Innovative approach to genetic counseling services for the deaf population.

Genetic service providers have stressed the importance of genetic counseling that is nondirective and specific to the personal needs of consultants. Successful genetic counseling for deaf persons often requires special provisions for complex family histories, syndromic conditions, and diversity in communication methods and cultural orientation. The Gallaudet University Genetic Services Center (GSC) was established in 1984 to provide genetic education and counseling services to the deaf community. The GSC staff developed and implemented a standardized system of data collection (family and medic

Familial factors and genetic predisposition to cancer: population studies.

Hereditary cancer-prone families provide one of the most powerful and potentially cost-effective models for cancer prevention and control. The public health impact of this problem is enormous when considering that between 5 to 10% of the total cancer burden is hereditary. At least 200 well-defined hereditary cancer syndromes have been identified. In spite of its impact, attention to family history of cancer is often given short shrift and, consequently, clinical expertise in identifying hereditary forms of cancer is exceedingly limited. This may change as a result of recent advances in biomole

Genetic disorders in children and young adults: a population study.

The data base of an ongoing population-based registry with multiple sources of ascertainment was used to estimate the present population load from genetic disease in more than 1 million consecutive live births. It was found that, before approximately age 25 years, greater than or equal to 53/1,000 live-born individuals can be expected to have diseases with an important genetic component. This total was composed of single-gene disorders (3.6/1,000), consisting of autosomal dominant (1.4/1,000), autosomal recessive (1.7/1,000), and X-linked recessive disorders (0.5/1,000). Chromosomal anomalies

A genetic profile of the South African Ashkenazi Jewish population.

The South African Ashkenazi Jewish population is described in terms of the prevailing frequencies of the genes at 25 red cell enzyme and serum protein loci and 4 placental enzyme loci. Variation was encountered in 23 of these systems. The Tay-Sachs allele which occurs at polymorphic frequencies in Ashkenazi populations was found at a frequency of 0.022, which suggests that approximately 1 baby with Tay-Sachs disease could be expected out of every 2,000 born in this community should preventive measures not be taken. The atypical serum pseudocholinesterase cholinesterase allele was encountered a

A population genetic study of the Vania Soni in Western India.

A total of 267 blood samples from persons belonging to the Shrimali Vania Soni caste group in Gujarat State, Western India have been analyzed for 6 blood group, 4 serum protein and 19 red cell enzyme systems, for haemoglobin and beta-thalassaemia and for red-green colour blindness. A number of rare genetic variants were detected, including a unique electrophoretically fast variant of superoxide dismutase. Genetic distance comparisons with other caste groups in Gujarat State show that Vania Soni from Surat are a distinctive group clustering with another subdivision of the Vania. The remaining V

Occurrence of acute leukemia in females in a genetically isolated population.

Acute lymphoblastic leukemia was found in six young females in a genetically isolated Syrian Jewish Community in Brooklyn, N.Y. over a 15-year period. This represents a frequency 30 times greater than the expected rate. In this community the frequency of first cousin marriages is estimated to be seven percent. Three cases were related on their maternal side, the parents of a fourth case were themselves related as were both sets of grandparents of a fifth case. In two families a history of infertility was present. On average, fathers of the cases were 5.5 years older at birth of the cases than

Esterase D: some population and formal genetical data.

562 healthy blood donors and 65 families with 149 offspring were types for esterase D (EsD). The observed allele frequencies are in good agreement with those determined in other Caucasian populations. The rare variant EsD3 was found in a blood donor who transmitted it to his daughter. The observed segregation ratios in our family material showed no deviation from expectation.

A new perspective on population genetics: Deciphering the relationship between genetic variants and disease prevalence in Psoriasis.

