Abrupt increased serum creatinine in a hyperferritinemia patient treated with deferoxamine after cord blood transplantation: a case report with literature review.

BACKGROUND: Erythrocyte transfusion is an indispensable component of supportive care after hematopoietic stem cell transplantation (HSCT). However, HSCT recipients are susceptible to the development of acute kidney injury (AKI) with multifactorial causes. We report a case of a rapid elevation in serum creatinine associated with deferoxamine after cord blood transplantation (CBT). CASE PRESENTATION: A 36-year-old Japanese male diagnosed with relapsed Philadelphia-positive acute lymphoblastic leukemia received CBT. At day 88 post-CBT, multidrug-resistant Pseudomonas aeruginosa (MDRP) was isolat

Multifunctional chito-oligosaccharide-and deferoxamine-modified GelMA-OHA hydrogel dressing with antibacterial, antioxidant, and pro-angiogenic activities for infected wound healing.

Infected wounds resist healing due to multidrug-resistant bacterial colonization and persistent inflammation, which render conventional antibiotic therapies largely ineffective. Consequently, non-antibiotic strategies that coordinate infection control with tissue regeneration are urgently required. We design a dual-crosslinked hydrogel composed of gelatin methacryloyl (GelMA) and oxidized hyaluronic acid (OHA), further functionalized with chitosan oligosaccharide (COS) and deferoxamine (DFO), yielding the multifunctional GOCD hydrogel. Dynamic Schiff base linkages confer controlled release of

Deferoxamine attenuates early brain injury after subarachnoid hemorrhage by inhibiting ferroptosis and preserving mitochondrial function and oxidative phosphorylation via Nrf2 signaling.

BACKGROUND: Early brain injury (EBI) occurring within the first 72 h is a critical determinant of outcome after subarachnoid hemorrhage (SAH), yet effective treatments remain limited. Ferroptosis and mitochondrial dysfunction are central mechanisms in EBI. This study investigated whether deferoxamine (DFO), a clinically approved iron chelator, alleviates EBI after SAH via ferroptosis inhibition and mitochondrial restoration. METHODS: A rat SAH model was established by endovascular perforation. Neurological scores, blood-brain barrier integrity, and brain edema were assessed. TUNEL staining ev

Combination of Deferoxamine and Deferiprone demonstrates superior efficiency compared to monotherapies in management of cardiac iron overload: Evidence from meta-analysis.

OBJECTIVE: Cardiac iron overload is a significant but treatable complication linked to various diseases. Iron chelators, such as Deferasirox (DFX), Deferiprone (DFP), and Deferoxamine (DFO), are commonly prescribed either as monotherapy or in combination for the treatment of systemic iron overload. However, the management of cardiac iron overload remains challenging owing to the absence of a standardized chelation regimen exclusively for the management of the condition in patients. This meta-analysis intends to evaluate the comparative efficacy of different iron chelation regimens for the trea

Deferoxamine alleviates T-2 toxin-induced articular chondrocyte ferroptosis via the Nrf2/xCT/GPX4 axis: Implications for Kashin-Beck disease pathology.

Kashin-Beck disease (KBD) is a debilitating endemic osteochondropathy. Environmental toxins, particularly T-2 toxin, and selenium deficiency are established etiological factors. This study aimed to investigate whether T-2 toxin induces ferroptosis in articular chondrocytes and explore the therapeutic potential and mechanism of the iron chelator, Deferoxamine (DFO). A total of 32 three-week-old male Sprague-Dawley rats were randomly allocated into four groups: Control, T-2 toxin (100 ng/g·BW/d), T-2 toxin + low DFO (10 mg/kg), and T-2 toxin + high DFO (100 mg/kg). After a 4-week intervention, k

Azo dye-based on-site assay paper kit for rapid and selective naked-eye and smartphone-assisted detection of deferoxamine drug.

Ferric ion {Fe(III)} is an integral part of many structural and functional components in the body and plays critical roles in physiological and pathological processes. High Fe(III) concentrations damage many systems and organs in the body. The most common method used to prevent excessive accumulation of Fe(III) ions is chelation therapy and the most commonly used drug is deferoxamine (DFO). In this study, an azo dye-based test-paper probe was developed for the colorimetric determination of DFO. The bi-dental azo-dye, named DAZS (diazosalicylaldehyde), was synthesized with a 58 % yield as due t

Prevention of Iron Overload and Long Term Maintenance of Normal Iron Stores in Thalassaemia Major Patients using Deferiprone or Deferiprone Deferoxamine Combination.

