Epidemiological model of typhoid fever and its use in the planning and evaluation of antityphoid immunization and sanitation programmes.

An epidemiological model has been constructed for typhoid fever in a stable population in order to study the transmission of infection at different levels of endemicity. It involves a number of parameters representing the proportions of epidemiological subgroups in the population-such as the susceptible, the infected, and the immune-and rates of transition between the groups. Numerical values based on available evidence were assigned to the parameters, to provide a realistic simulation of stable endemicity.Changes were then introduced in the values of some of the parameters in order to study t

The weighted DMFT (W-DMFT) model: A conceptual framework for severity-adjusted caries assessment in epidemiology.

To critically examine the structural limitations of the classical Decayed-Missing-Filled Teeth (DMFT) index and to propose a biologically structured, severity-adjusted conceptual framework through the Weighted DMFT (W-DMFT) model. This study presents a conceptual-analytic framework without quantitative field data. The methodological constraints of the DMFT index were examined across four domains: biological progression, functional impact, radiographic detectability, and restorative assessment. Based on the enamel-dentin-pulp hierarchy, a structured weighting system (0-3) was developed to refle

The evolution of sampling in epidemiology: from classical probability models to AI-enhanced recruitment and active learning.

Sampling is a foundational element of epidemiological research because it determines the validity, efficiency, and generalizability of population-based inferences. Classical probability and non-probability sampling methods have long underpinned public health study design by providing robust frameworks for unbiased estimation and causal inference. However, the rapid expansion of digital health data, electronic health records, and large-scale online cohorts has exposed important limitations in these largely static sampling frameworks. This narrative review traces the evolution of sampling in epi

Habitat-Mediated Spillover Risk of Andes Hantavirus in Oligoryzomys longicaudatus: A Mechanistic Eco-Epidemiological Model.

Andes hantavirus (ANDV) is a rodent-borne orthohantavirus associated with hantavirus cardiopulmonary syndrome in southern South America. Its maintenance and spillover risk depend on the ecology of its principal reservoir, Oligoryzomys longicaudatus, and on environmental changes that alter habitat availability, host abundance, and human-rodent interfaces. We developed a mechanistic eco-epidemiological model that couples effective habitat cover to an SIR framework for ANDV transmission in O. longicaudatus. Habitat degradation and compensatory restoration modify rodent carrying capacity, natality

Integrated Downstream Analysis and Epidemiological Modelling of Hantavirus Infection: From Host Transcriptomics to Transmission Dynamics.

Hantaviruses are emerging zoonotic pathogens responsible for two severe clinical syndromes: (i) haemorrhagic fever with renal syndrome (HFRS) and (ii) hantavirus cardiopulmonary syndrome (HCPS), collectively causing more than 200,000 human cases annually worldwide. Despite their public-health importance, the molecular mechanisms governing the host response and the population-level dynamics of rodent-to-human spillover remain incompletely characterised. The timeliness of this framework is underscored by the April-May 2026 outbreak of Andes orthohantavirus aboard the MV Hondius cruise ship, the

The Relevance of the Predominant Clonal Evolution (PCE) Model for the Molecular Epidemiology and Subspecific Taxonomy of Trypanosoma cruzi.

The predominant clonal evolution (PCE) model is often misunderstood. Contrary to common belief, it is not restricted to strict mitotic clonality. Instead, it encompasses processes such as selfing, strong homogamy, and various forms of parthenogenesis, as widely acknowledged by researchers studying clonality. Moreover, the PCE model does not claim that genetic recombination is entirely absent or devoid of epidemiological and evolutionary significance. In this review, I will explore the reciprocal relationship between PCE and molecular epidemiology (strain typing) and discuss the implications of

A Compartmental Mathematical Model to Assess the Impact of Vaccination, Isolation, and Key Epidemiological Parameters on Mpox Control.

BACKGROUND: Monkeypox (Mpox) is a re-emerging zoonotic disease caused by the monkeypox virus (MPXV). Transmission occurs primarily through direct contact with lesions or contaminated materials, with sexual transmission playing a significant role in recent outbreaks. In 2022, Mpox triggered a major global outbreak and was declared a Public Health Emergency of International Concern (PHEIC) by the World Health Organization (WHO), prompting renewed interest in effective control strategies. METHODS: This study developed a compartmental SEIR-based model to assess the epidemiological impact of key i

Impacts of population ageing on bloodstream infection epidemiology and outcomes: A machine learning and statistical modelling study.

