Detection of equine parapoxvirus in the stable environment during pastern dermatitis outbreak in Finland.

BACKGROUND: Equine parapoxvirus (EqPPV) has emerged as a cause of recurring pastern dermatitis outbreaks in horses in Finland. First detected in 2013, EqPPV has since been associated with potentially hundreds of clinical cases and suspected zoonotic transmission. Based on serological data and field reports, EqPPV causes pastern dermatitis outbreaks in Finland every few years. This study aimed to investigate the environmental presence and plausible transmission dynamics of the virus to prepare for future outbreaks and give targeted guidelines to stable staff and veterinarians. RESULTS: After a

Leptospira: Molecular Detection of Pathogenic Species in Natural Sources.

This protocol describes a method for the rapid detection of leptospiral DNA in environmental water. In summary, the DNA is extracted from water samples and tested in a TaqMan-based real-time quantitative polymerase chain reaction (qPCR) for the presence of lipl32, a gene that is present only in pathogenic Leptospira spp. The gene target used in this assay is important in that it only detects pathogenic leptospires and not the saprophytic leptospires that may be present in environmental samples. © 2017 by John Wiley & Sons, Inc.

Disruption of DNA methylation-mediated cranial neural crest proliferation and differentiation causes orofacial clefts in mice.

Orofacial clefts of the lip and palate are widely recognized to result from complex gene-environment interactions, but inadequate understanding of environmental risk factors has stymied development of prevention strategies. We interrogated the role of DNA methylation, an environmentally malleable epigenetic mechanism, in orofacial development. Expression of the key DNA methyltransferase enzyme DNMT1 was detected throughout palate morphogenesis in the epithelium and underlying cranial neural crest cell (cNCC) mesenchyme, a highly proliferative multipotent stem cell population that forms orofaci

Selective detection of viable Cryptosporidium oocysts by PCR.

Although the diagnosis of cryptosporidiosis in diarrheic patients is relatively simple, as large amounts of oocysts are usually shed, environmental samples can contain only few oocysts which have a comparatively high epidemiological relevance. Very sensitive detection methods are therefore requested in environmental hygiene. Additionally these methods should allow a statement about the viability of the detected organisms. The combination of a DNA-digest and a viability assay followed by a PCR might reveal a very promising method. A DNA-digest destroys free DNA in the sample. In the following e

How nucleotide excision repair protects against cancer.

Eukaryotic cells can repair many types of DNA damage. Among the known DNA repair processes in humans, one type--nucleotide excision repair (NER)--specifically protects against mutations caused indirectly by environmental carcinogens. Humans with a hereditary defect in NER suffer from xeroderma pigmentosum and have a marked predisposition to skin cancer caused by sunlight exposure. How does NER protect against skin cancer and possibly other types of environmentally induced cancer in humans?

Male Germ Cell Telomeres and Chemical Pollutants.

In recent decades, male infertility has been correlated with the shortening of sperm telomeres. Telomeres regulate the reproductive lifespan by mediating the synapsis and homologous recombination of chromosomes during gametogenesis. They are composed of thousands of hexanucleotide DNA repeats (TTAGGG) that are coupled to specialized shelterin complex proteins and non-coding RNAs. Telomerase activity in male germ cells ensures that the telomere length is maintained at maximum levels during spermatogenesis, despite telomere shortening due to DNA replication or other genotoxic factors such as env

[Epigenetics in Parkinson's Disease].

The genetic information encoded in the DNA sequence provides a blueprint of the entire organism. The epigenetic modifications, in particular DNA methylation and histone modifications, determine how and when this information is made available and define the specific gene transcription pattern of a given cell. Epigenetic modifications determine the functional differences of genetically identical cells in multicellular organisms and are important factors in various processes from embryonic development to learning and memory consolidation. DNA methylation patterns are altered by environmental cond

Molecular mechanism of the unusual biphasic effects of wood preservative copper-ortho-phenanthroline and tetrachlorocatechol on cellular DNA damage.

