Brainstem ischemic damage following occlusion of the blood vessels in the rat's posterior cerebral circulation.

Progression of ischemic damage was investigated immunohistochemically in neural dendrites using microtubule-associated protein 2 (MAP2) as a dendritic marker in the rat's brainstem. Neuronal soma and dendrites were clearly stained by this protein but some structures such as axonal bundles, glia and endothelial cells were not visualized. When the anterior inferior cerebellar artery (AICA) was occluded unilaterally for 30 min, a wide ischemic lesion was detected in the occluded side of the brainstem and was observed as a loss of reaction to MAP2. After ischemia for 2 h, loss of reaction in the p

Single Cell Mechanics in Disease Progression.

Mechanical force transmission is essential for maintaining cellular structure and function. Forces generated by the extracellular matrix (ECM) and neighboring cells are transmitted via adhesion and junctional complexes to the cytoskeleton and nucleus, forming an integrated mechanical network. These forces at cell-substrate and cell-cell interfaces can be quantified using biophysical tools such as atomic force microscopy, magnetic tweezers, and Förster resonance energy transfer, which have become core technologies for characterizing cellular mechanical properties. Disruption of cellular mechani

Perinuclear theca during spermatozoa maturation leading to fertilization.

Mammalian spermatozoa acquire the capacity to fertilize the ovum and display motility during their passage through the epididymis. At the same time, they undergo changes in metabolic patterns, enzymatic activities, ability to bind to zona pellucida surface, and electrophoretic properties and, furthermore, stabilization of some sperm structures by the establishment of disulphide linkages takes place in several sperm structures. The cytoplasmic perinuclear theca (PT) is a unique extranuclear cytoskeletal element that surrounds the nucleus, which is proposed to be a structural scaffold to the spe

Gastric related neurons in the rat medial vestibular nucleus.

Some structural and functional peculiarities of the rat vestibular nuclei neurones involved in realisation of vestibular-gastrointestinal reflectory reactions were studied. After microinjection of a horseradish peroxidase solution in the 'gastric' area of the nucleus tractus solitarius, retrogradely-labelled cell bodies were found in caudal part of the medial vestibular nucleus. Electrical stimulation of these neurons resulted in the decrease of gastric tone. The respiratory arrest was registered simultaneously. The results suggest that activation of the identified vestibular neurons can induc

Involvement in surface antigen expression by a moonlighting FG-repeat nucleoporin in trypanosomes.

Components of the nuclear periphery coordinate a multitude of activities, including macromolecular transport, cell-cycle progression, and chromatin organization. Nuclear pore complexes (NPCs) mediate nucleocytoplasmic transport, mRNA processing, and transcriptional regulation, and NPC components can define regions of high transcriptional activity in some organisms at the nuclear periphery and nucleoplasm. Lineage-specific features underpin several core nuclear functions and in trypanosomatids, which branched very early from other eukaryotes, unique protein components constitute the lamina, kin

Atrial natriuretic factor in central nervous system regulatory mechanisms: effect of experimental alterations in water and salt homeostasis and blood pressure.

Atrial natriuretic factor (ANF) is widely distributed in the preoptic area and the hypothalamus, it is present there both in cell bodies and nerve terminals. Effect of experimental alterations in the salt and water balance was examined on preoptic-hypothalamic ANF levels measured in ten microdissected nuclei. Immunohistochemical analysis was also performed to confirm radioimmunological results. Following interventions were performed in adult male rats: adrenalectomy (5 days), daily 0.9% NaCl, aldosterone (5 micrograms/100 g) and dexamethasone (2 micrograms/ml drinking water) treatments in both

Importin alpha 1 is required for the nucleus entry of Fowl Adenovirus serotype 4 Fiber-1 protein.

Fiber-1 protein (F1) is the structural protein of Fowl Adenovirus serotype 4 (FAdV-4), which could recondite the receptors of host cytomembrane. In this study, we firstly determined that F1 protein located in nucleus of LMH cells after infection with FAdV-4. We additionally revealed that F1 protein had a classic NLS, and the NLS was required for F1 nucleus entry, which was intently associated to the 26th Pro in NLS. The time rule result indicated that some F1 proteins firstly positioned in the nucleus 6 h posttranfection, and it entirely located in the nucleus 12 h posttranfection, then it ord

Maternal embryonic leucine zipper kinase (MELK) optimally regulates the HIV-1 uncoating process.

Human immunodeficiency virus type-1 (HIV-1) attaches to target cells and releases the capsid, an essential component of the viral core that contains viral RNA, into the cytoplasm. After invading target cells, the core structure gradually collapses. The timing of the disassembly of the HIV-1 capsid is essential for efficient viral cDNA synthesis and transport into the nucleus. HIV-1 uncoating is controlled by the host factor maternal embryonic leucine zipper kinase (MELK); however, the quantitative and dynamic relationship between the HIV-1 uncoating process and HIV-1 infection remains unresolv

[Pathogenesis of transparency disorders of the optical environment of the eye. IV. Electron microscopy study of the vitreous body in chalcosis].

