Isolation and functional characterization of chicken intestinal intra-epithelial lymphocytes showing natural killer cell activity against tumour target cells.

Intestinal intra-epithelial lymphocytes (IEL) of SC or FP chickens were isolated and examined for their natural killer (NK)-cell activity against chicken tumour cell lines, LSCC-RP9 (RP9), LSCC-RP12 (RP12), MDCC-MSB-1 (MSB-1) and MDCC-CU36 (CU36). In general, IEL of satisfactory yield and of good viability were obtained with EDTA treatment of the gut tissues, followed by rapid passages of the resultant cells through nylon-wool columns and centrifugation on two-step Percoll density gradients (45% and 80%). In 4-hr and 16-hr 51Cr-release assays, the NK-cell activity of chicken IEL depended not o

Natural killer (NK) cell activity and reversal reaction in leprosy.

Two studies were conducted to assess natural killer (NK) cell activity in leprosy patients and healthy Ethiopian controls. The first study tested 26 untreated leprosy patients across the spectrum of the disease. It was found that lepromatous leprosy and all untreated, nonreactional patients had lower NK activity than healthy controls. However, patients presenting with reversal reaction (RR) had NK activity within the normal range. Heterogeneity was particularly marked in the NK activity of borderline patients. In the second study, NK cell activity was assessed in treated borderline tuberculoid

Quinidine-induced agranulocytosis.

A case of agranulocytosis in an 86-year-old man after 8 weeks treatment with quinidine sulfate is described. Acute phase serum from the patient demonstrated antineutrophil activity by the microgranulocytotoxicity assay. Review of the literature reveals that more than one mechanism could cause this idiosyncratic immune-mediated agranulocytosis.

Cell mediated lympholysis; a modified technique using 111indium-oxine-labelled targets.

The isotope 51Cr generally used in the cell mediated lympholysis (CML) assay suffers from the disadvantage of low specific activity, poor incorporation and high spontaneous release, limiting the CML assay to 4-6 h. We have labelled PHA derived human lymphoblasts with the isotope 111indium (using 111indium-oxine) and evaluated these cells as targets in CML. The level of 111In-oxine incorporation decreased rapidly in the presence of serum; in the absence of serum approximately 85% of the available isotope in the supernatant was incorporated into the blasts. Under the labelling conditions used, s

Effector mechanisms in the spontaneous autoimmune thyroiditis of obese strain (OS) chickens: analysis of cytotoxic cells.

Different forms of cellular cytotoxicity that may constitute potential effector mechanisms in the spontaneous autoimmune thyroiditis (SAT) arising in Obese strain (OS) chickens have been investigated. A microcytotoxicity assay (MCA) employing 51Cr-labelled-, thyroglobulin (Tg) coated-chicken red blood cells (Tg-CRBC) was used to detect cells mediating Tg-specific direct cellular cytotoxicity (DCC). Tg-CRBC presensitized with anti-Tg autoantibody (Tg-AAB) obtained from high titer OS sera served as targets for antibody-dependent cell-mediated cytotoxicity (ADCC). Tannic acid-only treated CRBC (T

A stochastic model of cytotoxic T cell responses.

We have constructed a stochastic stage-structured model of the cytotoxic T lymphocyte (CTL) response to antigen and the maintenance of immunological memory. The model follows the dynamics of a viral infection and the stimulation, proliferation, and differentiation of naïve CD8(+) T cells into effector CTL, which can eliminate virally infected cells. The model is capable of following the dynamics of multiple T cell clones, each with a T cell receptor represented by a digit string. MHC-viral peptide complexes are also represented by strings and a string match rule is used to compute the affinity

Transplant monitoring: patterns of the immune response.

Immune responses, specific to the stored donor tissue, were measured in 26 recipients over 4,000 days. CML positive crossmatches were associated with accelerated rejections (three cases). LDA positive crossmatches resulted in primary non-functioning kidneys (three cases). Post-transplantation. CML (16 hour inculation) was associated with rejection (p equals 0.001), highly predictive of rejection (p equals 0.001), and when positive after rejection treatment, was associated with a failure to respond (p equals 0.005 - 0.01). 51Cr-CDA was also associated with rejection (p equals 0.002). Killing of targets by CD8 T cells in the mouse spleen follows the law of mass action.

It has been difficult to correlate the quality of CD8 T cell responses with protection against viral infections. To investigate the relationship between efficacy and magnitude of T cell responses, we quantify the rate at which individual CD8 effector and memory T cells kill target cells in the mouse spleen. Using mathematical modeling, we analyze recent data on the loss of target cells pulsed with three different peptides from the mouse lymphocytic choriomeningitis virus (LCMV) in mouse spleens with varying numbers of epitope-specific CD8 T cells. We find that the killing of targets follows th

Interleukins 2 and 12 produce recovery of cytotoxic function in tributyltin-exposed human natural killer cells.

Cytotoxic function of human natural killer (NK) cells is modulated by a variety of cytokines. Interleukins (IL) 2 and 12 are both potent stimulators of NK cell cytotoxic function. Tributyltin (TBT) is used in a variety of consumer products and industrial applications. TBT is found in dairy products, meat, and fish. We and others have shown that there are measurable levels of TBT in human blood. Butyltins appear to increase the risk of cancer and viral infections in exposed individuals. We have demonstrated that the ability of NK cells to kill tumor cells is greatly diminished after a l-h expos

Comparison of the sensitivity of in vivo and in vitro assays for detection of antiviral cytotoxic T cell activity.

