Surveillance reveals a prevalent pediatric A(H1N1)pdm09 virus with hemagglutinin substitutions S137P-R142K-V152I that diminish vaccine efficacy.

Influenza A(H1N1)pdm09 viruses pose a significant disease burden on children worldwide, with high rates of hospitalization and substantial morbidity and mortality. Outpatient < 18 years of age with upper respiratory infections (URIs) were enrolled through active surveillance at Shanxi Children's Hospital (SCH) between 7/1/2024 and 6/30/2025. Nasal swabs were collected for the detection of influenza virus and other respiratory pathogens by PCR-based methods. Influenza strains were obtained through in vitro culture, and their antigenic characterization were determined by hemagglutinin-inhibition

Age-specific epigenetic drift in late-onset Alzheimer's disease.

Despite an enormous research effort, most cases of late-onset Alzheimer's disease (LOAD) still remain unexplained and the current biomedical science is still a long way from the ultimate goal of revealing clear risk factors that can help in the diagnosis, prevention and treatment of the disease. Current theories about the development of LOAD hinge on the premise that Alzheimer's arises mainly from heritable causes. Yet, the complex, non-Mendelian disease etiology suggests that an epigenetic component could be involved. Using MALDI-TOF mass spectrometry in post-mortem brain samples and lymphocy

Antibody titer has positive predictive value for vaccine protection against challenge with natural antigenic-drift variants of H5N1 high-pathogenicity avian influenza viruses from Indonesia.

UNLABELLED: Vaccines are used in integrated control strategies to protect poultry against H5N1 high-pathogenicity avian influenza (HPAI). H5N1 HPAI was first reported in Indonesia in 2003, and vaccination was initiated in 2004, but reports of vaccine failures began to emerge in mid-2005. This study investigated the role of Indonesian licensed vaccines, specific vaccine seed strains, and emerging variant field viruses as causes of vaccine failures. Eleven of 14 licensed vaccines contained the manufacturer's listed vaccine seed strains, but 3 vaccines contained a seed strain different from that

Contrasting evolutionary trajectories of pyrimethamine resistance in human and zoonotic malaria parasites.

BACKGROUND: Pyrimethamine resistance in Plasmodium falciparum is driven by mutations at dihydrofolate reductase (DHFR) residues N51I, C59R, S108N/T, and I164L, with parallel substitutions in P. vivax (N50I, S58R, S117N/T, S173L). To evaluate the influence of drug pressure on parasite evolution, we compared the dihydrofolate reductase-thymidylate synthase (dhfr-ts) genes of human and zoonotic malaria parasites exposed to different drug environments. METHODS: Blood samples were collected from patients with symptomatic malaria in Thailand (P. falciparum, n = 241; P. vivax, n = 115) and from symp

Progressive glycosylation of the haemagglutinin of avian influenza H5N1 modulates virus replication, virulence and chicken-to-chicken transmission without significant impact on antigenic drift.

Highly pathogenic H5N1 avian influenza virus (A/H5N1) devastated the poultry industry and continues to pose a pandemic threat. Studying the progressive genetic changes in A/H5N1 after long-term circulation in poultry may help us to better understand A/H5N1 biology in birds. A/H5N1 clade 2.2.1.1 antigenic drift viruses have been isolated from vaccinated commercial poultry in Egypt. They exhibit a peculiar stepwise accumulation of glycosylation sites (GS) in the haemagglutinin (HA) with viruses carrying, beyond the conserved 5 GS, additional GS at amino acid residues 72, 154, 236 and 273 resulti

Genetic relationship of human populations and ethnic differences in reaction to drugs and food.

Using gene frequency data for a large number of protein and blood group loci and restriction-site data for mitochondrial DNA, we studied the genetic variation within and between the three major races of man, Caucasoid, Negroid, and Mongoloid. In all the three major races, about 50 percent of structural loci seem to be polymorphic at the electrophoretic level, and the average heterozygosity is about 14 percent. The genetic variation between the three major races is small compared with that within the races. However, even this small interracial variation seems to be the result of geographic isol

Genetic ancestry and monogenic disease risk in the Scottish Traveller founder population.

