Cyanotoxin exposure as a novel risk factor for liver fibrosis and steatosis.

Cyanobacteria are ubiquitous environmental bacteria capable of producing cyanotoxins, including potent liver tumor promoters. There is limited knowledge of exposure to cyanotoxins and their potential role in the pathogenesis of liver cancer The relationship of cyanotoxin exposure with two hallmarks of progressive liver disease-liver fibrosis and steatosis-was evaluated in a cross-sectional study of 399 adult residents of the state of Hawaii and the US territory of Guam without a history of cancer. Microcystins (MC), nodularins (NOD), cylindrospermopsin (CYN), and anabaenopeptins (AB) were anal

Combined low calcium and lack magnesium is a risk factor for motor deficit in mice.

The populations of the Kii Peninsula in Japan and of Guam present high incidences of amyotrophic lateral sclerosis and Parkinsonism-dementia complex. It is thought that low levels of calcium (Ca) and magnesium (Mg) in the drinking water are involved in the pathogenesis of these diseases. The present study aimed to test the hypothesis that catalepsy, behavioral immobility and a Parkinsonian symptom results from functionally impaired dopaminergic neurons in mice fed low amounts of Ca and Mg (LCa/Mg). A group of mice fed a LCa/Mg diet for 6 weeks was compared to a control group on a standard diet

Reactions to less common species of fire ants.

BACKGROUND: There are four species of fire ants found in the United States in addition to the most common, Solenopsis invicta. Reactions have been reported from stings of each of these species, but large numbers of insects and adequate amounts of venom for study are very difficult to obtain. METHODS: Venom was obtained, the purified allergens were isolated, and the complete amino acid sequences were determined for two of the three allergens from S. richteri. Skin testing and RAST studies were performed on patients with reactions to native fire ant stings and analyzed in comparison with clinic

Determination of manganese concentrations in the spinal cords from amyotrophic lateral sclerosis patients by inductively coupled plasma emission spectroscopy.

Continuing analytical studies on environmental factors in the foci of amyotrophic lateral sclerosis (ALS) in the Kii Peninsula of Japan and Guam, and metal analysis using neutron activation analysis in central nervous system (CNS) tissues from ALS cases indicate that chronic exposure to metals such as aluminum and manganese, together with a deficiency of minerals such as calcium and magnesium, may play a causative role in the neurodegeneration seen in ALS. An accurate and simple method for detecting minerals and trace metals in small, fresh samples of CNS tissue is necessary in order to follow

Trichinella infection in travelers.

To define the incidence of trichinosis associated with foreign travel and characterize the epidemiologic and clinical features of cases acquired abroad, all case report forms submitted to the Centers for Disease Control through state health departments and the National Morbidity Reporting System from 1975 to 1989 were reviewed. Twenty-six cases of travel-associated trichinosis were identified during that period. Most reported cases (73%) occurred between 1982 and 1987. Affected patients were more likely to have traveled to Mexico and Asian countries (65%). Reported high-risk behaviors included

State-level prevalence of chronic conditions co-occurring with obesity among US adults.

AIMS: Recent estimates of chronic conditions (CCs) among adults with obesity are lacking. This study analysed state-representative data to assess the prevalence of CCs that co-occur with obesity. MATERIALS AND METHODS: We used 2021-2022 Behavioural Risk Factor Surveillance System data to assess the age-standardised prevalence of nine CCs among adults with obesity (body mass index ≥30 kg/m2) based on self-reported weight and height. We examined prevalence overall and by key sociodemographic factors, obesity class, and states and territories, using t-tests for subgroup comparisons. RESULTS: Th

Unveiling health inequalities and frontier gaps in elderly-onset rheumatoid arthritis: evolving impact of smoking and future challenges.

BACKGROUND: Elderly-onset rheumatoid arthritis (EORA) poses a growing public health burden worldwide. Significant health inequalities and frontier gaps persist across countries, while the impact of smoking on EORA has evolved over time. METHODS: Using data from the 2021 Global Burden of Disease data, we assessed the incidence, prevalence, mortality, and disability-adjusted life years (DALYs) of EORA and analyzed trends by calculating the estimated annual percentage changes. We explored associations with the sociodemographic index (SDI), evaluated frontier gaps, quantified health inequalities,

AMPK is abnormally activated in tangle- and pre-tangle-bearing neurons in Alzheimer's disease and other tauopathies.

Tauopathies represent a class of neurodegenerative disorders characterized by abnormal tau phosphorylation and aggregation into neuronal paired helical filaments (PHFs) and neurofibrillary tangles. AMP-activated protein kinase (AMPK) is a metabolic sensor expressed in most mammalian cell types. In the brain, AMPK controls neuronal maintenance and is overactivated during metabolic stress. Here, we show that activated AMPK (p-AMPK) is abnormally accumulated in cerebral neurons in 3R+4R and 3R tauopathies, such as Alzheimer's disease (AD), tangle-predominant dementia, Guam Parkinson dementia comp

Aberrant CHCHD2-associated mitochondriopathy in Kii ALS/PDC astrocytes.