Decrease in mortality and morbidity is observed in thalassaemia major patients with reduced iron load in comparison to heavy iron loaded patients. Effective and complete treatment of transfusional iron overload can be achieved by chelation protocols that can eliminate excess iron and maintain normal iron stores (NIS). The maintenance of NIS, i. e., serum ferritin (350 μg/L >), MRI T2* cardiac (>20 ms) and liver (>6.3 ms) relaxation time levels was monitored in 16 thalassaemia major patients (32-53 years, 12 splenectomized, 10 male, erythrocyte transfusions 120-323 ml/kg/year) for about 90 pati

Deferoxamine-mediated up-regulation of HIF-1α prevents dopaminergic neuronal death via the activation of MAPK family proteins in MPTP-treated mice.

Accumulating evidence suggests that an abnormal accumulation of iron in the substantia nigra (SN) is one of the defining characteristics of Parkinson's disease (PD). Accordingly, the potential neuroprotection of Fe chelators is widely acknowledged for the treatment of PD. Although desferrioxamine (DFO), an iron chelator widely used in clinical settings, has been reported to improve motor deficits and dopaminergic neuronal survival in animal models of PD, DFO has poor penetration to cross the blood-brain barrier and elicits side effects. We evaluated whether an intranasal administration of DFO

Sustained improvements in myocardial T2* over 2 years in severely iron-overloaded patients with beta thalassemia major treated with deferasirox or deferoxamine.

Long-term controlled studies are needed to inform on the clinical benefit of chelation therapy for myocardial iron removal in transfusion-dependent beta thalassemia patients. In a 1-year nonrandomized extension to the CORDELIA study, data collected from patients with myocardial siderosis provided additional information on deferasirox or deferoxamine (DFO) efficacy and safety. Myocardial (m)T2* increased from baseline 11.6 to 15.9 ms in patients receiving deferasirox for 24 months (n = 74; geometric mean [Gmean ] ratio of month 24/baseline 1.38 [95% confidence interval 1.28, 1.49]) and from 10.

Expression of proteins secreted by osteocytes in bone biopsies of patients with chronic kidney disease and iron overload, before and after treatment with deferoxamine.

INTRODUCTION: Individuals with CKD can develop anemia, as well as mineral and bone disorder. Anemia is treated with recombinant erythropoietin and iron supplementation, which can result iron overload in various organs. Little is known about the effects of iron overload and its treatment on bone remodeling and osteocyte expression of relevant proteins. The objective of this study was to evaluate the effects that iron overload and treatment with the iron chelator deferoxamine have on parameters of iron metabolism in bone tissue and on osteocyte protein expression. METHODS: This was an observati

Could Local Application of Hypoxia Inducible Factor 1-α Enhancer Deferoxamine Be Promising for Preventing of Medication-Related Osteonecrosis of the Jaw?

This experimental study investigates the prophylactic effect of deferoxamine (DFO) on medication-related osteonecrosis of the jaw (MRONJ). Thirty-six female Sprague Dawley rats received zoledronic acid (ZA) for eight weeks to create an osteonecrosis model. DFO was locally applied into the extraction sockets with gelatin sponge (GS) carriers to prevent MRONJ. The specimens were histopathologically and histomorphometrically evaluated. Hypoxia-inducible factor 1-alpha (HIF-1α) protein levels in the extraction sockets were quantified. New bone formation rate differed significantly between groups (

Effects of chelators (deferoxamine, deferiprone and deferasirox) on the growth of Klebsiella pneumoniae and Aeromonas hydrophila isolated from transfusion-dependent thalassemia patients.

Infections are among the leading causes of death for thalassemia major patients. The known predisposing factors of infection include prior splenectomy, iron overload and use of iron chelator such as deferoxamine (DFO). While encapsulated organisms frequently found in splenectomized patients were readily controlled by prophylactic vaccination and vigilant antibiotic treatment, ferrophilic organisms such as Yersinia and Klebsiella remain common pathogens in thalassemic patients. Yersinia infections are more prevalent in temperate regions and Klebsiella infections are commonly found in tropical a

Inhibition of growth of Histoplasma capsulatum yeast cells in human macrophages by the iron chelator VUF 8514 and comparison of VUF 8514 with deferoxamine.