OBJECTIVES: To investigate the effect of population ageing on pathogen distribution and clinical outcomes in bloodstream infections (BSIs). METHODS: We analysed 37,100 adult patients with BSI from two university hospitals in South Korea (2006-2023) and used statistical and machine learning (ML) approaches to assess temporal trends, age-related changes in causative pathogens, and 30-day mortality. RESULTS: The mean age of patients was 64.6 years (standard deviation 15.0), with 55.7% aged ≥65. Advanced age was independently associated with a high 30-day mortality via a shift in pathogen distri

Importance of confirmatory test characteristics in optimizing community-based screening for tuberculosis: an epidemiological modeling analysis.

BACKGROUND: The costs, operational barriers, and sensitivity of available tools to confirm a TB diagnosis limit current active case-finding (ACF) efforts for tuberculosis (TB). However, it is not well understood which of these limitations have the greatest epidemiological relevance and might therefore warrant prioritization in confirmatory test development. METHODS: To explore which features of the confirmatory testing step most influence the impact of ACF, we developed a state-transition model of a one-time, community-based ACF campaign, with a fixed budget of one million United States dolla

Fate-and-transport modeling of SARS-CoV-2 for rural wastewater-based epidemiology application benefit.

Wastewater-based epidemiology (WBE) for the detection of agents of concern such as severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) has been prevalent in literature since 2020. The majority of reported research focuses on large urban centers with few references to rural communities. In this research the EPA-Storm Water Management Model (EPA-SWMM) software was used to describe a small sewershed and identify the effects of temperature, temperature-affected decay rate, flow rate, flush time, fecal shedding rate, and historical infection rates during the spread of the Omicron variant o

EpiFusion: Joint inference of the effective reproduction number by integrating phylodynamic and epidemiological modelling with particle filtering.

Accurately estimating the effective reproduction number (Rt) of a circulating pathogen is a fundamental challenge in the study of infectious disease. The fields of epidemiology and pathogen phylodynamics both share this goal, but to date, methodologies and data employed by each remain largely distinct. Here we present EpiFusion: a joint approach that can be used to harness the complementary strengths of each field to improve estimation of outbreak dynamics for large and poorly sampled epidemics, such as arboviral or respiratory virus outbreaks, and validate it for retrospective analysis. We pr

Modelling the epidemiological and economic impact of digital adherence technologies with differentiated care for tuberculosis treatment in Ethiopia.

BACKGROUND: Digital adherence technologies (DATs) with associated differentiated care are potential tools to improve tuberculosis (TB) treatment outcomes and reduce associated costs for both patients and healthcare providers. However, the balance between epidemiological and economic benefits remains unclear. Here, we used data from the ASCENT trial to estimate the potential long-term epidemiological and economic impact of DAT interventions in Ethiopia. METHODS: We developed a compartmental transmission model for TB, calibrated to Ethiopia and parameterised with patient and provider costs. We

Dynamic Epidemiological Networks: A Data Representation Framework for Modeling and Tracking of SARS-CoV-2 Variants.

The large-scale real-time sequencing of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) genomes has allowed for rapid identification of concerning variants through phylogenetic analysis. However, the nature of phylogenetic reconstruction is typically static, in that the relationships between taxonomic units, once defined, are not subject to alterations. Furthermore, most phylogenetic methods are intrinsically batch mode in nature, requiring the presence of the entire data set. Finally, the emphasis of phylogenetics is on relating taxonomical units. These characteristics complicate

An Epidemiology Model for Estimating the Numbers of US Patients With Multiple Myeloma by Line of Therapy and Treatment Exposure.

OBJECTIVES: Estimates on the distribution of patients with multiple myeloma (MM) by line of therapy (LOT) are scarce and get outdated quickly as new treatments become available. The objective of this study was to estimate the number of patients with MM by LOT and the number of patients who have received at least 4 previous LOTs including proteasome inhibitors, immunomodulatory agents, and anti-CD38 monoclonal antibodies (mAbs). METHODS: A compartmental model was developed to calculate the number of patients by LOT. Two pathways were considered based on stem cell transplant eligibility, and at

An eco-epidemiological modeling approach to investigate dilution effect in two different tick-borne pathosystems.

Disease (re)emergence appears to be driven by biodiversity decline and environmental change. As a result, it is increasingly important to study host-pathogen interactions within the context of their ecology and evolution. The dilution effect is the concept that higher biodiversity decreases pathogen transmission. It has been observed especially in zoonotic vector-borne pathosystems, yet evidence against it has been found. In particular, it is still debated how the community (dis)assembly assumptions and the degree of generalism of vectors and pathogens affect the direction of the biodiversity-
