Hepatoprotective Effect of Silymarin Herb in Prevention of Liver Dysfunction Using Pig as Animal Model.

Silymarin, a flavonolignan-rich extract of Silybum marianum, is widely recognized for its hepatoprotective potential. While rodent studies predominate, pigs (Sus scrofa) offer a more translationally relevant model due to their hepatic architecture, bile acid composition, and transporter expression, which closely resemble those of humans. This narrative review synthesises current evidence on the chemistry, pharmacokinetics, biodistribution, and hepatoprotective activity of silymarin in porcine models. Available studies demonstrate that when adequate intrahepatic exposure is achieved, particular

Evidence for a Cu(II)-Catalysed CuAAC Reaction: A Combined Experimental and Computational Study.

The Cu-catalysed azide-alkyne cycloaddition (CuAAC) reaction is one of the most broadly applied catalytic processes, with applications throughout synthetic chemistry, drug discovery, chemical biology, and materials science. This field of research is built upon the mechanistic premise that the metal-centred events proceed via Cu(I) catalysis. Here, we report details of a CuAAC reaction that can also be catalysed by Cu(II) through a self-assembled dinuclear Cu(II) complex that incorporates the triazole product. Computational analysis supports the experimental findings and the autocatalytic role

Iridoids from Cornus officinalis Siebold & Zucc and their alpha-glucosidase inhibitory activity.

Four new iridoid glycosides, cornoidosides A-D (1-4), along with 35 analogues (5-39), were isolated from the dried fruits of Cornus officinalis Siebold & Zucc. The chemical structures of them were elaborated by extensive spectroscopic means, including 1D, 2D-NMR, HR-ESI-MS and computational chemistry. Compound 4 was the first dimeric iridoid consisting of sweroside and α-morroniside moiety from C. officinalis. The effect of 1-39 against α-glucosidase activity was tested in vitro. Compounds 7-β-O-ethylmorroniside (10), 7-ketologanin (14) and 8-epiloganin (15) (IC50: 37.8, 40.6 and 19.3 μM, resp

Personal care products disrupt the human oxidation field.

People generate hydroxyl radicals (OH) in the presence of ozone via the ozonolysis of skin-emitted alkenes. In this study, we found that the application of personal care products (PCPs) including fragrances and body lotions suppresses the human oxidation field. Body lotion hampers the generation of 6-methyl-5-hepten-2-one, a key OH precursor, while many volatile ingredients of PCPs enhance OH loss in the gas phase. Although fragrances contain terpenes capable of generating OH through ozonolysis, the much larger amount of ethanol solvent acts as a large OH sink. We combined a multiphase chemica

Using Three-Dimensional Information to Predict and Interpret the Facial Selectivities of Nucleophilic Additions to Cyclic Ketones.

In this study, we devised a new method to predict facial selectivity by quantifying steric and orbital factors for the nucleophile approaching both π-plane faces. Using this method, we quantified the total electron density and frontier orbital distributions of 163 cyclic ketones with various structures and quantitatively explained the surface selectivity of 323 reactions with eight nucleophiles (BH3, LiAlH4, NaBH4, LiAl(OMe)3H, MeLi, MeMgI, PhLi, and PnMgI). Importance analysis showed a large orbital effect for BH3, LiAlH4, and NaBH4 and the dominance of the steric effect for LiAl(OMe)3H, MeLi

Palladium-Catalyzed Aryldifluoromethylation of Aryl Halides with Aryldifluoromethyl Trimethylsilanes.

Diaryl difluoromethanes are valuable targets for medicinal chemistry because they are bioisosteres of diaryl ethers and can function as replacements for diaryl methane, ketone, and sulfone groups. However, methods to prepare diaryl difluoromethanes are scarce, especially methods starting from abundant aryl halides. We report the Pd-catalyzed aryldifluoromethylation of aryl halides with aryldifluoromethyl trimethylsilanes (TMSCF2 Ar). The reaction occurs when the catalyst contains a simple, but unusual, dialkylaryl phosphine ligand that promotes transmetallation of the silane. Computational stu

Formation of Benzene and Naphthalene through Cyclopentadienyl-Mediated Radical-Radical Reactions.

Resonantly stabilized free radicals (RSFRs) have been contemplated as fundamental molecular building blocks and reactive intermediates in molecular mass growth processes leading to polycyclic aromatic hydrocarbons (PAHs) and carbonaceous nanoparticles on Earth and in deep space. By combining molecular beams and computational fluid dynamics simulations, we provide compelling evidence on the formation of benzene via the cyclopentadienyl-methyl reaction and of naphthalene through the cyclopentadienyl self-reaction, respectively. These systems offer benchmarks for the conversion of a five-membered

Ketone Incorporation Extends the Emission Properties of the Xanthene Scaffold Beyond 1000 nm.

Imaging in the shortwave-infrared region (SWIR, λ = 1000-2500 nm) has the potential to enable deep tissue imaging with high resolution. Critical to the development of these methods is the identification of low molecular weight, biologically compatible fluorescent probes that emit beyond 1000 nm. Exchanging the bridging oxygen atom on the xanthene scaffold (C10' position) with electron withdrawing groups has been shown to lead to significant redshifts in absorbance and emission. Guided by quantum chemistry computational modeling studies, we investigated the installation of a ketone bridge at th

Caution: Chemical Instability of Natural Biomolecules During Routine Analysis.