Tetrachlorocatechol (TCC) is the primary metabolite of wood preservative pentachlorophenol (PCP) in the human body, while copper-ortho-phenanthroline (Cu(OP)₂) is another widely used environmentally friendly wood preservative. TCC and Cu(OP)₂ can coexist in the human body, potentially exerting combined toxicity. This study explores the molecular mechanisms underlying their biphasic effects on intracellular DNA, providing insights into their environmental health risks. Results show that at lower TCC-Cu(OP)₂ ratios (TCC:Cu(OP)₂ ≤ 3), significant intracellular DNA double-strand breaks occur, wher

Development of a New LAMP Assay for the Detection of Ancylostoma caninum DNA (Copro-LAMPAc) in Dog Fecal Samples.

Ancylostoma caninum is a zoonotic nematode which is able to affect animals and humans. Diagnosis in the definitive host and environmental detection are key to prevent its dissemination and achieve control. Herein, a new coprological LAMP method for the detection of A. caninum (Copro-LAMPAc) DNA was developed. DNA extraction was performed using a low-cost method and a fragment of the cox-1 gene was used for primer design. The analytical sensitivity, evaluated with serial dilutions of genomic DNA from A. caninum adult worms, was 100 fg. A specificity of 100% was obtained using genomic DNA from t

Detection, movement and persistence of invertebrate eDNA in groundwater.

Sampling groundwater biodiversity is difficult because of limited access and issues with species identification. Environmental DNA (eDNA) provides a viable alternative to traditional sampling approaches, however limited knowledge of the abundance and fate of DNA in groundwater hinders the interpretation of data from these environments. Groundwater environments are dark and have lower oxygen concentrations and microbial activity than surface waters. Consequently, assumptions about DNA fate in surface ecosystems may not apply to groundwaters. Here, we test the longevity and transport of eDNA in

Epigenetics and environmental chemicals.

PURPOSE OF REVIEW: Epigenetics investigates heritable changes in gene expression occurring without changes in DNA sequence. Several epigenetic mechanisms, including DNA methylation, histone modifications, and microRNA expression, can change genome function under exogenous influence. Here, we review current evidence indicating that epigenetic alterations mediate toxicity from environmental chemicals. RECENT FINDINGS: In-vitro, animal, and human investigations have identified several classes of environmental chemicals that modify epigenetic marks, including metals (cadmium, arsenic, nickel, chr

How can environmental fecal sampling support Echinococcus multilocularis surveillance in wild carnivores?

Foxes (Vulpes vulpes) and raccoon dogs (Nyctereutes procyonoides) are natural definitive hosts of Echinococcus multilocularis. The aim of this study was to compare E. multilocularis findings from the necropsy analysis of hunted animals with those from environmental fecal samples. Between November 2023 and March 2025, 84 carcasses (58 foxes, 26 raccoon dogs) were provided by volunteer hunters located in the area of two municipalities of the island Rügen in North-Eastern Germany. Analysis using the sedimentation counting technique and real-time PCR resulted in a combined positive detection of E.

Mycobacterium avium Subspecies Hominissuis-Induced Fatal Vasculitis in Zebra Finches (Taeniopygia guttata), USA.

Mycobacterium avium subsp. hominissuis (MAH) is a zoonotic pathogen with a broad host range and diverse clinical manifestations. We report here the first documented case of MAH-induced fatal vasculitis in zebra finch (Taeniopygia guttata). Histopathological examination revealed acid-fast bacilli within macrophages and endothelial cells, primarily affecting the heart and aorta. Mycobacterial DNA was detected in cloacal swabs from affected finches and environmental samples from their housing facility. PCR targeting the rpoB gene and insertion elements IS1245 and IS901, followed by sequencing, co

Generation of a specific marker to discriminate Gacillus anthracis from other bacteria of the Bacillus cereus group.

