INTRODUCTION: Circulating tumor DNA (ctDNA) sequencing complements tissue-based next-generation sequencing (NGS), offering noninvasive and serial testing. We explore the mutational landscape of renal cell carcinoma (RCC) using matched tissue and ctDNA data to assess complementarity and clinical significance of molecular alterations.
METHODS: From the Tempus multimodal database, we retrospectively analyzed de-identified data from patients with RCC with concurrent tissue (Tempus xT) and ctDNA testing (Tempus xF). Patients with xT and xF matched samples (collected ±90 days of one another) were i

Circulating tumor DNA (ctDNA) is a minimally invasive biomarker that enables molecular profiling and prognostic stratification in cancer. This meta-analysis evaluated the prognostic significance of ctDNA in metastatic prostate cancer (mPC). PubMed, Web of Science and Scopus were searched from inception to September 15, 2025 for studies assessing ctDNA and survival in prostate cancer. Hazard ratios (HRs) for progression-free survival (PFS) and overall survival (OS) were extracted; when unavailable, HRs were reconstructed from published survival curves. Meta-analyses were performed according to

Circulating tumor DNA (ctDNA) is an emerging biomarker in advanced bladder cancer, but its role in non-muscle-invasive bladder cancer (NMIBC) remains undefined. We conducted a retrospective study of 23 patients with NMIBC who underwent serial ctDNA monitoring using the commercially available Signatera assay at a single institution. ctDNA was detected in 35% of patients, with two illustrative cases highlighting its clinical utility. In one case, baseline ctDNA positivity prompted earlier reimaging that revealed locally advanced disease, leading to initiation of systemic therapy followed by plan

PURPOSE: Current clinical challenges in Hodgkin lymphoma (HL) include difficult-to-treat relapsed/refractory disease and considerable long-term toxicities of treatment. Since clinical risk factors lack discriminatory power, intensity of therapy is mainly based on tumor burden. Exploring HL genetics and tumor microenvironment (TME) might provide valuable insights for improved risk stratification.
MATERIALS AND METHODS: In this study, we applied circulating tumor DNA sequencing to 243 patients obtained from pivotal German Hodgkin Study Group trials to identify subtypes of HL. Independent valida

Circulating tumor DNA (ctDNA) is an auspicious tumor biomarker released into the bloodstream by tumor cells, offering abundant information concerning cancer genes. It plays a crucial role in the early diagnosis of cancer. However, due to extremely low levels in body fluids, achieving a simple, sensitive, and highly specific detection of ctDNA remains challenging. Here, we constructed a purification-free fluorescence biosensor based on quadratic amplification of ctDNA by combining nicking enzyme mediated amplification (NEMA) and catalytic hairpin assembly (CHA) reactions. After double isotherma

BACKGROUND/AIM: Synchronous colorectal cancer, which occurs in approximately 4.8-8.4% of all colorectal cancers, has a genetic profile with a higher rate of v-raf murine sarcoma viral oncogene homolog B1 (BRAF) mutation and microsatellite instability-high than solitary colorectal cancer. However, little information is available on heterogeneity among tumor lesions because of difficulty in performing genetic tests in all lesions in clinical practice.
CASE REPORT: A 44-year-old man presented with multiple recurrent lung metastases 42 months after the endoscopic resection of early stage synchron

Diffuse large B cell lymphoma (DLBCL) is the most common non-Hodgkin lymphoma, its diagnosis and prognosis evaluation mainly depends on tissue biopsy and imaging examination. As a part of liquid biopsy, circulating tumor DNA (ctDNA) is a novel noninvasive and real-time tumor-specific biomarker, which can reliably reflect the comprehensive tumor genetic profiles, and it plays an important role in assisting early diagnosis, monitoring the curative effect, prognosis evaluation and prediction of recurrence of DLBCL. This review summarized recent research progress of ctDNA in DLBCL.

