ACSS2 Suppresses Ferroptosis to Drive Breast Cancer Brain Metastasis.

UNLABELLED: Brain metastasis in patients with breast cancer represents a terminal disease stage, with a median survival typically measured in months. Tumors that colonize the brain must adapt to its unique microenvironment, such as high acetate levels. Primary brain tumor cells enhance acetate conversion to acetyl-CoA through phosphorylation of acetyl-CoA synthetase 2 (ACSS2) by cyclin-dependent kinase 5 (CDK5), a process regulated by the nutrient sensor O-GlcNAc transferase (OGT). In this study, we showed that brain-metastatic breast cancer cells exhibited elevated O-GlcNAc, OGT, and phosphor

Beyond arsenite: Arsenite-oxidizing prokaryotes drive sulfur compound oxidation.

Arsenite-oxidizing prokaryotes (AOPs) contribute significantly to the biogeochemical processes governing arsenic cycling. Since AOPs can oxidize As(III) to As(V), markedly reducing arsenic toxicity and decreasing its migration potential, they have been widely utilized in the restoration of As(III)-contaminated environment and in the design of bioreactors for treating As(III)-contaminated groundwater. However, it is still unclear whether AOPs possess additional catalytic activities that may interfere with their bioremediation capacity, highlighting a critical knowledge gap that warrants further

Aiolos and Eos drive distinct human TH17 functional states.

CD4+ T helper (TH)-17 cells play a pivotal role in mucosal immune defense and are implicated in autoimmune diseases and cancer. Although Th17 cell plasticity is well-studied in mice, the factors driving their transition between pro-inflammatory and immunomodulatory states in humans remain less understood. Our study explored the transcriptional and epigenetic landscapes of single-cell cultures of human memory TH17 cells, focusing on clones that produce either immunomodulatory IL-10 or pro-inflammatory IFNγ and IL-22. We found that IL-10+ TH17 cells exhibit a T cell exhaustion-like profile with

How molecular mechanisms of aging drive Alzheimer's disease pathology.

Aging is a "multidimensional engine" of biological dysfunction that can fundamentally reshape the pathology of Alzheimer's disease (AD), This review systematically elaborates on how aging synergistically promotes the core pathologies of AD: aging upregulates the activity of β-secretase 1 (BACE1)/γ-secretase, impairs the clearance function of glial cells and meningeal lymphatic drainage, and accelerates Aβ deposition; the imbalance of kinases/phosphatases, dysfunction of molecular chaperones, and aging exosome-mediated propagation of Tau "seeds" facilitate Tau pathology; hyperreactivity of micr

Androgens Drive SOX9 Upregulation in Injured Proximal Tubular Cells.

INTRODUCTION: Sex influences susceptibility, recovery, and long-term outcomes after acute kidney injury (AKI) in humans. Rodent models have been invaluable for elucidating AKI mechanisms. However, most studies have focused on males, assuming direct applicability to females, an assumption that remains largely untested. In males, the transcription factor SOX9 regulates injury-associated proximal tubule cell states, promoting adaptive repair and survival in the early phase while contributing to fibrosis in later phases. Given the well-described role of SOX9 in sex determination during development

Defence systems drive accessory genome interactions in Pseudomonas aeruginosa.

Bacterial genomes represent dynamic ecological systems in which highly dynamic accessory genome element compositions drive evolution. Emerging evidence suggests that bacterial defence systems, which protect against phages and other genetic elements, can interact cooperatively, competitively, and antagonistically to influence horizontal gene transfer, shape phage susceptibility, and diversify genomes across environments. Recent ecological studies reveal non-random co-occurrence and avoidance patterns among defence systems, suggesting that these patterns may emerge from ecological and evolutiona

Structural defects in amyloid-β fibrils drive secondary nucleation.

Formation of new amyloid fibrils and oligomers from monomeric protein on the surfaces of existing fibrils is an important driver of many disorders such as Alzheimer's and Parkinson's diseases. The structural basis of this secondary nucleation process, however, is poorly understood. Here, we ask whether secondary nucleation sites are found predominantly at rare growth defects: irregularities in the fibril core structure incorporated during their original assembly. We first demonstrate using the specific inhibitor of secondary nucleation, Brichos, that secondary nucleation sites on Alzheimer's d

Assessment of Respiratory Drive in the Mechanically Ventilated Patient.

Respiratory drive is a critical yet often underappreciated determinant of outcome during mechanical ventilation. Both insufficient and excessive drive can cause harm: Low drive promotes diaphragm atrophy, whereas high drive may contribute to lung and diaphragm injury. A key clinical challenge is the potential dissociation between drive and effort in patients with respiratory muscle weakness, where high drive may not translate to adequate muscular output. Simple bedside tools such as airway-occlusion pressure (P0.1) and whole-breath occlusion pressure (POCC) can identify patients at risk and gu

Dynamic airways drive aerosol deposition in the human lung.

Most computational studies of aerosol transport in the lung treat airways as rigid tubes and breathing as a prescribed inflow problem. This simplification overlooks a fundamental reality that in the living lungs, airflow is generated by deforming airways, and not imposed at the inlet. As a result, the consequent predictions of where the inhaled particles deposit can remain fundamentally flawed. Here, we introduce a subject-specific, physiology-driven framework that directly couples airway motion with airflow and particle transport. Using Computed Tomography (CT)-derived tracheobronchial geomet

Mucosal-Associated Invariant T Cells Drive Bile Duct Inflammation.

