Squalene in Camellia oleifera: Biosynthetic Pathways, Regulatory Networks, and Functional Perspectives.

Squalene is a triterpene with potent biological activities. Squalene (C30H50) is a linear polyunsaturated hydrocarbon composed of six isoprene units and six carbon-carbon double bonds. It serves as an essential precursor for sterols, steroid hormones, and vitamin D in humans and exhibits antioxidant, anti-tumor, and lipid-regulating properties. In plants, squalene is produced via the mevalonate (MVA) and 2-C-methyl-D-erythritol-4-phosphate (MEP) pathways. The key rate-limiting enzymes in these pathways include 3-hydroxy-3-methylglutaryl-CoA reductase (HMGR), farnesyl diphosphate synthase (FPS)

Preparation of a Brucella multiepitope fusion protein based on bioinformatics and its application in serological diagnosis of human brucellosis.

Brucellosis is a zoonotic infectious disease caused by the Brucella spp., and the enhancement of diagnostic techniques is imperative for effective disease control. Currently, the diagnosis of brucellosis predominantly relies on serological tests, bacterial culture, and molecular biology methods. Among these approaches, serological diagnosis is the most widely utilized due to its relative simplicity. However, existing diagnostic antigens encounter challenges, such as cross-reactivity. Consequently, the development of novel antigens with high specificity and sensitivity is essential to improve t

Engineered microenvironments and pancreatic islet-on-chips for screening sugar substitute and antidiabetic compounds.

Recent advancements in the food industry have rekindled interest in the safety of food additives, such as sugar substitutes and food pigments. Consequently, the main purpose of this study was to develop models that can more accurately predict the effects of these additives on the human body. In response to this demand, we have created an innovative pancreas islet-on-a-chip system featuring a concentration gradient generator and a perfusable 3D cell culture array. This setup facilitates the 3D culture of pseudo-islets under stable biochemical and biophysical conditions. When compared to static

Limited support for a direct connection between prebiotics and intestinal permeability - a systematic review.

The intestinal barrier is a selective interface between the body´s external and the internal environment. Its layer of epithelial cells is joined together by tight junction proteins. In intestinal permeability (IP), the barrier is compromised, leading to increased translocation of luminal contents such as large molecules, toxins and even microorganisms. Numerous diseases including Inflammatory Bowel Disease (IBD), Coeliac disease (CD), autoimmune disorders, and diabetes are believed to be associated with IP. Dietary interventions, such as prebiotics, may improve the intestinal barrier. Prebiot

Identification of Listeria monocytogenes Genes Contributing to Oxidative Stress Resistance under Conditions Relevant to Host Infection.

The Gram-positive bacterium Listeria monocytogenes survives in environments ranging from the soil to the cytosol of infected host cells. Key to L. monocytogenes intracellular survival is the activation of PrfA, a transcriptional regulator that is required for the expression of multiple bacterial virulence factors. Mutations that constitutively activate prfA (prfA* mutations) result in high-level expression of multiple bacterial virulence factors as well as the physiological adaptation of L. monocytogenes for optimal replication within host cells. Here, we demonstrate that L. monocytogenesprfA*

Deciphering structure, function and mechanism of Plasmodium IspD homologs from their evolutionary imprints.

Malaria is a life-threatening mosquito-borne blood disease caused by infection with Plasmodium parasites. Anti-malarial drug resistance is a global threat to control and eliminate malaria and therefore, it is very important to discover and evaluate new drug targets. The 2-C-methyl-D-erythritol 4-phosphate cytidylyltransferase (IspD) homolog is a second in vivo target for fosmidomycin within isoprenoid biosynthesis in malarial parasites. In the present study, we have deciphered the sequence-structure-function integrity of IspD homologs based on their evolutionary imprints. The function and cata

Dietary intake of sugar substitutes aggravates cerebral ischemic injury and impairs endothelial progenitor cells in mice.

BACKGROUND AND PURPOSE: In our current food supply, sugar substitutes are widely used in beverages and other food products. However, there is limited information about the link between dietary consumption of sugar substitutes and stroke to date. This study sought to determine the effect of various sugar substitutes on the cerebral ischemic injury and endothelial progenitor cells, which have been implicated to play an important role in vascular repair and revascularization in ischemic brain tissues, in mice. METHODS: After treatment with sucrose and various sugar substitutes (the doses are in

Genome sequence of Brucella abortus vaccine strain S19 compared to virulent strains yields candidate virulence genes.

The Brucella abortus strain S19, a spontaneously attenuated strain, has been used as a vaccine strain in vaccination of cattle against brucellosis for six decades. Despite many studies, the physiological and molecular mechanisms causing the attenuation are not known. We have applied pyrosequencing technology together with conventional sequencing to rapidly and comprehensively determine the complete genome sequence of the attenuated Brucella abortus vaccine strain S19. The main goal of this study is to identify candidate virulence genes by systematic comparative analysis of the attenuated strai

[Brucellosis in pregnancy].

Brucellosis is rare in pregnancy. Recently, an increase in the incidence of this disease has been observed in our area. We present 7 cases of brucellosis in pregnancy and review the literature on the effects of brucellosis on the outcome of pregnancy. Brucellosis is rare in the Middle East and Africa and the most common source of infection is unpasteurized milk products. Brucella is a coccobacillus, gram-negative bacterium, whose hosts are mostly animals. There is controversy about the relationship between brucellosis and the outcome of pregnancy. There is some evidence that there is a higher

Conformational stability of lysozyme in choline chloride-polyol aqueous deep eutectic solvents (ADES).

