Efficacy and mechanism of action of the tyrosine kinase inhibitors gefitinib, lapatinib and neratinib in the treatment of HER2-positive breast cancer: preclinical and clinical evidence.

An increasing number of tumors, including breast cancer, overexpress proteins of the epidermal growth factor receptor (EGFR) family. The interaction between family members activates signaling pathways that promote tumor progression and resistance to treatment. Human epidermal growth factor receptor type II (HER2) positive breast cancer represents a clinical challenge for current therapy. It has motivated the development of novel and more effective therapeutic EGFR family target drugs, such as tyrosine kinase inhibitors (TKIs). This review focuses on the effects of three TKIs mostly studied in

Prognostic role of annexin A2 and cancer-associated fibroblasts in advanced non-small cell lung cancer: Implication in epithelial-mesenchymal transition and gefitinib resistance.

BACKGROUND: Despite advances in treatment of non-small cell lung cancer (NSCLC), its prognosis remains dismal. Development of drug resistance is a major obstacle against success of targeted epidermal growth factor receptor (EGFR) -tyrosine kinase inhibitors (TKI) therapy. This study aimed to assess the prognostic role of annexin A2 (ANXA2) expression, within both tumor cells and stroma, as well as cancer associated fibroblasts (CAFs) in NSCLC and to investigate their potential role in induction of epithelial mesenchymal transition (EMT) and resistance to gefitinib. METHOD: Immunohistochemistr

Gefitinib response after acquisition of C797S following first-line osimertinib in EGFR L858R/E709K-mutated non-small cell lung cancer: A case report.

Osimertinib is the standard first-line therapy for epidermal growth factor receptor (EGFR)-mutated non-small cell lung cancer, but acquired resistance remains a major clinical issue. Here, we report a case of lung adenocarcinoma harboring compound EGFR L858R and E709K mutations that acquired C797S after progression on first-line osimertinib. Gefitinib achieved a durable partial response lasting approximately 15 months, despite E709K generally conferring reduced sensitivity to first-generation EGFR-tyrosine kinase inhibitors. This case highlights the importance of repeat molecular profiling and

Dual targeting of EGFR signaling and microtubule dynamics enhances replication-associated cellular stress and suppresses tumor growth in gefitinib-resistant non-small cell lung cancer.

Acquired resistance to epidermal growth factor receptor (EGFR)-targeted therapy remains a major obstacle in the treatment of EGFR-mutant non-small cell lung cancer (NSCLC). This study investigated whether repurposing mebendazole (MBZ), a microtubule-disrupting agent, could restore sensitivity to gefitinib in gefitinib-resistant HCC827 GR5 cells. Gefitinib monotherapy showed minimal cytotoxicity, reducing cell viability only marginally (92.7 ± 1.5%), whereas MBZ significantly decreased viability to 79.7 ± 0.6% at 0.1 µM and 29.7 ± 0.6% at 0.5 µM (p < 0.001). A novel EGFR inhibitor suppresses survivin expression and tumor growth in human gefitinib-resistant EGFR-wild type and -T790M non-small cell lung cancer.

Tyrosine kinase inhibitors of epidermal growth factor receptor (EGFR-TKIs) are currently used therapy for non-small cell lung cancer (NSCLC) patients; however, drug resistance during cancer treatment is a critical problem. Survivin is an anti-apoptosis protein, which promotes cell proliferation and tumor growth that highly expressed in various human cancers. Here, we show a novel synthetic compound derived from gefitinib, do-decyl-4-(4-(3-(4-(3-chloro-4-fluorophenylamino)-7-methoxyquinazolin-6-yloxy)propyl) piper-azin-1-yl)-4-oxobutanoate, which is named as SP101 that inhibits survivin express