BACKGROUND & AIMS: Mucosal-associated invariant T cells recognize microbial-derived vitamin B metabolites presented by MHC I-related molecules. Because bile from patients with chronic biliary inflammation contain mucosal-associated invariant T antigens, we investigated whether intrabiliary mucosal-associated invariant T antigen exposure activates mucosal-associated invariant T cells and induce pathogenic biliary inflammation. METHODS: Escherichia coli, phosphate-buffered saline, or the known mucosal-associated invariant T ligand 5-(2-oxopropylideneamino)-6-D-ribitylaminouracil were injected i

KRAS mutations take different trajectories to drive pancreatic transformation.

KRAS mutations are highly prevalent in pancreatic cancer and are critical for epithelial reprogramming during tumor initiation. In this issue of Developmental Cell, Grimont et al.1 demonstrate that the three most common KRAS mutations differentially activate downstream signaling pathways, resulting in distinct capacities to develop pancreatic pre-neoplastic lesions.

Chromatin unfolding via loops can drive clustered transposon insertion.

Transposons, DNA sequences capable of relocating within the genome, make up a significant portion of eukaryotic genomes and are often found in clusters. Within the cell nucleus, the genome is organized into chromatin, a structure with varying degrees of compaction due to three-dimensional folding. Transposon insertion or activation can lead to chromatin decompaction, increasing accessibility and potentially facilitating further nearby insertions. This positive feedback between chromatin unfolding and transposon insertion may result in transposon clustering. Here, we combine bioinformatics with

Extreme storm events drive beach connectivity through headland bypassing.

Headland bypassing is a mainly wave-driven coastal process that interconnects sediment compartments and allows the continuity of longshore sediment transport. In turn, waves are subject to the variability of atmospheric patterns and climate drivers. This study focuses on identifying the atmospheric systems and associated hydrodynamic conditions that have triggered bypassing pulses in the study area, Fingal Head (New South Wales, Australia), over the last 33 years. For this, clustering techniques were applied to identify 225 weather types that represent the daily atmospheric variability over th

Multiple cell-type interactions drive invariant NKT cell hepatitis.

BACKGROUND: α-Galactosylceramide (α-Galcer), a specific ligand for invariant natural killer T (NKT) cell activation, has been actively investigated in clinical trials such as antitumor therapy; however, treatment with α-Galcer is well known to induce acute hepatitis due to enriched NKT cells in the liver. The molecular mechanisms underlying NKT-mediated hepatitis still remain obscure. The object of this study was to investigate whether and how myeloid cells affect NKT-mediated hepatitis. METHODS: α-Galcer-induced NKT hepatitis was used in this study. microRNA-223 (miR-223) and neutrophil cyto

GM1 and OligoGM1 Drive Divergent α-Synuclein Aggregation Pathways.

Parkinson's disease (PD) is an age-related disorder characterized by amyloid deposits of the 140-amino-acid protein α-synuclein (AS), which contribute to neuronal dysfunction and degeneration, leading to motor and cognitive impairments. AS fibrillation, the process by which the native soluble protein misfolds into insoluble cross-β-sheet fibrils, involves transient prefibrillar species with distinct biophysical features. In vitro evidence suggests that these oligomeric intermediates, rather than mature fibrils, are the primary pathogenic agents in neurodegenerative diseases. Initial studies de

Radiation reprograms fibroblasts to drive prostate cancer therapy resistance.

Radiotherapy is a mainstay treatment for localized prostate cancer (PCa). Yet, radiation resistance remains a major clinical obstacle. Here, radiation induced a BMP/CD105-dependent metabolic shift in the tumor microenvironment that facilitates PCa cell survival. Using prostate tumor models and fibroblast cultures, we show that radiation enhances epithelial BMP ligand production, which promotes fibroblastic CD105 signaling. Metabolomic analysis upon radiation revealed that fibroblastic CD105 signaling elevated key enzymes involved in mitochondrial biogenesis (PGC1α) and ketogenesis (HMGCS2). Th

Transcription factors SP5 and SP8 drive primary cilia formation.

While specific transcription factors are known to regulate cell fate decisions, the degree to which they can stimulate formation of specific cell organelles is less clear. We used a multi-omics comparison of the transcriptomes of ciliated and non-ciliated embryonic cells to identify transcription factors upregulated in ciliated cells, and conditional genetics in mouse embryos and stem cells to demonstrate that SP5/8 regulate cilia formation and gene expression. In Sp5/8 mutant embryos primary and motile cilia are shorter than normal and reduced in number across cell types, contributing to situ

Progenitor T cells drive chronic pulmonary type 2 inflammation.

Type 2 inflammation unfolds over time and is coordinated by remarkably durable CD4+ Th2 cell responses, standing in contrast to the collapse of adaptive immunity seen in chronic infection and malignancy. In experimental models, short-lived Th2 effector cells and type 2 innate lymphocytes (ILC2) contribute to acute type 2 inflammation, yet the cellular architecture that drives chronic type 2 inflammation and prevents exhaustion remains poorly understood. To define the Th2 landscape in chronic type 2 inflammation, we established a mouse model of long-term pulmonary allergen exposure and found th