The Scottish Travellers are a traditionally nomadic community in Scotland that has historically been marginalised, and remained socially isolated from the settled Scottish population until recently. Little, however, is known about their genetic origins, population structure and risks of Mendelian disease. After an approach from the community to address this gap and increase representation, we analyzed array genotypes and whole-exome sequencing data from up to 125 Gypsy/Traveller individuals, alongside settled British and Irish references. We demonstrate that Scottish Travellers are genetically

Population structure of the banana weevil, an introduced pest in the Canary Islands, studied by RAPD analysis.

The banana weevil (BW), Cosmopolites sordidus (Coleoptera: Curculionidae), is one of the most important insect pests of bananas and plantains. The mobility and the origin of BW infestations at the Canary Islands (Tenerife, La Gomera and La Palma) have been analysed using Random Amplified Polymorphic DNA (RAPD) as molecular markers. Populations from Costa Rica, Colombia, Uganda and Madeira were also included for comparison. One hundred and fifteen reproducible bands from eight primers were obtained. The level of polymorphism in the populations from the Canary Islands (40-62%) was in the range o

Genetic differentiation of hypothalamus parentally biased transcripts in populations of the house mouse implicate the Prader-Willi syndrome imprinted region as a possible source of behavioral divergence.

Parentally biased expression of transcripts (genomic imprinting) in adult tissues, including the brain, can influence and possibly drive the evolution of behavioral traits. We have previously found that paternally determined cues are involved in population-specific mate choice decisions between two populations of the Western house mouse (Mus musculus domesticus). Here, we ask whether this could be mediated by genomically imprinted transcripts that are subject to fast differentiation between these populations. We focus on three organs that are of special relevance for mate choice and behavior:

Genetic variations and treatments that affect the lifespan of the NPC1 mouse.

Niemann-Pick type C (NPC) disease is a multisystem disorder caused primarily by a mutation in the npc1 gene. These studies evaluated the effect of genetic background, deletion of additional genes, and administration of several agents on the age at death in a murine model of this disorder. Such factors as differing strain background or genetic drift within a given background in the npc1(-/-) mouse significantly altered the age at death and the degree of organ disease. Genetic deletion of Siat9 (GM3 synthetase) or Nr1h2 [liver X receptor (LXR)beta] shortened the life of the npc1(-/-) animals. Da

Population genetics of Aedes aegypti (Diptera: Culicidae) in India reveals high connectivity and weak geographic and climatic structuring.

Understanding the genetic structure and diversity of Aedes aegypti (Linnaeus, 1762) populations is critical for predicting transmission dynamics and implementing effective vector control strategies. This study assessed the genetic diversity, population structure, and influence of geography and climate on Ae. aegypti populations across India. Of 308 genotyped individuals, 257 high-quality genotypes from 13 collection sites across 6 climatic regions were retained for analysis using 10 microsatellite loci. High genetic diversity was observed across all populations, consistent with substantial eff

Genome-wide genetic diversity of Holstein Friesian cattle reveals new insights into Australian and global population variability, including impact of selection.

Past breeding strategies for dairy cattle have been very effective in producing rapid genetic gain to achieve industry targets and raise profitability. Such gains have been largely facilitated by intense selection of sires combined with the use of artificial insemination. However, this practice can potentially limit the level of genetic diversity through inbreeding and selection plateaus. The rate of inbreeding in Australia is increasing, primarily as a result of semen importation from a small number of prominent bulls from the USA. The effect of this genetic influx in the Australian dairy cat

Cryopreservation of Rodent Ovarian Tissues and Oocytes.