Naturally occurring deep eutectic solvents (NADES) offer a tunable microenvironment for biomolecular stabilization. Yet, the atomistic mechanisms governing the protein-solvent interactions remain poorly understood. Here, we present microsecond-long atomistic molecular dynamics simulations investigating lysozyme (Lys) stability in Choline Chloride (ChCl)-polyol (erythritol, xylitol, and sorbitol) aqueous deep eutectic solvents as a function of water wt. % (fw=0,25,50,75). Lys maintains its native structure across all conditions, with stable Rg, RMSD, RMSF, and secondary structure populations. T

Guar Gum β-Manno-Oligosaccharides Alleviate Ulcerative Colitis in Mice Models via Gut Microflora Modulation.

Gut dysbiosis is a key feature in ulcerative colitis, a globally prevalent subtype of inflammatory bowel disease. Microbiota-directed dietary supplements can restore the dysbiotic gut and alleviate ulcerative colitis. We aimed to delineate whether prebiotic guar gum β-manno-oligosaccharides [GG-β-MOS(i) 500 mg/kg, (ii) 1000 mg/kg] favors gut microflora and microflora associated characteristics (MACs) and can prevent dextran sodium sulfate (DSS) induced ulcerative colitis in mice model. DSS challenged adult male C57BL/6 mice were used as model for colon inflammation evaluation. Food and water c

Design and synthesis of sulfur-containing pseudilin analogs and their biological activity evaluation.

Pseudilin is a natural product that inhibits the enzyme 4-diphosphocytidyl-2C-methyl-D-erythritol synthase (IspD) in the methylerythritol phosphate (MEP) pathway, and using it as a template for designing lead structure is an effective strategy in developing new active compounds. In this work, a series of sulfur-containing pseudilin analogs were designed by replacing the pyrrole rings in pseudilin with pyrazole rings through bioisosteric replacement strategy, while introducing sulfur-containing groups. The target compounds were synthesized through a four-step reaction sequence, in which the key

Protein-Ribofuranosyl Interactions Activate Orotidine 5'-Monophosphate Decarboxylase for Catalysis.

The role of a global, substrate-driven, enzyme conformational change in enabling the extraordinarily large rate acceleration for orotidine 5'-monophosphate decarboxylase (OMPDC)-catalyzed decarboxylation of orotidine 5'-monophosphate (OMP) is examined in experiments that focus on the interactions between OMPDC and the ribosyl hydroxyl groups of OMP. The D37 and T100' side chains of OMPDC interact, respectively, with the C-3' and C-2' hydroxyl groups of enzyme-bound OMP. D37G and T100'A substitutions result in 1.4 kcal/mol increases in the activation barrier ΔG⧧ for catalysis of decarboxylation

Metabolomics signatures of acutely ill and short-term weight recovered women with anorexia nervosa.

Although metabolomics studies are recently spreading and have allowed the characterization of putative biomarkers in many diseases, they are relatively scanty in anorexia nervosa (AN). In this explorative study we analyzed the fecal metabolomics profiles of women with AN in the underweight phase (n = 24) and after short-term weight restoration (n = 16) and compared them with 20 healthy women. An untargeted metabolomic procedure allowed the characterization of 224 metabolites involved in energy, lipid, and amino acid metabolism. A partial least square discriminant analysis identified 14 metabol

Dissection of Chemical Composition and Associated Gene Expression in the Pigment-Deficient Tea Cultivar 'Xiaoxueya' Reveals an Albino Phenotype and Metabolite Formation.

The tea cultivar 'Xiaoxueya', a temperature-sensitive albino mutant, is a rare tea germplasm because of its highly enriched amino acid content and brisk flavour. In comparison with green leaf tissues of 'Xiaoxueya', albino leaves show significant deficiency in chlorophylls and carotenoids and severely disrupted chloroplasts. Furthermore, the accumulation of quality-related secondary metabolites is altered in 'Xiaoxueya' albino leaf, with significantly increased contents of total amino acids, theanine, and glutamic acid and significantly decreased contents of alkaloids, catechins, and polypheno

Evaluation and modelling of osmotic pre-treatment of peach using alternative agents in a multiple-component solution.

BACKGROUND: Peach is a perishable fruit of great importance for the Greek food export market which is prone to rapid deterioration after harvesting. Its short shelf life can be substantially prolonged by applying an appropriate osmotic dehydration (OD) pre-treatment, which is a mild, non-thermal procedure. The aim of this study was to describe mass transfer and quality retention induced by the OD process. RESULTS: Peach pieces were immersed in tertiary solutions of glycerol, erythritol, steviol glucosides and mineral salts at pre-selected OD conditions (25-45 °C for 3-240 min, wfruit /wsoluti

Role of FODMAPs in Patients With Irritable Bowel Syndrome.

Irritable bowel syndrome (IBS) is a condition characterized by abdominal pain, bloating, flatus, and altered bowel habits. The role of dietary components in inducing IBS symptoms is difficult to explore. To date, foods are not considered a cause but rather symptom-triggering factors. Particular interest has been given to the so-called FODMAPs (fermentable oligo-, di-, and monosaccharides and polyols). We aimed to summarize the evidence from the most common approaches to manage suspected food intolerance in IBS, with a particular interest in the role of FODMAPs and the effects of a low FODMAP d
