Interaction of pulsed low frequency electromagnetic field (PEMF) with mitochondria.

Pulsed electromagnetic field (PEMF) therapy is a non-invasive treatment that delivers electric and magnetic fields to tissues via inductive coils inducing salutary effects. Previously PEMF was reported to accelerate gas transport in the liquid phase. Drawing analogies from electrical elements, we hypothesized that PEMF can modulate transmembrane potential of biological membranes. Given that mitochondria interact with gaseous molecules such as oxygen and nitric oxide (NO) and contain voltage-sensitive channels, this study focused on the effects of a single PEMF device with a low input-energy an

Application of a high-throughput respirometry technique for assessing respiratory metabolism of terrestrial and aquatic invertebrates.

Respiratory metabolism is one of the key ecological traits of organisms. As such, it is studied by ecologists from a wide range of disciplines, from macroecological studies to evolutionary biology, with the global changes context making the need for its understanding more pressing. Unfortunately, properly measuring respiration is a time- and cost-intensive endeavor, and gathering large datasets can be prohibitive due to the difficulty of measuring it. Here, we describe a fluorescence-based high-throughput closed-system respirometry technique for terrestrial and aquatic invertebrates, able to m

Blockade of Na+/H+ exchanger type 3 causes intracellular acidification and hyperexcitability via inhibition of pH-sensitive K+ channels in chemosensitive respiratory neurons of the dorsal vagal nucleus in rats.

Extracellular pH (pHe) and intracellular pH (pHi) are important factors for the excitability of chemosensitive central respiratory neurons that play an important role in respiration and obstructive sleep apnea. It has been proposed that inhibition of central Na(+)/H(+) exchanger 3 (NHE-3), a key pHi regulator in the brainstem, decreases the pHi, leading to membrane depolarization for the maintenance of respiration. However, how intracellular pH affects the neuronal excitability of respiratory neurons remains largely unknown. In this study, we showed that NHE-3 mRNA is widely distributed in res

Pseudomonas aeruginosa-secreted respiratory toxin HQNO triggers fatty acid accumulation in respiring Staphylococcus aureus, decreasing SaeRS-dependent transcriptional regulation.

Staphylococcus aureus and Pseudomonas aeruginosa are the two pathogens that colonize the airway of cystic fibrosis patients. As patients age, P. aeruginosa outcompetes S. aureus to become the predominant organism in the airway, which overlaps with worsening symptoms. This inverse correlation is partly due to the ability of P. aeruginosa to secrete secondary metabolites and virulence factors that are antagonistic to the host cells and other bacteria present. Several of these secondary metabolites inhibit S. aureus respiration. SaeRS is a two-component regulatory system that promotes the transcr

Abnormal cardiac autonomic control in sickle cell disease following transient hypoxia.

Abnormalities in autonomic control in sickle cell anemia (SCA) patients have been reported by multiple researchers. However their potential causal association with sickle cell crisis remains unknown. We employed hypoxia, a known trigger to sickle cell crisis, to perturb the autonomic systems of the subjects. Cardiac autonomic control was non-invasively assessed by tracking the changes in heart rate variability (HRV) that occur following brief exposure to a hypoxia stimulus. Time varying spectral analysis of HRV was applied to estimate the cardiac autonomic response to the transient episode of

The use of germicidal ultraviolet light, vaporized hydrogen peroxide and dry heat to decontaminate face masks and filtering respirators contaminated with a SARS-CoV-2 surrogate virus.

BACKGROUND: In the context of the ongoing severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic, the supply of personal protective equipment remains under severe strain. To address this issue, re-use of surgical face masks and filtering facepiece respirators has been recommended; prior decontamination is paramount to their re-use. AIM: We aim to provide information on the effects of three decontamination procedures on porcine respiratory coronavirus (PRCV)-contaminated masks and respirators, presenting a stable model for infectious coronavirus decontamination of these typicall

Interaction of plant cell signaling molecules, salicylic acid and jasmonic acid, with the mitochondria of Helicoverpa armigera.

