Ovarian injury caused by chemotherapy can lead to early menopause, infertility, and even premature senility in female cancer patients, impairing the quality of life and overall health of the cancer survivors seriously. However, there is still a lack of effective protection strategies against such injury. Cellular senescence can be induced by chemotherapeutic agents in multiple organs and may corrode the structure and function of normal tissues. We hypothesized that the widely used first-line chemotherapy drug, doxorubicin, could increase senescent cell burden in normal ovarian tissue during th

Calcium research, since its pivotal discovery in the early 1800s through the heating of limestone, has led to the identification of its multi-functional roles. These include its functions as a reducing agent in chemical processes, structural properties in shells and bones, and significant role in cells relating to this review: cellular signaling. Calcium signaling involves the movement of calcium ions within or between cells, which can affect the electrochemical gradients between intra- and extracellular membranes, ligand binding, enzyme activity, and other mechanisms that determine cell fate.

Osteoarthritis (OA) is a degenerative joint disease characterized by low-grade inflammation and cartilage degradation. Dendrobine (DEN) is reported to inhibit inflammation and oxidative stress in some diseases, but its role in chondrocyte senescence and OA progress has not yet been elucidated. Our study aimed to explore the protective effects of DEN on OA both in vitro and in vivo. We found that DEN inhibited extracellular matrix (ECM) degradation and promoted ECM synthesis. Meanwhile, DEN inhibited senescence-associated secretory phenotype (SASP) factors expression and senescence phenotype in

The locomotor system comprises skeletal muscles and bones with active metabolism and cellular turnover. Chronic locomotor system disorders gradually arising with aging are inversely associated with the correct function of bone and muscles. Senescent cells appear more frequently in advanced ages or pathological conditions, and the accumulation of senescent cells in muscle tissue negatively correlates with muscle regeneration, which is crucial for maintaining strength and preventing frailty. Senescence in the bone microenvironment, osteoblasts, and osteocytes affects bone turnover favoring osteo

Cellular senescence is a dynamic process driven by epigenetic and genetic changes. Although some transcriptomic signatures of senescent cells have been discovered, how these senescence-related signals change over time remains largely unclear. Here, we profiled the transcriptome dynamics of human dermal fibroblast (HDF) cells in successive stages of growth from proliferation to senescence. Based on time-series expression profile analysis, we discovered four trajectories (C1, C2, C3, C4) that are dynamically expressed as senescence progresses. While some genes were continuously up-regulated (C4)

Senescence-associated β-galactosidase (hereafter SA-β-gal) staining has now been employed for more than 20 years to identify the presence of senescent cells (Dimri et al., Proc Natl Acad Sci U S A 92:9363-9367, 1995). These cells, characterized by a permanent cell-cycle arrest (Hayflick and Moorhead, Exp Cell Res 25:585-621, 1961) and the production of a distinct secretory phenotype of cytokines, chemokines, and proteases (Coppe et al., PLoS Biol 6:2853-2868, 2008), have received much attention in recent years for their impacts on diverse biological processes. Here we describe a method to iden

Cellular senescence, a term originally used to define the characteristics of normal human fibroblasts that reached their replicative limit, is an important factor for ageing, age-related diseases including cancer, and cell reprogramming. These outcomes are mediated by senescence-associated changes in gene expressions, which sometimes lead to the secretion of pro-inflammatory factors, or senescence-associated secretory phenotype (SASP) that contribute to paradoxical pro-tumorigenic effects. p53 functions as a transcription factor in cell-autonomous responses such as cell-cycle control, DNA repa

Cellular senescence is a stress-responsive cell fate characterized by stable growth arrest and acquisition of a proinflammatory senescence-associated secretory phenotype. Senescent cells accumulate with advancing age and contribute to dysfunction and disease across multiple organ systems. In skeletal muscle (SkM), emerging evidence indicates that senescence arises across diverse cell populations that collectively reshape the SkM niche. Through the SASP and altered intercellular signaling, senescent cells promote chronic inflammation, fibrosis, impaired regeneration, and metabolic dysfunction t

Coronary artery disease (CAD) is a major global cause of morbidity and mortality, and population ageing is reshaping its clinical phenotype, disease trajectory, and therapeutic complexity. Chronological age alone cannot explain the heterogeneity of CAD outcomes in older individuals, highlighting the need to integrate biological ageing into cardiovascular disease research and management. Cellular senescence, characterised by stable cell-cycle arrest, metabolic reprogramming, and acquisition of a senescence-associated secretory phenotype (SASP), provides a mechanistic bridge between ageing and C

Parkinson's disease (PD) is a progressive neurological disorder that primarily affects motor movement but also causes a range of non-motor symptoms. Characteristic hallmarks include progressive loss of dopaminergic neurons in the substantia nigra pars compacta and the buildup of intracellular protein aggregates known as Lewy bodies containing the protein alpha-synuclein. While the etiology of PD is multifactorial, aging is widely recognized as the most significant risk factor for the disorder. Cellular senescence, a hallmark of aging, has in recent years emerged as a potential driver of neurod

