Effects of the gut microbiota on placental angiogenesis and intrauterine growth in gnotobiotic mice.

Environmental causes of intrauterine growth restriction (IUGR) remain poorly characterized. Here, we compare germ-free (GF) and conventionally raised (CONV-R) mice to assess the effects of the gut microbiota on placental/fetal development at embryonic day (E)11.5 (end of placentation) and E17.5 (near term). Pregnancy- and microbiota-associated changes in gene expression occur along the gut, including those related to angiogenesis, while bacterial composition and fermentation activity remain stable. Placental weights at E11.5 and fetal weights at E17.5 are significantly reduced in GF animals. C

Intestinal colonization resistance in the context of environmental, host, and microbial determinants.

Microbial communities that colonize the human gastrointestinal (GI) tract defend against pathogens through a mechanism known as colonization resistance (CR). Advances in technologies such as next-generation sequencing, gnotobiotic mouse models, and bacterial cultivation have enhanced our understanding of the underlying mechanisms and the intricate microbial interactions involved in CR. Rather than being attributed to specific microbial clades, CR is now understood to arise from a dynamic interplay between microbes and the host and is shaped by metabolic, immune, and environmental factors. Human Rotavirus Replicates in Salivary Glands and Primes Immune Responses in Facial and Intestinal Lymphoid Tissues of Gnotobiotic Pigs.

Human rotavirus (HRV) is a leading cause of viral gastroenteritis in children across the globe. The virus has long been established as a pathogen of the gastrointestinal tract, targeting small intestine epithelial cells and leading to diarrhea, nausea, and vomiting. Recently, this classical infection pathway was challenged by the findings that murine strains of rotavirus can infect the salivary glands of pups and dams and transmit via saliva from pups to dams during suckling. Here, we aimed to determine if HRV was also capable of infecting salivary glands and spreading in saliva using a gnotob

Commensal Microbiota Regulate Vertebrate Innate Immunity-Insights From the Zebrafish.

Microbial communities populate the mucosal surfaces of all animals. Metazoans have co-evolved with these microorganisms, forming symbioses that affect the molecular and cellular underpinnings of animal physiology. These microorganisms, collectively referred to as the microbiota, are found on many distinct body sites (including the skin, nasal cavity, and urogenital tract), however the most densely colonized host tissue is the intestinal tract. Although spatially confined within the intestinal lumen, the microbiota and associated products shape the development and function of the host immune sy

The microbiota protects from viral-induced neurologic damage through microglia-intrinsic TLR signaling.

Symbiotic microbes impact the function and development of the central nervous system (CNS); however, little is known about the contribution of the microbiota during viral-induced neurologic damage. We identify that commensals aid in host defense following infection with a neurotropic virus through enhancing microglia function. Germfree mice or animals that receive antibiotics are unable to control viral replication within the brain leading to increased paralysis. Microglia derived from germfree or antibiotic-treated animals cannot stimulate viral-specific immunity and microglia depletion leads

The effects of micronutrient deficiencies on bacterial species from the human gut microbiota.

Vitamin and mineral (micronutrient) deficiencies afflict 2 billion people. Although the impact of these imbalances on host biology has been studied extensively, much less is known about their effects on the gut microbiota of developing or adult humans. Therefore, we established a community of cultured, sequenced human gut-derived bacterial species in gnotobiotic mice and fed the animals a defined micronutrient-sufficient diet, followed by a derivative diet devoid of vitamin A, folate, iron, or zinc, followed by return to the sufficient diet. Acute vitamin A deficiency had the largest effect on

Skin Vaccination against Rotavirus Using Microneedles: Proof of Concept in Gnotobiotic Piglets.

Live-attenuated oral rotavirus (RV) vaccines have lower efficacy in low income countries, and additionally are associated with a rare but severe adverse event, intussusception. We have been pursuing the development of an inactivated rotavirus vaccine (IRV) using the human rotavirus strain CDC-9 (G1P[8]) through parenteral immunization and previously demonstrated dose sparing and enhanced immunogenicity of intradermal (ID) unadjuvanted IRV using a coated microneedle patch in comparison with intramuscular (IM) administration in mice. The aim of this study was to evaluate the immune response and

Experimental induction of bacterial gastritis and gastric ulcer disease in gnotobiotic swine inoculated with porcine Helicobacter-like species.

OBJECTIVE: To determine whether 2 isolates of recently isolated swine-origin Helicobacter pylori-like bacteria are pathogenic in pigs and compare the signs of gastric disease induced by these isolates with those detected in H pylori- and Helicobacter heilmannii-infected pigs. ANIMALS: 36 neonatal gnotobiotic pigs. PROCEDURE: Groups of separately housed pigs were inoculated orally with swine-origin Helicobacter-like isolates 2662 or 1268, H pylori (human gastric pathogen), or a gastric homogenate from gnotobiotic swine containing H heilmannii. Noninoculated pigs were used as control animals.

