Pharmacology of 0-chlorobenzylidene malononitrile (CS).

1. The effects of o-chlorobenzylidene malononitrile (CS) have been studied on several isolated organs and tissues, anaesthetized animals and cat encéphale isolé preparations.2. On the isolated guinea-pig ileum an initial dose of CS produced a small, non-maintained contraction. Subsequent doses had reduced effects. There was no effect on peristalsis when the substance was given intraluminally.3. No significant effects of CS were detected on the rat phrenic nerve-diaphragm preparation, the isolated perfused rabbit heart or on the contractor response of the indirectly stimulated cat tibialis musc

Functional magnetic resonance imaging of tonic pain and vasopressor effects in rats.

In functional magnetic resonance imaging (fMRI) studies, an elevation in blood pressure (BP) in individuals with a poor autoregulatory response may increase cerebral blood flow, potentially enhancing the blood oxygenation level dependent response. To investigate the role of BP changes, the cerebral activation to either tonic pain or the infusion of the vasopressor norepinephrine was correlated with the accompanying BP changes in alpha-chloralose anesthetized rats. Immediately after formalin (2%) injection into the forepaw, fMRI detected an activation that was correlated with the BP increase an

Prolonged enhancement of the micturition reflex in the cat by repetitive stimulation of bladder afferents.

1. Prolonged modulation of the parasympathetic micturition reflex was studied in cats anaesthetized by alpha-chloralose. Reflex discharges were recorded from a thin pelvic nerve filament to the bladder and evoked by stimulation of the remaining ipsilateral bladder pelvic nerves or urethral branches of the pudendal nerve. 2. Stimulation of bladder or urethral afferents at Adelta intensity evoked micturition reflexes with a latency of 90-120 ms. Such reflexes were much enhanced following repetitive conditioning stimulation of the same afferents at 20 Hz for 5 min. 3. The reflex enhancement laste

Depending on the mode of application morphine enhances or depresses somatocardiac sympathetic A- and C-reflexes in anesthetized rats.

The effects of morphine on the reflex discharges in sympathetic efferents recorded from branches of the inferior cardiac nerve (ICN) were studied in rats anesthetized with alpha-chloralose and urethane. Somatocardiac sympathetic A- and C-reflexes were elicited by single shock electrical stimulation of myelinated (A) and unmyelinated (C) afferent fibers of the tibial nerve, respectively. Application of morphine either into the femoral vein or into the subarachnoid space of the cisterna magna enhanced both the A- and C-reflexes in a dose-dependent manner, while application of morphine into the i

The effect of anaesthetics on the uptake and release of gamma-aminobutyrate and D-aspartate in rat brain slices.

1 The effect of various concentrations of thiopentone, pentobarbitone, methohexitone, hydroxydione, alphaxalone/alphadolone, ketamine, alpha-chloralose, and urethane on the transport of radiolabelled gamma-aminobutyric acid (GABA) and D-aspartate was investigated. 2 Uptake of the amino acids was weakly inhibited, if at all, by the anaesthetics and it is unlikely that such effects contribute significantly to their physiological function. 3 The spontaneous efflux of GABA and D-aspartate was not detectably altered by any of the drugs tested. 4 Thiopentone, pentobarbitone, methohexitone and hydrox

Inhibition of spinocervical tract discharges from localized areas of the sensorimotor cortex in the cat.

1. Intracortical microstimualtion (ICMS) was applied within the sensorimotor cortex of cats anaesthetized with chloralose. 2. The effects of the ICMS were examined on the number of impulses in spinocervical tract (SCT) cells (recorded extracellularly in the contralateral lumbosacral spinal cord) evoked by peripheral stimulation. 3. Inhibition of SCT discharges was produced by ICMS in two distinct regions of the sensorimotor cortex. 4. One inhibitory regions was in part of cytoarchitectonic area 4 gamma in the upper bank of the cruciate sulcus. It sometimes extended caudally into area 4 delta,

Effects of diphenylhydantoin on cardiac arrhythmias induced by carotid occlusion in the cat.

The effects of diphenylhydantoin (DPH) on cardiac arrhythmias induced by occlusion of the common carotid arteries were studied in vagotomized cats under light chloralose anesthesia. It was found that the mean effective dose of DPH via the i.v. route was 11.0 mg/kg. When DPH was injected into the vertebral artery, the carotid artery or the 4th cerebral ventricle, the mean effective dose was 0.8, 1.2 and 0.32 mg/kg, respectively, When DPH was injected directly into the posterior hypothalamic regions, the mean effective dose was even smaller, i.e., 0.14 mg/kg. On the other hand, when DPH was inje

The adrenergic mechanism in the nictitating membrane.

