Dendrobium officinale polysaccharide improves hair regrowth in androgenetic alopecia mice and is associated with coordinated changes in local steroid metabolism, ESR1 activation, and keratin-related follicular responses.

BACKGROUND: Androgenetic alopecia (AGA) is characterized by androgen-dependent follicular miniaturization and progressive disturbance of follicular structure and cycling. Local steroid metabolism, including 5α-reductase-related androgen production and aromatase-related estrogen conversion, may contribute to this process. METHODS: A dihydrotestosterone (DHT)-induced mouse model of AGA was used to evaluate the in vivo effects of Dendrobium officinale polysaccharide (DOP). Dose screening was followed by histological assessment, quantitative proteomics, biochemical assays, multiplex immunofluores

Single-cell and haplotype-resolved transcriptomics reveal molecular signatures of orchid floral structures.

Orchidaceae is among the largest families of flowering plants, yet the cell heterogeneity and molecular basis underlying specialized orchid floral structures remain largely unknown. Here, we report a spatially resolved single-cell transcriptomic census of Dendrobium Chao Praya Smile flower buds, identifying 14 distinct cell types. Compared with the generation of conventional pollen grains, Dendrobium meiotic cells are uniquely enriched with specific enzymatic activities associated with mucilage-mediated pollinium formation. We identified specialized cell populations that drive tepal morphology

Dendrobine promotes bone formation via the canonical Wnt/β-catenin signaling pathway and prevents postmenopausal osteoporosis.

BACKGROUND: In early postmenopausal women with sex steroid deficiency, an imbalance between bone resorption and bone formation causes an accelerated decline in bone mineral density due to a longer lifespan of osteoclasts and a shorter lifespan of osteoblasts. Given this context, dendrobine, as a primary bioactive alkaloid isolated from Dendrobium nobile Lindl., has potential efficacy on osteoporosis due to the traditional use of Dendrobium nobile Lindl. in musculoskeletal disease. OBJECTIVE: To investigate the anti-osteoporotic efficacy of dendrobine in increasing osteoblastogenesis and to el

Investigation of cytokinin-deficient phenotypes in Arabidopsis by ectopic expression of orchid DSCKX1.

The plant hormone cytokinin plays a major role in regulating plant growth and development. Here we generated cytokinin-reduction Arabidopsis plants by overexpressing a heterologous cytokinin oxidase gene DSCKX1 from Dendrobium orchid. These transgenic plants exhibited reduced biomass, rapid root growth, decreased ability to form roots in vitro, and reduced response to cytokinin in growing calli and roots. Furthermore, the expression of KNAT1, STM, and CycD3 genes was significantly reduced in the transgenic plants, suggesting that cytokinin may function to control the cell cycles and shoot/root
