Emodin Improves Intestinal Health and Immunity through Modulation of Gut Microbiota in Mice Infected by Pathogenic Escherichia coli O1.

The effect of emodin on the intestinal mucosal barrier of a mouse E. coli O1-induced diarrhea model was observed. Following successful establishment of a diarrhea model, the mice were treated with drugs for seven days. Intestinal lesions and the shape and the number of goblet cells were assessed via hematoxylin-eosin and periodic-acid-Schiff staining, while changes in inflammatory factors, ultrastructure of the small intestine, expression of MUC-2, and changes in the intestinal microbiota were analyzed via RT-PCR, electron microscopy, immunofluorescence, and 16S rRNA sequencing. Examination sh

Aloe-Emodin Relieves High-Fat Diet Induced QT Prolongation via MiR-1 Inhibition and IK1 Up-Regulation in Rats.

BACKGROUND/AIMS: High-fat diet (HFD) causes cardiac electrical remodeling and increases the risk of ventricular arrhythmias. Aloe-emodin (AE) is an anthraquinone component isolated from rhubarb and has a similar chemical structure with emodin. The protective effect of emodin against cardiac diseases has been reported in the literature. However, the cardioprotective property of AE is still unknown. The present study investigated the effect of AE on HFD-induced QT prolongation in rats. METHODS: Adult male Wistar rats were randomly divided into three groups: control, HFD, and AE-treatment groups

Involvement of PI3K/Akt, ERK and p38 signaling pathways in emodin-mediated extrinsic and intrinsic human hepatoblastoma cell apoptosis.

As a natural anthraquinone derivative, 1,3,8-trihydroxy-6-methylanthraquinone, known as emodin, has recently been reported to possess potential chemopreventive capacity, but the underlying molecular mechanism of its hepatocyte toxicity remains poorly clarified. The present research indicated that emodin targeted HepG2 cells without being cytotoxic to primary human hepatocyte cells in comparison with chrysophanol and rhein. The anti-proliferative effect of emodin was ascribed to occurrence of apoptosis, which characterized by higher ethidium bromide signal, brighter DAPI fluorescence, cleavages

Emodin ameliorated lipopolysaccharide-induced fulminant hepatic failure by blockade of TLR4/MD2 complex expression in D-galactosamine-sensitized mice.

Emodin has been reported to possess anti-inflammatory and anti-oxidant activities. The aim of this study was to explore the effect and mechanism of emodin on lipopolysaccharide (LPS)-induced fulminant hepatic failure (FHF) in D-galactosamine (D-GalN)-sensitized mice. Our results showed that pretreatment with emodin inhibited the elevation of plasma aminotransferases, alleviated the hepatic histopathological abnormalities and improved the survival rate of LPS/D-GalN-primed mice. Moreover, emodin markedly attenuated the increased serum and hepatic tumor necrosis factor-α (TNF-α) production, and

A diatom/PdNPs nanocomposite carbon paste electrode for sensitive voltammetric determination of aloe emodin: electrodeposition, characterization, and oxidation mechanism.

Aloe emodin (AE, 1,8-dihydroxy-3-(hydroxymethyl)anthraquinone) is a naturally occurring anthraquinone derivative with well-documented anticancer, antiviral, and anti-inflammatory properties, making it an important target for sensitive analytical determination. In this study, a novel electrochemical sensor based on a diatom/palladium nanoparticle nanocomposite-modified carbon paste electrode (DA/PdNPs/CPE) was developed for the highly sensitive and selective determination of AE in herbal samples. The carbon paste electrode was first modified with purified diatomaceous earth (DA), and palladium

Emodin inhibits colon cancer tumor growth by suppressing tumor cell glycolysis through inhibition of NAT10-mediated PGK1 ac4C modification.

INTRODUCTION: Inhibition of glycolysis represents a potent therapeutic approach for colon cancer. Emodin, a natural plant-derived compound with anticancer properties, has shown efficacy in cancer treatment. NAT10 promotes cancer progression by catalyzing ac4C production on mRNAs, but it remains unclear whether emodin regulates glycolysis in colon cancer cells by targeting NAT10. This study aimed to investigate the role of emodin in glycolysis regulation in colon cancer and its underlying mechanisms. METHODS: Glycolysis was assessed by measuring cell proliferation, glucose uptake, lactate prod

Emodin Decreases Tumor-Associated Macrophages Accumulation and Suppresses Bladder Cancer Development by Inhibiting CXCL1 Secretion from Cancer-Associated Fibroblasts.

Tumor-associated macrophages (TAMs) and cancer-associated fibroblasts (CAFs) are the most abundant stromal cells in the bladder cancer (BC) microenvironment (TME). However, the detailed mechanisms underlying TAM-CAF communication and their contributions to BC progression remain incompletely understood. Emerging evidence shows that Emodin exerts anti-tumor effect on several tumor models by targeting TME. To date, the impact of Emodin on BC has not been previously reported. Our study firstly demonstrated that Emodin significantly inhibited tumor growth and reduced TAM accumulation in a murine BC

Emodin attenuates estrogen-deficient sarcopenia by suppressing CYP1B1 to restore CD36-driven fatty acid handling in muscle stem cells.

