development and progression of endometriotic lesions. To evaluate the therapeutic potential of exosomes derived from menstrual blood-derived stem cells, exosomes from non-endometriotic MenSCs (NE-MenSCs), both unmodified (Exo) and transfected with miR-4289, were applied as treatments to MenSCs from the endometriosis cell line (E-Me

Synergistic apoptotic effects of berberine and hesperidin co-encapsulated in PLGA nanoparticles on lymph node carcinoma of prostate cell line.

Natural origin therapeutic agents have recently gained attention as a promising cancer treatment approach. However, achieving effective therapeutic results depends on improving the pharmacokinetic properties of these compounds using new delivery systems. In this study, the simultaneous encapsulation of berberine and hesperidin in poly(lactic-co-glycolic acid)(PLGA) nanoparticles utilizing the double emulsion-solvent evaporation technique and the evaluation of their effects on the viability and induction of apoptosis in prostate cancer cells were investigated. The produced nanoparticles were ch

Injectable Thermosensitive Hydrogel Encapsulating Mesenchymal Stem Cell Membrane-Based Biomimetic Liposomes for Local Rheumatoid Arthritis Treatment.

In rheumatoid arthritis (RA), the immune system mistakenly attacks joints by releasing harmful inflammatory substances that cause swelling and gradual joint damage. As inflammation progresses, patients experience severe pain and reduced mobility. Therefore, localized anti-inflammatory and analgesic therapeutic interventions may offer a viable strategy for the clinical management of RA. To effectively regulate joint inflammation, it is essential to prolong the retention time of therapeutic agents within the joint cavity and enhance the target delivery capability. To achieve this, we loaded mese

Lipid Nanoparticle-Encapsulated TALEN-Encoding mRNA Inactivates Hepatitis B Virus Replication in Cultured Cells and Transgenic Mice.

Chronic infection with the hepatitis B virus (HBV) results in over 1 million deaths annually. Although currently licensed treatments, including pegylated interferon-α and nucleoside/nucleotide analogs, can inhibit viral replication, they rarely eradicate covalently closed circular DNA (cccDNA) reservoirs. Moreover, vaccination does not offer therapeutic benefit to already infected individuals or non-responders. Consequently, chronic infection is maintained by the persistence of cccDNA in infected hepatocytes. For this reason, novel therapeutic strategies that permanently inactivate cccDNA are

Microfluidics-based PLGA nanoparticles of ratiometric multidrug: From encapsulation and release rates to cytotoxicity in human lens epithelial cells.

Multidrug nanomedicine is an effective therapeutic approach for the treatment of chronic diseases and cancers. However, co-encapsulation and release of drug combination at a fixed ratio by nanoparticles, particularly for long acting ocular formulations, remains challenging. Herein, poly (lactic-co-glycolic acid) nanoparticles ratiometrically co-encapsulating hydrophilic dual drugs, mitomycin C and doxorubicin, was obtained (D/M PLGANPs) by combining microfluidics and the Design of Experiments approaches. The formulation variable of lactide-to-glycolide ratios (L/G 50:50, 75:15 and 85:15) was u

Differentiation of PC12 cell line into neuron by Valproic acid encapsulated in the stabilized core-shell liposome-chitosan Nano carriers.

Valproic acid (VPA) usage in high dose is teratogen with low bioavailability. Hence to improve its efficacy and reduce its side effect it was encapsulated by the Nano liposomes and stabilized by the chitosan at different concentrations. The cellular uptake, biocompatibility, loading and encapsulation efficiency of the six-different formulations (1:1, 2:1, and 4:1 of chitosan-phospholipids: VPA), PC12 differentiation to neuron cells assays (gene-expression level by qRT-PCR) were conducted for the efficacy assessment of the Nano carriers. The encapsulation efficiency (EE) results revealed that t

CD44-targeted vesicles encapsulating granzyme B as artificial killer cells for potent inhibition of human multiple myeloma in mice.

Multiple myeloma (MM) is a malignant blood cancer homing in bone marrow that is particularly hard to treat. The effective treatment for MM shall be not only MM-selective but also capable of homing to bone marrow. Herein, we report on hyaluronic acid-directed reduction-responsive chimaeric polymersomes encapsulating a key player in the NK cells, granzyme B (HA-RCP-GrB) as an artificial killer cell for targeted protein therapy of MM. Interestingly, HA-RCP-GrB displayed high MM-targetability and anti-MM activity with a remarkably low IC50 of 8.1 nM toward CD44 overexpressing LP1 human MM cells. T

Human umbilical cord mesenchymal stem cell derived exosomes encapsulated in functional peptide hydrogels promote cardiac repair.

Stem cell-derived exosomes have been recognized as a potential therapy for cardiovascular disease. However, the low retention rate of exosomes after transplantation in vivo remains a major challenge in clinical applications. The aim of this study is to investigate whether human umbilical cord mesenchymal stem cell derived exosomes (UMSC-Exo) encapsulated in functional peptide hydrogels could increase the retention and stability of exosomes and improve heart function in a rat myocardial infarction model. Our results demonstrated that the PA-GHRPS peptide protected H9C2 cells from H2O2-induced o

Vascular Endothelial Cell Injury Is an Important Factor in the Development of Encapsulating Peritoneal Sclerosis in Long-Term Peritoneal Dialysis Patients.

