Efficacy of weekly versus daily cholecalciferol for repleting serum vitamin D (25(OH)D) deficiency: A systematic review and meta-analysis of randomized controlled trials.

BACKGROUND/RATIONALE: Weekly cholecalciferol can replace daily supplementation to reduce pill burden in patients with complex medication regimens and hypovitaminosis D, but evidence supporting this switch is unclear. OBJECTIVE: We aimed to determine whether weekly cholecalciferol was superior to daily cholecalciferol to replete patients with hypovitaminosis D. METHODS: We conducted a systematic review of randomized controlled trials involving participants with baseline hypovitaminosis D (<30 ng/ml) comparing weekly versus daily cholecalciferol dosing and where serum cholecalciferol was measu

Effects of weekly cholecalciferol supplementation of lactating mothers on vitamin D status, and infant growth and gross motor development: a randomized controlled trial in rural Ethiopia.

BACKGROUND: Vitamin D plays crucial roles in bone metabolism, growth, and immune response during critical life stages. There is little evidence on the effects of vitamin D supplementation among lactating mothers and their infants in sub-Saharan Africa. OBJECTIVE: We aimed to evaluate the efficacy of maternal vitamin D supplementation during lactation on maternal and infant vitamin D status, biomarkers of bone turnover, and infant anthropometry and motor development in rural Ethiopia. METHODS: In an individually randomized placebo-controlled trial, 126 mothers <2 wk postpartum were allocated

Effect of 25-OH-cholecalciferol supplementation on acute phase reactants, cytokines, and colostrum quality of Holstein cows during the transition period and plasma metabolites of newborn calves.

This study investigated the effects of 25-hydroxycholecalciferol (HyD) supplementation on (1) plasma acute phase reactants, cytokines, and Ig concentrations in colostrum in multiparous Holstein cows from 3 wk prepartum to 21 DIM and (2) plasma concentrations of fat-soluble vitamins, Ig concentrations, minerals, and metabolites of newborn calves. Forty-two cows were assigned to receive either cholecalciferol (D3) or HyD. From 21 d before the expected calving until parturition, the D3 group received 0.475 mg/d (0.035 mg/kg DM) of D3, and the HyD group received 1.45 mg/d (0.108 mg/kg DM) of HyD.

A pragmatic, randomised, open-label, multi-centre UK registry-facilitated trial evaluating the effect of high-dose cholecalciferol on mortality in patients on dialysis: the SIMPLIFIED trial.

BACKGROUND: Vitamin D deficiency is common in patients with end stage kidney disease (ESKD) and is a strong predictor of death from cardiovascular disease, infections and cancer. Currently only 68% of patients in the UK survive 3 years or more on dialysis. Vitamin D replacement typically utilises activated (1α-hydroxylated) vitamin D compounds allowing for impaired renal activation. However, this approach increases blood calcium concentrations and may paradoxically accelerate vitamin D catabolism. Contemporary treatment guidelines recommend replacement with inactive (unhydroxylated) vitamin D3

A multisite, randomized, controlled field study of a prepartum cholecalciferol injection on milk production, uterine health, blood minerals, and vitamin D metabolites in multiparous dairy cows.

The objective of the present study was to evaluate the effect of a prepartum injection with 10 × 106 IU cholecalciferol on milk production, uterine health, serum total calcium concentration (tCa) and vitamin D metabolites in a multisite setting, on farms feeding a prefresh diet with a positive DCAD. The study was conducted on 11 farms in northwestern Germany. Cows at the end of their first or greater lactation (n = 446) were randomly assigned to a treatment or a control group. Seven days before calving, cows of the treatment group received 10 × 106 IU (10 mL) of cholecalciferol, whereas cows o

Effect of antepartum vitamin D3 (cholecalciferol) and postpartum oral calcium administration on serum total calcium concentration in Holstein cows fed an acidogenic diet in late gestation.

Several strategies are available to control periparturient hypocalcaemia in dairy cows. Three complementary strategies were applied in this study: feeding a low DCAD (acidogenic) ration during late gestation, oral vitamin D3 (cholecalciferol) administration in late gestation, and oral Ca administration immediately after parturition. Multiparous Holstein cows (n = 240) were fed an acidogenic ration in late gestation and randomly assigned to one of three treatment groups. Group A (n = 80) were fed the acidogenic diet without supplementary Ca or cholecalciferol. Group Ca + A (n = 80) received 50

1alpha(OH)D3 One-alpha-hydroxy-cholecalciferol--an active vitamin D analog. Clinical studies on prophylaxis and treatment of secondary hyperparathyroidism in uremic patients on chronic dialysis.

Chronic uremia is characterized by decreased levels of plasma 1,25(OH)2D3 due to decreased renal 1-hydroxylase activity and by decreased renal phosphate excretion. The consequence is an increased synthesis and secretion of parathyroid hormone--secondary hyperparathyroidism--due to the low levels of plasma calcium, low levels of plasma 1,25(OH)2D3 and high levels of phosphate. The association between renal bone disease and chronic renal failure is well described. Epidemiological studies have indicated that an association also exists between secondary hyperparathyroidism and increased mortality
