Gene therapy: how to target the kidney. Promises and pitfalls.

The success of gene therapy strongly depends on an efficient delivery system to allow local transfer and expression of the therapeutic gene in the target organ or tissue. Vector systems have been improved and many show promise. There are two different categories of delivery vehicles: non-viral and viral vectors, both with advantages and disadvantages that must be taken into consideration in view of the final aim. Compared to other solid organs, the kidney offers the main advantage of access by different routes that dictate different sites of transfection. Thus, the choice of the delivery vehic

A screen for targets of the Xenopus T-box gene Xbra.

Brachyury (T), a member of the T-box gene family, is essential for the formation of posterior mesoderm and notochord in vertebrate development. Expression of the Xenopus homologue of Brachyury, Xbra, causes ectopic ventral and lateral mesoderm formation in animal cap explants and co-expression of Xbra with Pintallavis, a forkhead/HNF3beta-related transcription factor, induces notochord. Although eFGF and the Bix genes are thought to be direct targets of Xbra, no other target genes have been identified. Here, we describe the use of hormone-inducible versions of Xbra and Pintallavis to construct

Chromatin immunoselection defines a TAL-1 target gene.

Despite the major functions of the basic helix-loop-helix transcription factor TAL-1 in hematopoiesis and T-cell leukemogenesis, no TAL-1 target gene has been identified. Using immunoprecipitation of genomic fragments bound to TAL-1 in the chromatin of murine erythro-leukemia (MEL) cells, we found that 10% of the immunoselected fragments contained a CAGATG or a CAGGTG E-box, followed by a GATA site. We studied one of these fragments containing two E-boxes, CAGATG and CAGGTC, followed by a GATA motif, and showed that TAL-1 binds to the CAGGTG E-box with an affinity modulated by the CAGATG or th

Neural-targeted gene therapy for rodent and primate hemiparkinsonism.

Expression of the rate-limiting enzyme for catecholamine biosynthesis, tyrosine hydroxylase (TH), via retroviral and plasmid expression vectors improved the efficacy of conditionally immortalized nigral neural cells in ameliorating rodent and nonhuman primate models of Parkinson's disease through neural transplantation. No improvement in rotational behavior occurred when sham transplants or nondopaminergic transplants were performed. Transplantation of the temperature-sensitive immortalized parental nigral neural line with a TH expression vector resulted in improvement for at least 2 months. I

Targeting Neurovascular Pathways: Gene Therapy as a Novel Treatment for Vascular Dementia After Stroke.

Stroke is the second leading cause of mortality worldwide, resulting from an interruption of blood flow to the brain, which subsequently diminishes oxygen supply. Although strokes can occur at any age, incidence rises significantly after 55. Vascular dementia, a progressive condition associated with cerebral infarction, is a long-term sequela of stroke that predominantly affects older populations. To date, there is no cure for this form of dementia. Recent experimental studies in rodent models, however, demonstrate promising outcomes in combating neuronal degeneration through gene treatments u

Targeting Microglial Phagocytosis for Alzheimer's Disease Management: Natural, Pharmacological, Nanoparticle, and Gene Therapy Approaches.

Persistent swelling in the brain, internal tau bundles, and external Amyloid-Beta (Aβ) deposits are characteristics of Alzheimer's Disease (AD), an ongoing neurodegenerative illness. Microglia are the main immune cells in the CNS (Central Nervous System). They keep the brain stable by keeping an eye on the immune system and removing apoptotic cells and protein clusters through a process called phagocytosis. However, in AD, microglia exhibit dysregulated phagocytic activity, resulting in either insufficient Aβ clearance or exacerbated inflammatory responses, both of which contribute to neurodeg

Development of In Vitro Potency Assays for Gene Therapy Products Targeting cGMP-Related Retinopathies.

The emergence of efficient viral vectors derived from adeno-associated viruses (AAV) has led many groups to develop gene therapies for inherited retinal diseases. To evaluate the potency of new gene therapy vectors in a preclinical context, it is common to use animal models, such as gene-deficient or mutant animal models of a given human disease, and then assess transgene expression and resulting restoration of vision with functional or behavioral assays. While such animal models are invaluable to the preclinical testing process, they cannot be readily used as batch release tests during manufa

Extracellular matrix targeting peptide functionalized plant-derived lipid nanoparticles for gene therapy in osteoarthritis.

BACKGROUND: The pathogenesis of osteoarthritis (OA) involves the degradation of the extracellular matrix (ECM) of cartilage, which is mainly mediated by matrix metalloproteinases (MMPs). Small interfering RNA (siRNA)-based gene therapy has shown promise in treating various diseases, including OA, but its effectiveness is hindered by poor stability and low uptake efficiency. Additionally, the avascular nature of cartilage poses a challenge for drug delivery. This study aims to develop a novel plant-derived nanoparticle system for targeted, efficient delivery of siRNA to intra-articular chondroc

Pyrimethamine-based targeting of HEXA gene mutations in Tay-Sachs disease: a computational analysis.