Natural products (NPs) constitute a significant source of active biomolecules widely used in medicine, pharmacology and cosmetics. However, NPs structural characterization has the drawback of their chemical instability during the extraction steps and their likely transformation during the analytical protocol. In particular, tamariscol and conocephalenol are two compounds largely used in the cosmetic industry for their odorant properties. Thus, in the present study, we focused on the evolution of these two metabolites (extracted from Frullania tamarisci and Conocephalum conicum, respectively),

Single-Step Synthesis of Dithieno[3,2-b:2',3'-e]Pyridines and Their Acid-Dependent Optical and Photosensitising Properties.

Dithieno[3,2-b:2',3'-e]pyridine (DTP) is a heterocycle consisting of a central pyridine ring fused with two thiophene rings and can be regarded as a thiophene-based analogue of the classic N-heterocycle acridine. Unlike acridine-based dyes, the fundamental chemistry of DTP has received relatively little attention. Here, we report the straightforward synthesis of a library of DTP molecules and an investigation of their electronic properties using UV/vis absorbance and emission spectroscopy supported by computational insights, with particular focus on the influence of protonation and methylation

A ligand-controlled regiodivergent three-component Heck-Tsuji reaction utilizing umpolung hydrazones enabled by palladium catalysis.

Developing sustainable, selective strategies for the multicomponent functionalization of allenes remains an ongoing challenge in synthetic chemistry. We report a ligand-controlled, regiodivergent palladium-catalyzed three-component Heck-Tsuji reaction of allenes, aryl iodides, and umpolung hydrazones. Leveraging the umpolung reactivity of hydrazones as sustainable carbanion surrogates enables the direct arylalkylation of allenes, forming two C-C bonds in a single step. The ligand environment governs reaction outcomes, switching selectivity between branched and linear adducts using simple, comm

Comparative Binding Affinities of Platinum-Based Drugs toward Purine Alkaloids and Nucleobases.

Platinum-based anticancer drugs are among the most commonly used agents in cancer therapy. In particular, carboplatin, cisplatin, and oxaliplatin have been used in both monotherapies and combination therapies for many years. The main targets of these compounds are nucleobases in DNA. By creating cross-links, they inhibit cell development and lead to apoptosis. Recent studies indicate that platinum-(II) drugs can interact with other compounds with structures similar to nucleobases. For this reason, the current study analyzed the interactions of carboplatin, cisplatin, and oxaliplatin with nucle

On the mechanism of the dyotropic expansion of hydrindanes into decalins.

A combined computational/experimental approach has revealed key mechanistic aspects in a recently reported dyotropic expansion of hydrindanes into decalins. While computer simulations had already anticipated the need for acid catalysis for making this reaction feasible under the mild conditions used in the laboratory, this work places the dyotropic step not into the reaction flask but at a later step, during the work up instead. With this information in hand the reaction has been optimized by exploring the performance of different activating agents and shown to be versatile, particularly in st

Machine learning with physicochemical relationships: solubility prediction in organic solvents and water.

Solubility prediction remains a critical challenge in drug development, synthetic route and chemical process design, extraction and crystallisation. Here we report a successful approach to solubility prediction in organic solvents and water using a combination of machine learning (ANN, SVM, RF, ExtraTrees, Bagging and GP) and computational chemistry. Rational interpretation of dissolution process into a numerical problem led to a small set of selected descriptors and subsequent predictions which are independent of the applied machine learning method. These models gave significantly more accura

Redox-Active Reagents for Photocatalytic Generation of the OCF3 Radical and (Hetero)Aryl C-H Trifluoromethoxylation.

The trifluoromethoxy (OCF3 ) radical is of great importance in organic chemistry. Yet, the catalytic and selective generation of this radical at room temperature and pressure remains a longstanding challenge. Herein, the design and development of a redox-active cationic reagent (1) that enables the formation of the OCF3 radical in a controllable, selective, and catalytic fashion under visible-light photocatalytic conditions is reported. More importantly, the reagent allows catalytic, intermolecular C-H trifluoromethoxylation of a broad array of (hetero)arenes and biorelevant compounds. Experim

Infrared Signature of Structural Isomers of Gas-Phase M+(N2O)n (M = Cu, Ag, Au) Ion-Molecule Complexes.

Gas-phase metal ion-ligand complexes offer model environments to study molecular interactions that are key to many catalytic processes. Here, we present a combined experimental and computational study of M+(N2O)n [M = Cu, Ag, Au; n = 2-7] complexes. The spectra provide clear evidence for both nitrogen- and oxygen-bound ligands giving rise to a wide range of structural isomers for each complex studied. The evolution of the complex structures observed as well as spectral trends for each metal center are interpreted in terms of a molecular orbital binding picture and resulting calculated ligand b

Four-membered heterocycles with a carbon-heteroatom exocyclic double bond at the 3-position: puckering potential and thermodynamic properties.

Despite the specific importance of four-membered heterocycles with a carbon-heteroatom double bond at the 3-position in organic and medicinal chemistry, little attention has been given up to now to theoretical computational studies on these molecules. However, the overall geometry, and degree of ring puckering especially, could significantly influence the reactivity and biological properties of these four-membered ring compounds. In this paper, focus is made on the influence of different substituents on the equilibrium geometry, ring puckering potential, and thermodynamic quantities. It was fo