Bacillus anthracis is a soil pathogen capable of causing anthrax that is closely related to several environmental species, including B. cereus, B. mycoides, and B. thuringiensis. DNA homology studies showed that B. anthracis, B. cereus, B. mycoides, and B. thuringiensis are closely related, with a high sequence homology. To establish a method to specifically detect B. anthracis in situations such as environmental contamination, we initially performed RAPD-PCR with a 10-mer random primer and confirmed the presence of specific PCR bands only in B. anthracis species. One region specific for B. an

Nucleic acid aptamers protect against lead (Pb(II)) toxicity.

Lead (Pb(II)) is a pervasive heavy metal toxin with many well-established negative effects on human health. Lead toxicity arises from cumulative, repeated environmental exposures. Thus, prophylactic strategies to protect against the bioaccumulation of lead could reduce lead-associated human pathologies. Here we show that DNA and RNA aptamers protect C. elegans from toxic phenotypes caused by lead. Reproductive toxicity, as measured by brood size assays, is prevented by co-feeding of animals with DNA or RNA aptamers. Similarly, lead-induced neurotoxicity, measured by behavioral assays, are also

Rapid Identification of a Cooling Tower-Associated Legionnaires' Disease Outbreak Supported by Polymerase Chain Reaction Testing of Environmental Samples, New York City, 2014-2015.

We investigated an outbreak of eight Legionnaires' disease cases among persons living in an urban residential community of 60,000 people. Possible environmental sources included two active cooling towers (air-conditioning units for large buildings) <1 km from patient residences, a market misting system, a community-wide water system used for heating and cooling, and potable water. To support a timely public health response, we used real-time polymerase chain reaction (PCR) to identify Legionella DNA in environmental samples within hours of specimen collection. We detected L. pneumophila serogr

DNA methylation in Huntington's disease: Implications for transgenerational effects.

Huntington's disease (HD) is a devastating, neurodegenerative disorder caused by a CAG repeat mutation in the HTT gene. A growing body of evidence suggests that epigenetic modifications play a key role in HD pathogenesis. Expression of the disease protein, huntingtin, leads to extensive transcriptional dysregulation due to disruption of histone-modifying complexes and altered interactions with chromatin-related factors. Such epigenetic mechanisms also readily respond to environmental factors, which are now thought to influence the risk, onset and progression of neurodegenerative disorders, inc

Efficient recovery and DNA extraction for algae-associated microbial communities.

The extraction of high-quality microbial DNA from environmental samples is critical for many downstream applications, including short- and long-read metagenomic sequencing. However, environmental DNA is prone to low recovery, degradation, and contamination by enzymatic inhibitors, with the extent of these issues largely dependent on the DNA purification method. The embedding of bacterial cells in a mucoid matrix within biofilms further complicates the process, making the study of algal symbionts particularly challenging. This study benchmarked five methods to recover microbial cells from biofi

A comparison between petrous bone and tooth, femur and tibia DNA analysis from degraded skeletal remains.

Skeletal remains are the only biological material that remains after long periods; however, environmental conditions such as temperature, humidity, and pH affect DNA preservation, turning skeletal remains into a challenging sample for DNA laboratories. Sample selection is a key factor, and femur and tooth have been traditionally recommended as the best substrate of genetic material. Recently, petrous bone (cochlear area) has been suggested as a better option due to its DNA yield. This research aims to evaluate the efficiency of petrous bone compared to other cranium samples (tooth) and postcra

Neuroepigenetics of the Environmental Exposome: Molecular and Clinical Implications in Neurodegeneration.

BACKGROUND: Neuroepigenetics studies the molecular mechanisms that regulate gene expression in the nervous system without altering the DNA sequence. The environmental exposome has emerged as a key factor influencing neuronal plasticity and brain homeostasis, contributing to vulnerability to neurodegeneration. REVIEW SUMMARY: This narrative review analyzes the role of neuroepigenetic reprogramming as a central mechanism linking chronic environmental exposures with neurodegenerative diseases. Evidence indicates that alterations in DNA methylation, histone remodeling, and microRNA regulation con
