The vagus nerve: a tonic inhibitory influence associated with inflammatory bowel disease in a murine model.

BACKGROUND & AIMS: The recently proposed Inflammatory Reflex describes an interaction between the vagus nerve and peripheral macrophages, resulting in attenuation of proinflammatory cytokine release in response to systemic exposure to bacterial endotoxin. The purpose of this study was to determine whether a similar vagus/macrophage axis modulates the inflammatory responses in the colon in mice. METHODS: We assessed the Disease Activity Index (DAI), macroscopic and histologic damage, serum amyloid-P level, and myeloperoxidase activity in colitis induced by administration of dextran sodium sulf

Innervation of the cat lip by two groups of parasympathetic vasodilator fibres.

1. Electrical stimulation of the peripheral cut ends of the chorda tympani nerve proper (CTNP) and the chorda-lingual nerve (CLN) elicited a blood flow increase in the ipsilateral lower lip, tongue and submandibular gland in a stimulus intensity-dependent manner in anaesthetized cats. 2. Pretreatment with hexamethonium (1.0 mg kg-1, i.v.), an autonomic ganglionic blocker, significantly reduced the CTNP-induced blood flow increases in all of the above three sites as well as the CLN-induced blood flow in the lower lip, but it had no effects on the CLN-induced blood flow increases in the tongue a

Control of interdigestive and intraduodenal meal-stimulated pancreatic secretion in rats.

Mechanisms of neural (vagal and cholinergic) and hormonal [cholecystokinin (CCK)] control of pancreatic exocrine secretion were studied in basal interdigestive conditions and after stimulation by an intraduodenal meal in rats equipped with a semichronic pancreatic fistula. Bile was recirculated into the duodenum, and a solution of trypsin and electrolytes was continuously infused. Pancreatic secretion was compared in control experiments, after vagotomy, and after venous infusion of cholinergic and CCK antagonists. Basal pancreatic secretion was decreased by atropine, pirenzepine, and hexametho

A cholinergic link in the reflex release of vasopressin by hypotension in the rat.

Inhalation of amyl nitrite in the water-loaded rat under ethanol anaesthesia produced a brief fall of blood pressure followed by a prolonged antidiuretic response. The antidiuretic response to amyl nitrite was accompanied by increased urinary excretion of vasopressin, it was blocked by a specific vasopressin antagonist and by a barbiturate and it was absent in the Brattleboro rat with congenital diabetes insipidus. These results show that the antidiuretic response to the hypotension induced by amyl nitrite is due to the release of vasopressin and that this release is mediated by a neuroendocri

Nicotine-induced relaxation in isolated canine cerebral arteries.

Nicotine in concentrations ranging from 5 times 10- minus 6 to 10- minus 4 M caused a transient relaxation in strips of isolated canine basilar, middle cerebral and posterior cerebral arteries contracted with prostaglandin F2a (PGF). Transmural neural stimulation did not produce changes in the tension of these arteries. Tyramine contracted the arterial strips. Relaxation induced by nicotine was markedly attenuated by hexamethonium (10- minus 5 M), cocaine (10- minus 5M), bretylium (2 times 10- minus 5 M), sotalol (5 times 10- minus 5 M), propranolol (10- minus 6 M) and removal of Ca-++ from th

Sir Horace Smirk. Pioneer in drug treatment of hypertension.

Sir Horace Smirk deserves much of the credit for establishing the benefits of reducing blood pressure. He was one of the main early proponents of the idea that it was the raised intra-arterial pressure itself that caused many of the cardiac and vascular complications of hypertension. His training in both pharmacology and internal medicine enabled him to devise practical methods for successful treatment of hypertension with ganglion blocking drugs such as hexamethonium. The major clinical benefits that followed such drug treatment proved a great stimulus to the development of drugs with fewer d

Effects of various anesthetic and autonomic drugs on refraction in monkeys.

Resting refractive correction in ketamine-, pentobarbital-, or halothane-anesthetized rhesus and cynomolgus monkeys was approximately 1-3 diopters myopic, with little difference under the various anesthetic regimens. Topical cyclopentolate or atropine, or systemic hexamethonium eliminated much of the myopia, while epinephrine, phenylephrine and thymoxamine had little effect. Anesthesia-induced myopia in monkeys thus seems comparable to tonic accommodation ("night myopia") in the human. Accommodation induced by electrical stimulation of the Edinger-Westphal nucleus averaged approximately 50% le

Effects of cholinomimetic drugs and their antagonists injected into vertebral artery of unanaesthetized dogs.

In unanaesthetized dogs, cholinomimetic drugs and their antagonists, catecholamines and 5HT were injected into vertebral artery placed in a skin loop. Acetylcholine (1 mg), pilocarpine (1-2 mg), nicotine (250-500 microng), eserine (100 microng) and neostigmine (250 microng) produced sleep apart from a few other peripheral effects. Sleep for longer periods followed injections of tubocurarine (1 mg), atropine (50 microng) and hexamethonium (500 microng). Adrenaline (50 microng) and noradrenaline (50-250 microng) did not produce significant effects on behaviour and sleep. 5HT (250-500 microng) al

Co-dependence of the neural and humoral pathways in the mechanism of remote ischemic conditioning.

The cardioprotection afforded by remote ischaemic conditioning (RIC) is mediated via a complex mechanism involving sensory afferent nerves, the vagus nerve, and release of a humoral blood-borne factor. However, it is unknown whether release of the protective factor depends on vagal activation or occurs independently. This study aimed to evaluate the co-dependence of the neural and humoral pathways of RIC, focussing on the vagus nerve and intrinsic cardiac ganglia. In the first study, anesthetised rats received bilateral cervical vagotomy or sham-surgery immediately prior to RIC (4 × 5 min limb

Spontaneous pressure-flow patterns in the kidney of conscious rats.