The authors proved, based on electron microscopic analysis that the vitreous body of the rabbit creates under physiological conditions a regular network of collagenous fibrils with a different cell population. The cells can be divided into three basic types: 1. Large cells with a lobular nucleus and well differentiated cytoplasm and multiple organelles which ensure a high metabolic activity. 2. The second type are cells of an elongated shape with an oval nucleus surrounded by a thin layer of cytoplasm with a small amount of organelles. 3. The third type are large globular cells with a homogene

Rapid Dissection and Dissociation of the Mouse Olfactory Epithelium for Single-Nucleus Suspensions.

The murine olfactory epithelium is the initial entry point of the olfactory system, housing various cell types that include olfactory sensory neurons, their regenerating progenitors, and support cells. Olfactory sensory neurons transduce chemical odorants into neural signals, yet the mechanisms underlying how these cells develop and turnover, create synapses with the olfactory bulb, and regulate their odorant receptors remain areas of intense study. Located on the dorsal aspect of the nasal cavity, the olfactory epithelium adheres to intricate bony structures known as turbinates. This anatomy

Dynamic as well as stable protein interactions contribute to genome function and maintenance.

The cell nucleus is responsible for the storage, expression, propagation, and maintenance of the genetic material it contains. Highly organized macromolecular complexes are required for these processes to occur faithfully in an extremely crowded nuclear environment. In addition to chromosome territories, the nucleus is characterized by the presence of nuclear substructures, such as the nuclear envelope, the nucleolus, and other nuclear bodies. Other smaller structural entities assemble on chromatin in response to required functions including RNA transcription, DNA replication, and DNA repair.

Barriers to mesenchymal stromal cells for low back pain.

Intervertebral disc degeneration is the main cause of low back pain. In the past 20 years, the injection of mesenchymal stromal cells (MSCs) into the nucleus pulposus of the degenerative disc has become the main approach for the treatment of low back pain. Despite the progress made in this field, there are still many barriers to overcome. First, intervertebral disc is a highly complex load-bearing composite tissue composed of annulus fibrosus, nucleus pulposus and cartilaginous endplates. Any structural damage will change its overall biomechanical function, thereby causing progressive degenera

An unusual modification of endoplasmic reticulum in epithelial cells of the mouse ascending colon.

A rarely occurring structure that is apparently a modification of endoplasmic reticulum was observed in the epithelial cells of the ascending colon of the mouse. The structure consists of a stack of 3 to 15 parallel cisterna-like elements separated by about 67 nm. The stacked cisternae usually are located adjacent to the basal end of the nucleus or in the region between nucleus and basal cell membrane. Top and bottom cisternae of many of the stacks have patent lumens and their outer membranes are lined by ribosomes. Most frequently, middle cisternae have attenuated lumens in their central regi

Translational and Posttranslational Dynamics in a Model Peptidergic System.

The cell bodies of hypothalamic magnocellular neurones are densely packed in the hypothalamic supraoptic nucleus, whereas their axons project to the anatomically discrete posterior pituitary gland. We have taken advantage of this unique anatomical structure to establish proteome and phosphoproteome dynamics in neuronal cell bodies and axonal terminals in response to physiological stimulation. We have found that proteome and phosphoproteome responses to neuronal stimulation are very different between somatic and axonal neuronal compartments, indicating the need of each cell domain to differenti

Neurodegeneration after repeated noise trauma in the mouse lower auditory pathway.

High intensity noise exposure leads to a permanent shift in auditory thresholds (PTS), affecting both peripheral (cochlear) tissue and the central auditory system. Studies have shown that a noise-induced hearing loss results in significant cell loss in several auditory structures. Degeneration can be demonstrated within hours after noise exposure, particularly in the lower auditory pathway, and continues to progress over days and weeks following the trauma. However, there is limited knowledge about the effects of recurring acoustic trauma. Repeated noise exposure has been demonstrated to incre

Design, Synthesis, and Anticancer Activity of Drug-like Iron Chelators/G-Quadruplex Binders as Synergic Dual Targeting Agents.

Iron homeostasis is strictly related to numerous physiological pathways including cell cycle progression and cell growth. The newest anticancer strategies focus on either depleting the cells with a suitable chelator or increasing their loading by administering iron complexes to induce ferroptosis. Iron depletion inhibits cell proliferation, while iron overload induces the damage of guanine nucleobases in G-quadruplex structures via ROS generation, leading to genome instability. Here, we demonstrated that designing a molecular chimera embodying structural requirements for both iron chelation an

Nucleocytoplasmic transport of nucleocapsid proteins of enveloped RNA viruses.

Most viruses with non-segmented single stranded RNA genomes complete their life cycle in the cytoplasm of infected cells. However, despite undergoing replication in the cytoplasm, the structural proteins of some of these RNA viruses localize to the nucleus at specific times in the virus life cycle, primarily early in infection. Limited evidence suggests that this enhances successful viral replication by interfering with or inhibiting the host antiviral response. Nucleocapsid proteins of RNA viruses have a well-established, essential cytoplasmic role in virus replication and assembly. Intriguin
