Oxochromium(V) species formed with 2,3-dehydro-2-deoxy-N-acetylneuraminic or N-acetylneuraminic (sialic) acids: an in vitro model system of oxochromium(V) species potentially stabilized in the respiratory tract upon inhalation of carcinogenic chromium(VI) compounds.

The human respiratory tract is rich in sialoglycoconjugates. These polyfunctional, oxygen-rich molecules stabilize potentially genotoxic oxoCr(V) species, which has relevance to the metabolic fate of carcinogenic Cr(VI) compounds when inhaled as dusts, mists, and fumes during occupational exposure. A series of oxoCr(V) species formed from solutions of Cr(VI) and the reductant, glutathione (GSH), in the presence of an excess of small molecule ligands that model sialoglycoconjugates, either 2,3-dehydro-2-deoxy-N-acetylneuraminic acid (I) or N-acetylneuraminic acid (II), have been characterized a

Chromium.

Chromium occurs primarily in the trivalent state (III), which is the most stable form, or in the hexavalent state (VI), which is a strong oxidizing agent. Elemental chromium (0) does not occur naturally on earth. Trivalent chromium (III) is an essential trace metal necessary for the formation of glucose tolerance factor and for the metabolism of insulin. Commercial applications of chromium compounds include tanning (III), corrosion inhibition, plating, glassware-cleaning solutions, wood preservatives (VI), manufacture of safety matches, metal finishing (VI), and the production of pigments (III

Differential toxicity and clearance kinetics of chromium(III) or (VI) in mice.

The acute and subacute toxicities of several chromium(III) and chromium(VI) compounds were determined in NZC and (CxO) mice injected i.p. The distal median lethal doses (more than 10 days after treatment) averaged (17.9 +/- 1.8) X 10(-6) g chromium/g body weight regardless of the oxidation state of the chromium compound injected (chromium(III) sulphate may be an exception), but acute toxicity (3 days) was much greater with chromium(VI) compounds. Acid digests of entire male mice that were administered i.p. one-sixth of the distal LD50, either once or repeatedly at weekly intervals, were analys

Role of paramagnetic chromium in chromium(VI)-induced damage in cultured mammalian cells.

Chromium(VI) compounds are known to be potent toxic and carcinogenic agents. Because chromium(VI) is easily taken up by cells and is subsequently reduced to chromium(III), the formation of paramagnetic chromium such as chromium(V) and chromium(III) is believed to play a role in the adverse biological effects of chromium(VI) compounds. The present report, uses electron spin resonance (ESR) spectroscopy; the importance of the role of paramagnetic chromium in chromium(VI)-induced damage in intact cultured cells is discussed, based upon our studies with antioxidants including vitamin E (alpha-toco

The carcinogenicity of chromium.

The carcinogenicity of chromium compounds is reviewed with specific attention to the gaps in knowledge for risk estimation and research needs. The most important problems at present are whether trivalent chromium compounds cause cancer, and whether there is a difference in cancer causing effects between the soluble and the slightly soluble hexavalent compounds in the practical exposure situation. Dose estimates for risk estimation based on epidemiological investigations are also lacking. Present evidence indicates that the trivalent chromium compounds do not cause cancer although high concentr

Impact of Environmental and Lifestyle Use of Chromium on Male Fertility: Focus on Antioxidant Activity and Oxidative Stress.

Male reproductive tissues are strongly susceptible to several environmental and lifestyle stressors. In general, male reproductive health is highly sensitive to oxidative stress, which results in reversible and/or irreversible changes in testosterone-producing cells, spermatogenesis, and sperm quality. Chromium compounds are widely used in the +3 and +6 valence states, as food supplements, and in the industrial field, respectively. Chromium (III) compounds, i.e., Cr(III)-tris-picolinate, [Cr(pic)3], known as chromium picolinate, are used as nutritional supplements for the control of diabetes,

Cancer Risks of Hexavalent Chromium in the Respiratory Tract.

Hexavalent chromium (Cr(VI)) compounds are recognized as carcinogens in the respiratory tract, giving rise to cancers of the lung, nose and nasal sinuses, especially in certain occupational environments. Inhalation exposure of Cr(VI)-containing particles, dusts and fumes commonly occurs in chromium-related occupational environments, such as chromium production, plating, welding of chromium-containing metals and alloys, electroplating, chromium-containing pigments and paints. Epidemiological surveys of chromium compounds have shown strong associations between exposure to Cr(VI) and mortality du

[Chromosome aberrations in a lymphocyte culture from persons in contact with chromium].

A detailed analysis was carried out of chromosome aberrations induced by chromium compounds in a lymphocyte culture of persons which were in contact with the compounds. It is shown that the frequency of cells with chromosome aberrations depends on the age and duration of contact with chromium and does not depend on the sex. It was marked that hexavalent chromium compounds have a higher cytogenetic activity. Mechanisms of mutagenic action of heavy metal ions are discussed (taking chromium as an example).

Status of antioxidant defense system in chromium-induced Swiss mice tissues.