The cotton bollworm, Helicoverpa armigera is a polyphagous pest in Asia, Africa, and the Mediterranean Europe. Salicylic acid (SA) and jasmonic acid (JA) are the cell signaling molecules produced in response to insect attack in plants. The effect of these signaling molecules was investigated on the oxidative phosphorylation and oxidative stress of H. armigera. SA significantly inhibited the state III and state IV respiration, respiratory control index (RCI), respiratory complexes I and II, induced mitochondrial swelling, and cytochrome c release in vitro. Under in vivo conditions, SA induced s

DSCAM deficiency causes loss of pre-inspiratory neuron synchroneity and perinatal death.

Down syndrome cell adhesion molecule (DSCAM) is a neural adhesion molecule that plays diverse roles in neural development. We disrupted the Dscam locus in mice and found that the null mutants (Dscam(-/-)) died within 24 h after birth. Whole-body plethysmography showed irregular respiration and lower ventilatory response to hypercapnia in the null mutants. Furthermore, a medulla-spinal cord preparation of Dscam(-/-) mice showed that the C4 ventral root activity, which drives diaphragm contraction for inspiration, had an irregular rhythm with frequent apneas. Optical imaging of the preparation u

Production of thiosulphate during sulphide oxidation by mitochondria of the symbiont-containing bivalve Solemya reidi.

Isolated mitochondria of the bivalve Solemya reidi Bernard oxidize sulphide and couple this oxidation to ADP phosphorylation. The products of mitochondrial sulphide oxidation were analyzed by HPLC using monobromobimane derivatization. Concurrent measurements of respiration were made using sulphide-insensitive oxygen electrodes. S. reidi mitochondria oxidized sulphide exclusively to thiosulphate. The reaction occurred in two steps. One sulphide molecule was first oxidized to sulphite. A second molecule of sulphide was then added oxidatively to form the free product thiosulphate. This oxidation

Chloramphenicol-induction of a second cyanide-and azide-insensitive mitochondrial pathway in Ustilago maydis.

Growth of wild type Ustilago maydis in presence of chloramphenicol results in increased resistance of cyanide-insensitive respiration to hydroxamic acid derivatives. The biphasic nature of the Dixon plots for hydroxamate inhibition of mitochondrial alternative respiration suggested involvement of two cyanide-sensitive systems. This hypothesis was confirmed by the analysis of kinetic data. One of the two systems exhibits normal sensitivity to hydroxamates and appears similar to that present in mitochondria from cells grown in control medium. In contrast, the inducible system is characterised by

Bioenergetics in fibroblasts of patients with Huntington disease are associated with age at onset.

OBJECTIVE: We aimed to assess whether differences in energy metabolism in fibroblast cell lines derived from patients with Huntington disease were associated with age at onset independent of the cytosine-adenine-guanine (CAG) repeat number in the mutant allele. METHODS: For this study, we selected 9 pairs of patients with Huntington disease matched for mutant CAG repeat size and sex, but with a difference of at least 10 years in age at onset, using the Leiden Huntington disease database. From skin biopsies, we isolated fibroblasts in which we (1) quantified the ATP concentration before and af

Retinal Pigment Epithelium Injury in Pentosan Polysulfate Exposure: Morphologic Changes, Phagocytic Deficits, and Mitochondrial Dysfunction.

Pentosan polysulfate (PPS) is a semisynthetic sulfated polysaccharide that was approved by the United States Food and Drug Administration (FDA) for treatment of interstitial cystitis (IC). A 2018 study by our group described a vision-threatening macular toxicity associated with long-term use of PPS. However, given the relatively recent characterization of PPS maculopathy, we have limited knowledge of its pathophysiology. The present study therefore investigated the pathophysiology of PPS maculopathy in a cell culture model, assessing impacts of PPS exposure on morphology and mitochondrial func

Mitochondrial dysfunction in bipolar disorder: Evidence, pathophysiology and translational implications.

