The principal capsular polysaccharide of the opportunistic fungal pathogen Cryptococcus neoformans consists of an alpha-1,3-linked mannose backbone decorated with a repeating pattern of glucuronyl and xylosyl side groups. This structure is critical for virulence, yet little is known about how the polymer, called glucuronoxylomannan (GXM), is faithfully synthesized and assembled. We have generated deletions in two genes encoding predicted parallel beta-helix repeat proteins, which we have designated PBX1 and PBX2. Deletion of either gene results in a dry-colony morphology, clumpy cells, and dec

Fungal morphogenesis is a major driver of disease outcome. For the opportunistic fungal pathogen Cryptococcus neoformans, extreme morphological heterogeneity within the lung environment drives dissemination, immune evasion, and drug resistance. One such cell type is the small, oval, titanide (2-4 µm). This poorly understood cell type is prevalent in lung histology as well as host-relevant in vitro conditions. In conditions that generate cellular heterogeneity, titanides comprise 4%-10% of cells 24 hours post-induction and by 72 hours are the dominant cell type. Despite their prevalence and the

BACKGROUND AND IMPORTANCE: Cerebral phaeohyphomycosis caused by Cladophialophora bantiana carries a high mortality rate of approximately 70%. Prompt, complete resection is associated with improved patient outcomes, with surgeons typically relying on imaging for appropriate margins of resection. 5-aminolevulinic acid (5-ALA) has traditionally been used in patients with gliomas; however, when used in our case of a patient with C. bantiana mimicking glioma, 5-ALA illuminated the capsule, allowing for optimal excision.

AIMS: The SAGA complex is a conserved transcriptional co-activator essential for eukaryotic gene regulation. In fungi of the Ascomycota, the core protein Spt20 contributes to the structure and function of SAGA. This study aimed to identify and characterize SPT20 in Cryptococcus neoformans, the WHO top-ranked critical priority group species on their Fungal Priority Pathogen list.
MATERIALS AND METHODS: Identification of C. neoformans SPT20 revealed the presence of a tRNA gene within its 5' UTR. Precisely deleting the SPT20 ORF preserved the tRNA gene while enabling analysis of Spt20 function.

Cryptococcus neoformans is an encapsulated opportunistic fungal pathogen that is found in multiple niches in the environment and that can cause fatal meningoencephalitis in susceptible patients, mainly HIV+ individuals. Cryptococcus also infects environmental hosts such as nematodes, insects and plants. In particular, C. neoformans can kill the lepidopteran Galleria mellonella, which offers a useful tool to study microbial virulence and drug efficacy. Galleria mellonella immunity relies on innate responses based on melanization, accumulation of antimicrobial peptides, and cellular responses as

An understanding of the connections between metabolism and elaboration of virulence factors during host colonization by the human-pathogenic fungus Cryptococcus neoformans is important for developing antifungal therapies. Lipids are abundant in host tissues, and fungal pathogens in the phylum basidiomycota possess both peroxisomal and mitochondrial β-oxidation pathways to utilize this potential carbon source. In addition, lipids are important signaling molecules in both fungi and mammals. In this report, we demonstrate that defects in the peroxisomal and mitochondrial β-oxidation pathways infl

BACKGROUND: Recurrent vulvovaginal candidiasis affects nearly 138 million women globally each year. In the United States, fluconazole is considered the standard of care for acute vulvovaginal candidiasis, but until recently there was no US Food and Drug Administration-approved drug for the treatment of recurrent vulvovaginal candidiasis. Oteseconazole is a novel oral selective inhibitor of fungal lanosterol demethylase (sterol 14α-demethylase cytochrome P450, an enzyme required for fungal growth) approved for the treatment of recurrent vulvovaginal candidiasis.

Two patients developed renal mucormycosis following transplantation of kidneys from the same donor, a near-drowning victim in a motor vehicle crash. Genotypically, indistinguishable strains of Apophysomyces elegans were recovered from both recipients. We investigated the source of the infection including review of medical records, environmental sampling at possible locations of contamination and query for additional cases at other centers. Histopathology of the explanted kidneys revealed extensive vascular invasion by aseptate, fungal hyphae with relative sparing of the renal capsules suggesti

This study evaluated the synergistic interactions between amphotericin B (AMB) and azithromycin (AZM), daptomycin (DAP), linezolid (LNZ), minocycline (MINO), fluconazole (FLZ), flucytosine (5FC), linezolid (LZD), or tigecycline (TIG) against clinical isolates of Cryptococcus neoformans var. grubii before and after capsule induction. High synergism (>75%) was observed for the combinations, AMB+5FC, AMB+TIG, AMB+AZM, AMB+LZD and AMB+MINO but only in the strains after capsule induction. The results show that the presence of the capsule may lower the minimum inhibitory concentrations (MICs) of ant

