Shared genetic architecture between metabolic traits and Alzheimer's disease: a large-scale genome-wide cross-trait analysis.

A growing number of studies clearly demonstrate a substantial link between metabolic dysfunction and the risk of Alzheimer's disease (AD), especially glucose-related dysfunction; one hypothesis for this comorbidity is the presence of a common genetic etiology. We conducted a large-scale cross-trait GWAS to investigate the genetic overlap between AD and ten metabolic traits. Among all the metabolic traits, fasting glucose, fasting insulin and HDL were found to be genetically associated with AD. Local genetic covariance analysis found that 19q13 region had strong local genetic correlation betwee

Genetic polymorphisms in TLR1, TLR2, TLR4, and TLR10 of Helicobacter pylori-associated gastritis: a prospective cross-sectional study in Thailand.

The toll-like receptors (TLRs) mediate the recognition of Helicobacter pylori and initiate the innate immune response to infection. We hypothesized those genetic polymorphisms in the TLR1, TLR2, TLR4, and TLR10 influence bacterial infection, affecting susceptibility H. pylori to disease outcomes. Genomic DNA was extracted and genotypes of TLR1 (rs4833095), TLR2 (rs3804099 and rs3804100), TLR4 (rs10759932), and TLR10 (rs10004195) polymorphism were detected by the TagMan single-nucleotide epolymorphisms genotyping assay using the real-time PCR hybridization probe method. The TLR1 (rs4833095), C

Chimerization at the AQP2-AQP3 locus is the genetic basis of melarsoprol-pentamidine cross-resistance in clinical Trypanosoma brucei gambiense isolates.

Aquaglyceroporin-2 is a known determinant of melarsoprol-pentamidine cross-resistance in Trypanosoma brucei brucei laboratory strains. Recently, chimerization at the AQP2-AQP3 tandem locus was described from melarsoprol-pentamidine cross-resistant Trypanosoma brucei gambiense isolates from sleeping sickness patients in the Democratic Republic of the Congo. Here, we demonstrate that reintroduction of wild-type AQP2 into one of these isolates fully restores drug susceptibility while expression of the chimeric AQP2/3 gene in aqp2-aqp3 null T. b. brucei does not. This proves that AQP2-AQP3 chimeri

Cross sectional study of prevalence, genetic diversity and zoonotic potential of Cryptosporidium parvum cycling in New Zealand dairy farms.

BACKGROUND: The estimation of the prevalence and zoonotic potential of Cryptosporidium parvum cycling in bovine populations requires the use of genotyping, as several morphologically similar non-parvum genetic variants of unproven clinical and public health impact are found in cattle. However, robust C. parvum prevalence estimates in cattle are lacking and comparative data of bovine and human isolates collected from the same regions are scarce. Thus, the relative contribution of the C. parvum oocysts released by farmed animals to animal and human cryptosporidiosis burden is, in general, poorly

OPRM1 genetic polymorphisms are associated with the plasma nicotine metabolite cotinine concentration in methadone maintenance patients: a cross sectional study.

Majority of the heroin-dependent patients smoke cigarettes. Although it has been reported that the OPRM1 genetic polymorphism is associated with the brain mu-opioid receptor binding potential in cigarette smokers, there is no direct evidence showing the impact of plasma cotinine, a nicotine metabolite, on treatment responses to methadone maintenance treatment (MMT). In this study, we aimed to test the hypothesis that the genetic polymorphisms in the OPRM1 are associated with the methadone treatment responses and the severity of cigarette smoking directly measured by the plasma concentration of

Cross-trait genomic and sequential analyses of multiple omics datasets identified shared genetic components for the gut-eye axis.

BACKGROUND: The gut–eye axis (GEA) has been proposed as a framework for understanding comorbidity between gastrointestinal and ocular diseases. This study aimed to investigate their shared genetic architecture, pleiotropy, and putative biological pathways potentially influenced by environmental exposures and gut microbiota. METHODS: This study integrated large-scale genome-wide association study summary data on five gastrointestinal and eight ocular diseases to assess genetic correlations and genetic overlap. Pleiotropic variants were identified, followed by functional and tissue-specific anal

Genetic and cross-species single-cell analyses prioritize CD83 in antigen-presenting cells as a candidate therapeutic target in asthma.

BACKGROUND: Asthma remains a heterogeneous inflammatory airway disease, and current therapies, including biologics, benefit only selected patient subsets. We aimed to identify genetically supported therapeutic targets with clear mechanistic and translational relevance in asthma. METHODS: We integrated cis-eQTL and cis-pQTL instruments for 585 druggable genes with asthma genome-wide association data from FinnGen, EBI, and UK Biobank, comprising more than 1.1 million participants in total, using Mendelian randomization, colocalization, and SMR/HEIDI analyses. We then combined cell-type-specific

Integrative cross-tissue transcriptome-wide association and metabolomic analysis reveals novel genetic risk loci for aortic aneurysm.