Germplasm cryobanking of rodents is a valuable tool for protecting scientifically important human disease models from genetic drift, genetic contamination, and loss of breeding colonies due to disease or catastrophic disasters to the housing facilities, as well as avoiding stress associated with domestic and international live animal shipment. Furthermore, germplasm cryopreservation enhances management efficiency by saving vivarium space, reducing workload for staff, financial burden of maintaining live animals, reducing the number of animals used to maintain a breeding colony, facilitating tr

Transmission of the human mitochondrial genome.

The segregation and transmission of mitochondrial genomes in humans are complicated processes, but are particularly important for understanding the inheritance and clinical abnormalities of mitochondrial disorders. This review describes three aspects of mitochondrial genetics. First, that the segregation and transmission of mitochondrial (mt)DNA molecules are likely to be determined by their physical association within the organelles and by the dynamics of mitochondrial structure and subcellular organization. Second, that the transmission of heteroplasmic mtDNA sequence changes from one genera

Island-wide population genomic analysis of the coastal shrub, beach naupaka (Scaevola taccada L.), on O'ahu.

Plant species face myriad threats to their survival, including climate change and habitat destruction. On O'ahu in Hawai'i, Scaevola taccada L., a coastal dune shrub, is highly impacted by human activities and environmental threats. Despite its ubiquity across Pacific islands, the extent and distribution of genetic diversity within individual islands is unclear. We surveyed genomic diversity using double digest restriction-associated DNA sequencing (ddRAD-seq) in eight populations of S. taccada on O'ahu. Using principal components (PCA) and STRUCTURE analyses, we assessed whether genetic diver

Repeated genetic adaptation under demographic limits in kiwifruit.

Population genetic theory predicts that species with narrow geographic distributions experience stronger genetic drift, higher inbreeding, and greater mutational burden than their widespread relatives, yet how demographic history shapes the genomic basis of adaptation remains poorly understood. We hypothesized that (i) restricted-range species exhibit reduced genetic diversity, elevated inbreeding, and greater mutation load owing to smaller long-term effective population sizes; (ii) despite these demographic constraints, shared environmental pressures can still drive repeated adaptation; (iii)

Temporal shifts in polygenic traits track major epidemics in Western Eurasia.

Infectious diseases are recognized as one of the strongest selective forces, exerting a profound influence on the genetic makeup of human populations over time. Recently, large-scale genome-wide association studies (GWAS) on immunological traits have underscored the notion that genetic predisposition to infectious diseases likely stems from the contribution of several thousand loci across the human genome. To model the polygenic inheritance of these traits, multiple variants can be combined into polygenic risk scores (PRS), which estimate an individual's genetic predisposition for a trait. By

Decoding HuH-7: a comprehensive genetic and molecular portrait of a widely used hepatocellular carcinoma model.

INTRODUCTION: Immortalized cell lines play a crucial role in biomedical research by enabling reproducible experiments and enhancing our understanding of complex diseases. HuH-7, originally derived from a human hepatocellular carcinoma, is particularly valuable for studying liver cancer dynamics, viral hepatitis, and drug metabolism. However, concerns about cell line misidentification and genetic drift in cell lines highlight the importance of rigorous authentication to maintain the reliability of research outcomes, despite their widespread use. METHODS: In this study, we present a detailed (c

Low 2012-13 influenza vaccine effectiveness associated with mutation in the egg-adapted H3N2 vaccine strain not antigenic drift in circulating viruses.

BACKGROUND: Influenza vaccine effectiveness (VE) is generally interpreted in the context of vaccine match/mismatch to circulating strains with evolutionary drift in the latter invoked to explain reduced protection. During the 2012-13 season, however, detailed genotypic and phenotypic characterization shows that low VE was instead related to mutations in the egg-adapted H3N2 vaccine strain rather than antigenic drift in circulating viruses. METHODS/FINDINGS: Component-specific VE against medically-attended, PCR-confirmed influenza was estimated in Canada by test-negative case-control design. I
