Perturbation of azurophilic granule integrity drives NLRP3-independent IL-1β processing and release in neutrophils.

Interleukin 1-beta (IL-1β) is an inflammatory cytokine produced by myeloid cells in response to infection or sterile tissue damage. Secretion of bioactive IL-1β from macrophages (Mφ) or dendritic cells (DC) downstream of activated NLRP3/caspase-1 inflammasomes is the best characterized model; this is mediated by caspase-1 cleavage of proIL-1β and Gasdermin D. Gasdermin D pores that form in the plasma membrane mediate IL-1β release and pyroptotic cell death. NLRP3 inflammasome assembly is triggered by perturbation of ionic, metabolic or organelle homeostasis via diverse stimuli. A recent report

S-phase targeted treatment triggers caspase-dependent lytic immunogenic cell death with pyroptotic features in cancers.

Immunogenic cell death (ICD) is a type of cell death that can enhance anti-tumour immune responses of chemotherapies and targeted therapies by releasing DAMPs and cytokines that activate dendritic cells and T cells, thereby engaging the patient's immune system to combat the cancer. Pyroptosis and necroptosis are strongly immunogenic because they release DAMPs and inflammatory signals through pore-forming proteins, whereas apoptosis can be tolerogenic. This immunogenic response is contingent on a functional immune system. Unfortunately, most conventional chemotherapies and many targeted therapi

Alpha-1 Antitrypsin Mediates Spontaneous Resolution of Acute Gouty Inflammation Via Inhibiting Caspase 3/GSDME-dependent Macrophage Pyroptosis.

Research on spontaneous resolution of acute gout remains limited. Macrophages pyroptosis is crucial for the inflammation of acute gout, while current research mainly focus on Caspase 1/Gasdermin D axis. We aimed to investigate the involvement of other Gasdermin proteins in MSU crystal-induced macrophages, and to explore the role of Caspase 3-interacting protein alpha-1 antitrypsin (AAT) in regulating macrophage pyroptosis. Here, clinical evidence demonstrated elevated Gasdermin E (GSDME) in peripheral blood mononuclear cells (PBMCs) and CD68+ synovial macrophages from patients with acute gout.

Molecular Regulation of Pyroptosis in Alzheimer's Disease: Linking Neuroinflammation, Cell Death, and Therapeutic Targeting.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by profound cognitive decline, wherein chronic neuroinflammation plays a pivotal pathogenic role. Central to this inflammatory milieu is pyroptosis, a highly inflammatory form of programmed lytic cell death mediated by gasdermin proteins. This comprehensive review provides an in-depth synthesis of the cellular and molecular mechanisms underlying pyroptosis in AD. We detail the distinct roles of microglia as primary initiators responding to amyloid-beta (Aβ) and tau aggregates, alongside the specific vulnerabilit

Pyroptosis in glioma: Current management and future application.

Glioma, the predominant form of central nervous system (CNS) malignancies, presents a significant challenge due to its high prevalence and low 5-year survival rate. The efficacy of current treatment methods is limited by the presence of the blood-brain barrier, the immunosuppressive microenvironment, and other factors. Immunotherapy has emerged as a promising approach, as it can overcome the blood-brain barrier. A tumor's immune privilege, which is induced by an immunosuppressive environment, constricts immunotherapy's clinical impact in glioma. Pyroptosis, a programmed cell death mechanism fa

Pyroptosis: A New Insight Into Eye Disease Therapy.

Pyroptosis is a lytic form of programmed cell death mediated by gasdermins (GSDMs) with pore-forming activity in response to certain exogenous and endogenous stimuli. The inflammasomes are intracellular multiprotein complexes consisting of pattern recognition receptors, an adaptor protein ASC (apoptosis speck-like protein), and caspase-1 and cause autocatalytic activation of caspase-1, which cleaves gasdermin D (GSDMD), inducing pyroptosis accompanied by cytokine release. In recent years, the pathogenic roles of inflammasomes and pyroptosis in multiple eye diseases, including keratitis, dry ey

Targeting Pyroptosis in Oral and Maxillofacial Diseases.