BACKGROUND: Aortic aneurysm (AA) is a life-threatening cardiovascular condition with a strong genetic component, however, its molecular mechanisms remain poorly understood. Although genome-wide association studies (GWAS) have identified numerous risk loci, most prior studies have investigated genetic and metabolic factors separately, leaving the causal pathways from genetic variants to disease largely unexplored. METHODS: We established an integrative framework combining cross-tissue transcriptome-wide association studies (TWAS) with metabolomic mediation analysis. First, we integrated GWAS d

Antioxidant Enzymes Genetic Variants Associated with Urticaria/Angioedema Induced by Cross-Reactive Hypersensitivity to Nonsteroidal Anti-Inflammatory Drugs.

Background/Objectives: Nonsteroidal anti-inflammatory drugs (NSAIDs) are among the most consumed drugs worldwide and the main cause of drug hypersensitivity reactions (HSRs). The most common NSAID-HSR class is cross-hypersensitivity (CR), with patients reacting to NSAIDs from different chemical groups without specific immunological recognition, with NSAID-induced acute urticaria/angioedema (NIUA) being the most frequent clinical phenotype. Although CR-HSRs are triggered by arachidonic acid (AA) alterations following cyclooxygenase (COX)-1 inhibition and cysteinyl-leukotrienes synthesis by 5-ly

Experimental evolution reveals genetic and transcriptomic mechanisms underlying benzalkonium chloride tolerance and fluoroquinolone cross-resistance in Listeria monocytogenes.

UNLABELLED: Preventing Listeria monocytogenes persistence in food processing environments is challenged by tolerance to quaternary ammonium compound sanitizers such as benzalkonium chloride (BAC). To define the genetic and molecular routes to BAC tolerance and antibiotic cross-resistance, we performed an evolve-and-resequence experiment on three L. monocytogenes strains (three replicate lineages each) passaged under sublethal BAC for ~400-500 generations. All nine BAC-exposed lineages increased BAC minimum inhibitory concentrations (MICs), and several showed elevated tolerance to a commercial

Genetic Regulation of Monocyte MicroRNAs and Their Implication in Musculoskeletal Diseases: A Cross-Ancestry Expression Quantitative Trait Loci and Imputation Study.

This study investigated the genetic regulation of microRNA (miRNA) expression in monocytes and its potential role in musculoskeletal diseases. We mapped expression quantitative trait loci (eQTLs) for miRNAs using data from 281 Caucasian (CAU) and 170 African American (AA) individuals, constructed ancestry-specific models to impute miRNA expression from genotype data, and applied these models to test associations with osteoporosis and sarcopenia. Analysis identified 468 and 2653 independent eQTLs for 61 miRNAs in CAU and 25 in AA, respectively, the majority of which were ancestry-specific. Asso

Genome-wide cross-trait analysis reveals shared genetic architecture between inflammatory bowel disease and ankylosing spondylitis.

Patients with inflammatory bowel disease (IBD) and ankylosing spondylitis (AS) are clinically closely related. They suffer from comorbidities, possibly attributed to a shared genetic structure. By utilizing the genome-wide association study (GWAS) data of IBD and AS as the study object, the overall and local genetic associations were first assessed by employing the linkage disequilibrium score regression (LDSC), GNOVA, and high-definition likelihood (HDL). Subsequently, SUPERGNOVA was employed to identify region-specific genetic signals across chromosomes; finally, conditional and conjunctiona

Clinical and Genetic Factors Associated with Multivessel Coronary Artery Disease: A Cross-Sectional Study of CYP2C19 Polymorphisms and Metabolic Comorbidities.

BACKGROUND: Multi-vessel coronary artery disease (MVCD), defined as stenosis affecting ≥2 major coronary arteries, confers poor clinical outcomes in patients with coronary artery disease (CAD). Cytochrome P450 2C19 (CYP2C19) CYP2C19 participates in lipid metabolism and inflammatory regulation, which may be associated with individual variations in the severity of coronary artery lesions. This study aimed to explore the clinical and genetic factors associated with MVCD focusing on CYP2C19 polymorphisms and chronic metabolic diseases. METHODS: This single‑center cross‑sectional study enrolled 41

A genetically-distinct Pigeon Paramyxovirus-I isolate fails to cross-react with neutralizing antibodies raised by LaSota vaccine in chicken.

Pigeon paramyxovirus-1 (PPMV-1) is a major cause of mortality in domestic and free-living pigeons worldwide and has the potential to infect chickens, posing risks to the poultry sector. This study investigated a severe outbreak of PPMV-1 in a racing pigeon loft in Srinagar, India, despite routine vaccination with the LaSota strain. Molecular screening of 30 clinical samples identified NDV in 18 birds by real-time qPCR targeting the matrix gene. Histopathology revealed characteristic viscerotropic lesions, including epithelial necrosis and congestion in the proventriculus and duodenum, sinusoid

Cross-Border Circulation and Molecular Surveillance of HIV-1 in the Azov and Donbas Regions: A Study of Genetic Diversity and Drug Resistance.

As critical geopolitical and migratory hubs in Eastern Europe, the Azov and Donbas regions represent an epidemiological melting pot for HIV-1 trafficking, exacerbating the global trend of rising dolutegravir (DTG) resistance through the cross-border dissemination of drug-resistant strains. This study presents a comprehensive molecular epidemiological analysis of HIV-1 in these regions in 2025 (N = 1666), focusing on drug resistance and cross-border transmission networks using phylogenetic and molecular network approaches. The study cohort was predominantly female (53.33%) and had heterosexual
