The Impact of Injury Severity, Sex, and Nociceptor Activity on Post-Traumatic Temporomandibular Joint Osteoarthritis in Mice.

BACKGROUND: Intrinsic and extrinsic trauma to the temporomandibular joint (TMJ) increases the incidence of painful temporomandibular disorders. TMJ trauma also causes increased degeneration of the TMJ structure, leading to TMJ osteoarthritis (TMJOA). Clinically, it is important to identify risk factors of post-traumatic TMJOA, such as sex, injury severity, and persistent post-traumatic pain. OBJECTIVES: We determined the effects of potential risk factors on post-traumatic TMJ degeneration using a mouse model. METHODS: A repeated forced-mouth opening (FMO) model was used to determine the effe

Transcranial direct current stimulation alleviates depression-like behaviors by activating the ACC-ZI neural circuit.

BACKGROUND: Depression is a common mental disorder that imposes a significant burden on individuals and society. Transcranial direct current stimulation (tDCS) has attracted much attention as a non-invasive treatment method, but its underlying neurobiological mechanisms remain elusive. METHODS: We assessed the behavioral effects of prefrontal-targeted tDCS at different current intensities and polarities with the chronic restraint stress (CRS) model of depression. Neuronal activation induced by tDCS was mapped using c-Fos staining, while optical fiber photometry and whole-cell patch-clamp reco

Role of Supramammillary Nucleus Glutamatergic Neurons in Modulating Consciousness Transitions during Propofol Anesthesia in Mice.

BACKGROUND: Glutamatergic neurons in the supramammillary nucleus (SuM) have previously been shown to be pivotal parts of the wakefulness-sleep regulation system. However, whether they play a role in propofol-mediated modulation of consciousness remains unclear. This study hypothesized that glutamatergic neurons in the SuM contribute to the regulation of altered states of consciousness under propofol anesthesia in mice. METHODS: The effects of propofol anesthesia on neuronal activity were measured by calcium fiber photometry recording. Lesions and chemogenetic activation were used to investiga

Regulation of REM and NREM sleep by preoptic glutamatergic neurons.

The preoptic area of the hypothalamus is key for the control of sleep onset and sleep homeostasis. Although traditionally considered exclusively somnogenic, recent studies identified a group of preoptic glutamatergic neurons that promote wakefulness. Specifically, our previous investigations demonstrated that chemogenetic stimulation of glutamatergic neurons within the medial-lateral preoptic area (MLPO_VGLUT2) promotes wakefulness, fragments non-rapid eye movement sleep (NREMs), and suppresses REM sleep (REMs). This evidence is further supported by recent work showing that preoptic glutamater

Systemic Glucose Regulation by a Hindbrain Inhibitory Circuit in a Mouse Model of Type 1 Diabetes.

INTRODUCTION: Previous work showed that increasing the electrical activity of inhibitory neurons in the dorsal vagal complex (DVC) is sufficient to increase whole-body glucose concentration in normoglycemic mice. Here we tested the hypothesis that deactivating GABAergic neurons in the dorsal hindbrain of hyperglycemic mice decreases synaptic inhibition of parasympathetic motor neurons in the dorsal motor nucleus of the vagus (DMV) and reduces systemic glucose levels. METHODS: Chemogenetic activation or inactivation of GABAergic neurons in the nucleus tractus solitarius (NTS) was used to asses

Regulation of states of consciousness by supramammillary nucleus glutamatergic neurones during sevoflurane anaesthesia in mice.

BACKGROUND: The supramammillary nucleus (SuM), located in the caudal hypothalamus, includes wake-promoting glutamatergic neurones. Their potential role in regulating states of consciousness during general anaesthesia remains unknown. METHODS: We used in vivo fibre photometry, c-Fos staining, chemogenetic and optogenetic manipulations, and electroencephalography/electromyography to explore the roles of glutamatergic SuM neurones (SuMVglut2 neurones) at different phases of sevoflurane anaesthesia. Rabies-mediated retrograde and anterograde tract tracing were used to investigate the monosynaptic

ipRGC properties prevent light from shifting the SCN clock during daytime.

