Spatial and longitudinal monitoring of cartilage and intervertebral disc extracellular matrix formation using proteoglycan building blocks and click chemistry: specificity, quantification, and detection upon subcutaneous implantation.

BACKGROUND/OBJECTIVE: The development of regenerative therapies for osteoarthritis and intervertebral disc degeneration requires non-destructive methods to spatiotemporally monitor extracellular matrix (ECM) synthesis. This study aimed to establish a method for the use of bioorthogonal click chemistry for longitudinal and spatially resolved tracking of newly synthesized proteoglycans in cartilaginous tissues, including tools to assess specificity, enable quantification and for future application for noninvasive real-time tracking of ECM production by imaging. METHODS: Bovine articular cartila

A coumarin-tethered 1,2,3-triazole-based fluorescent "turn-ON" probe via metal-free click chemistry for selective sensing of serum albumin: quantitative detection of HSA in urine.

A series of novel highly substituted coumarin-based 1,2,3-triazoles (CTZs) were synthesised by a metal-free organo-click reaction. Under environmentally benign, atom-economical conditions, the desired products were obtained in moderate to excellent yields. All derivatives exhibited good binding affinity toward bovine serum albumin (BSA), with binding constants on the order of Kb ∼ 104 L mol-1. Among them, compound 3f, a nitro analogue of CTZ, demonstrated a selective fluorescence "turn-on" response upon binding to both BSA and human serum albumin (HSA). Fourier transform infrared (FT-IR) and c

Preparation of chiral stationary phase based on a [3+3] chiral polyimine macrocycle by thiol-ene click chemistry for enantioseparation in normal-phase and reversed-phase high performance liquid chromatography.

Polyimine macrocycles are a new class of organic macrocycles with cyclic structures, well-defined molecular cavities, and multiple cooperative binding sites, which have recently aroused considerable research interest in molecular recognition and separation. Herein, we report the bonding of a [3+3] chiral polyimine macrocycle (H3L, C78H78N6O3) on thiol-functionalized silica gel using thiol-ene click chemistry to prepare a chiral stationary phase (CSP) for high performance liquid chromatography (HPLC). The fabricated column exhibited excellent chiral separation capability under both normal-phase

Ultrasound-assisted Cu(I)-catalyzed azide-alkyne click cycloaddition as polymer-analogous transformation in chitosan chemistry. High antibacterial and transfection activity of novel triazol betaine chitosan derivatives and their nanoparticles.

In this work, we involved ultrasound-assisted click CuAAC in chitosan chemistry. Ultrasound-mediated CuAAC between propargylic ester of betaine and azido chitosan derivative proceeds fast in water under aerobic conditions and gives rise novel water-soluble triazole betaine chitosan derivatives. Using ionic gelation technique, we prepared and characterized nanoparticles from the synthesized chitosan derivatives. We studied antibacterial and transfection activity of the novel chitosan derivatives and their nanoparticles. The nanoparticles with size ca. 100 nm and ζ-potential ca. +65 mV proved to

Engineering thiol-ene click chemistry for the preparation of a chiral stationary phase based on a [4+6]-type homochiral porous organic cage for enantiomeric separation in normal-phase and reversed-phase high performance liquid chromatography.

In this study, a new chiral stationary phase (CSP) was fabricated by covalent bonding of a [4+6]-type homochiral porous organic cage (POC) CC19-R onto thiolated silica via a thiol-ene click reaction. The CC19-R was synthesized via Schiff-base reaction between 2-hydroxybenzene-1,3,5-tricarbaldehyde and (1R, 2R)-(-)-1,2-diaminocyclohexane. The enantioseparation capability of the resulting CC19-R-based CSP was systematically evaluated upon separating various chiral compounds or chiral pharmaceuticals in normal phase HPLC (NP-HPLC) and reversed phase HPLC (RP-HPLC), including alcohols, organic aci

Click and Bio-Orthogonal Reactions with Mesoionic Compounds.