The contractions of the nictitating membrane in response to postganglionic nerve stimulation have been studied in experiments in which cats' heads were perfused. When eserine was added to the perfusion fluid there was a rapid increase in the size of the contractions. In the presence of eserine the contractions produced by injecting acetylcholine into the perfusion fluid were greatly increased. When atropine was injected into the perfusion fluid the contractions caused by postganglionic nerve stimulation returned to the size before eserine was added. In experiments with cats anaesthetized with

The efficient method to get better raw brain signal on rat anesthetics experiment.

This paper introduces an efficient methodology for conducting rat anesthesia experiments, aimed at enhancing the quality of raw brain signals obtained. The proposed approach enables the acquisition of animal brain signals during experiments without the confounding influence of muscle noise. Initially, the use of alpha-chloralose (a-c) in conjunction with Isoflurane is introduced to induce anesthesia in rats. Subsequently, Dexdomitor is administered to prevent muscular movements during the collection of brain signals, further refining the signal quality. Experimental outcomes conclusively demon

Effects of fentanyl, diazepam, and the combination of both on arterial baroreflex and sympathetic nerve activity in intact and baro-denervated dogs.

The combination of fentanyl and diazepam significantly decreases systemic vascular resistance and blood pressure. We attempted to elucidate the reason the combination of these drugs can reduce blood pressure. In alpha-chloralose-anesthetized dogs, we investigated the effects of fentanyl and diazepam on mean arterial pressure (MAP) and arterial baroreflex control of renal sympathetic nerve activity (RSNA) in both intact (Study 1) and baroreflex-denervated dogs (Study 2). Study 1 included five dogs that received fentanyl 10 micrograms/kg followed by diazepam 0.4 mg/kg after a 10-min interval. Fi

Sequential relationship between actions of CRF and PGE2 in the brain on splenic sympathetic nerve activity in rats.

An infusion of corticotropin-releasing factor (CRF, 0.1-5.0 micrograms/10 microliters/rat) into the third cerebral ventricle (i.c.v.) for 10 min increased the splenic sympathetic nerve activity in a dose-dependent manner in urethane + alpha-chloralose anesthetized rats. No changes in the arterial blood pressure and body temperature were observed. The CRF (1.0 micrograms/10 microliters)-induced enhancement of the splenic sympathetic nerve activity was completely blocked by an i.c.v. pretreatment with a CRF antagonist, alpha-helical CRF9-41 (alpha-hCRF, 10 micrograms/10 microliters). The increas

Selective control of sympathetic pathways to the kidney, spleen and intestine by the ventrolateral medulla in rats.

1. Electrical activity of multifibre renal, splenic, mesenteric and greater splanchnic nerves and 13th thoracic white rami was recorded in artificially respired, urethane-anaesthetized rats. Discharge of neurones in the rostral ventrolateral medulla was blocked by unilateral microinjections of the inhibitory amino acid glycine and effects on the electrical activity of these sympathetic nerves were compared. 2. Blockade of the rostral ventrolateral medulla caused greater decreases in discharge of renal than splenic nerves and had no consistent effect on mesenteric nerves. This blockade also dec

Patterns of activity in sympathetic postganglionic nerves to skeletal muscle, skin and kidney during stimulation of the medullary raphe area of the rat.

In chloralose/urethane anaesthetised rats multi and single fibre activity was recorded in renal sympathetic nerves and in sympathetic nerves to skeletal muscle and to the skin of the hind limb. The activity was pulse-modulated and inhibited by stimulating baroreceptors with an increase in blood pressure following i.v. phenylephrine. The study investigated the effect of stimulating medial regions of the lower brainstem on activity in these sympathetic nerves. Comparing renal nerve and skin sympathetic nerve activity two main patterns of response were seen, either an inhibition of both or excita

Carotid body control of bronchial circulation in sheep.

The bronchial vascular response to chemoreceptor stimulation was studied in sheep anesthetized with alpha-chloralose, paralyzed with pancuronium, and artificially ventilated with room air. Blood flow through the common bronchial artery was measured with an electromagnetic flow-meter. Pressures were measured in the left atrium and aorta using appropriately placed catheters connected to pressure transducers. The carotid body was vascularly isolated and perfused at aortic pressure with blood of varied PO2 and PCO2. The viability of our preparation was assessed by the occurrence of a bradycardia i