The suprachiasmatic nucleus (SCN), the central circadian pacemaker, receives photic input exclusively from intrinsically photosensitive retinal ganglion cells (ipRGCs)1,2. However, light mainly shifts the SCN clock during night-time3-5. Here we induced phase shifts in the SCN clock during the daytime in mice by activating ipRGCs using chemogenetics or violet light. Our data reveal that the inability to induce daytime shifts with light in most animals is not only attributed to the SCN, as has been proposed for decades, but also requires the limitation of ipRGC firing via depolarization block. Cerebellar Bergmann glia integrate noxious information and modulate nocifensive behaviors.

The cerebellum is activated by noxious stimuli and pathological pain but its role in noxious information processing remains unknown. Here, we show that in mice, cutaneous noxious electrical stimuli induced noradrenaline (NA) release from locus coeruleus (LC) terminals in the cerebellar cortex. Bergmann glia (BG) accumulated these LC-NA signals by increasing intracellular calcium in an integrative manner ('flares'). BG flares were also elicited in response to an intraplantar capsaicin injection. Chemogenetic inactivation of LC terminals or BG in the cerebellar cortex or BG-specific knockdown of

Cerebellar climbing fibers impact experience-dependent plasticity in the mouse primary somatosensory cortex.

In the cerebellum, climbing fibers (CFs) provide instructive signals for supervised learning at parallel fiber to Purkinje cell synapses. It has not been tested so far whether CF signaling may also influence plasticity in other brain areas. Here, we show that optogenetic CF activation suppresses potentiation of whisker responses in L2/3 pyramidal cells in primary somatosensory cortex (S1) of awake mice that is observed after repeated whisker stimulation. Using two-photon imaging and chemogenetics, we find that CFs control plasticity by modulating SST- and VIP-positive interneurons in S1 cortex

Targeting the hypothalamic a11 nucleus to treat parkinsonian-like nociceptive impairments.

Pain is a common non-motor symptom in Parkinson's disease (PD), yet treatment options remain limited due to incomplete understanding of underlying mechanisms. Using the 6-hydroxydopamine (6-OHDA) rat model, we combined pharmacological, behavioural, chemogenetic, electrophysiological, and immunohistochemical approaches to investigate dopaminergic modulation of nociception by the hypothalamic A11 projecting to the dorsal horn of the spinal cord (DHSC). We demonstrate that A11 dopaminergic neurons are the sole source of dopamine in the DHSC. Activation of both D1 and D2 receptors alleviated mecha

Leptin signaling in the dorsomedial hypothalamus couples breathing and metabolism in obesity.

Mismatch between CO2 production (Vco2) and respiration underlies the pathogenesis of obesity hypoventilation. Leptin-mediated CNS pathways stimulate both metabolism and breathing, but interactions between these functions remain elusive. We hypothesized that LEPRb+ neurons of the dorsomedial hypothalamus (DMH) regulate metabolism and breathing in obesity. In diet-induced obese LeprbCre mice, chemogenetic activation of LEPRb+ DMH neurons increases minute ventilation (Ve) during sleep, the hypercapnic ventilatory response, Vco2, and Ve/Vco2, indicating that breathing is stimulated out of proporti

Prenatal Exposure to Ketamine Leads to Anxiety-Like Behaviors and Dysfunction in Bed Nucleus of Stria Terminalis.

BACKGROUND: Both the clinical and preclinical studies have suggested embryonic or infant exposure to ketamine, a general anesthetic, pose a great threat to the developing brain. However, it remains unclear how ketamine may contribute to the brain dysfunctions. METHODS: A mouse model of prenatal exposure to ketamine was generated by i.m. injection and continuous i.p. infusion of pregnant mice. Open field test and elevated plus maze test were used to analyze the behavioral alterations induced by ketamine. Immunostaining by c-Fos was used to map the neuron activity. Astrocyte Gi-GPCR signaling corrects compulsive-like grooming and anxiety-related behaviors in Sapap3 knockout mice.

Astrocytes are morphologically complex cells that serve essential roles. They are widely implicated in central nervous system (CNS) disorders, with changes in astrocyte morphology and gene expression accompanying disease. In the Sapap3 knockout (KO) mouse model of compulsive and anxiety-related behaviors related to obsessive-compulsive disorder (OCD), striatal astrocytes display reduced morphology and altered actin cytoskeleton and Gi-G-protein-coupled receptor (Gi-GPCR) signaling proteins. Here, we show that normalizing striatal astrocyte morphology, actin cytoskeleton, and essential homeosta

Electroacupuncture Relieves Neuropathic Pain by Suppressing ACC Pyramidal Activity via mGluR5 Inhibition in CCI Mice.

