A transferrin-responsive fluorescent probe based on Zn-doped carbon dots for selective Fe3+ detection in serum.

BACKGROUND: The detection of Fe3+ and Cu2+ in biological fluids is crucial for clinical diagnostics, yet remains challenging due to severe matrix interference in complex environments like serum. Current methods often lack real-time capability and struggle with selectivity, while fluorescent probes are frequently compromised by unpredictable biomolecular interactions. There is a pressing need for robust sensors that can overcome these physiological barriers. This study addresses this gap by developing a dual-functional probe designed to maintain reliable performance within biological systems, e

Visualization of endogenous hypochlorous acid during ferroptosis based on Rhodamine B fluorescent probe.

Hypochlorous acid (HClO) plays critical roles in ferroptosis process and the specific molecular mechanism related to it is still obscure. To reveal HClO fluctuation in the ferroptosis cells, a fluorescence turn-on probe RDM-577 that can detect intracellular HClO was skillfully engineered. The probe exhibited a substantial fluorescence enhancement in response to HClO (F/F0 = 35), enabling sensitive detection of HClO in both in vitro and in vivo environments. Additionally, it demonstrated remarkable performance characteristics, such as an exceptionally low detection limit (5.8 nM). Consequently,

NADH-Activated Near-Infrared Fluorescent Probe for Efficient Screening of ADH/ALDH2 Dual Agonists for Alcohol Detoxification.

Alcohol-induced liver injury remains a major global health concern, while effective strategies that facilitate alcohol metabolism are still limited. Alcohol dehydrogenase (ADH) and aldehyde dehydrogenase 2 (ALDH2) play central roles in hepatic ethanol metabolism, their catalytic reactions are stoichiometrically coupled to nicotinamide adenine dinucleotide (NADH) production. However, many existing NADH probes suffer from short excitation/emission wavelengths, high background interference, and limited dynamic ranges, restricting their application in enzyme-related screening. Herein, we report a

Construction of viscosity-sensitive RNA fluorescent probes and their application in inflammtation imaging.

Abnormal structure or function of the nucleolus can lead to dynamic imbalances in intracellular viscosity, which is a core pathological feature of major diseases, including inflammation and cancer. However, existing fluorescent probes face challenges in synchronously and accurately detecting nucleolar RNA dynamics and viscosity changes. Therefore, in this study, probes NK-1 and NK-2, which specifically target the nucleolus, were developed and synthesized. Both probes incorporate a donor-π-acceptor (D-π-A) structure with a positively charged quinolinium salt as the core, enabling specific RNA r

Photocaged Fluorogenic Glycoprobes for Light-Activatable Sensing and Imaging of Galactosidase Activities in Live Cells.

The accurate sensing and imaging of enzymatic activities in live cells is key to exploring their intrinsic biological and pathological functions. While previously developed molecular probes are directly activated in the presence of a target enzyme, here, we developed a photocaging strategy for enzymatic substrates to enable tandem light-activatable, fluorogenic enzyme recognition as an approach with improved sensing fidelity. Galactosidases were used as the target enzyme in a proof-of-concept approach. We synthesized α- and β-galactose-conjugated, rhodol-based glycoprobes, in which the C6-posi

The development of functional organic molecules for advanced biological and environmental applications is of growing interest, particularly for metal ion sensing and therapeutic delivery. Rhodamine-based fluorescent compounds have been widely studied due to their well-defined absorption and emission properties. In this study, a rhodamine-appended fluorescent probe (RhB-A) was designed and synthesized, exhibiting strong fluorescence upon excitation at 400 nm. The probe demonstrates exceptional selectivity for Fe3+ ions, with significant fluorescence quenching even in the presence of 16 competin

Uracil-Peptide-Pyrene Conjugates as Cu2+-Sensitive Probes Targeting DNA, RNA, and Biorelevant Model Membranes.

We report a series of uracil-triazole-pyrene peptidomimetics (2-4) designed to achieve modular geometric control through peptide linkers of varying length and flexibility. All three compounds exhibited submicromolar affinity for ds-DNA, while compound 4 also showed strong binding to ds-RNA, demonstrating the advantage of combining a uracil recognition unit with an extended pyrene aromatic surface compared to the Pyr-Trp reference ligand and the phenanthridine analog 4'. Structural variations strongly affected Cu(II) coordination. Although Pyr-Trp bound Cu(II) approximately 1000-fold more stron

An Organelle-Orthogonal Dual-Response Probe for In Situ Monitoring of Mitochondrial H2O2 and Lipid Droplet Polarity in Ferroptosis.

Ferroptosis, an iron-dependent form of regulated cell death, is characterized by organelle dysfunction and disrupted communication due to reactive oxygen species (ROS). A major obstacle has been the lack of methods for real-time, simultaneous monitoring of key organellar parameters, which has kept the spatiotemporal role of H2O2 in organelle interactions unclear. Here, we designed a dual-function near-infrared fluorescent probe, PSi-NXZD/HCy-H2O2, that enables synchronous, dual-channel tracking of lipid droplet polarity and mitochondrial H2O2 in living cells. Application of this probe unveiled

Unveiling Pathological Lipid Droplet Accumulation of Dopaminergic Neurons in Parkinson's Disease via a Super-Retentive Fluorescent Probe.