Bipolar disorder (BD) is a chronic psychiatric illness characterized by severe and biphasic changes in mood. Several pathophysiological mechanisms have been hypothesized to underpin the neurobiology of BD, including the presence of mitochondrial dysfunction. A confluence of evidence points to an underlying dysfunction of mitochondria, including decreases in mitochondrial respiration, high-energy phosphates and pH; changes in mitochondrial morphology; increases in mitochondrial DNA polymorphisms; and downregulation of nuclear mRNA molecules and proteins involved in mitochondrial respiration. Mi

A microfluidic platform for isolating functional mitochondria and restoring bioenergetic activity in hypoxia-injured cardiomyocytes.

Hypoxic stress is known to severely impair mitochondrial function in cardiomyocytes, leading to disruption of intracellular bioenergetic homeostasis, reduced ATP production, and compromised cellular performance. Because cardiomyocytes rely heavily on mitochondrial oxidative metabolism to meet their high energy demands, mitochondrial dysfunction represents a central mechanism underlying hypoxia-induced cardiac injury. In this study, a microfluidic-based platform was developed for mitochondrial isolation and to investigate whether the delivery of functionally preserved mitochondria could enhance

Calbindin 2-specific deletion of arginase 2 preserves visual function after optic nerve crush.

We previously found that global deletion of the mitochondrial enzyme arginase 2 (A2) limits optic nerve crush (ONC)-induced neuronal death. Herein, we examined the cell-specific role of A2 in this pathology by studies using wild type (WT), neuronal-specific calbindin 2 A2 KO (Calb2cre/+ A2 f/f), myeloid-specific A2 KO (LysMcre/+ A2f/f), endothelial-specific A2 KO (Cdh5cre/+ A2f/f), and floxed controls. We also examined the impact of A2 overexpression on mitochondrial function in retinal neuronal R28 cells. Immunolabeling showed increased A2 expression in ganglion cell layer (GCL) neurons of WT

Nr4a3 deficiency disrupts MEK1-ERK1/2-Drp1 signaling, driving adiposity and glucose intolerance in male mice.

Obesity is strongly associated with elevated blood glucose levels, glucose intolerance, insulin resistance, and type 2 diabetes. The Nr4a family of orphan nuclear receptors is essential for proliferation, cell survival, mitochondrial function, and fuel utilization in a tissue-dependent manner. Nr4a3 overexpression has been shown to decrease blood glucose levels and improve glucose tolerance. Here, we present the effects of full body Nr4a3 deletion in mice fed a standard chow diet. We demonstrate that male and female Nr4a3 knockout mice fed a standard chow diet have elevated nonfasting blood gl

Proanthocyanidin-containing polyphenol extracts from fruits prevent the inhibitory effect of hydrogen sulfide on human colonocyte oxygen consumption.

Hydrogen sulfide (H2S), a metabolic end product synthesized by the microbiota from L-cysteine, has been shown to act at low micromolar concentration as a mineral oxidative substrate in colonocytes while acting as an inhibitor of oxygen consumption at higher luminal concentrations (65 µM and above). From the previous works showing that polyphenols can bind volatile sulfur compounds, we hypothesized that different dietary proanthocyanidin-containing polyphenol (PACs) plant extracts might modulate the inhibitory effect of H2S on colonocyte respiration. Using the model of human HT-29 Glc-/+ cell c

The role of stem respiration in cold-acclimation of winter-dormant Quercus robur trees under large air temperature fluctuations.

Cold acclimation of leaf respiration often increases respiration rates at a given temperature after temperature declines. Whether a similar response occurs in stems and contributes to frost tolerance remains unknown. We evaluated stem CO2 efflux (Es) and cold acclimation in dormant Quercus robur trees in a climate chamber with day/night air temperatures progressively decreasing week-by-week from 20/10 °C to 5/-10 °C and then progressively increasing back to 20/10 °C over 9 weeks. Cold hardening was evidenced by lower cell damage at sub-freezing temperatures compared with trees kept in a greenh
