INTERFERENCE BETWEEN VIRUSES IN TISSUE CULTURE.

The influence of one virus on the growth of another in tissue culture was investigated. The 17DD High strain of yellow fever virus was found capable of completely suppressing the growth of both the Asibi strain of the same virus and of the heterologous West Nile virus, even when these were added to the cultures in large amounts. The 17DD High strain of yellow fever virus and the West Nile virus produced either partial or complete suppression of growth of the Venezuelan equine encephalomyelitis virus, depending upon the quantity of the latter inoculated into the cultures. Owing to lack of metho

Culture-Agnostic and Culture-Specific Analyses of Semantic Fluency in Korean and English Speakers With Alzheimer's Disease.

PURPOSE: This study investigated cross-linguistic and cultural influences on semantic fluency in monolingual Korean- and English-speaking individuals with Alzheimer's disease (AD) by contrasting culture-agnostic and culture-specific analytic frameworks. We focused on how culturally grounded schemas, such as the East Asian zodiac, shape clustering and switching patterns and on their associations with global cognition. METHOD: We analyzed semantic fluency in 36 individuals with AD and 30 language-matched older adults using culture-agnostic and culture-specific frameworks. We examined total and

Probing the Ovarian Microenvironment: Methods for Isolation, 2D Culture, and Organoid Culture of Mouse Ovarian Somatic Cells.

The ovary consists of heterogeneous populations of somatic cells, both within the follicle and the surrounding stroma, which are critical to support ovarian function and for the generation of high-quality gametes. We report methods for isolating somatic cells from mouse ovaries, including endothelial, epithelial, steroidogenic, stromal, and immune cells. When these primary ovarian somatic cells are plated and cultured in a traditional 2D culture system, the cellular heterogeneity, organization, as well as cell-cell and cell-matrix interactions typically found in the ovary are lost. Thus, we al

Microfluidic Fibroblast Cell Culture Chip for Embryo Co-Culture: Analysis of Preimplantation Embryo Viability and Development Potential.

Preimplantation embryo development requires a tightly regulated microenvironment that is not fully reproduced by conventional static culture. We developed a polydimethylsiloxane-based microfluidic embryo co-culture platform integrating compartmentalized architecture with dynamic perfusion to simulate physiological conditions. The study included two stages. First, NIH/3T3 mouse fibroblasts were evaluated as helper cells under three culture conditions after transition to embryo culture medium. Helper-cell viability in the dynamic chip was 84.72%, compared with 71.91% after manual medium replacem

Rethinking in vitro embryo culture: new developments in culture platforms and potential to improve assisted reproductive technologies.

The preponderance of research toward improving embryo development in vitro has focused on manipulation of the chemical soluble environment, including altering basic salt composition, energy substrate concentration, amino acid makeup, and the effect of various growth factors or addition or subtraction of other supplements. In contrast, relatively little work has been done examining the physical requirements of preimplantation embryos and the role culture platforms or devices can play in influencing embryo development within the laboratory. The goal of this review is not to reevaluate the solubl

[Human breast cancer cells under serum-free culture. Its hormone-dependency and application to the primary culture].

Assessment of hormone-dependency of breast cancer cells in human to estradiol (E2) and progesterone (Pg) has been made by making serum-free culture of the cells, and primary culture of breast cancer cells is made by this method. The results are as follows: 1. E2 promotes proliferation of estrogen receptor (ER)-positive cells in low concentrations but rather inhibits it in high concentrations. This inhibitory effect is also observed in ER-negative cells. If the mode of this inhibitory effect is compared with the cellular dynamics of Tamoxifen (TAM) which binds predominantly to ER, the cellular

Clinical laboratory comparison of the 10-ml isolator blood culture system with BACTEC radiometric blood culture media.

The efficiency of the 10-ml Isolator (E. I. du Pont de Nemours & Co., Inc.) for recovery of pathogens from blood was compared with that of BACTEC 6B and 7C media (Johnston Laboratories) by using 4,195 cultures from 1,662 patients. During the first phase of the study, BACTEC bottles were inoculated with 3 ml of blood; in the second phase, bottles were inoculated with 5 ml. The objectives were to compare results with similar blood volumes used for the detection of anaerobes as well as similar overall volumes and to determine the relative sensitivity of BACTEC media inoculated with the minimum an

Comparison of culture and culture-free methods for comprehensive identification of mycobacteria: a single-center prospective study.

The genus Mycobacterium, including Mycobacterium tuberculosis and over 200 nontuberculous mycobacteria (NTM), shows wide variability in clinical outcomes and drug susceptibility. Although culture-based identification remains the gold standard, slow mycobacterial growth delays diagnosis and treatment. In this study, we evaluated a novel culture-free method for subspecies-level identification directly from sputum. In this single-center prospective cohort study at Osaka Toneyama Medical Center, we analyzed 125 sputum samples from 115 patients with NTM pulmonary disease and 10 with non-NTM respira

"What am I?" microbiology of culture-positive, biofire® blood culture identification 2 panel-negative bloodstream infections.

BACKGROUND: The BioFire® blood culture identification 2 panel (BCID2) detects 33 of the most common bloodstream infection (BSI) pathogens, yet it can fail to detect the causal agent of sepsis in up to 13 % of cases, mostly due to infections caused by off-panel target microorganisms. A better understanding of the microbiology of culture-positive, BCID2-negative BSI is needed. METHODS: Single-center, retrospective study of microbiology and outcomes in 275 cases of BCID2-negative BSI between 2022 and 2024. RESULTS: A total of 257 adult patients with 275 cases of culture-positive, BCID2-negative

Shigella PCR-positive Gastroenteritis in Children: Insights From a Comparison Between Culture-positive and Culture-negative Cases.

BACKGROUND: Detection of Shigella in stool by polymerase chain reaction (PCR) is rapid and highly sensitive but presents challenges in distinguishing between disease-associated and asymptomatic Shigella carriage. This study compared children with gastroenteritis who had Shigella identified in their stool by PCR-testing and a positive stool culture to those with Shigella identified by PCR-testing but a negative culture. METHODS: All stool samples of outpatients and inpatients (<18 years) tested by multiplex-PCR at the Soroka University Medical Center laboratory between January 2020 and July 20

Establishment of an experimental model of canine malignant mesothelioma organoid culture using a three-dimensional culture method.

Canine malignant mesothelioma (cMM) is a rare and drug-resistant malignant tumor. Due to few patients and experimental models, there have not been enough studies to demonstrate the pathogenesis of the disease and novel effective treatment for cMM. Since cMM resembles human MM (hMM) in histopathological characteristics, it is also considered a promising research model of hMM. Compared with conventional 2-dimensional (2D) culture methods, 3-dimensional (3D) organoid culture can recapitulate the properties of original tumor tissues. However, cMM organoids have never been developed. In the present

Human multi-organ chip co-culture of bronchial lung culture and liver spheroids for substance exposure studies.

Extrapolation of cell culture-based test results to in vivo effects is limited, as cell cultures fail to emulate organ complexity and multi-tissue crosstalk. Biology-inspired microphysiological systems provide preclinical insights into absorption, distribution, metabolism, excretion, and toxicity of substances in vitro by using human three-dimensional organotypic cultures. We co-cultured a human lung equivalent from the commercially available bronchial MucilAir culture and human liver spheroids from HepaRG cells to assess the potential toxicity of inhaled substances under conditions that permi
