Nascent preplatelets and B4GALT1 glycosylation contribute to 5-FU-induced bone marrow recovery.

Megakaryocytes (MKs) are essential for hemostasis, vascular integrity, and hematopoietic stem and progenitor cell (HSPC) support, but their role in bone marrow (BM) recovery has remained underexplored. Here, using a 5-fluorouracil (5-FU) injury model, we identify a transient intramedullary space enriched in extracellular matrix (ECM) proteins such as perlecan, von Willebrand factor, and heparanase, as well as functionally responsive GPIbα+ platelet particles (termed preplatelets). During 5-FU-induced injury, this compartment undergoes dynamic changes, and its resolution depends on the presence

The importance of endothelial protection: the emerging role of defibrotide in reversing endothelial injury and its sequelae.

Hepatic veno-occlusive disease/sinusoidal obstruction syndrome (VOD/SOS), a potentially life-threatening complication of hematopoietic cell transplantation (HCT), results from prolonged sinusoidal endothelial cell activation and profound endothelial cell damage, with sequelae. Defibrotide, the only drug approved in the United States and Europe for treating VOD/SOS post-HCT, has European Commission orphan drug designation for preventing graft-versus-host disease (GvHD), associated with endothelial dysfunction. This endothelial cell protector and stabilizing agent restores thrombo-fibrinolytic b

Towards gene therapy of postoperative adhesions: fiber and transcriptional modifications enhance adenovirus targeting towards human adhesion cells.

Postoperative abdominal/pelvic peritoneal adhesions are a major source of morbidity (bowel obstruction, infertility, ectopic gestation as well as chronic pelvic pain) in women. In this study, we screened various transduction and transcription modifications of adenovirus (Ad) to identify those that support maximal Ad-mediated gene delivery to human adhesion fibroblasts, which in turn would enhance the efficacy of this novel treatment/preventative strategy for postoperative adhesions. We transduced primary cultures of human peritoneal adhesion fibroblasts with fiber-modified Ad vectors Ad5-RGD-l

Prophylactic Antiheparanase Activity by PG545 Is Antiviral In Vitro and Protects against Ross River Virus Disease in Mice.

Recently we reported on the efficacy of pentosan polysulfate (PPS), a heparan sulfate mimetic, to reduce the recruitment of inflammatory infiltrates and protect the cartilage matrix from degradation in Ross River virus (RRV)-infected PPS-treated mice. Here, we describe both prophylactic and therapeutic treatment with PG545, a low-molecular-weight heparan sulfate mimetic, for arthritogenic alphaviral infection. We first assessed antiviral activity in vitro through a 50% plaque reduction assay. Increasing concentrations of PG545 inhibited plaque formation prior to viral adsorption in viral strai

Clinical and genetic characteristics for the Urofacial Syndrome (UFS).

The Urofacial (Ochoa) Syndrome (UFS) is a rare autosomal recessive disorder and over 100 patients have been reported thus far. UFS is characterized by the abnormal facial expression and dysfunctional voiding. The patients show a peculiar distortion of the facial expression (grimacing as if in pain or sadness when they tried to smile or laugh) along with urinary tract infection, enuresis, vesicoureteral reflux and hydronephrosis without any underlying neurological lesion and previous urinary obstruction. Some patients are also noted with nocturnal lagophthalmos. Until 2010, HPSE2, the gene enco

Skipjack tuna peptides exert sleep-promoting effects by regulating neurotransmitter metabolism in pentobarbital sodium-induced mice.

It is estimated that sleep disorders affect approximately 35% of the global population, thus becoming the second most prevalent mental health disorder. In this work, the sleep-promoting effect of skipjack tuna peptide GHHAA (S4) was evaluated systematically in pentobarbital sodium (PS)-induced mice, and its mechanism was explored through measuring the content changes of sleep-related neurotransmitters and c-Fos expression levels in mouse brain regions. In comparison with the control group, S4 was able to dose-dependently reduce the sleep latency (6.33 ± 1.15 min) and prolong sleep time (60.33

A universal stress protein upregulated by hypoxia has a role in Burkholderia cenocepacia intramacrophage survival: Implications for chronic infection in cystic fibrosis.

Universal stress proteins (USPs) are ubiquitously expressed in bacteria, archaea, and eukaryotes and play a lead role in adaptation to environmental conditions. They enable adaptation of bacterial pathogens to the conditions encountered in the human niche, including hypoxia, oxidative stress, osmotic stress, nutrient deficiency, or acid stress, thereby facilitating colonization. We previously reported that all six USP proteins encoded within a low-oxygen activated (lxa) locus in Burkholderia cenocepacia showed increased abundance during chronic colonization of the cystic fibrosis (CF) lung. Impairment of Flow-Sensitive Inwardly Rectifying K+ Channels via Disruption of Glycocalyx Mediates Obesity-Induced Endothelial Dysfunction.

OBJECTIVE: To determine if endothelial dysfunction in a mouse model of diet-induced obesity and in obese humans is mediated by the suppression of endothelial Kir (inwardly rectifying K+) channels. Approach and Results: Endothelial dysfunction, observed as reduced dilations to flow, occurred after feeding mice a high-fat, Western diet for 8 weeks. The functional downregulation of endothelial Kir2.1 using dominant-negative Kir2.1 construct resulted in substantial reductions in the response to flow in mesenteric arteries of lean mice, whereas no effect was observed in arteries of obese mice. Circulating heparin oligosaccharides rapidly target the hippocampus in sepsis, potentially impacting cognitive functions.