Click and bio-orthogonal reactions are dominated by cycloaddition reactions in general and 1,3-dipolar cycloadditions in particular. Among the dipoles routinely used for click chemistry, azides, nitrones, isonitriles, and nitrile oxides are the most popular. This review is focused on the emerging click chemistry that uses mesoionic compounds as dipole partners. Mesoionics are a very old family of molecules, but their use as reactants for click and bio-orthogonal chemistry is quite recent. The facility to derivatize these dipoles and to tune their reactivity toward cycloaddition reactions makes

Click3D: Click reaction across deep tissues for whole-organ 3D fluorescence imaging.

Click chemistry offers various applications through efficient bioorthogonal reactions. In bioimaging, pretargeting strategies have often been used, using click reactions between molecular probes with a click handle and reporter molecules that make them observable. Recent efforts have integrated tissue-clearing techniques with fluorescent labeling through click chemistry, allowing high-resolution three-dimensional fluorescence imaging. Nevertheless, these techniques have faced a challenge in limited staining depth, confining their use to imaging tissue sections or partial organs. In this study,

Carbonic anhydrase inhibitors developed through 'click tailing'.

In recent years there has been renewed activity in the literature concerning the 1,3-dipolar cycloaddition reaction (1,3-DCR) of organic azides (R-N₃) with alkynes (R'-C≡CH) to form 1,2,3-triazoles, i.e. the Huisgen synthesis. The use of catalytic Cu(I) leads to a dramatic rate enhancement (up to 10(7)-fold) and exclusive synthesis of the 1,4-disubstituted 1,2,3-triazole product. The reaction, now referred to as the copper-catalyzed azide-alkyne cycloaddition (CuAAC), meets the stringent criteria of a click-reaction in that it is modular, wide in scope, high yielding, has no byproducts, operat

Non-specific cysteine modifications on proteins induced by trans-cyclooctene.

Tetrazine-trans-cyclooctene (Tz-TCO) click chemistry has been widely adopted for various applications due to its faster reaction kinetics and higher biocompatibility compared to conventional copper-catalyzed click chemistry. However, most click chemistry groups do not appear to be fully bioorthogonal, as several of them-including tetrazine-have been found to directly modify proteins. Here, we report the unexpected discovery of non-specific protein modifications by TCO during the development of a tetrazine-based proximity labeling method. TCO probes readily conjugate to cysteine residues on pro

Recent Advancements in CuAAC Click Approaches for the Synthesis of 1,2,3-Triazole Hybrid Compounds as Anticancer Agents.

The field of chemical biology has been revolutionized by click chemistry due to its remarkable efficiency, selectivity, and gentle reaction conditions. Among the various click reactions, the copper(I)-catalyzed azide-alkyne cycloaddition (CuAAC) has emerged as the most widely used technique. The formation of 1,2,3-triazoles through copper(I)-catalyzed reactions between azides and terminal acetylenes serves as an effective method for creating important medicinal frameworks. This process is highly reliable, completely specific, and utilizes biocompatible reagents. Click chemistry has emerged as

A Modular and Convergent "Stick and Click" Conjugation Platform Enables Fast Antibody Conjugate Library Synthesis.

The preparation of antibody drug conjugates (ADC) most often relies on a linear sequence to elaborate the small molecule component, followed by a final bioconjugation step to attach it to its immunoglobulin partner. This linear and iterative approach is incompatible with expedient parallel synthesis and process automation. Here, we describe the design and implementation of a general modular platform for the assembly of ADCs that enables facile variation in the nature of the payload, the linker composition, and the type of bioconjugation technique used. A library of antibody conjugates bringing

Multispecific DNA coatings for self-assembly.

DNA-coated particles are promising building blocks for functional and finite-sized assemblies because they can be programmed to exhibit orthogonal interactions through the sequence-specific hybridization of DNA strands. To fully exploit this programmability, it is important to develop particles with coatings that incorporate multiple distinct DNA sequences in tunable ratios and to understand how the coating composition influences self-assembly. Here, we compare two strategies for grafting multiple DNA sequences in tunable and well-defined ratios onto micron-sized colloidal particles. We found

Ultrasound-assisted green synthesis of 1,4-disubstituted 1,2,3-triazoles using natural polymer supports.