In the quest to identify the genetic underpinnings of complex diseases, we developed a novel approach called Causal Genotype Combination Patterns (CGCP) to uncover characteristic genetic signatures of common diseases. In this study, we applied the CGCP method to a whole-exome sequencing dataset of 781 psoriasis cases and 676 healthy controls from the Chinese Han population. Our analysis revealed 620 genotype combinations specific to psoriasis, covering 4.7% to 10% of all cases, with each genotype having a frequency of at least 1%. These genotypes converged into 134 genes, including 41 previous

Development of a Multiple Epitopes-Based Dengue Vaccine: An Immunoinformatics Approach and Insights From Pakistani Population Genetics.

Dengue fever poses a grave threat to public health worldwide, claiming numerous fatalities each year with tropical regions being particularly hard hit by dengue outbreaks. The multiple-epitope construct is tailored to the dengue virus's geographical prevalence and the genetics of the Pakistani population. 14 experimentally validated MHC-I and MHC-II epitope sequences, were employed to generate the variants by taking into account the conservancy in all serotypes. Subsequently, the binding affinities of each derived variant against the human leukocyte antigen alleles most common among the Pakist

Population genetics of two chromatic morphs of the Chagas disease vector Rhodnius pallescens Barber, 1932 in Panamá.

Rhodnius pallescens is the principal vector of Chagas disease in Panama. Recently a dark chromatic morph has been discovered in the highlands of Veraguas Province. Limited genetic studies have been conducted with regards to the population structure and dispersal potential of Triatominae vectors, particularly in R. pallescens. Next generation sequencing methods such as RADseq and complete mitochondrial DNA (mtDNA) genome sequencing have great potential for examining vector biology across space and time. Here we utilize a RADseq method (3RAD), along with complete mtDNA sequencing, to examine the

Reverse Genetics of RNA Viruses: ISA-Based Approach to Control Viral Population Diversity without Modifying Virus Phenotype.

Reverse genetic systems are essential for the study of RNA viruses. Infectious clones remain the most widely used systems to manipulate viral genomes. Recently, a new PCR-based method called ISA (infectious subgenomic amplicons) has been developed. This approach has resulted in greater genetic diversity of the viral populations than that observed using infectious clone technology. However, for some studies, generation of clonal viral populations is necessary. In this study, we used the tick-borne encephalitis virus as model to demonstrate that utilization of a very high-fidelity, DNA-dependent

Microsatellite loci cross-species transferability in Aedes fluviatilis (Diptera:Culicidae): a cost-effective approach for population genetics studies.

BACKGROUND: Aedes fluviatilis is a neotropical mosquito species thought to be a potential vector of Yellow Fever viruses and can be infected with Plasmodium gallinaceum in laboratory. A better understanding of its genetic structure is very important to understand its epidemiologic potential and how it is responding to urbanization. The objective of this study was to survey the transferability of microsatellites loci developed for other Aedes to Ae. fluviatilis. FINDINGS: We tested in Ae. fluviatilis 40 pairs of primers known to flank microsatellite regions in Aedes aegypti, Aedes albopictus a

Population Genetics and Antimicrobial Susceptibility of Canine Campylobacter Isolates Collected before and after a Raw Feeding Experiment.

In recent years, increasing numbers of consumers have become interested in feeding raw food for their pet dogs as opposed to commercial dry food, in the belief of health advantages. However, raw meat and internal organs, possibly contaminated by pathogens such as Campylobacter spp., may pose a risk of transmission of zoonoses to the pet owners. Campylobacter jejuni is the leading cause of bacterial gastroenteritis in humans but C. upsaliensis has also been associated with human disease. In this study we investigated the effect of different feeding strategies on the prevalence of Campylobacter

Population genetics of 30 insertion-deletion polymorphisms in two Chinese populations using Qiagen Investigator® DIPplex kit.

Insertion-deletion polymorphisms (INDELs) are short length diallelic polymorphisms caused by the insertion or deletion of several bases. INDEL markers can serve as useful supplementary or stand-alone assays for human identification. The Qiagen Investigator(®) DIPplex kit multiplexes 30 autosomal INDELs plus amelogenin for forensic use. The objective of this study was to estimate genetic diversity of 30 INDEL markers in the Han (the largest ethnic group of China, n=565) and She population (almost the smallest ethnic group of China, n=119), and to evaluate their usefulness in forensic genetics.