The oral and maxillofacial region is a complex microenvironment continuously exposed to microbial challenge, mechanical stress, and oncogenic stimuli. Accumulating human evidence firmly implicates pyroptosis in the pathogenesis of major oral and maxillofacial diseases, with cleaved gasdermin D (GSDMD)/gasdermin E (GSDME) detected in clinical specimens of periodontitis, peri-implantitis, apical periodontitis, oral lichen planus, and oral submucous fibrosis. The central mechanism involves inflammasome assembly (eg, NLRP3), leading to caspase activation, gasdermin cleavage (eg, GSDMD, GSDME), and

Bacterial detection by NAIP/NLRC4 elicits prompt contractions of intestinal epithelial cell layers.

The gut epithelium serves to maximize the surface for nutrient and fluid uptake, but at the same time must provide a tight barrier to pathogens and remove damaged intestinal epithelial cells (IECs) without jeopardizing barrier integrity. How the epithelium coordinates these tasks remains a question of significant interest. We used imaging and an optical flow analysis pipeline to study the dynamicity of untransformed murine and human intestinal epithelia, cultured atop flexible hydrogel supports. Infection with the pathogen Salmonella Typhimurium (STm) within minutes elicited focal contractions

Clinical and pathophysiological characteristics of non-acute decompensation of cirrhosis.

BACKGROUND & AIMS: The heterogenous presentation patterns in decompensated cirrhosis confer variable outcomes. While acute decompensation (AD) is well-characterized, the presentation patterns and outcomes of non-acute decompensation (NAD) remain unclear. The aim of this study was to characterize clinical and pathophysiological features of NAD and identify predictors of progression in NAD. METHODS: In this prospective study, patients across the cirrhosis spectrum were enrolled from two centers in India between 2020-2023: compensated cirrhosis (CC; n = 29), NAD (n = 311), AD (n = 201), and heal

GSDMD and GSDME exhibit distinct roles in enteric coronavirus PDCoV-induced pyroptosis and inflammatory responses.

Porcine deltacoronavirus (PDCoV), an emerging enteric coronavirus with zoonotic potential, typically causes intestinal villous epithelial cell damage with inflammation. Pyroptosis is a recently identified inflammatory form of programmed cell death that has been found to be associated with the pathogenesis of many viruses. However, the effects of PDCoV infection on pyroptosis and the role of pyroptosis in its pathogenesis remain unclear. In this study, we report that PDCoV infection triggers pyroptosis, as demonstrated in porcine ileum epithelial cell lines and intestinal tissues of PDCoV-infec

Compound loss of GSDMD and GSDME function is necessary to achieve maximal therapeutic effect in colitis.

Gasdermin D (GSDMD) and gasdermin E (GSDME) perpetuate inflammation by mediating the release of cytokines such as interleukin-1β (IL-1β) and IL-18. However, not only are the actions of GSDMD in colitis still controversial, but its interplay with GSDME in the pathogenesis of this disease has not been investigated. We sought to fill these knowledge gaps using the dextran sodium sulfate (DSS) experimental mouse colitis model. DSS ingestion by wild-type mice caused body weight loss as the result of severe gut inflammation, outcomes that were significantly attenuated in Gsdmd -/- or Gsdme -/- mice

Serum Biomarkers of a Pro-Neuroinflammatory State May Define the Pre-Operative Risk for Postoperative Delirium in Spine Surgery.

Advances in spine surgery enable technically safe interventions in older patients with disabling spine disease, yet postoperative delirium (POD) poses a serious risk for postoperative recovery. This study investigates biomarkers of pro-neuroinflammatory states that may help objectively define the pre-operative risk for POD. This study enrolled patients aged ≥60 scheduled for elective spine surgery under general anesthesia. Biomarkers for a pro-neuroinflammatory state included S100 calcium-binding protein β (S100β), brain-derived neurotrophic factor (BDNF), Gasdermin D, and the soluble ectodoma

Inflammasome Activation Induces Pyroptosis in the Retina Exposed to Ocular Hypertension Injury.