Histoplasma capsulatum requires intracellular iron to survive and multiply within human and murine macrophages (M phi). Thus, iron chelators may be useful compounds in the treatment of histoplasmosis. In the present study we compared the efficacies of five different iron chelators with deferoxamine (DEF) for their capacity to inhibit the growth of H. capsulatum yeast cells in culture medium and within human M phi. Of the agents tested, only one, VUF 8514, a 2,2'-bipyridyl analog, was found to be effective. VUF 8514 inhibited the growth of yeast cells in tissue culture medium and within M phi i

Disrupting cell death: ferroptosis and pyroptosis inhibition in hepatic ischemia-reperfusion injury via modulation of GPX4 and cGAS pathways by deferoxamine and octreotide.

Hepatic ischemia-reperfusion injury (HIRI) is a significant complication of liver transplantation, often precipitating postoperative liver dysfunction/failure. Given the rising global prevalence of end-stage liver disease, which necessitates liver transplantation, protecting against HIRI is crucial. Here, we investigated the effects of octreotide (OCT), a pyroptosis inhibitor, and deferoxamine (DEF), a ferroptosis suppressor, individually and combined in the HIRI model. Their impact on cyclic GMP-AMP synthase/stimulator-of-interferon (cGAS/STING) genes and glutathione peroxidase 4 (GPX4) activ

The effects of n-acetylcysteine and/or deferoxamine on manic-like behavior and brain oxidative damage in mice submitted to the paradoxal sleep deprivation model of mania.

Bipolar disorder (BD) is a severe psychiatric disorder associated with social and functional impairment. Some studies have strongly suggested the involvement of oxidative stress in the pathophysiology of BD. Paradoxal sleep deprivation (PSD) in mice has been considered a good animal model of mania because it induces similar manic-like behavior, as well as producing the neurochemical alterations which have been observed in bipolar patients. Thus, the objective of the present study was to evaluate the effects of the antioxidant agent's n-acetylcysteine (Nac) and/or deferoxamine (DFX) on behavior

Effect of deferoxamine and allopurinol on non-protein-bound iron concentrations in plasma and cortical brain tissue of newborn lambs following hypoxia-ischemia.

Reduction of non-protein-bound iron (NPBI) using iron chelators may attenuate hypoxia-ischemia-induced reperfusion injury of the brain. This study investigated whether administration of low-dose deferoxamine and allopurinol, both having NPBI-chelating properties, reduced hypoxia-ischemia-induced NPBI formation in plasma effluent from the brain and in cerebral cortical tissue. Twenty-one newborn lambs underwent severe hypoxia-ischemia. Upon reperfusion and reoxygenation the lambs received either a placebo (n = 7), or deferoxamine 2.5 mg/kg (n = 7) or allopurinol 20 mg/kg (n = 7). The post-hypox

Systemic lipid remodeling associated with deferoxamine treatment, dietary intervention and combinatorial effect in ameliorating high cholesterol diet-induced steatotic liver in mice.

INTRODUCTION: Metabolic dysfunction-associated steatotic liver disease (MASLD) is becoming increasingly more prevalent and poses a major growing health concern worldwide. Iron deposition have been observed in the liver of MASLD patients and iron can lead to the formation of reactive oxygen species. Although lipids were altered in the livers of MASLD patients, the underlying mechanisms implicated in this disease have not yet been fully elucidated. Efficacy of Deferoxamine Mesylate in Serum and Serum-Free Media: Adult Ventral Root Schwann Cell Survival Following Hydrogen Peroxide-Induced Cell Death.

Schwann cell (SC) transplantation shows promise in treating spinal cord injury as a pro-regenerative agent to allow host endogenous neurons to bridge over the lesion. However, SC transplants face significant oxidative stress facilitated by ROS in the lesion, leading to poor survival. deferoxamine mesylate (DFO) is a neuroprotective agent shown to reduce H2O2-induced cell death in serum-containing conditions. Here we show that DFO is not necessary to induce neuroprotection under serum-free conditions by cell survival quantification and phenotypic analysis via immunohistochemistry, Hif1α and col