BACKGROUND: Electroacupuncture (EA) is widely used for analgesia, but its central mechanisms remain unclear. We investigated whether EA alleviates neuropathic pain by suppressing metabotropic glutamate receptor 5 (mGluR5) signaling in the anterior cingulate cortex (ACC). METHODS: In naïve mice, we manipulated ACC pyramidal neurons using adeno-associated viral (AAV) vectors encoding calcium/calmodulin-dependent protein kinase II (CaMKII)-driven opsins-channelrhodopsin-2 (ChR2) or halorhodopsin (NpHR3.0)-followed by blue- or yellow-light stimulation to assess behavioral responses. [Social isolation induces social avoidance in mice: the mediating role of the medial prefrontal cortex glutamatergic neurons].

OBJECTIVES: To investigate social isolation (SI)-induced abnormal behaviors and changes in neuronal activity in the medial prefrontal cortex (mPFC) of mice. METHODS: Male C57BL/6J mice kept under normal conditions or SI mice housed in individual cages for 2 weeks were subjected to intracerebral microinjection of a control virus, a chemogenetic activation virus, or an optogenetic activation virus (n=8). The mice were assessed for behavioral changes and examined for neuronal changes in mPFC using c-fos immunofluorescence staining and co-staining with specific antibodies against CaMKIIα (for exc

GABAergic ventrolateral preoptic projection to dorsomedial hypothalamus recapitulates post-ischemic neuroprotection by hypothermia.

Therapeutic hypothermia by exogenous cooling induces potent neuroprotection. Post-stroke, therapeutic hypothermia so far did not translate into clinically applicable therapies due to hypothermia-associated side-effects compromising patient outcome. The hypothalamus contains two major thermoregulatory centers in the ventrolateral preoptic area (vlPOA) and dorsomedial hypothalamus (DMH), which are connected via gamma-aminobutyric acid (GABA)-ergic fibers. Using chemogenetic and optogenetic approaches, we explored the role of this GABAergic projection in regulating body temperature responses, cer

Dorsal raphe nucleus MC4R-GABAergic neurons regulate feeding and anxiety.

OBJECTIVE: The melanocortin receptor 4 (MC4R) plays a key role in the CNS regulation of metabolism. In addition to its role within the hypothalamus, other brain areas, including the dorsal raphe nucleus (DRN), express MC4R. However, the identity and role of these neurons in metabolism regulation are not fully understood. We performed studies to address these questions. METHODS: We generated Mc4r-cre;Vgat-FlpO and Mc4r-cre;Vglut2-FlpO mice to determine the contribution of these MC4R neuronal populations in DRN. We then chemogenetically activated or inhibited the GABAergic and glutamatergic pop

Off-targets effects of CNO on somatosensory and anxiety-related behaviors in rats.

Designer receptors exclusively activated by designer drugs (DREADDs) are a powerful chemogenetic tool for controlling targeted cell populations and manipulating brain functions. As a typical chemogenetic actuator in DREADD systems, clozapine-N-oxide (CNO) is widely utilized in neurobiological and pharmacological research. However, the acute off-target effects of CNO on somatosensory (e.g., itch, pain) stimulation-induced and anxiety-related behaviors in rodents have not been systematically investigated. Through classical behavioral paradigms, including evoked itch-scratching, noxious thermal p

Impairment of hippocampal astrocyte-mediated striatal dopamine release and locomotion in Alzheimer's disease.

BACKGROUND: Clinical and translational research has identified deficits in the dopaminergic neurotransmission in the striatum in Alzheimer's disease (AD) and this could be related to the pathophysiology of psychiatric symptoms appearing even at early stages of the pathology. HYPOTHESIS: We hypothesized that AD pathology in the hippocampus may influence dopaminergic neurotransmission even in the absence of AD-related lesion in the mesostriatal circuit. METHODS: We chemogenetically manipulated the activity of hippocampal neurons and astrocytes in wild-type and hemizygous TgF344-AD (Tg) rats, a