Chimeric antigen receptor T (CAR T) cell immunotherapy has shown remarkable efficacy in non-Hodgkin's lymphoma (NHL) patients. Minimal residual disease (MRD) monitoring in NHL is essential after CAR T cell therapy, which can be achieved by monitoring circulating tumor DNA (ctDNA). The mutation of TP53 in NHL has been suggested to be associated with a poor prognosis. However, whether TP53-mutated ctDNA can be used as a biomarker remains undetermined. In this study, a total of 40 patients with mutated TP53 who received CAR T cell treatment were analyzed, and specific probes targeting 29 differen

PURPOSE: Recent advances in molecular diagnostic technologies allow for the evaluation of solid tumor malignancies through noninvasive blood sampling, including circulating tumor DNA profiling (ctDNA). Pancreatic ductal adenocarcinoma (PDAC) has a poor prognosis, often because of late presentation of disease. Diagnosis is often made using endoscopic ultrasound or endoscopic retrograde cholangiopancreatography, which often does not yield enough tissue for next-generation sequencing. With this study, we sought to characterize the ctDNA genomic alteration landscape in patients with advanced PDAC

Identifying locoregional gastric cancer patients who are at high risk for relapse after resection could facilitate early intervention. By detecting molecular residual disease (MRD), circulating tumor DNA (ctDNA) has been shown to predict post-operative relapse in several cancers. Here, we aim to evaluate MRD detection by ctDNA and its association with clinical outcome in resected gastric cancer. This prospective cohort study enrolled 46 patients with stage I-III gastric cancer that underwent resection with curative intent. Sixty resected tumor samples and 296 plasma samples were obtained for t

Although targeted deep sequencing of cell-free DNA (cfDNA) was recently used to investigate tumor somatic mutations in particular subtypes of non-Hodgkin lymphomas (NHLs), the immense genetic heterogeneity across subtypes poses a hurdle to design a universal gene panel applicable for diverse subtypes of NHLs. We designed a panel targeting 66 genes associated with NHLs and performed targeted deep sequencing to analyze plasma cfDNA from patients with various subtypes of NHLs. Genetic profiling in plasma cfDNA using the method resulted in 88.0% sensitivity and >99% specificity in detecting mutati

The NSABP B-51 trial left unresolved whether postmastectomy radiotherapy benefits ypN0 patients after neoadjuvant chemotherapy. Meanwhile, the PREDICT-DNA study showed that ctDNA clearance after neoadjuvant therapy identifies patients with favorable outcomes irrespective of pathologic response. We propose integrating these datasets: ctDNA status may serve as a tiebreaker for radiotherapy decisions, particularly in patients with residual disease, offering a molecularly guided framework for de-escalation.

INTRODUCTION: Neoadjuvant chemotherapy has potential benefit in patients with localized colon cancer with proficient mismatch repair (pMMR). However, patient selection is a major challenge. We explored whether pre-operative circulating tumor DNA (pre-op ctDNA) would be helpful in selecting high-risk patients for neoadjuvant chemotherapy.
METHODS: Pre-op ctDNA levels were determined using blood samples from patients undergoing potentially curative surgery for localized pMMR colon cancer. Associations between pre-op ctDNA and pathological stage, positive (R1) resection margins and disease recur

BACKGROUND: Recurrence of colorectal cancer (CRC) after curative-intent treatment is largely driven by minimal residual disease (MRD). Circulating tumor DNA (ctDNA) offers a noninvasive approach to detect MRD and tailor adjuvant therapy and surveillance.
METHODS: This scoping review synthesized prospective cohorts, interventional/randomized trials, and real-world registries on ctDNA-based MRD testing in resected CRC, comparing tumor-informed vs plasma-only (mutation- and/or methylation-based) assays.
RESULTS: Postoperative ctDNA positivity strongly predicts recurrence and often precedes radi

Circulating tumor DNA (ctDNA) has emerged as a clinically actionable biomarker for the management of colorectal cancer (CRC). Improvements in analytical accuracy and sequencing depth have expanded the role of ctDNA from early cancer detection to include molecular profiling of advanced disease, postoperative risk stratification, and real-time evaluation of therapeutic response and resistance. In screening and early detection settings, ctDNA-based assays integrating mutation analysis, methylation profiling, and fragmentomic features have demonstrated high specificity for CRC and multi-cancer ear