Hexosaminidase-Tay Sachs disease (TSD) is a fatal, progressive neurodegenerative disorder, brought on by an accumulation of GM2 gangliosides that results from a deficiency. Treatment requires the restoration of the brain's HEXA enzyme, but the blood-brain barrier makes this difficult by preventing the preponderance of molecules from entering the brain. This study uses computational approaches analyzing the effects of genetic changes on the HEXA gene. 158 missense mutations were collected from different databases and analyzed using in-silico techniques like Meta-SNP and ConSurf. The mutations (

Targeting the stringent response alters gene expression and extracellular vesicle RNA in Staphylococcus aureus.

Staphylococcus aureus relies on the (p)ppGpp-mediated stringent response to adapt to nutrient limitation and other environmental stress, with important consequences for metabolism, antibiotic tolerance, and virulence. In this study, we identified that disruption of (p)ppGpp synthesis altered the small RNA (sRNA) cargo of extracellular vesicles (EVs) produced by S. aureus. We characterized EV-associated sRNAs from wild-type and stringent response mutant (rshsyn) strains, revealing the presence of SprX2 in mutant EVs. The rshsyn mutant also showed modestly enhanced survival under high concentrat

Efficient Silencing of Androgen Receptor Gene via UTR-Targeting siRNAs for Androgenetic Alopecia Therapy.

Androgenetic alopecia (AGA) is predominantly driven by excessive local activity of dihydrotestosterone (DHT), leading to follicular miniaturization and progressive hair loss. The need for novel treatment strategies for AGA is emphasized by the side effects and postoperative sequelae of current therapeutic approaches, including pharmacological interventions and surgical procedures. Small-interfering RNAs (siRNAs) have emerged as promising therapeutic candidates due to their target specificity, the enhanced efficacy, and long-term effect. Here, we screened a series of siRNA sequences targeting n

A specific, stable, and accessible LAMP assay targeting the HSP70 gene of Trypanosoma cruzi.

Diagnostic delays prevent most Chagas disease patients from receiving timely therapy during the acute phase when treatment is effective. qPCR-based diagnostic methods provide high sensitivity during this phase but require specialized equipment and complex protocols. More simple and cost-effective tools are urgently needed to optimize early Chagas disease diagnosis in low-income endemic regions. Here, we present a loop-mediated isothermal amplification (LAMP) that targets a highly conserved region in the HSP70 gene of Trypanosoma cruzi, the causative agent of Chagas disease. This assay demonstr

Novel Therapies in Advanced Pancreatic Cancer: A Peak Beyond the Realms of Gene Targeting.

Pancreatic cancer presents a formidable challenge in oncology, marked by its aggressive behavior and dismal prognosis. Despite a surge in global diagnoses, particularly in affluent nations, mortality data remain scarce in low-income countries due to limited access to diagnostic tools. The five-year survival rate remains dismally low, hovering around 6-8%, emphasizing the urgent need for innovative treatment strategies. In patients with resectable and borderline resectable pancreatic cancer, both neoadjuvant/perioperative mFOLFIRINOX and gemcitabine-nab-paclitaxel (GEM-Abraxane) have shown comp

Targeting the IL-23 Receptor Gene: A Promising Approach in Inflammatory Bowel Disease Treatment.

Inflammatory bowel disease (IBD), which includes Crohn's Disease (CD) and ulcerative colitis (UC), is characterized by chronic inflammation of the gastrointestinal tract. A key component of the inflammatory pathway in IBD is interleukin 23 (IL-23), which promotes the differentiation and maintenance of Th17 cells. These cells are major contributors to intestinal inflammation and the release of pro-inflammatory cytokines. A dysregulated IL-23/Th17 axis can lead to excessive gut inflammation. Notably, IL-23 affects Th17 cell responses differently in UC and CD, fostering IL-17 production in UC and

Exploration of drug repurposing for Mpox outbreaks targeting gene signatures and host-pathogen interactions.

Monkeypox (Mpox) is a growing public health concern, with complex interactions within host systems contributing to its impact. This study employs multi-omics approaches to uncover therapeutic targets and potential drug repurposing opportunities to better understand Mpox's molecular pathogenesis. We developed an in silico host-pathogen interaction (HPI) network and applied weighted gene co-expression network analysis (WGCNA) to explore interactions between Mpox and host proteins. Subtype-specific host-pathogen protein-protein interaction networks were constructed, and key modules from the HPI a

Gene targeting in amyotrophic lateral sclerosis using causality-based feature selection and machine learning.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a rare progressive neurodegenerative disease that affects upper and lower motor neurons. As the molecular basis of the disease is still elusive, the development of high-throughput sequencing technologies, combined with data mining techniques and machine learning methods, could provide remarkable results in identifying pathogenetic mechanisms. High dimensionality is a major problem when applying machine learning techniques in biomedical data analysis, since a huge number of features is available for a limited number of samples. The aim of this

Calcitonin gene-related peptide-targeting drugs for migraine: how pharmacology might inform treatment decisions.

Migraine is the second most disabling disorder across all age groups worldwide. Since 2018, two classes of drugs that inhibit the actions of calcitonin gene-related peptide (CGRP), which is implicated in migraine pathophysiology, have become available: gepants (CGRP receptor antagonists) and monoclonal antibodies directed against CGRP or its receptor. Despite phase 3 clinical trials and some real world evidence, knowledge of the pharmacology and related clinical effects of these drugs is low, and trial data are not necessarily generalisable to all populations. Additionally, several pharmacodyn
