Structure-activity relationship of heme and its analogues in membrane damage and inhibition of fusion.

Under pathological conditions, such as sickle cell disease and malaria, heme concentration increases considerably, and it induces membrane damage. As sickled and normal erythrocytes contain high cholesterol: phospholipid ratio, we investigated the role of lipid composition, chain length, and unsaturation on the partitioning and leakage of hemin in phospholipid vesicles. To establish structure-activity relationship in membrane damage, experiments with two other analogues, protoporphyrin-IX and hematoporphyrin (HP) were also carried out. Hemin and its analogues localize differently in membranes

Effects of combining lysozyme, ultrasound, and heat treatments on the inactivation of Bacillus subtilis spores.

The inactivation of bacterial spores remains a critical challenge in food sterilization. This study aimed to investigate the effects of combining ultrasound with heat and lysozyme treatment on the key structures of Bacillus subtilis spores. Spores were subjected to 600 W ultrasound combined with heat treatments at 75 ℃ and 90 ℃ and lysozyme at concentrations of 0.2% and 0.4% for 30 min. The combined treatments exhibited a synergistic effect on spores inactivation, with the most pronounced effect observed with the 600W-90 ℃-0.4% lysozyme treatment, achieving a reduction of 4.7 log CFU/mL. This

Structure and molecular composition of Toxoplasma gondii pellicle.

Toxoplasma gondii is one of the most successful protozoan parasites in the world, chronically infecting around 25-30 % of the human population. As a member of the phylum Apicomplexa, Toxoplasma has unique cellular structures, the best known being the apical complex and, no less notable, a trilaminar pellicle structure formed by the plasma membrane on top of a set of flattened membranous sacs (alveoli) called inner membrane complex. As an obligated intracellular pathogen, T. gondii pellicle contains an arsenal of proteins involved in host cell recognition and adhesion, which are crucial for cel

Electron-cytochemical localization of alkaline phosphatase to G cells of Necturus maculosus antrum.

Electron-cytochemical localization of alkaline phosphatase activity was performed on G cells of Necturus maculosus antral mucosa. Alkaline phosphatase activity was localized to the nuclear membrane, the Golgi/endoplasmic reticulum, and the limiting membranes of G cell peptide-secretion vesicles. There was no specific localization of alkaline phosphatase activity to the plasma membrane. Treatment of the tissues with levamisole (an alkaline phosphatase inhibitor) did not markedly reduce the specific alkaline phosphatase activity. Specific lead deposition was reduced by removal of the substrate f

Effect of Satureja montana Essential Oil on Model Lipid Membranes.

Satureja montana essential oil is a natural substance able to inhibit the growth of several pathogens. This antimicrobial effect is often attributed to its ability to penetrate cellular structures and disrupt them. Although these properties are recognized as playing a key role in the mechanism of action of this substance, many unresolved issues still exist, and fundamental studies focused on such aspects are scarce. In this framework, we investigated the interaction of SEO with lipid monolayers, which represent simplified models of cell membranes, using the Langmuir monolayer technique, comple

Acidic Environment Significantly Alters Aggregation Pathway of Human Islet Amyloid Polypeptide at Negative Lipid Membrane.

The misfolding and aggregation of human islet amyloid polypeptide (hIAPP) at cell membrane has a close relationship with the development of type 2 diabetes (T2DM). This aggregation process is susceptible to various physiologically related factors, and systematic studies on condition-mediated hIAPP aggregation are therefore essential for a thorough understanding of the pathology of T2DM. In this study, we combined surface-sensitive amide I and amide II spectral signals from the protein backbone, generated simultaneously in a highly sensitive femtosecond broad-band sum frequency generation vibra

Distinct membrane interaction mechanisms of gramicidin A and S in mammalian cells: A combined experimental and molecular dynamics study.