Mechanical stress and hypoxia during episodes of ocular hypertension (OHT) trigger glial activation and neuroinflammation in the retina. Glial activation and release of pro-inflammatory cytokines TNFα and IL-1β, complement, and other danger factors was shown to facilitate injury and loss of retinal ganglion cells (RGCs) that send visual information to the brain. However, cellular events linking neuroinflammation and neurotoxicity remain poorly characterized. Several pro-inflammatory and danger signaling pathways, including P2X7 receptors and Pannexin1 (Panx1) channels, are known to activate in

The dual role of pyroptosis in prostate cancer: new perspectives from molecular mechanisms to clinical translation.

Pyroptosis, a gasdermin-mediated form of programmed inflammatory cell death, plays a complex and context-dependent dual role in the initiation, progression, and therapeutic response of prostate cancer. This review methodically elucidates canonical and non-canonical pyroptotic molecular pathways, emphasizing their dysregulation in prostate cancer. On one hand, the epigenetic silencing of key effector proteins, such as Gasdermin E (GSDME), weakens pyroptosis’s tumor-suppressive and immune-activating functions, thereby promoting therapeutic resistance. On the other hand, sustained pyroptosis-asso

Targeting methionine-induced pyroptosis enhances anti-tumor immunity.

Pyroptosis is an inflammatory form of programmed cell death that plays a critical role in tumor immunity. However, the metabolic signals involved in this process remain undefined. In the present study, we uncover that methionine metabolism plays an important role in the pyroptosis of bone marrow-derived macrophages (BMDMs). The depletion of methionine adenosyltransferase 2A (Mat2a) in macrophages leads to cell death by activating GSDME, resulting in suppression of tumor growth in vivo. Furthermore, pharmacological inhibition of MAT2A induces macrophage pyroptosis by activating Gasdermin E (GSD

Pyroptosis: the dawn of a new era in endometrial cancer treatment.

Endometrial cancer (EC) is a malignancy of the inner epithelial lining of the uterus. While early-stage EC is often curable through surgery, the management of advanced, recurrent and metastatic EC poses significant challenges and is associated with a poor prognosis. Pyroptosis, an emerging form of programmed cell death, is characterized by the cleavage of gasdermin proteins, inducing the formation of extensive gasdermin pores in the cell membrane and the leakage of interleukin-1β (IL-1β) and interleukin-18 (IL-18), consequently causing cell swelling, lysis and death. It has been found to be im

Inflammasome activation and pyroptosis mediate coagulopathy and inflammation in Salmonella systemic infection.

Inflammasome activation is a critical defense mechanism against bacterial infection. Previous studies suggest that inflammasome activation protects against Salmonella oral infection. Here we find inflammasome activation plays a critical role in the pathogenesis of Salmonella systemic infection. We show that in a systemic infection model by i.p. injection of Salmonella, deficiency of caspase-1 or gasdermin-D prolonged survival time, reduced plasma concentrations of the proinflammatory cytokines IL-1β, IL-6 and TNFα. These deficiencies also protected against coagulopathy during Salmonella infect

Role of Caspase 1 in Ischemia/Reperfusion Injury of the Myocardium.

Acute occlusion of a coronary artery can result in myocardial infarction-a leading cause of premature death. Prompt restoration of blood flow to the myocardium can prevent excessive death of cardiomyocytes and improve clinical outcome. Although the major mechanism of cell death after reperfusion is necrosis, it is now recognized that many other cell death pathways may be involved in ischemia-reperfusion (I/R) injury. Pyroptosis is one such cell death pathway that is caspase-1-dependent and induced in response to cellular insult. The activated caspase-1 protease cleaves and activates specific c
