The relative effectiveness of extinction and counter-conditioning in diminishing children's fear.

Two behavioural strategies for reducing learned fear are extinction and counter-conditioning, and in this study we compared the relative effectiveness of the two procedures at diminishing fear in children. Seventy-three children aged 7-12 years old (M = 9.30, SD = 1.62) were exposed to pictures of two novel animals on a computer screen during the fear acquisition phase. One of these animals was paired with a picture of a scared human face (CS+) while the other was not (CS-). The children were then randomly assigned to one of three conditions: counter-conditioning (animal paired with a happy fa

Mild hypothermia can disrupt extinction learning but not original conditioning.

Using a retrograde amnesia procedure, the susceptibility of the extinction of fear conditioning was assessed in two experiments. Extinction of a passive-avoidance task was impaired by a body-cooling treatment (e.g., hypothermia; [7]) which was too mild to induce amnesia for the avoidance training, suggesting that the memory for extinction is more susceptible to body cooling than the memory for the initial fear conditioning. Decreasing the severity of the treatment decreased its ability to disrupt extinction. Thus, the study demonstrates a difference in the vulnerability to amnesia of fear cond

New treatment options for substance abuse from a public health viewpoint.

Naltrexone is the first safe and effective pharmaceutical adjunct for use in the treatment of alcohol abuse. Theoretically it could be effective also as a means for terminating methadone maintenance and in the treatment of other forms of substance abuse. Two general types of protocols have been used with naltrexone. One protocol is similar to the protocol appropriate for use with disulfiram; it is designed to preclude use of the substance while on naltrexone. The other protocol is based on preclinical research showing that opioid antagonists can cause extinction of alcohol drinking; it is desi

Diazepam blocks the interfering effect of post-training behavioral manipulations on retention of a shuttle avoidance task.

Rats were submitted to a training and a test session of shuttle avoidance. Exposure to a session of extinction of this task either 2 or 24 h after training interfered with retention test performance. Exposure to an open field 2, but not 24 h after the avoidance training also interfered with retention. Diazepam blocked the deleterious effect of extinction and of the open field on retention of the avoidance task. Diazepam alone had no effect when given after avoidance training; it did, however, also interfere with retention when given prior to training. It is likely, therefore, that diazepam can

MDMA-assisted psychotherapy for PTSD: Growing evidence for memory effects mediating treatment efficacy.

The application of MDMA in conjunction with psychotherapy has in recent years seen a resurgence of clinical, scientific, and public interest in the treatment of posttraumatic stress disorder (PTSD). Clinical trials have shown promising safety and efficacy, but the mechanisms underlying this treatment form remain largely unestablished. This article explores recent preclinical and clinical evidence suggesting that the treatment's efficacy may be influenced by the mnemonic effects of MDMA. We review data on the effects of MDMA on fear extinction and fear reconsolidation and the utility of these p

Maternal separation facilitates extinction of social fear in adult male mice.

Early life stress, such as child abuse or neglect, is a risk factor for the development of psychopathologies characterized by abnormal social and emotional behaviors. In rodents, long-lasting changes in stress coping and emotional behavior can be induced by separating pups from their mother. We used maternal separation (MS; 3h daily on postnatal days 1-14) to test whether early life stress alters acquisition and extinction of social fear in adult male mice as studied in a specific model of social fear, i.e., in the social fear conditioning paradigm. We show that MS facilitated extinction of so

Sensitive periods in fear learning and memory.

Adolescence represents a uniquely sensitive developmental stage in the transition from childhood to adulthood. During this transition, neuronal circuits are particularly susceptible to modification by experience. In addition, adolescence is a stage in which the incidence of anxiety disorders peaks in humans and over 75% of adults with fear-related disorders met diagnostic criteria as children and adolescents. While postnatal critical periods of plasticity for primary sensory processes, such as in the visual system are well established, less is known about potential critical or sensitive period

The influence of sleep on fear extinction in trauma-related disorders.

In Posttraumatic Stress Disorder (PTSD), fear and anxiety become dysregulated following psychologically traumatic events. Regulation of fear and anxiety involves both high-level cognitive processes such as cognitive reattribution and low-level, partially automatic memory processes such as fear extinction, safety learning and habituation. These latter processes are believed to be deficient in PTSD. While insomnia and nightmares are characteristic symptoms of existing PTSD, abundant recent evidence suggests that sleep disruption prior to and acute sleep disturbance following traumatic events bot

Motivational and discriminative aspects of context in renewal and reinstatement of operant behavior in mice.

Renewal, reinstatement, and other forms of recovery of learned behavior after extinction have been interpreted in terms of the concept of context. Context is defined here as the field of stimuli that is interrelated with the behavior of an individual in the present. Two important classes of stimuli that comprise context are discriminative stimuli and motivating operations. Two experiments examined interactions between discriminative and motivational aspects of context in the renewal and reinstatement of operant behavior following extinction in mice. In Experiment 1, mice received all operant a

Acquisition, generalization and renewal of instrumental pain-related avoidance behavior in high-versus low-anxious individuals.