Gramicidins, a class of ionophores, with a well-known antimicrobial activity against bacteria, and more recently against cancer cells, present two distinct structure types: linear peptides for Gramicidin A, B and C and cyclic peptide for Gramicidin S. Independent of their structure, the first stage of their action mechanism involves their interaction with the plasma membrane of the cells. Considering this, we were interested to study how the peptide structure affects cell properties (cell viability, LDH release, etc) and how it correlates with membrane fluidity parameters (GP , ΔSr, Rs) and mo

Fabrication of 3D monolayer nanofiber structures via capillary induced bending and fusion.

Electrospun nanofibers offer tunable topography and broad material versatility for advanced cell culture studies and cell-based applications; however, the fabrication of free-standing, highly porous, and ultrathin nanofiber architectures remains challenging. Here, we report a solvent evaporation and capillary-force-assisted fusion of two monolayers of gelatin nanofibers deposited on opposite sides of a through-hole membrane. During solvent evaporation, capillary forces drive the two nanofiber layers into contact in each hole, enabling localized nanofiber fusion in parallel of chemical crosslin

Effects of aspirin, indomethacin, and sodium salicylate on human erythrocyte membranes as detected with electron spin resonance spectroscopy.

Electron spin resonance spectroscopy of probed samples was used to determine the structural changes in human erythrocyte membranes prior to and at intervals following ingestion of either 10 grains acetylsalicylic acid, 10 grains sodium salicylate, or 50 mg indomethacin by both male and female subjects. Analysis of erythrocytes from female subjects indicated a time-dependent disordering of the membrane over the eight hour period following aspirin ingestion while the cells of male subjects showed a slight membrane ordering over the same time period. Erythrocytes drawn from females at the beginni

The Plasmodium knowlesi MAHRP2 ortholog localizes to structures connecting Sinton Mulligan's clefts in the infected erythrocyte.

During development within the host erythrocyte malaria parasites generate nascent membranous structures which serve as a pathway for parasite protein transport to modify the host cell. The molecular basis of such membranous structures is not well understood, particularly for malaria parasites other than Plasmodium falciparum. To characterize the structural basis of protein trafficking in the Plasmodium knowlesi-infected erythrocyte, we identified a P. knowlesi ortholog of MAHRP2, a marker of the tether structure that connects membranous structures in the P. falciparum-infected erythrocyte. We

Salivary duct carcinoma in the submandibular region.

A case of salivary duct carcinoma arising in the submandibular region of an 83-year-old man is presented. Histologically, the tumour consisted of solid cell nests with ductal structures. Tumour cell nests showed central comedonecrosis. Immunohistochemically, the tumour cells were positive for keratin and epithelial membrane antigen but negative for S-100 protein and calponin. Clinical features as well as pathological examinations based on haematoxylin-eosin and immunohistochemical stainings were important in the diagnosis of this case.

Normal and abnormal embryonic development of the anorectum in human embryos.

In the literature, some controversy still exists about the normal and abnormal development of the human anorectum. Therefore, a three-dimensional and histological study was performed on human embryos. In early anorectal development (< or = 49 days postfertilization), the cloaca plays a crucial role, separated from the amniotic cavity by its cloacal membrane. In the cloaca, the yolk sac/primitive hindgut and allantois/primitive urogenital sinus enter. During the embryonic caudal folding process, incorporation of these structures occurs, including their surrounding extraembryonic mesoderm, which

Differential distribution of salivary agglutinin and amylase in the Golgi apparatus and secretory granules of human salivary gland acinar cells.

The secretory granules of salivary glands often display complex internal substructures, yet little is known of the molecular organization of their contents or the mechanisms involved in packaging of the secretory proteins. We used post-embedding immunogold labeling with antibodies to two secretory proteins, agglutinin and alpha-amylase, to determine their distribution in the Golgi apparatus and secretory granules of the human submandibular gland acinar cells. With monoclonal antibodies specific for carbohydrate epitopes of the agglutinin, reactivity was found in the trans Golgi saccules, trans

A model for signal transmission in an ear having hair cells with free-standing stereocilia. I. Empirical basis for model structure.

