Comparative Transcriptomic Analysis Revealing the Potential Mechanisms of Erythritol-Caused Mortality and Oviposition Inhibition in Drosophila melanogaster.

Erythritol has shown excellent insecticidal performance against a wide range of insect species, but the molecular mechanism by which it causes insect mortality and sterility is not fully understood. The mortality and sterility of Drosophila melanogaster were assessed after feeding with 1M erythritol for 72 h and 96 h, and gene expression profiles were further compared through RNA sequencing. Enrichment analysis of GO and KEGG revealed that expressions of the adipokinetic hormone gene (Akh), amylase gene (Amyrel), α-glucosidase gene (Mal-B1/2, Mal-A1-4, Mal-A7/8), and triglyceride lipase gene (

Transcriptome Analysis of an Aedes albopictus Cell Line Single- and Dual-Infected with Lammi Virus and WNV.

Understanding the flavivirus infection process in mosquito hosts is important and fundamental in the search for novel control strategies that target the mosquitoes' ability to carry and transmit pathogenic arboviruses. A group of viruses known as insect-specific viruses (ISVs) has been shown to interfere with the infection and replication of a secondary arbovirus infection in mosquitoes and mosquito-derived cell lines. However, the molecular mechanisms behind this interference are unknown. Therefore, in the present study, we infected the Aedes albopictus cell line U4.4 with either the West Nil

Genomic evidence for limited entomophagy in ancient Europeans.

To meet the rising food demands of our growing population, the Food and Agriculture Organization proposed edible insects as sustainable sources of animal protein. Although hundreds of million people already consume insects around the tropics, western societies remain averse to entomophagy. To trace whether ancient Europeans consumed insects, we here apply two complementary genomic approaches. Metagenomic screening on 745 ancient anatomically modern human dental calculus returned limited insect DNA traces, with read abundances well below those observed in Neanderthals, western chimpanzees, and

Divergent immune strategies of Colorado potato beetle larvae against the entomopathogenic fungi Metarhizium robertsii and Beauveria bassiana: A comparative transcriptomic analysis.

The entomopathogenic fungi Beauveria bassiana and Metarhizium robertsii are natural pathogens of the Colorado potato beetle (CPB) and hold promise for biocontrol. However, the insect's immune responses to fungal infections remain poorly understood. In this study, we performed transcriptomic analyses of haemocytes and fat body tissue from CPB larvae topically infected with B. bassiana and M. robertsii at the stage of pathogen encapsulation. Quantitative PCR validation of gene expression, along with complementary physiological and biochemical assays, was also conducted. Reference gene annotation

The invasive MED/Q Bemisia tabaci genome: a tale of gene loss and gene gain.

BACKGROUND: Sweetpotato whitefly, Bemisia tabaci MED/Q and MEAM1/B, are two economically important invasive species that cause considerable damages to agriculture crops through direct feeding and indirect vectoring of plant pathogens. Recently, a draft genome of B. tabaci MED/Q has been assembled. In this study, we focus on the genomic comparison between MED/Q and MEAM1/B, with a special interest in MED/Q's genomic signatures that may contribute to the highly invasive nature of this emerging insect pest. RESULTS: The genomes of both species share similarity in syntenic blocks, but have signif

Membrane association of importin α facilitates viral entry into salivary gland cells of vector insects.

The importin α family belongs to the conserved nuclear transport pathway in eukaryotes. However, the biological functions of importin α in the plasma membrane are still elusive. Here, we report that importin α, as a plasma membrane-associated protein, is exploited by the rice stripe virus (RSV) to enter vector insect cells, especially salivary gland cells. When the expression of three importin α genes was simultaneously knocked down, few virions entered the salivary glands of the small brown planthopper, Laodelphax striatellus Through hemocoel inoculation of virions, only importin α2 was found

Trans-complementation of polyhedrin by a stably transformed Sf9 insect cell line allows occ- baculovirus occlusion and larval per os infectivity.