Amerindians show no association of PC-1 gene Gln121 allele and obesity: a thrifty gene population genetics.

PC-1 Gln121 gene is a risk factor for type 2 diabetes, obesity and insulin resistance in European/American Caucasoids and Orientals. We have aimed to correlate for the first time this gene in Amerindians with obesity and their corresponding individuals genotypes with obesity in order to establish preventive medicine programs for this population and also studying the evolution of gene frequencies in world populations. Central obesity was diagnosed by waist circumference perimeter and food intake independent HDL-cholesterol plasma levels were measured. HLA genes were determined in order to more

Population genetics of Schistosoma japonicum within the Philippines suggest high levels of transmission between humans and dogs.

BACKGROUND: Schistosoma japonicum, which remains a major public health problem in the Philippines and mainland China, is the only schistosome species for which zoonotic transmission is considered important. While bovines are suspected as the main zoonotic reservoir in parts of China, the relative contributions of various non-human mammals to S. japonicum transmission in the Philippines remain to be determined. We examined the population genetics of S. japonicum in the Philippines in order to elucidate transmission patterns across host species and geographic areas. METHODOLOGY/PRINCIPAL FINDIN

A community genetics approach to population screening in India for mental retardation--a model for developing countries.

Some 0.55 million people living in semi-urban and slum populations were screened for mental retardation by trained primary health centre (PHC) doctors, nurses and community health volunteers (CHVs). The staff were provided with prior training on the detection, prevention and diagnosis of mental retardation, prenatal diagnosis, and reproductive responsibilities. Field visits were employed to confirm diagnosed developmental disabilities, and demographic data incorporating social maps of 14 PHCs were prepared. Cases with high-risk genetic factors detected by PHC staff were referred to the Centre

Updated analysis of the Bronze Age population genetics from Bezdanjača Cave - the case of I2a1a male individuals.

This case report presents an updated interpretation of genetic and chronological data from human remains discovered in Bezdanjača Cave, a Bronze Age burial site located in the Lika region of Croatia. The cave contains a complex necropolis with at least 57 graves and up to 200 individuals, which indicates its use as a collective burial site during the Middle and Late Bronze Age. Based on ancient DNA analysis, 13 males were identified among 38 analyzed individuals, with the majority belonging to the Y-chromosome haplogroup R1b, commonly associated with Bronze Age populations. However, two indivi

Phylogeography and Population Genetics of the Kentucky Endemic Cave Beetle Genus Neaphaenops Jeannel (1920) (Coleoptera: Carabidae: Trechini).

Cave beetles are an excellent group for studying evolutionary history and speciation within cave habitats. However, they are underrepresented in such research, particularly in North America, a hotspot of cave biodiversity, where beetles account for a significant proportion of documented taxa. This study aims to elucidate the evolutionary history and possible connectivity or barriers to gene flow among populations of the central Kentucky endemic cave beetle, Neaphaenops tellkampfii (Erichson), and to test the validity of four proposed subspecies. We collected specimens from 62 caves, extracted

Population-Specific Mutational Spectrum of Autosomal Recessive Nonsyndromic Hearing Loss in Croatian Roma: Implications for Clinical Genetics.

BACKGROUND/OBJECTIVES: Hearing impairment is a highly prevalent sensory disorder resulting from a variety of causes. A high proportion of autosomal recessive non-syndromic hearing impairment is linked to the GJB2 (OMIM 121011) gene which encodes for a gap junction protein, connexin-26. Alterations of genes that encode for connexins can lead to changes in cell ion content and cause hearing impairment. METHODS: GJB2 gene polymorphisms (c.71G>A, p.Trp24*rs104894396; c.457G>A, p.Val153Ile, rs111033186; c.380G>A, p.Arg127His, rs111033196; c.109G>A, p.Val37Ile, rs72474224; and c.269T>C, p.Leu90Pro,