Sarcopenia is a progressive, estrogen-deficiency-related syndrome characterized by the loss of muscle mass and regenerative capacity, particularly in postmenopausal women. We identify Emodin, a natural anthraquinone compound, as a potent metabolic modulator capable of alleviating sarcopenia through the CYP1B1/CD36 axis. Bioinformatics analysis revealed CYP1B1 as a top-ranked target of Emodin. In ovariectomized mice, Emodin administration significantly restored grip strength, muscle fiber architecture, and redox homeostasis. Mechanistically, Emodin suppressed CYP1B1 expression, thereby enhancin

Aloe-Emodin Targeting FOXC2 Disrupts NETs Formation and EMT-Driven Postoperative Peritoneal Adhesion Through TGF-β1-Smad2/3 Pathway.

Postoperative peritoneal adhesion (PPA) develops through TGF-β1-driven fibrotic remodeling, characterized by neutrophil extracellular trap (NETs)-induced aberrant epithelial-to-mesenchymal transition (EMT) deposition. Although aloe-emodin (AE) exhibits anti-fibrosis potential, its molecular mechanisms remain elusive. Forkhead box protein C2 (FOXC2) is a critical regulator of fibrotic tissue formation, yet its role in PPA is unknown. Here, it is demonstrated that FOXC2 expression is elevated in human ileostomy tissue, PPA rodent model, and TGF-β1-exposed peritoneal mesothelial cells (PMCs), whe

Aloe-emodin alleviates cerebral ischemia-reperfusion injury by regulating microglial polarization and pyroptosis through inhibition of NLRP3 inflammasome activation.

BACKGROUND: Microglial activation plays a crucial role in injury and repair after cerebral ischemia, and microglial pyroptosis exacerbates ischemic injury. NOD-like receptor protein 3 (NLRP3) inflammasome activation has an important role in microglial polarization and pyroptosis. Aloe-emodin (AE) is a natural anthraquinone compound originated from rhubarb and aloe. It exerts antioxidative and anti-apoptotic effects during cerebral ischemia/reperfusion (I/R) injury. However, whether AE affects microglial polarization, pyroptosis, and NLRP3 inflammasome activation remains unknown. PURPOSE: This

Emodin suppresses activation of hepatic stellate cells through p38 mitogen-activated protein kinase and Smad signaling pathways in vitro.

The aim of this study was to evaluate the hypothesis that emodin inhibits extracellular matrix (ECM)-related gene expression in activated hepatic stellate cells (HSCs) by blocking canonical or/and noncanonical components of transforming growth factor β1 (TGFβ1) intracellular signaling. Here, we demonstrate that emodin suppressed the gene expression of HSCs activation markers type I collagen, fibronectin, and α-smooth muscle actin, as well as HSCs proliferation. Mechanistically, emodin suppresses TGFβ1, TGFβ receptor II, TGFβ receptor I, and Smad4 gene expression, as well as Smad luciferase act

Surface-Directed Regulation of Intracellular Trafficking and Apoptotic Pathways by Fluorescent SBA-15 Nanocarriers Governs the Intracellular Fate of Emodin.

Emodin (EO) is a naturally occurring anthraquinone with promising anticancer activity; however, its clinical application is limited by poor aqueous solubility and low bioavailability. Herein, we demonstrate that surface engineering of fluorescent SBA-15 nanocarriers governs the intracellular fate of EO by modulating drug confinement, release behavior, intracellular trafficking, and apoptotic responses. SBA-15 was functionalized with aminopropyl groups and a fluorescent moiety to generate traceable nanocarriers with distinct drug-carrier interactions. Physicochemical characterization (SAXS, BET

Inhibitory Effects of Emodin, Thymol, and Astragalin on Leptospira interrogans-Induced Inflammatory Response in the Uterine and Endometrium Epithelial Cells of Mice.

Leptospirosis is a systemic infection that causes, among others, acute kidney injury, acute liver disease, muscle pain, vasculitis, bleeding disorders, and reproductive loss. In an effort to reduce uterine inflammatory responses induced by Leptospira, we evaluated the anti-inflammation effects of emodin, thymol, and astragalin in a mouse model. Our results showed that treatment with emodin, thymol, and astragalin alleviated uterine inflammation induced by leptospira infection via suppression of pro-inflammatory cytokine expression and prevented tissue damage. Furthermore, we used primary endom

Isolation of a new aloe-emodin dianthrone diglucoside from senna and its potentiating effect on the purgative activity of sennoside A in mice.

Two aloe-emodin dianthrone diglucosides (I and II) were isolated from the leaves of Cassia angustifolia Vahl by successive column chromatography with Amberlite XAD-2, silica gel, Polyamide C-200 and Sephadex LH-20. The stereostructures of I and II were elucidated as trans and meso isomers at 10-10', respectively, from the patterns of the ultraviolet absorption spectra and circular dichroism curves. This is the first report of isolation of diglucoside I from senna. Despite the lack of purgative activity, diglucoside I exerts a potentiating effect of about 1.3 times on the purgative activity of

De novo biosynthesis of the plant-derived aloe-emodin in Aspergillus nidulans via artificial pathway assembly and temporal flux control.

Traditional botanical extraction of medicinal anthraquinones like aloe-emodin is limited by low yields, long growth cycles, and environmental dependence. Microbial biosynthesis provides a sustainable alternative, but de novo reconstruction of aloe-emodin remains difficult due to unresolved pathway steps and poor metabolic flux distribution in engineered hosts. In this study, we engineered Aspergillus nidulans for the de novo biosynthesis of aloe-emodin via modular assembly of an artificial pathway. We first established an emodin-producing chassis by reconstructing a minimal mdp-derived upstrea