Lipid droplets are highly dynamic organelles in nerve cells and are essential for the function of the central nervous system. The abnormal accumulation of lipid droplets in dopaminergic neurons, found in Parkinson's disease (PD) cells and animal models, presents as a promising target for PD diagnosis. However, previously reported fluorescent probes are unsuitable for in vivo long-term tracking of lipid droplets, limiting their applications in animal models of PD. Here, we developed three lipid droplet-targeted probes, named s-CBTA, b-CBTA, and LD-b-PBTA, which share similar chemical structures

Rational design of environment-sensitive fluorescent probes for butyrylcholinesterase and their application in biological imaging.

Butyrylcholinesterase (BChE) is closely associated with Alzheimer's disease (AD), with its expression significantly elevated in the brains of AD patients. This enzyme has emerged as a potential biomarker for monitoring AD progression. Therefore, developing a reliable chemical tool to detect BChE both in vitro and in vivo is of considerable interest. In this study, we rationally designed a series of environmentally sensitive fluorescent probes targeting BChE, based on a highly potent, selective, and reversible BChE inhibitor. Among these, the most promising probe ESP4 demonstrated an ultrafast

In situ NIR-IIb imaging of endogenous H2S signaling for high-resolution abiotic stress visualization in plants.

In vivo analysis of plant stress responses remains a major challenge in precision agriculture, limiting dynamic optimization of crop growth under variable environmental conditions. Fluorescence imaging enables nondestructive tracking of stress biomarkers, but its accuracy is compromised by the low abundance of endogenous signaling molecules, tissue autofluorescence interference, and limited signal penetration depth. Here, we develop a ratiometric near-infrared IIb fluorescent probe for sensitive monitoring of endogenous hydrogen sulfide (H2S). This nanoprobe integrates lanthanide nanoparticles

Nonconjugated Alkyl Chains in Ionic Molecular Rotor Fluorophores: An Effective Modulating Moiety for Amyloid-β Probes.

Developing molecular structural tuning strategies for ionic molecular rotor (MR)-based β-amyloid (Aβ) fluorescent probes is essential for balancing high sensitivity, signal-to-background ratio (SBR) in wash-free mode, and blood-brain barrier (BBB) permeability. Compared with the widely used adjustment strategy on the electron donor, π-bridge, and electron acceptor, the nonconjugated hydrophobic part that can directly affect the lipophilicity and dispersion of the whole ionic MRs has been omitted. Herein, we developed a series of diarylbutadiene ionic MRs with variable alkyl chains substituted

Spatiotemporal analysis of reactive oxygen species using specific activity-based sensing fluorescent imaging probes.

Oxidative stress, driven by reactive oxygen species (ROS), is implicated in numerous diseases. ROS are vital for redox signalling and homeostasis but can damage proteins, lipids and nucleic acids, compromising cellular health and contributing to disease progression. ROS functions are governed by their spatiotemporal distribution within cells and organisms; however, their mechanisms of action remain insufficiently understood, highlighting the compelling need for accurate ROS detection methods. In this context, fluorescence imaging has emerged as a powerful tool for real-time ROS detection. Effe

Dual-channel fluorescent probes enable synchronous tracking of peroxynitrite and cysteine in mitochondrial redox dynamics.

Peroxynitrite (ONOO-) and cysteine (Cys) are pivotal components of reactive nitrogen and sulfur species that together shape a complex redox regulatory network in living systems. High-resolution, real-time imaging of these species is essential for unraveling their roles in mitochondrial redox dynamics. However, the development of fluorescent probes capable of simultaneously detecting ONOO- and Cys with high fidelity remains limited. In this work, we introduce a single-excitation, dual-response fluorescent probe system (probes A and B) that enables synchronous visualization of ONOO- and Cys. The

Interface-Stabilized Logic Nanoprobe Enables High-Fidelity Orthogonal Sensing of MMP-1 and HNE in Thiol-Rich Biological Environments.

Reliable multiplexed sensing in thiol-rich biological environments remains challenging because nonspecific ligand displacement, background leakage, and partial channel corruption can compromise the analytical meaning of multichannel fluorescence outputs. Here, we report an interface-stabilized logic-gated fluorescent nanoprobe for high-fidelity orthogonal sensing of matrix metalloproteinase-1 (MMP-1) and human neutrophil elastase (HNE). Two fluorogenic peptide substrates were co-immobilized on gold nanoparticles through an Au-C≡C nano-biointerface to generate independent FITC and 5-TAMRA fluor

From solvent-tailored carbon dots to a practical sensor array: fingerprint discrimination and quantification of azo dyes in beverages.

Azo dyes are widely used as synthetic food colorants, but non-compliant use poses health risks, necessitating rapid detection methods. Here, dual-emissive bromine-doped carbon dots (Br-CDs) were synthesized via solvent engineering by tuning the ethanol/water ratio, which precisely modulates their surface states and differential affinity toward azo dyes. Based on these Br-CDs, a dual-channel fluorescence sensor array was developed for discrimination and detection of azo dyes. This array achieves fingerprint-like discrimination of five common azo dyes (visual detection limit: 2 μM) and accuratel