Cryptococcus neoformans, a prominent opportunistic pathogen, is equipped with unique mechanisms to evade host immune defenses, notably through its capsule and the secretion of extracellular vesicles (EVs). Despite significant understanding of its pathogenesis, the precise role of EVs in virulence and their molecular components remain underexplored. This review synthesizes current research on the virulence factors encapsulated within EVs of Cryptococcus, highlighting their contribution to fungal survival and pathogenicity. By analyzing the biochemical composition of these vesicles, we found the

Monocytes are considered to play a central role in the pathogenesis of Cryptococcus neoformans infection. Monocytes and monocyte-derived macrophages and dendritic cells are key components for the control of infection, but paradoxically they can also contribute to detrimental host responses and may even support fungal proliferation and dissemination. Simultaneously, the C. neoformans polysaccharide capsule can impair the functions of monocytes. Although monocytes are often seen as simple precursor cells, they also function as independent immune effector cells. In this review, we summarize these

The fungal Zds proteins are regulators of the serine/threonine phosphatase 2A (PP2A) and the protein kinase A. Here, we characterize a Zds-like gene ZDS3 that plays a broad range of roles in the basidiomycetous pathogenic yeast Cryptococcus neoformans. ZDS3 harbors the conserved activation domain ZDS_C of Zds proteins. By gene disruption, ZDS3 is shown to play roles in capsule production, cell wall integrity, growth at a high temperature, resistance to H(2)O(2) stress, osmotic pressures and glucose-dependent invasive growth on the agar. As a consequence, the disruption of ZDS3 resulted in comp

Cryptococcosis, caused by Cryptococcus neoformans, is the most common opportunistic fungal disease in HIV/AIDS patients. The prognosis of AIDS patients with Cryptococcus infection is very poor. One of the major characteristics in cryptococcosis patients is the presence of high concentrations of the cryptococcal capsule polysaccharide (CCP) in the serum and cerebrospinal fluid. CCP enhances HIV replication in H9 T-cells, but the mechanism is unknown. In this study, we tested whether extracellular glucuronoxylomannan (GXM), a major component of CCP, enhances HIV entry using replication-incompete

A 73-year-old woman from Guyana had nodular keloidal lesions on her face which had persisted for 56 years. A biopsy of the lesion revealed lobomycosis, a rare fungal infection. Microscopically, numerous fungi were seen in giant cells and macrophages. The macrophages had abundant pink reticulated cytoplasm. These macrophages in lobomycosis, considered by some in the past to be granular myoblastoma cells, were rich in glycoproteins and appeared to be "Gaucher-like cells" in electronmicrographs. Further, a transition of macrophages containing organisms to Gaucher-like cells was noted. Antibodies

Invasive fungal infections are emerging as a global public health problem. The lack of effective antifungal drugs is the bottleneck of clinical antifungal treatment. To identify novel antifungal agents with new mechanisms of action, JIB-04, a Jumonji histone demethylase inhibitor, was identified to possess broad-spectrum antifungal activity by a cell-based screen. Particularly, JIB-04 effectively inhibited Jumonji demethylase activity and ergosterol biosynthesis of Cryptococcus neoformans cells, leading to in vitro and in vivo anti-Cryptococcus activity. It also significantly inhibited the vir

UNLABELLED: Cryptococcus is a genus of saprophytic fungi with global distribution. Two species complexes, Cryptococcus neoformans and Cryptococcus gattii, pose health risks to humans and animals. Cryptococcal infections result from inhalation of aerosolized spores and/or desiccated yeasts from terrestrial reservoirs such as soil and trees. More recently, C. gattii has been implicated in infections in marine mammals, suggesting that inhalation of cells from the air-water interface is also an important, yet understudied, mode of respiratory exposure. Based on historical records and epidemiologic

Cryptococcus neoformans (C. neoformans) infections pose a critical threat to global health, with high mortality rates observed among immunocompromised individuals. Rapid, cost-effective diagnostics are essential for preventive treatment and reducing mortality in resource-scarce settings, yet the development of selective fluorescent probes for C. neoformans remains challenging. The Prp8 intein, a unique self-splicing protein in C. neoformans, represents an ideal target for developing fluorescent probes and antifungal drugs. In this study, a series of fluorescent probes integrating Prp8 intein i