Sepsis induces heparanase-mediated degradation of the endothelial glycocalyx, a heparan sulfate-enriched endovascular layer critical to vascular homeostasis, releasing highly sulfated domains of heparan sulfate into the circulation. These domains are oligosaccharides rich in heparin-like trisulfated disaccharide repeating units. Using a chemoenzymatic approach, an undecasaccharide containing a uniformly 13C-labeled internal 2-sulfoiduronic acid residue was synthesized on a p-nitrophenylglucuronide acceptor. Selective periodate cleavage afforded a heparin nonasaccharide having a natural structu

Proteoglycans as miscommunication biomarkers for cancer diagnosis.

Cancer cells reside in a microenvironment comprising of fibroblasts, endothelial cells, pericytes, macrophages, and other immune cells. All these cells coevolve with the cancer cells into a clinically manifested tumor. The immune system of the host should eliminate the tumor but fails to do so until it develops into a deadly disease. Based on these facts, cancer is a system disorder caused by miscommunications among cancer cells, its microenvironment, and host immune system. Therefore, identifying communication-related biomarkers will be important for cancer diagnosis and treatment. Proteoglyc

Putative biological mechanisms of efficiency of substrate reduction therapies for mucopolysaccharidoses.

Mucopolysaccharidoses (MPS) are inherited metabolic diseases caused by mutations in genes coding for lysosomal enzymes involved in the degradation of glycosaminoglycans (GAGs). Dysfunction of any of these enzymes results in the accumulation of GAGs, which leads to severe clinical symptoms and significantly shortened life span. Several kinds of therapies have been proposed to treat MPS, including bone marrow or stem cell transplantation, enzyme replacement therapy, and gene therapy. Another option is substrate reduction therapy (SRT), in which synthesis of GAGs is inhibited. Recent studies empl

Actions of heparin that may affect the malignant process.

BACKGROUND: Heparin has many actions that may affect the malignant process, especially metastasis. METHODS: The author conducted an extensive review of the available medical literature about heparin activity that may apply to important factors involved in the malignant process. RESULTS: Thrombin is generated by tumors, and the resultant fibrin formation impedes natural killer cell activity. Microthrombi arrest tumor cells in capillaries. Heparin prevents the formation of thrombin and neutralizes its activity. Angiogenesis has an important role in metastasis; heparin minimizes angiogenesis vi

Heparan sulfate sulfation modification in airway mucosal barrier: biosynthetic regulation, pathological implications, and therapeutic targets in allergic respiratory diseases.

Heparan sulfate (HS) is a highly sulfated glycosaminoglycan essential for airway mucosal barrier function. Its biological activities are determined by the "sulfation code" generated through N-deacetylase/N-sulfotransferases (NDST1-4) and O-sulfotransferases. Emerging evidence indicates that HS sulfation operates within a narrow homeostatic range - a "rheostat" - where both insufficient and excessive sulfation disrupt barrier integrity and promote inflammation. This review proposes the HS sulfation rheostat model as a unifying framework for allergic respiratory diseases. We examine the HS biosy

Endothelial glycocalyx in perioperative medicine current understanding and future direction.

The endothelial glycocalyx (EG) is a dynamic, gel-like layer that lines the luminal surface of blood vessels, playing a crucial role in vascular biology. Previously considered a passive barrier, it is now recognized as a key regulator of vascular tone, permeability, inflammation, and coagulation. Composed mainly of proteoglycans, glycoproteins, and glycosaminoglycans, the EG serves as a semipermeable interface between blood and the endothelium, maintaining microvascular flow and modulating nitric oxide (NO) production while protecting against oxidative and inflammatory damage. This review high

From Physiology to Clinical Practice in Pancreatic Cancer-Related Thromboembolism-A Review.

Pancreatic cancer (PC) is a highly lethal malignancy linked to the highest rate of thromboembolic complications (TEC) among all solid tumors. TECs occur in approximately 5-40% of PC patients. The most common type of TEC in PC is venous thromboembolism (VTE). The mechanisms leading to frequent TEC in PC are complex and involve interactions between tumor-derived procoagulant factors and the prothrombotic tumor microenvironment (TME). Secretion of tissue factor and proinflammatory cytokines by tumor cells and the TME, overexpression of heparanase and podoplanin, impaired fibrynolysis and increase

EXTL3 dysfunction identified as a driver of aberrant bone development in severe familial ankylosing spondylitis.

OBJECTIVES: Pathological ossification leading to spinal ankylosis is a hallmark of ankylosing spondylitis (AS), yet its molecular mechanisms remain poorly understood. We investigated the genetic basis and functional consequences of severe familial AS to identify pathways driving structural progression. METHODS: Genome-wide linkage analysis and whole-exome sequencing were performed in a multiplex family with severe AS. A heterozygous knock-in mouse carrying the identified EXTL3 variant was generated and characterized using microCT, histology, transcriptomics, primary osteoblast and chondrocyte