BACKGROUND AND OBJECTIVES: Encapsulating peritoneal sclerosis (EPS) is a rare but serious and life-threatening complication of peritoneal dialysis (PD). However, the precise pathogenesis remains unclear; in addition, predictors and early diagnostic biomarkers for EPS have not yet to be established. METHODS: Eighty-three peritoneal membrane samples taken at catheter removal were examined to identify pathological characteristics of chronic peritoneal deterioration, which promotes EPS in patients undergoing long-term PD treatment with low occurrence of peritonitis. RESULTS: According to univari

Liposome-encapsulated antigens induce a protective CTL response against Listeria monocytogenes independent of CD4+ T cell help.

Protection against intracellular pathogens is usually mediated by cytotoxic T lymphocytes (CTL). Induction of a protective CTL response for vaccination purposes has proven difficult because of the limited access of protein antigens or attenuated pathogens to the MHC class I presentation pathway. We show here that pH-sensitive PE/CHEMS liposomes can be used as a vehicle to efficiently deliver intact proteins for presentation by MHC class I. Mice immunized with listerial proteins encapsulated in such liposomes launched a strong CTL response and were protected against a subsequent challenge with

Therapeutic potential of a polymer-encapsulated L-DOPA and dopamine-producing cell line in rodent and primate models of Parkinson's disease.

Encapsulation of cells within polymer membranes prior to transplantation provides a novel means of achieving continuous, site-specific delivery of therapeutic molecules to the CNS. The use of encapsulated dopamine-secreting cells that can be transplanted directly into the striatum has particular appeal for the treatment of Parkinson's disease. This article provides a brief and timely review of the progress that has been made over the past decade using encapsulated PC12 cells as a means of delivering dopamine and L-DOPA to the striatum in rodent and primate models of Parkinson's disease. Melatonin inhibits the formation of chemically induced experimental encapsulating peritoneal sclerosis through modulation of T cell differentiation by suppressing of NF-κB activation in dendritic cells.

Encapsulating peritoneal sclerosis (EPS) is a severe complication of peritoneal dialysis (PD). Surgery is a therapeutic strategy for the treatment of complete intestinal obstruction. However, complete intestinal obstruction in long-term PD results in high mortality and morbidity rates after surgery. Immunopathogenesis participates in EPS formation: CD8, Th1, and Th17 cell numbers increased during the formation of EPS. The anti-inflammatory and immunomodulatory effects of melatonin may have beneficial effects on this EPS. In the present study, we determined that melatonin treatment significantl

Amyloid-Beta Peptides and Activated Astroglia Impairs Proliferation of Nerve Growth Factor Releasing Cells In Vitro: Implication for Encapsulated Cell Biodelivery-Mediated AD Therapy.

Alzheimer's disease (AD) treatment is constrained due to the inability of peripherally administered therapeutic molecules to cross the blood-brain barrier. Encapsulated cell biodelivery (ECB) devices, a tissue-targeted approach for local drug release, was previously optimized for human mature nerve growth factor (hmNGF) delivery in AD patients but was found to have reduced hmNGF release over time. To understand the reason behind reduced ECB efficacy, we exposed hmNGF-releasing cells (NGC0211) in vitro to human cerebrospinal fluid (CSF) obtained from Subjective Cognitive Impairment (SCI), Lewy

Intrarectal delivery of chitosan hydrogel-encapsulated mesenchymal stem cell-conditioned media attenuates disease severity in experimental ulcerative colitis.

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory disease characterized by mucosal immune dysregulation and epithelial injury. While mesenchymal stem cells (MSCs) hold regenerative and immunomodulatory promise, their use entails safety and logistical challenges. MSC-derived conditioned medium (MSC-CM) may offer a safer, cell-free alternative. Here, we present a novel intrarectal delivery strategy that pairs MSC-CM with a mucoadhesive chitosan hydrogel (Cs-Hyd), aiming for localized mucosal therapy in colitis. METHODS: Experimental colitis was induced in mice via DSS exposure. Huma

Oral Microcapsules Encapsulating Endometrial Regenerative Cell-Derived Exosomes Promote Intestinal Epithelial Barrier Repair and Ameliorate Experimental Colitis.

BACKGROUND: Endometrial regenerative cell-derived exosomes (ERC-Exos) exhibit tissue repair and anti-inflammatory properties for ulcerative colitis (UC) treatment, but oral administration is limited by gastrointestinal degradation. This study first developed chitosan-sodium alginate microcapsules (CSE) for ERC-Exos delivery and explored the role of milk fat globule-EGF factor 8 (MFGE8). METHODS: MFGE8-knockdown ERC-Exos (MFGE8 -/- -ERC-Exos) were constructed via lentiviral transfection. Colitis-targeted exosomes (CSE) were prepared, and their in vivo stability and targeting ability were verif

Human ES cell-derived MSC spheroids encapsulated in polydopamine nanoparticle-modified GelMA hydrogel enhance diabetic wound healing.

Mesenchymal stromal cell (MSC)-based strategies hold promises for treating diabetic foot ulcers. However, the hostile microenvironment of the lesions characterized by hypoxia, oxidative stress, and chronic inflammation impairs the survival and efficacy of transplanted MSCs. Here, we developed a composite bio-dressing by integrating MSC spheroid (Sp) derived from human embryonic stem cells via trophoblast intermediates (T-MSC) with gelatin methacryloyl hydrogel (Gm) and polydopamine nanoparticles (Pn). The GmPn matrix exhibited excellent adaptability, adhesion, porosity, biodegradability, and b