The advent of click chemistry has significantly accelerated the synthesis of 1,2,3-triazoles, key structures found in numerous biologically active and pharmaceutical compounds. Click reactions can be notably enhanced by employing ultrasonic irradiation and supported catalysts. Herein, we present an eco-friendly catalyst composed of natural biopolymers, chitosan and shilajit, designed to facilitate a click chemistry protocol under ultrasonic conditions at room temperature. The catalyst was characterized via FTIR, XRD, TGA, EDX, and SEM analyses, confirming the successful immobilization of coppe

Size-matched alkyne-conjugated cyanine fluorophores to identify differences in protein glycosylation.

Currently, there are few methods to detect differences in posttranslational modifications (PTMs) in a specific manner from complex mixtures. Thus, we developed an approach that combines the sensitivity and specificity of click chemistry with the resolution capabilities of 2D-DIGE. In "Click-DIGE", posttranslationally modified proteins are metabolically labeled with azido-substrate analogs, then size- and charge-matched alkyne-Cy3 or alkyne-Cy5 dyes are covalently attached to the azide of the PTM by click chemistry. The fluorescently-tagged protein samples are then multiplexed for 2DE analysis.

Drug Target Identification Using an iTRAQ-Based Quantitative Chemical Proteomics Approach-Based on a Target Profiling Study of Andrographolide.

Identifying the cellular binding targets of drugs and other bioactive small molecules is a crucial step for understanding their molecular mechanisms of action as well as potential off-target effects. The field of chemical proteomics is an emerging discipline in chemical biology using synthetic chemistry and high-throughput detection techniques to study small molecule-protein interactions. In this chapter, we describe a quantitative chemical proteomics protocol combining bioorthogonal click chemistry and quantitation by isobaric tags for relative and absolute quantification (iTRAQ) to identify

Enzyme-free and isothermal detection of microRNA based on click-chemical ligation-assisted hybridization coupled with hybridization chain reaction signal amplification.

An enzyme-free and isothermal microRNA (miRNA) detection method has been developed based on click-chemical ligation-assisted hybridization coupled with hybridization chain reaction (HCR) on magnetic beads (MBs). The click-chemical ligation between an azide-modified probe DNA and a dibenzocyclooctyne-modified probe DNA occurred through the hybridization of target miRNA (miR-141). HCR on MBs was performed by the addition of DNA hairpin monomers (H1 and H2). After magnetic separation and denaturation/rehybridization of HCR products ([H1/H2] n ), the resulting HCR products were analyzed by the flu

Recent development in charged stationary phases for ion exchange chromatography and mixed-mode chromatography.

Ion exchange chromatography (IEC) and mixed-mode chromatography are highly effective for the separation of polar and ionic compounds, including inorganic ions, proteins, peptides, pharmaceuticals, nucleotides, and organic acids, owing to their unique ion exchange or mixed-mode retention mechanisms. As the core component of chromatographic system, the stationary phase directly determines separation selectivity and column efficiency. Therefore, the rational design and development of novel stationary phases remain a focal in modern separation science. In response to the increasing complexity of s

Synthesis of Click-Ready Aminooxy-Terminated Poly(ε-caprolactone) Oligomers for Oxime Ligation.

Oxime ligation, a chemoselective coupling between carbonyl compounds and aminooxy groups, enables conjugation under mild and aqueous conditions. Here, we integrate this reaction with degradable polymers through a one-pot synthesis of aminooxy-functionalized poly(ε-caprolactone) (PCL) oligomers. The bifunctional initiator 6-(tert-butyloxycarbonylaminooxy)-1-hexanol was employed for simultaneous ε-caprolactone ring-opening polymerization and in situ Boc-deprotection catalyzed by methanesulfonic acid (MSA). Optimized conditions afforded >95% conversion for both reactions, while maintaining well-d