Oral infection of insect larvae with baculovirus is an advantageous methodology for producing high levels of recombinant proteins and for achieving plague control. However, many recombinant baculoviruses express a foreign protein in lieu of the polyhedrin and hence do not form occlusion bodies (occ-), resulting in extremely reduced per os infectivity in larvae. To overcome this limitation, stably transformed insect cell lines expressing polyhedrin capable of occluding occ- recombinant baculovirus by trans-complementation were developed to obtain oral inoculum for insect larvae infection. First

Nuclear receptors in the mosquito Aedes aegypti: annotation, hormonal regulation and expression profiling.

In anautogenous mosquitoes, egg development requires blood feeding and as a consequence mosquitoes act as vectors of numerous devastating diseases of humans and domestic animals. Understanding the molecular mechanisms regulating mosquito egg development may contribute significantly to the development of novel vector-control strategies. Previous studies have shown that in the yellow fever mosquito Aedes aegypti, nuclear receptors (NRs) play a key role in the endocrine regulation of reproduction. However, many mosquito NRs remain uncharacterized, some of which may play an important role in mosqu

Intestinal Taste Receptor Expression and Its Implications for Health: An Integrative Analysis in Female Rats after Chronic Insect Supplementation.

Taste receptors are found in the gastrointestinal tract, where they are susceptible to dietary modulation, a key point that is crucial for diet-related responses. Insects are sustainable and good-quality protein sources. This study analyzed the impact of insect consumption on the modulation of taste receptor expression across various segments of the rat intestine under healthy or inflammatory conditions. Female Wistar rats were supplemented with Tenebrio molitor (T) or Alphitobius diaperinus (B), alongside a control group (C), over 21 days under healthy or LPS-induced inflammation. The present

Fusion protein based strategies for developing insect-resistant crops.

Insect pest control in crop production incurs substantial economic costs annually on a global scale. Although broad-spectrum chemical pesticides were once considered the most effective solution, their overreliance has led to adverse effects on beneficial insects, human health, and the environment, as well as the development of pesticide-resistant insect populations. Consequently, there is an urgent need for alternative pest management strategies that minimize pesticide use and reduce unintended impacts on natural enemies, thereby maintaining ecological balance. Host plant resistance plays a pi

Phenotype-genotype association for the identification of resistance to stem borers (Diatraea spp., Lepidoptera: Crambidae) in sugarcane.

Sugarcane (Saccharum spp.) is a major crop for sugar production, but its productivity is affected by stem borers (Diatraea spp.). These insects cause significant losses by damaging the stem and disrupting the transport of photosynthates and nutrients. Although biological control has been effective in Colombia, the identification of genetic resources for the resistance would provide complementary long-term crop protection. This study aimed to identify markers associated with the resistance to insects. For this purpose, a population of 348 genotypes was evaluated by infesting 30 stalks per genot

Sequence characterization of an unusual lysozyme gene expressed in the intestinal tract of the reduviid bug Triatoma infestans (Insecta).

Antibacterial proteins like lysozyme are important components of the insect non-specific immune response against bacteria. The complementary deoxyribonucleic acid (cDNA) encoding a new lysozyme from Triatoma infestans, named lysozyme2, has been amplified by polymerase chain reaction and the rapid amplification of cDNA ends technique. The gene is expressed in the small intestine of the insect. The deduced protein sequence shows up to 70% similarity to lysozymes from other species. Furthermore, the protein exhibits significant structural concordance to other insect lysozymes. A striking feature

Lactobacillus apisdorsatae sp. nov. from the giant honeybee (Apis dorsata), Lactobacillus pollinatoris sp. nov. and Lactobacillus xylocopae sp. nov., from the carpenter bee (Xylocopa violacea).

Insects host diverse and distinctive communities of gut mutualistic bacteria, including lactic acid bacteria of the Lactobacillus genus. Recently, most research in the insect microbial taxonomy has focused on identifying novel Lactobacillaceae species in managed insects such as Apis mellifera and few species of Bombus spp. from North America, Europe, and China. However, the gut microbiota of wild insects remains largely unexplored. Yet, insects represent the largest animal class and a vast, untouched source of novel microbial symbionts, especially in wild populations. This study focuses on the

Cross-kingdom RNA interference mediated by insect salivary microRNAs may suppress plant immunity.