Cellular senescence has emerged as a key contributor to brain aging and neurodegenerative diseases, which are increasing in prevalence as life expectancy rises. Senescent cells accumulate in multiple brain cell populations and secrete the senescence-associated secretory phenotype (SASP), promoting chronic neuroinflammation, mitochondrial dysfunction, blood-brain barrier disruption, and progressive neuronal damage. This review examines the role of cellular senescence in brain aging and neurodegenerative diseases and discusses senotherapeutic strategies aimed at either eliminating senescent cell

Despite rapid advances in anticancer therapy, cancer incidence and mortality remain substantial worldwide. Although immune checkpoint inhibitors (ICIs), including anti-PD-1/PD-L1 and anti-CTLA-4 therapies, have transformed cancer treatment by harnessing host antitumor immunity, their efficacy remains limited to a subset of patients, largely because of the complexity of the tumor microenvironment. This review examines drug-induced immunogenic cell death (ICD) and drug-induced senescence as complementary strategies for overcoming these limitations and enhancing immunotherapeutic responses. We fu

Endothelial senescence is a critical contributor to vascular aging and age-related vasculopathies. Our previous work identified AGGF1 as a regulator of cell cycle progression and anti-inflammatory signaling. However, whether AGGF1 modulates endothelial senescence remains unclear. Here, we demonstrate that AGGF1 is downregulated in both replicative and DOX-induced senescent HUVECs. AGGF1 knockdown accelerated cellular senescence as evidenced by senescence markers, including increased SA-β-gal activity, enhanced formation of γH2AX foci, elevated IL-6 levels, and impaired proliferation, whereas A

Background. Cellular senescence is a negative prognostic indicator in glioblastoma (GB). However, the specific cell types exhibiting senescence and the molecular mechanisms by which senescent cells (SCs) contribute to GB pathogenesis remain unknown. Methods. We performed multi-omics integration of publicly available GB patient-derived datasets to identify SCs and their functional impact on GB pathogenesis. We created a transcriptomic definition of SCs to verify their presence in GB datasets. Next, we analyzed transcriptomic profiles of GB and low-grade gliomas to reveal key features of GB aggr

Aging-associated loss of chromatin compaction is linked to derepression of retrotransposable elements (RTEs) in mouse and human tissues. Whether such RTE transcription contributes to the microglia activation that is common in aged brains is unknown. Here, we show that DAXX, a histone chaperone and RTE repressor, is downregulated during aging, preserves microglia homeostasis and inhibits cellular senescence. Loss of Daxx in young-adult microglia drives a reactive phenotype marked by chromatin decompaction at RTEs, loss of homeostatic markers, cell cycle re-entry and behavioral changes. This sta

Aging involves the accumulation of molecular alterations within cells and the extracellular matrix, resulting in cellular senescence and declining physiological functions. This study investigates the correlation between the biophysical environment and cellular aging, specifically examining how mechanical and biochemical cues affect cellular senescence and tissue degeneration. Cells were cultured on acrylamide hydrogels of different stiffnesses (4 and 19 kPa), and their mechanical properties were characterized by measuring Young's modulus via compression tests. Cell proliferation, morphology, g

Cellular senescence precipitates a decline in physiological activities and metabolic functions, often accompanied by heightened inflammatory responses, diminished immune function, and impaired tissue and organ performance. Despite extensive research, the mechanisms underpinning cellular senescence remain incompletely elucidated. Emerging evidence implicates circadian rhythm and hypoxia as pivotal factors in cellular senescence. Circadian proteins are central to the molecular mechanism governing circadian rhythm, which regulates homeostasis throughout the body. These proteins mediate responses

BACKGROUND: Cellular senescence (CS) is a hallmark of ageing and age-related diseases. While dietary interventions are often explored to reduce CS, less is known about dietary ingredients that induce it. This study systematically reviews the evidence on dietary ingredients that promote CS in animal models and humans.
METHODS: Following PRISMA guidelines (PROSPERO: CRD42022338885), PubMed and Embase were searched for studies on dietary ingredients administered via the gastrointestinal tract affecting CS markers in animal models or adults. Risk of bias was assessed using SYRCLE's and Cochrane's

Benign prostatic hyperplasia (BPH) is a prevalent condition among aging men, characterized by prostate enlargement and urinary symptoms. Despite its common occurrence, the molecular mechanisms, particularly those related to cellular senescence, are not well understood. This study aims to pinpoint and analyze cellular senescence-related differentially expressed genes (CSRDEGs) in BPH. We integrated data from GEO datasets GSE119195 and GSE7307, addressing batch effects to ensure data reliability. Differential gene expression analysis revealed key CSRDEGs, which were further analyzed through Gene

Hepatocellular carcinoma (HCC) is the most common form of liver cancer and remains a global health challenge. The biological process of HCC is very complex, involving the imbalance of tumor suppressor genes and oncogenes, abnormal activation of molecular signaling pathways, and the differentiation of HCC cells. Standard clinical approaches for HCC treatment encompass surgery, chemotherapy, and radiation therapy. However, treatment options for advanced HCC are constrained, primarily due to an incomplete understanding of its underlying mechanisms. Cellular senescence is a crucial mechanism that