Excessive avoidance is a key contributor to chronic pain-related disability. In the acute stage, pain-related avoidance prevents further harm, but when it persists beyond healing time it becomes maladaptive. Exposure therapy is the preferred treatment, yet avoidance often returns. Changing contexts (e.g., from therapy to home) may contribute to avoidance returning, referred to as renewal. This study aimed to experimentally investigate the moderating effects of trait anxiety on renewal of fear and avoidance, as well as on fear and avoidance during acquisition, generalization and response preven

Schedule control with a synchronous reinforcement treadmill preparation: A replication and extension.

Pinkston et al. (2024) provided 17 participants with their high-preference music for increasing and decreasing their walking speeds on a treadmill. The results showed that high-preference music produced schedule control of walking speed for 14 of 17 (82.35%) participants. In addition, Pinkston et al. found that 78.57% of participants whose walking showed schedule control also displayed variable responding during an extinction component. As an extension of Pinkston et al., we presented 25 participants with their high-preference music for walking on a treadmill during a five-component mixed sche

The role of avoidance in modulating single prolonged stress effects on emotional memory in male and female rats.

The incidence of post traumatic stress disorder (PTSD) is greater in women than men, but mechanisms via which this difference manifests remain under explored. The single prolonged stress (SPS) rodent model of traumatic stress has been used to identify mechanisms through which traumatic stress leads to deficits in retaining extinction (a core PTSD symptom), but has been mostly utilized in male model systems. Recent studies have observed that SPS leads to changes in persistent fear memory in female rats, though these results are variable. This variability could be driven by changes in behavioral

Recruitment of neurons in basolateral amygdala after intense training produces a stronger memory trace.

Typical amnestic treatments are ineffective when administered to subjects trained in aversively-motivated tasks using relatively high foot-shock intensities. This effect has been found when treatments that disrupt neuronal activity are administered to different regions of the brain, including the amygdala. However, the molecular mechanisms induced by this intense training are unknown. We made a detailed mapping of c-Fos-expressing neurons in four regions of the amygdala after moderate and intense one-trial inhibitory avoidance training. Rats were sacrificed 90 min after training or after appro

Conditioned contribution of peripheral cocaine actions to cocaine reward and cocaine-seeking.

Cocaine has actions in the peripheral nervous system that reliably precede--and thus predict--its soon-to-follow central rewarding effects. In cocaine-experienced animals, the peripheral cocaine signal is relayed to the central nervous system, triggering excitatory input to the ventral tegmental origin of the mesocorticolimbic dopamine system, the system that mediates the rewarding effects of the drug. We used cocaine methiodide, a cocaine analog that does not cross the blood-brain barrier, to isolate the peripheral actions of cocaine and determine their central and behavioral effects in anima

Further evaluation of response interruption and redirection as treatment for stereotypy.

The effects of 2 forms of response interruption and redirection (RIRD)-motor RIRD and vocal RIRD-were examined with 4 boys with autism to evaluate further the effects of this intervention and its potential underlying mechanisms. In Experiment 1, the effects of motor RIRD and vocal RIRD on vocal stereotypy and appropriate vocalizations were compared for 2 participants. In Experiment 2, the effects of both RIRD procedures on both vocal and motor stereotypy and appropriate vocalizations were compared with 2 additional participants. Results suggested that RIRD was effective regardless of the proce

Neuronal correlates of extinction learning are modulated by sex hormones.

In emotional learning tasks, sex differences, stress effects and an interaction of these two moderators have often been observed. The sex hormones estradiol (E2) and progesterone (P4) vary over the menstrual cycle. We tested groups with different sex hormone status: 39 men, 30 women in the luteal phase (LU, high E2+P4) and 29 women taking oral contraceptives (OC, low E2+P4). They received either 30 mg cortisol or placebo prior to instructed differential fear conditioning consisting of neutral conditioned stimuli (CS) and an electrical stimulation (unconditioned stimulus; UCS). One figure (CS+)

Reducing chronic anxiety by making the threatening event predictable: an experimental approach.

Panic disorder is characterized by both specific, phased fear and generalized, chronic anxiety. Standard extinction procedures are efficient in reducing specific fear. However, methods based on human conditioning research - that are capable of reducing chronic anxiety have not yet been thoroughly investigated. This study evaluates a new way of reducing chronic anxiety by signaling aversive events (or by making them more predictable). Using an experimental approach with healthy participants, specific fear and chronic anxiety were operationalized in a within-subjects fear-potentiated startle par

Trait mindfulness reduces the generalization of conditioned fear: a preliminary pilot study.

Trait mindfulness refers to an individual’s inherent capacity to maintain sustained awareness and focus on present experiences. This characteristic is naturally present in the population, even without intentional mindfulness training, and is significantly negatively correlated with negative emotions, anxiety, and depression. Fear is an adaptive response to threat, but excessive generalization of fear is a key underlying factor in anxiety disorders. Fear generalization occurs when an individual, having learned to fear a specific threat, extends this fear response to stimuli that share similarit