Amyotrophic Lateral Sclerosis/Parkinsonism-Dementia Complex (ALS/PDC), a rare and complex neurological disorder, is predominantly observed in the Western Pacific islands, including regions of Japan, Guam, and Papua. This enigmatic condition continues to capture medical attention due to affected patients displaying symptoms that parallel those seen in either classical amyotrophic lateral sclerosis (ALS) or Parkinson's disease (PD). Distinctly, postmortem examinations of the brains of affected individuals have shown the presence of α-synuclein aggregates and TDP-43, which are hallmarks of PD and

Retroviral Elements in Pathophysiology and as Therapeutic Targets for Amyotrophic Lateral Sclerosis.

The study of the role of retroviruses in amyotrophic lateral sclerosis (ALS) dates back to the 1960s shortly after transposable elements themselves were first discovered. It was quickly realized that in wild mice both horizontal and vertical transmissions of retroviral elements were key to the development of an ALS-like syndrome leading to the postulate that endogenous retroviruses (ERVs) contribute significantly to the pathogenicity of this disease. Subsequent studies identified retroviral reverse transcriptase activity in brains of individuals with ALS from Guam. However, except for a single

The iron-binding protein lactotransferrin is present in pathologic lesions in a variety of neurodegenerative disorders: a comparative immunohistochemical analysis.

Lactotransferrin is a glycoprotein that specifically binds and transports iron. This protein is also believed to transport other metals such as aluminum. Several lines of evidence indicate that iron and aluminum are involved in the pathogenesis of many dementing diseases. In this context, the analysis of the iron-binding protein distribution in the brains of patients affected by neurodegenerative disorders is of particular interest. In the present study, the distribution of lactotransferrin was analyzed by immunohistochemistry in the cerebral cortex from patients presenting with Alzheimer's di

Qualitative risk assessment of Japanese encephalitis virus introduction and transmission in Pacific Island countries and territories.

BACKGROUND: The emergence of Japanese encephalitis virus (JEV) in Australia raises questions about transmission potential across neighbouring Pacific Island countries and territories (PICs), where surveillance infrastructure and diagnostic capacity are limited. METHODOLOGY: We conducted a qualitative risk assessment of 22 PICs structured around three components: (1) 'release assessment', evaluating JEV introduction pathways based on geographic proximity, migratory waterbird flyway connectivity, and windborne mosquito dispersal potential; (2) 'exposure assessment', evaluating ecological transm

"Smoking weed it gets you over the hump": Cannabis co-use as a facilitator of decreased opioid use among people who inject drugs in Los Angeles, California.

INTRODUCTION: Opioid overdose mortality rates have surged dramatically in the last decade due largely to fentanyl in the illicit US drug supply. As of June 2024, 38 states, three territories, namely US Virgin Islands, Guam and the Northern Mariana Islands, and the District of Columbia, allow the medical use of cannabis products. However, there remains limited qualitative community-based evidence on the role of cannabis co-use among opioid using and injecting populations. In this study, we present data from people who inject drugs (PWID)'s co-use of cannabis-opioid. METHODS: We conducted 30 on

Age-specific case data reveal varying dengue transmission intensity in US states and territories.

Dengue viruses (DENV) are endemic in the US territories of Puerto Rico, American Samoa, and the US Virgin Islands, with focal outbreaks also reported in the states of Florida and Hawaii. However, little is known about the intensity of dengue virus transmission over time and how dengue viruses have shaped the level of immunity in these populations, despite the importance of understanding how and why levels of immunity against dengue may change over time. These changes need to be considered when responding to future outbreaks and enacting dengue management strategies, such as guiding vaccine dep

Bioimpedence to Assess Breast Density as a Risk Factor for Breast Cancer in Adult Women and Adolescent Girls.

Although high mammographic density is one of the strongest predictors of breast cancer risk, X-ray based mammography cannot be performed before the recommended screening age, especially not in adolescents and young women. Therefore, new techniques for breast density measurement are of interest. In this pilot study in Guam and Hawaii, we evaluated a radiation-free, bioimpedance device called Electrical Breast DensitometerTM (EBD; senoSENSE Medical Systems, Inc., Ontario, Canada) for measuring breast density in 95 women aged 31-82 years and 41 girls aged 8-18 years. Percent density (PD) was esti

Guadeloupean parkinsonism: a cluster of progressive supranuclear palsy-like tauopathy.

An unusually high frequency of atypical Parkinson syndrome has been delineated over the last 5 years in the French West Indies. Postural instability with early falls, prominent frontal lobe dysfunction and pseudo-bulbar palsy were common and three-quarters of the patients were L-dopa unresponsive. One-third of all patients seen had probable progressive supranuclear palsy (PSP). This new focus of atypical parkinsonism is reminiscent of the one described in Guam and may be linked to exposure to tropical plants containing mitochondrial complex I inhibitors (quinolines, acetogenins, rotenoids). The first NINDS/NIBIB consensus meeting to define neuropathological criteria for the diagnosis of chronic traumatic encephalopathy.

Chronic traumatic encephalopathy (CTE) is a neurodegeneration characterized by the abnormal accumulation of hyperphosphorylated tau protein within the brain. Like many other neurodegenerative conditions, at present, CTE can only be definitively diagnosed by post-mortem examination of brain tissue. As the first part of a series of consensus panels funded by the NINDS/NIBIB to define the neuropathological criteria for CTE, preliminary neuropathological criteria were used by 7 neuropathologists to blindly evaluate 25 cases of various tauopathies, including CTE, Alzheimer's disease, progressive su