Based on epidemiological studies, chromium(VI) compounds are considered as more toxic and carcinogenic than chromium(III) compounds. The deleterious effects of chromium(VI) compounds are diversified affecting almost all the organ systems in a wide variety of animals. The present study, describes the cytotoxic effects of chromium trioxide, a well-known chromium(VI) compound in three tissues (liver, kidney, lungs) of male Swiss mice during post-treatment phase (5th-8th week after treatment). Lipid peroxidation, an index of oxidative stress, was determined as thiobarbituric acid-reactive substanc

An oxygen dependence in chromium mutagenesis.

Evidence is provided that mutagenicity in Salmonella by a chromium(VI) salt and a chromium(III) compound has a differential dependence on the presence of oxygen. The mutagenic chromium(III) compound, cis-dichlorobis(2,2'-bipyridyl)chromium(III), reverted Salmonella strains, TA102 and TA2638, only under aerobic conditions. Potassium dichromate (chromium VI) required the presence of oxygen to revert the Salmonella strain TA102 but induced a moderate reversion frequency in TA2638 under anaerobic conditions. The data also support a role for oxygen radicals in chromium-mediated mutagenesis and sugg

Health surveillance and biological effect monitoring for chromium-exposed workers.

Although workers may be exposed to chromium metal, Cr(III) compounds, and Cr(VI) compounds at the workplace, only Cr(VI) compounds are of primary concern in terms of possible health hazards. A special health surveillance program must focus on known health impairments and target organs. Medical surveillance in combination with biological monitoring can help to protect the workers' health. Biological monitoring for chromium exposure in urine, blood, and erythrocytes provides different types of information. Whereas chromium measurement in urine and whole blood or plasma is indicative of recent to

Chromium: a review of environmental and occupational toxicology.

The following topics are covered in this brief review on the environmental and occupational toxicology of chromium: occurrence, production and uses of chromium and chromium compounds; experimental toxicology; chromium toxicity for man; hygienic and ecologic aspects of chromium contamination of the environment. The review provides a conclusive evidence which suggests that chromium, especially its hexavalent form, is both toxic and carcinogenic, but its trivalent form is physiologically essential in the metabolism of insulin. It is also emphasized that among the major sources of environmental ch

Acute and chronic systemic chromium toxicity.

Although chromium and compounds containing it have been recognized as having potential severe adverse effects on health for more than 160 years, understanding of the systemic toxicology and true hazard of these compounds is still not complete. A review of the current state of knowledge is attempted in this paper, with appropriate attention given to the complications of multiple valence states and solubility. Selected chromium compounds, particularly hexavalent ones, are carcinogens, corrosives, delayed contact sensitizers, and have the kidney as their primary target organ. But chromium is also

[The change of valence of chromium by acid treatment].

There are many kinds of chromium compounds in nature, and most of them exist as Cr6+- and Cr3+-compounds. Generally, these two kinds of compounds are stable in normal circumstances. When the chromium in biomaterial or in atmospheric substances is analyzed, the samples are usually ashed with strong acids at high temperatures to destroy the organic components, and chromium compounds are changed to water soluble salts in this ashing process. And then chromium is determined in various methods. The relation between the used acids and the behavior of chromium were investigated, and the following fac

Acute toxicity by water containing hexavalent or trivalent chromium in native Brazilian fish, Piaractus mesopotamicus: anatomopathological alterations and mortality.

This study evaluated the toxicity of hexavalent and trivalent compounds of chromium to the pacu, Piaractus mesopotamicus, in acute exposures of 96 h through mortality and histopathological responses. Hexavalent potassium dichromate was more toxic than trivalent compounds of chromium chloride, chromium oxide and chromium carbochelate. Sufficient mortalities occurred only with potassium dichromate to yield an LC50 value at 124.2 mg L(-1). Hexavalent chromium caused reversible and irreversible lesions, which may affect organ functionality. Histopathological evaluation showed that trivalent chromi

Methods for determining soluble and insoluble Cr III and Cr VI compounds in welding fumes.

An analytical procedure for simultaneous determination of soluble and insoluble Cr III and Cr VI compounds in welding fumes has been proposed. In the welding fume samples collected on a membrane filter, total chromium was determined with atomic absorption spectrophotometry (AAS). Glass filters with collected samples were divided into two parts. In one part of the sample, soluble and insoluble chromium was determined by means of AAS. The separation of soluble chromium III and VI was carried out on diphenylcarbazide resin. In the second part of the sample total chromium VI was determined by mean

Particulate hexavalent chromium is cytotoxic and genotoxic to the North Atlantic right whale (Eubalaena glacialis) lung and skin fibroblasts.

Hexavalent chromium compounds are present in the atmosphere and oceans and are established mutagens and carcinogens in human and terrestrial mammals. However, the adverse effects of these toxicants in marine mammals are uncertain. Previously, we reported that North Atlantic right whales, one of the most endangered great whales, have tissue chromium levels that are high, levels that may pose a risk to the whale's health. Furthermore, the study suggested that inhalation may be an important exposure route. Exposure to chromium through inhalation is mainly because of particulate compounds. However