Communication between insects and plants relies on the exchange of bioactive molecules that traverse the species interface. Although proteinic effectors have been extensively studied, our knowledge of other molecules involved in this process remains limited. In this study, we investigate the role of salivary microRNAs (miRNAs) from the rice planthopper Nilaparvata lugens in suppressing plant immunity. A total of three miRNAs were confirmed to be secreted into host plants during insect feeding. Notably, the sequence-conserved miR-7-5P is specifically expressed in the salivary glands of N. lugen

Lipid metabolism in insect disease vectors.

More than a third of the world population is at constant risk of contracting some insect-transmitted disease, such as Dengue fever, Zika virus disease, malaria, Chagas' disease, African trypanosomiasis, and others. Independent of the life cycle of the pathogen causing the disease, the insect vector hematophagous habit is a common and crucial trait for the transmission of all these diseases. This lifestyle is unique, as hematophagous insects feed on blood, a diet that is rich in protein but relatively poor in lipids and carbohydrates, in huge amounts and low frequency. Another unique feature of

Molecular Detection of Hemoparasites in Hematophagous Insects Collected from Livestock Farms in Northeastern Thailand.

Vector-borne parasitic diseases (VBPDs) are diseases that have a major impact on the health and productivity of livestock. VBPDs, including trypanosomosis, theileriosis, and anaplasmosis, are caused by blood parasites and Rickettsiales infections that can be transmitted by biting insects. The control and elimination of VBPDs is difficult and challenging. This study contributed to better understanding the impact of hematophagous insects on carrying pathogens to livestock by investigating the detection of Trypanosoma evansi, Theileria spp. and Anaplasma spp. in hematophagous insects collected fr

Neuropeptides in the antennal lobe of the yellow fever mosquito, Aedes aegypti.

For many insects, including mosquitoes, olfaction is the dominant modality regulating their behavioral repertoire. Many neurochemicals modulate olfactory information in the central nervous system, including the primary olfactory center of insects, the antennal lobe. The most diverse and versatile neurochemicals in the insect nervous system are found in the neuropeptides. In the present study, we analyzed neuropeptides in the antennal lobe of the yellow fever mosquito, Aedes aegypti, a major vector of arboviral diseases. Direct tissue profiling of the antennal lobe by matrix-assisted laser deso

Knockdown of the ionotropic γ-aminobutyric acid receptor (GABAR) RDL gene decreases fipronil susceptibility of the small brown planthopper, Laodelphax striatellus (Hemiptera: Delphacidae).

Insect γ-aminobutyric acid receptors (GABARs) are important molecular targets of cyclodiene and phenylpyrazole insecticides. Previously GABARs encoding rdl (resistant to dieldrin) genes responsible for dieldrin and fipronil resistance were identified in various economically important insect pests. In this study, we cloned the open reading frame cDNA sequence of rdl gene from fipronil-susceptible and fipronil-resistant strains of Laodelphax striatellus (Lsrdl). Sequence analysis confirmed the presence of a previously identified resistance-conferring mutation. Different alternative splicing vari

A genetically engineered Pseudomonas fluorescens strain possesses the dual activity against phytopathogenic fungi and insects.

A Pseudomonas fluorescens strain was isolated and showed antagonistic activity against phytopathogenic fungi and found to possess a gene responsible for production of antibiotic 2, 4-diacetylphloroglucinol. For the extension of biocontrol range, a gene for an Androctonus australis Hector insect toxin 1 (AaHIT1), one of the most toxic known insect-selective peptides, was designed and synthesized according to the preferred codon usage of Pseudomonas fluorescens, cloned and transformed into the strain by pSUP106 vector, a broad-host-range plasmid. Functional proteomic insights into the lateral oviduct-associated secretions of a bean bug.

The secretions within the lateral oviduct lumen of the insect female reproductive tract (FRT) play crucial roles in reproduction, yet their protein composition and functional mechanisms remain poorly understood. In this study, we revealed that the lateral oviduct of the bean bug, Riptortus pedestris, is composed of an outer muscle layer and an inner epithelium with a microvillar border. Proteomic analysis identified 159 FRT-highly expressed proteins, with functional characterization focusing on 10 putative secretory proteins, including 2 ovary-specific (RpLoap2 and RpLoap4), 7 oviduct-specific