No adequate theory for the signal-transmission properties of the peripheral auditory system exists for any vertebrate ear. Because the mammalian ear seems to pose conceptual and technical problems that complicate the development of an adequate theory, it is worthwhile to investigate simpler ears. The ear of the alligator lizard is simpler than mammalian ears in several respects: the motion of the basilar membrane is approximately independent of longitudinal position and is approximately linearly related to the sound pressure at the tympanic membrane; in a large region of the cochlea the hair c

Protocol for generation of mouse and human endometrial assembloids†.

Interactions between the endometrial epithelium and stroma are fundamental to development, physiology, and diseases of the uterus. Current endometrial organoid methods frequently rely on basement membrane extracts, which support epithelial organoid growth in the absence of physiological mesenchyme/stroma. These systems typically generate cystic epithelial structures with apical-in polarity, limiting access to the luminal surface and preventing direct contact with the stroma. To overcome these limitations, we developed a protocol for generating basement membrane extract-free assembloids compose

The distinct trimeric structure of the immunodominant chlamydial antigen Major Outer Membrane Protein.

The chlamydial Major Outer Membrane Protein (MOMP) is a promising subunit vaccine candidate due to its abundance in the outer membrane (OM), the presence of four surface-exposed variable domains (VDs) containing neutralizing and serotyping epitopes, and confirmed T-cell epitopes located in its constant domains (CDs). However, recombinant and denatured forms of MOMP have failed to elicit immune responses comparable to native preparations, indicating that conformation is critical for immunogenicity. Here, we present two cryo-EM structures of native Chlamydia muridarum MOMP, isolated from infecti

Renal tubular basement membrane changes in tubulointerstitial damage in patients with glomerular diseases.

Injury to renal tubules and interstitium occur in various glomerular diseases, leading to functional impairment. Tubular basement membrane (TBM) is an important component in maintaining tubular epithelial cell integrity. Because ultrastructural changes in these structures had not been studied in detail, the authors analyzed 30 patients with various types of glomerular diseases, including minimal change disease (MCD), focal segmental glomerular sclerosis, IgA nephropathy, diffuse proliferative glomerulonephritis, membranous nephropathy, and lupus nephritis, by light, electron, and immunofluores

Repair of a Nasal Tip Defect Using Graft and Patch Versions of a Deep-Frozen Amniotic Membrane: A Case Report.

This report describes the clinical course of a patient who underwent radical excision of a basal cell carcinoma and subsequent reconstruction using a Zitelli modification of the bilobed flap, which resulted in a nasal tip defect. The patient did not wish to undergo another surgical nose reconstruction; thus, the author used a two-stage application method with deep-frozen amniotic membrane to treat the nasal tip defect. The first application-the graft version-was applied to exposed cartilaginous structures, initiating tissue restoration. Seven days later, the author observed complete absorption

Identification of structures containing polyphosphate in Helicobacter pylori.

For the first time polyphosphate (poly P) granules have been detected in Helicobacter pylori organisms colonizing the gastric antrum as well as in organisms isolated from the same tissue. Poly P granules showed typical sublimation characteristics during exposure to the electron beam and chipped out of ultrathin sectioning. A prominent phosphorus signal was identified using elemental specific electron microscopy such as electron energy loss spectroscopy (EELS) and was localized to at least three different locations: the cytoplasm, the flagellar pole and in association with the cell membrane. Further ultrastructural findings in epithelial cells of hairy leukoplakia.

Epstein-Barr virus and multivesicular structures appearing in lesions of hairy leukoplakia have been examined ultrastructurally in serial sections. It was found that both structures probably represent various levels of sectioning of transversely cut cell processes. The so-called viral nucleocapsid having a single membrane results when the plane of section is at right angles to one of the membranes but tangential to the other. While the so-called viral particle having two membranes would result if the plane of section is perpendicular to both the membranes of adjacent cells. The internal electr
