Histopathology of the human inner ear in Alström's syndrome.

Alström's syndrome is an autosomal recessive syndromic genetic disorder caused by mutations in the ALMS1 gene. Sensorineural hearing loss occurs in greater than 85% of patients. Histopathology of the inner ear abnormalities in the human has not previously been fully described. Histopathology of the inner ear in Alström's syndrome is presented in 2 genetically confirmed cases. The predominant histopathologic correlates of the sensorineural loss were degeneration of the organ of Corti, both inner and outer hair cells, degeneration of spiral ganglion cells, and atrophy of the stria vascularis and

[Introspection of oxidant/antioxidant imbalance in the inner ear].

Free radicals are atoms, molecules or ions with unpaired electrons. In biological systems, free radicals can have a dual role, being beneficial in some situations and deleterious in others. Free radicals are required for normal cellular metabolism, but they lead to cellular degeneration if overproduced. To prevent the excessive buildup of free radicals, cells have developed an elaborate series of antioxidant enzymes that counteract oxidative stress and protect cells by maintaining the proper balance of oxidation and anti-oxidation. Therefore, when there is an oxidant/anti-oxidant imbalance, no

Role of AQP1 in inner ear in motion sickness.

Inner ear is critical for the development of motion sickness (MS). The present work was designed to test the role of aquaporins (AQPs) in inner ear in MS. After repetitive stimulus of rotation, the MS symptom was steadily alleviated in mice. After repetitive stimulus of rotation, several AQPs mRNA levels including AQP1, AQP2, AQP3, AQP4, AQP6, AQP7, and AQP9 in the inner ears of mice were analyzed. It was found that AQP1 mRNA level was increased remarkably, which was reconfirmed by Western blotting analysis. In addition, the relationship between AQP1 expression and MS sensitivity was studied a

A preliminary theoretical model of hydrodynamics in the inner ear.

Head movement should create a transient pressure imbalance across the membranous inner ear. We used basic concepts of fluid dynamics to develop a theoretical model of the inner ear. According to this model, two contiguous fluidic systems-the perilymphatic system and the endolymphatic system-are in hydrostatic equilibrium across a compliant membrane. Our model demonstrates that changes in resistance or compliance in one system results in a transient distortion of the membranous inner ear until equilibrium between the two systems is restored. The concept of hydrodynamic pressure changes in the i

Histopathology of the inner ear in unoperated acoustic neuroma.

Although hearing loss is the most common presenting symptom in patients with acoustic neuroma, the pathophysiology of hearing loss associated with acoustic neuroma is unknown. Although primary dysfunction of the auditory nerve is intuitively logical, available histopathologic and clinical data suggest that although neural degeneration is common, it alone does not adequately account for hearing loss in many cases. The purpose of this study was to evaluate 11 cases of unoperated unilateral acoustic neuromas. Temporal bones were identified by means of a search mechanism provided by the National T

Virtual endoscopy of the inner ear and the auditory canal.

To assess the role of virtual endoscopy (VE) in the examination of intracisternal structures and of the inner ear, we studied the anatomy of the labyrinth and internal auditory canal using the original CT slices and VE on the unaffected side in three female and three male patients, age range 3-46 years, with contralateral retrocochlear hearing loss. We also examined seven patients with different pathological findings. VE was performed using an advanced postprocessing program with high-resolution 3D data sets of CT (1-1.5 mm thickness, pitch 1.25) and MRI-CISS-3D (constructive interference in s

Regulation of inner ear fluid in the rat by vasopressin.

The anti-diuretic hormone vasopressin has been shown to be important in regulating inner ear fluid. The diuretic hormone, CNP, and its receptor, ANP-B receptor, may also function in the regulation of inner ear fluid. To determine whether vasopressin directly affects the fluid level, we infused this hormone to rat and assay of V2-AVP receptor mRNA by semiquantitative RT-PCR demonstrated a significantly lower level of this transcript in vasopressin-infused animals than in saline-infused animals. The levels of CNP and ANP-B receptors mRNA, however, were the same in both groups of rats. Results su

Allergy for the otologist. External canal to inner ear.

Allergy may affect the outer, the middle, or the inner ear. Although the otologic manifestations of allergy are not by themselves diagnostic, the history, including family history and associated symptoms in other target organs, will often help lead to the correct diagnosis and institution of therapy. Patients with significant and chronic symptoms, including those with labyrinthine symptoms of allergy, will respond well to specific immunotherapy and/or dietary elimination.

[Delayed damage of ionizing radiation on the inner ear].

To investigate the feature and mechanism of delayed effect of ionizing radiation on the inner ear, the function and structure of the inner ear of the guinea pigs were observed during 8 month's period after exposure to fractional doses of gamma radiation (2 Gy per day) with a total dose of 60 Gy. Morphological study was performed using light microscopy, transmission and scanning electron microscopy while the function was measured with electrocochleography (ECochG) and electronystagmography (ENG). The results showed that the mean CAP response threshold was 35.67 +/- 6.78 dB before radiation, and

[The phylogenetic imprint on the embryonic development of the inner ear].