Sensitivities of in vitro and in vivo assays for the detection of lymphocytic choriomeningitis virus (LCMV)-specific CTL were compared. Measurement of primary cytotoxicity was the least sensitive of all the tested assays. However, when the same 51Cr release assays were performed after in vitro restimulation, this in vitro method was found to be more sensitive than all of the five in vivo assays tested. Assessment of CTL-mediated protection against LCMV replication in spleens was most sensitive among the in vivo tests, followed by CTL-mediated resistance to intracerebral or intraperitoneal chal

Virus-specific memory T cells are Pgp-1+ and can be selectively activated with phorbol ester and calcium ionophore.

Memory lymphocytic choriomeningitis virus (LCMV)-immune cytotoxic T-lymphocyte precursors (CTLp) can be stimulated to proliferate and to mediate specific cytotoxic activity following incubation with phorbol myristate acetate (PMA), calcium ionophore (CaI), and interleukin 2 (IL-2). This protocol can be used to selectively induced virus-specific CTL activity under both bulk culture and limiting dilution conditions, in the absence of added antigen. There is no concurrent stimulation of alloreactive CTLp. Proliferation of the effector Lyt-2+ population in medium containing PMA and CaI requires L3

Cell-mediated cytotoxicity in recovery from poxvirus infections.

The availability of mutant and gene targeted knockout mice with defects in components of cellular cytotoxicity mediated by either the Fas or the exocytosis pathway permitted an analysis of their role in recovery from poxvirus infections. Ectromelia (EV), a natural mouse pathogen causing mousepox, the closely related orthopoxviruses cow pox (CPV) and vaccinia virus (VV), each encode serpins that inhibit Fas mediated apoptosis and lysis of target cells. Nevertheless, distinct differences were seen when the three viruses were inoculated into perforin-deficient mice: highly resistant C57Bl/6 mice

Three distinct mechanisms underlying human γδ T cell-mediated cytotoxicity against malignant pleural mesothelioma.

INTRODUCTION: Malignant pleural mesothelioma (MPM) is a rare and highly aggressive thoracic tumor with poor prognosis and limited therapeutic options. Although immune checkpoint inhibitors exhibit a promising effect in some patients with unresectable MPM in clinical trials, the majority of MPM patients show only modest response rates to the currently available treatments. It is thus imperative to develop novel and innovative therapeutic modalities for MPM, including immune effector cell-based therapies. METHODS: γδ T cells were expanded using tetrakis-pivaloyloxymethyl 2-(thiazole-2-ylamino)

Vaccine adjuvants to engage the cross-presentation pathway.

Adjuvants are indispensable components of vaccines for stimulating optimal immune responses to non-replicating, inactivated and subunit antigens. Eliciting balanced humoral and T cell-mediated immunity is paramount to defend against diseases caused by complex intracellular pathogens, such as tuberculosis, malaria, and AIDS. However, currently used vaccines elicit strong antibody responses, but poorly stimulate CD8 cytotoxic T lymphocyte (CTL) responses. To elicit potent CTL memory, vaccines need to engage the cross-presentation pathway, and this requirement has been a crucial bottleneck in the

Inadequate epidermal homing leads to tissue damage in human cutaneous leishmaniasis.

Leishmaniasis is a model disease for the study of immunoregulatory mechanisms associated with host resistance and susceptibility. In this article, Felix Tapia and colleagues propose that defects in the signaling properties of the epidermis can result in the generation of either a chronic granulomatous response, which is unable to eliminate the parasite, or a proinflammatory mucocutaneous response and tissue damage.

Proinflammatory and antiinflammatory cytokines in multiple sclerosis and central nervous system acquired immunodeficiency syndrome.

While certain cytokines such as interleukin-1 (IL-1), tumor necrosis factor-alpha (TNF-alpha), IL-6, and transforming growth factor-beta (TGF-beta) may be involved in pro- and anti-inflammatory events in the central nervous system (CNS) of patients with multiple sclerosis (MS) and CNS acquired immunodeficiency syndrome (AIDS), there is not uniform consensus as to whether they are elevated or even detectable in all compartments of the body such as serum, cerebrospinal fluid (CSF), and tissue. Furthermore, if they are elevated in these diseases, there are no data as to whether they regulate the

Study of granulocyte function of peripheral blood in recurrent erysipelas.

The Boyden chamber technique, the NBT reduction and Candida killing test were carried out on 51 patients with recurrent erysipelas to investigate the non-specific immune response. A statistically significant difference was not observed between the patients and the controls. It is suggested, that the infected individual is not able to increase his resistance via the more effective functioning of the non-specific line, which in effect promotes the possibly multiple occurrence of the disease.

The role of calmodulin in human natural killer cell activity.

Calcium is known to play an essential role in the lytic mechanism of natural killer cells (NK), which form a subset of large granular lymphocytes. Many of the intracellular effects of calcium are mediated through the calcium-binding protein calmodulin. In this study we have demonstrated that the specific calmodulin inhibitors (naphthalene-sulphonamides) inhibit NK activity in humans at IC50's of 6.9 microM for W7 and 5.2 microM for W13. Comparison of the potency of these compounds with their less active counterparts suggests that NK activity is calmodulin-dependent.

[Lymphocyte macrophage activation factor. Comparison of the mode of production in man and mice].

A macrophage activating factor (MAF) has been previously described in mixed lymphocyte cultures (MLC) between the donor and the recipient of a mouse skin allograft. This factor can render macrophages cytotoxic against mouse mastocytoma targert cells. The present work demonstrates the production of a similar factor in the human, using macrophages from uremic patients treated with peritoneal dialysis. There is, however, an apparent difference between mouse and human MAF. In the mouse the production of MAF in primary MLC is inconsistent, late and weak, while it is regularly found in large amounts