The natural course of Clostridium perfringens--induced pneumatosis cystoides intestinalis.

Primary pneumatosis cystoides intestinalis (PCI) is an uncommon, usually benign condition whose natural course is poorly understood and which can sometimes produce significant changes in a patient's cecum and sigmoid colon. In this study, PCI was produced by monocontaminating the peritoneal cavities of adult germfree rats with Clostridium perfringens. These animals were then observed for up to 26 weeks. PCI took up to two weeks to develop, lasted at least ten weeks in most animals, and presumably disappeared from 42% of 26 animals killed during the final 16 weeks. PCI was usually benign, but i

Derivation of gnotobiotic ferrets: perinatal diet and hand-rearing requirements.

A procedure is described which has resulted in successful gnotobiotic derivation of the domestic ferret. The most critical element of this hand-rearing procedure was found to be diet, with ferret milk being required for at least the first 7 days. Puppy milk replacer was phased in during the next 10 days, and enriched cow's milk sufficed thereafter. Around-the-clock sip-feeding with fire-polished Pasteur pipettes was necessary at intervals gradually increasing from 1 to 1.5 hours at birth to 3 hours by day 21. Temperature regulation was accomplished with an electric heating pad placed eccentric

Pneumonic pasteurellosis induced experimentally in gnotobiotic and conventional calves inoculated with Pasteurella haemolytica.

Experimental pneumonia caused by Pasteurella haemolytica was induced in 2-week-old gnotobiotic (n = 4) and conventional (n = 6) calves by endobronchial inoculation into the right caudal lung lobe of 7.9 x 10(10) +/- 0.6 x 10(10) (mean +/- SD) colony-forming units of P haemolytica in the 6-hour log phase of growth. The calves were studied for 24 hours or less. Regression lines for the relationship between clinical index and time for the gnotobiotic group and conventional group of calves were compared, and the clinical index was found to be significantly (P less than or equal to 0.005) more rapi

An unusual localization of developmental stages of Cryptosporidium parvum Tyzzer, 1912 in the cells of small intestine of a gnotobiotic piglet.

In a gnotobiotic piglet infected with 50.10(6) oocysts of Cryptosporidium parvum, developmental stages of the parasite were found in the duodenum, anterior jejunum and middle jejunum on the first day post infection (1 DPI). During the studies of the ultrastructure, trophozoites and meronts of C. parvum were encountered in the microvillous zone of enterocytes, and the outer membrane of parasites was a continuation of the membrane of microvilli. Unusual was the attachment of C. parvum trophozoite on the opening of a goblet cell, as well as a meront localized in the cytoplasm in the enterocyte of

Resistance of mice with limited intestinal flora to enteric colonization by Clostridium botulinum.

Infant botulism is an age-dependent illness that is caused by the toxin produced by Clostridium botulinum infecting the intestinal tract. Because of composition of the intestinal microflora determines the resistance of mice to enteric colonization by C. botulinum, attempts were made to identify the kinds of bacteria that prevent this in vivo growth. Orogastric challenges of 50 spores of C. botulinum type A were given to adult germfree mice, which are highly susceptible, and to gnotobiotic adult mice carrying the eight species comprising the Charles River Altered Schaedler flora or two or three

Control of mousepox epizootics in St Louis and Chicago.

In September 1980, mousepox was diagnosed in mice from the Jewish Hospital in St Louis, Missouri. The disease was eliminated by vaccination with vaccinia virus. In October 1980, mousepox was suspected in mice located in an off-campus research facility of the University of Chicago. Based on history, clinical signs, serology, and characteristic pathological lesions, mousepox was diagnosed. The disease was eliminated by killing or moving all mice from the facility. During serological surveillance of mice from other University of Chicago campus facilities, mice from an on-campus facility were foun

Partition and absorption of valatile fatty acids in the alimentary canal of the rat.

The partition and absorption of volatile fatty acids (VFA) have been studied in the rat using a non purified diet comprising mainly a mixture of cereals and soya-bean meal. 2. The concentration of acetic and lactic acid was compared in non secretory (cul-de-sac) and secretory regions of the stomach. The greatest concentrations of both acids were found in the cul-de-sac region where the pH was highest (pH 5.0). 3. Concentration and absorption of VFA were negligible in the small intestine. 4. High VFA concentrations were found in the caecal contents, while lactic acid concentrations were lower.