The AA. present a review of the embryologic development of the internal ear linked with evolutive phylogenic events, which will determine the morphogenesis of this organ, of ancestral origin. The significance of those phenomena in order to bring about several physiological attributes as the possibility of vestibular hearing and even to set off pathologic conditions like the otosclerosis are considered.

[Sensorineural impairment in the inner ear in epidemic parotitis].

During morbilli and mumps, classical forms of acquired deafness develop in childhood. The aim of the study was to investigate effects of mumps virus on cochlear function. Of 237 examined patients 71 had parotitis and 166 meningitis-meningoencephalitis. Audiometric analyses were carried out at the beginning of the treatment and after 6 months. Hearing loss above 30 dB was found in 109 patients (45.99%). Mild hearing impairment of sensorineural type was registered in the majority of patients, while total one-sided hearing loss was found in 2.53% of subjects. No correlation between clinical manif

Stapedius reflex in patients with an inner ear prosthesis.

The stapedius reflex elicited by electrical stimulation was investigated in 21 deaf individuals supplied with an inner ear prosthesis. All patients were implanted with the Vienna cochlear implant using intra or extracochlear electrodes. Analog stimulation with sinus bursts of various durations (25, 50, 100, 300, 500 ms) and frequencies (125, 500, 1000, 2000 Hz) was used. The time course of the contralateral acoustic reflex was monitored using a fast-response impedance meter. The stapedius reflex was observed in about half of the cases. Reflex threshold was observed at a stimulation level close

The effects of DFMO on polyamine metabolism in the inner ear.

Difluoromethylornithine (DFMO) is a novel antineoplastic agent that was associated with an unexpected hearing loss in Phase II clinical trials. DFMO interferes with polyamine synthesis by inhibition of the enzyme ornithine decarboxylase (ODC). The objective of the current study was to establish a methodology to determine the effect of DFMO on polyamine levels and ODC activity in the cochlea. Guinea pigs received DFMO in their drinking water and were tested for auditory brainstem response threshold shifts. The organ of Corti, the lateral wall, and the acoustic nerve were assayed for both ODC ac

Preservation of hearing in cholesteatomas with inner ear invasion.

Two cases of inner ear destruction by cholesteatoma are presented. There was total demineralization of the cochlea in one and significant vestibular destruction in the other. Eradication and exteriorization of the disease was accomplished with preservation of hearing in both cases. Safe management was enhanced by aggressive infection control and accurate surgical planning aided by current imaging techniques. The factors which protect auditory function in these lesions remain unclear. However, the site and extent of the disease as well as local tissue reaction leading to separation of the infla

[Malformation of the inner ear. Cerebrospinal otorrhea in stapedectomy].

Otoliquorrhoea occurred during stapedectomy (gusher) and signified a case of inner ear malformation which was not apparent on preoperative tomograms but could be demonstrated on postoperative CT scans. There was no bony separation between the irregularly dilated inner auditory canal and the vestibulum. The cochlea was not fully rotated. Intraoperative problems in such malformations may lead to further postoperative hearing impairment. Hence, we report on the clinical, operative and CT findings in order to avoid an operation in patients with similar signs and symptoms.

Pathology of neuroepithelial suprastructures of the human inner ear.

Neuroepithelial suprastructures in abnormal human inner ears were studied by light microscopy, scanning electron microscopy, and x-ray diffraction. The most common abnormality was calcification, which selectively affected the gelatinous membranes (otoconial, cupular, and tectorial) and the secretory tissues (stria vascularis and utricular dark cells). The structures most frequently affected were the otoconial membranes. The minerals involved were apatite, octacalcium phosphate, and vaterite, replacing the normal layer of calcium carbonate in the form of calcite crystals. The first two of these

[Therapeutic considerations in vascular diseases of the inner ear].

I propose an analogy between vascular diseases of the inner ear and those of other organs like brain, heart or peripheral vessel diseases in which functional and organic alteration can be objectively demonstrated. Symptomatic disturbances of inner ear circulation are monoform in its clinical appearance and the term of 'otangina' is proposed for the functional result. Three types of vascular distribution are discussed for the different regulatory entities, (I) the proximal cerebral vascular type, (II) the predistal innervated regulatory type and the (III) capillary regulatory type. Experimental studies on the role of vitamin A in the inner ear.

With the sole exception of the hair cells of the inner ear, where information is lacking, all special somatic afferent receptor cells have been shown to be dependent upon vitamin A for normal function. In view of the paucity of information on the role of vitamin A in the inner ear, three experiments were performed to examine this relationship. Temporal bone histopathology was studied in rats deprived of vitamin A. In a second experiment, vitamin A-deficient rats were maintained with vitamin A acid and the histopathology was studied under the light microscope. In the third experiment, a microfl

Affinity of Sendai virus for the inner ear of mice.

The direct immunofluorescent antibody method revealed that Sendai virus, which was thought to be most prevalent in respiratory diseases in mice, infected the inner ear of mice when the virus was injected intracerebrally. Specific immunofluorescence that indicated the presence of antigens of Sendai virus was observed in both perilymphatic and endolymphatic structures. The routes of virus spread are discussed. This experimental study, using newborn mice and Sendai virus, may be useful for the investigation of the pathogenesis of viral labyrinthitis not only because of the high frequency of inner