We have developed a model that permits a quantitative analysis of the contribution of different mechanisms to the spontaneously occurring pressure-flow patterns of a vasculature. In this study we evaluated the spontaneous relationship between arterial pressure (P) and renal blood flow (F) in resting conscious rats during control conditions, autonomic ganglionic blockade (hexamethonium), and nonselective alpha-adrenoreceptor blockade (phentolamine). In a total of 250 trials in 29 rats, we measured the average P and F for each cardiac cycle over 13-min periods (approximately 4,000 cardiac cycles

Neural pathways and pharmacological modulation of defecation reflex in rats.

1. A new method was developed for quantitative studies on defecation reflex in urethane-anesthetized rats which involves the continuous infusion of saline (0.1 ml/min) to distend a balloon placed in the rectum. At threshold values, the balloon was expelled during an active rectal contraction. 2. Balloon expulsion was greatly delayed or even abolished by i.v. hexamethonium. Cord transection at the upper cervical level increased defecation threshold but a functional, hexamethonium-sensitive response was still elicited in spinal rats. 3. Various parameters of the defecation response were modulate

Parasympathetic-mediated reflex salivation and vasodilatation in the cat submandibular gland.

The aim of the present study was to compare and characterize the secretory and vasodilator effects induced by chorda lingual nerve (CLN) stimulation (i.e., direct parasympathetic stimulation) and reflex parasympathetic stimulation in the submandibular gland (SMG) of sympathectomized cats. The increase in blood flow and salivary secretion in response to electrical stimulation of the central cut ends of the vagus and inferior alveolar nerves, as well as to stimulation of the CLN at a site approximately 5 mm distal to the intersection of the CLN and the SMG duct (site D) was completely abolished

Cholinergic interneurons in the feeding system of the pond snail Lymnaea stagnalis. II. N1 interneurons make cholinergic synapses with feeding motoneurons.

The N1 neurons are a population of interneurons active during the protraction phase of the feeding rhythm. All the N1 neurons are coupled by electrical synapses which persist in a high Mg/low Ca saline which blocks chemical synapses. Individual N1 spikes produce discrete electrotonic postsynaptic potentials (PSPS) in other N1 cells, but the coupling is not strong enough to ensure 1:1 firing. Bursts of N1 spikes generate compound PSPS in the feeding motoneurons. The sign (excitation or inhibition) of the N1 input corresponds with the synaptic barrage recorded during the protraction phase. Fluid secretion by gallbladder mucosa in experimental cholecystitis is influenced by intramural nerves.

The normal fluid absorption across the gallbladder mucosa is, in experimental cholecystitis, changed to an active net fluid secretion. This fluid secretion, studied in anesthetized cats, is not abolished by extrinsic gallbladder denervation and is unaffected by atropine but is strongly reduced by intraarterial tetrodotoxin or intraluminal administration of lidocaine hydrochloride. Intravenous somatostatin or hexamethonium administration also reduce this secretion. Indomethacin, known to abolish this fluid secretion, did not further reduce it when administered after nerve blocking agents in the

Effects of morphine and methionine-enkephalin on the smooth muscle tonus and the contraction induced by transmural stimulation in the carp (Cyprinus carpio) intestinal bulb.

The effects of morphine and methionine-enkephalin (met-enkephalin) on the smooth muscle tonus and the contraction induced by transmural stimulation were investigated in the isolated intestinal bulb of carp in vitro. Morphine (30 nM-3 microM) and met-enkephalin (3 nM-5 microM) caused dose-dependent non-sustained contraction. Naloxone (10 nM) inhibited the contraction induced by morphine or met-enkephalin in a competitive manner. Tetrodotoxin (400 nM) or atropine (500 nM) did not inhibit the contraction induced by morphine or met-enkephalin. Cooling of the bath fluid from 20 to 10 degrees C decr

On the nature of the contractile motor responses of the rat stomach elicited by serotonin or substance P in vitro.

SP or 5-HT elicited contractile motor responses from strip preparations of the rat antrum and pylorus, effects which were efficiently antagonized by blockade directed against each substance (SP-receptors: tachyphylaxis or a SP analogue; 5-HT2 receptors: ketanserin). Indirect effects of SP or 5-HT, i.e. causing the release of other spasmogens such as histamine (from mast cells) and prostaglandins, were ruled out in separate experiments using pyrilamine or indomethacin. When the contractile effects of SP or 5-HT motor responses were studied after TTX no inhibition was demonstrated indicating tha

Effects of peripherally administered GABA and other amino acids on cardiopulmonary responses in anesthetized rats and dogs.

Intravenously administered GABA (1.0-1000 micrograms/kg) induced hypotension, bradycardia and tachypnea in both dogs and rats. The increased respiratory rate was not caused by changes in blood pH, pCO2, or pO2. None of the effects of GABA were modified by the following agents: propranolol, atropine, diphenhydramine, or hexamethonium. Taurine, glycine, beta-alanine, glutamic acid and aspartic acid at doses of 1000 micrograms/kg, i.v., had no significant effects on any of the variables measured. Since these doses of GABA probably do not cross the blood-brain barrier, it is postulated that the ob

Intrinsic gamma aminobutyric acid receptors modulate the release of catecholamine from canine adrenal gland in situ.

Immunohistochemical analysis documented the presence of gamma-aminobutyric acid (GABA)-containing fibers and GABA-containing chromaffin cells in canine adrenal glands. A dense network of fibers was visualized at the boundary between medullary and cortical cells, and, in the medullary tissue, GABA-containing fibers surrounded chromaffin cells. Some of these fibers enter the adrenal medulla together with splanchnic cholinergic nerves. The functional role of the GABAergic system in the regulation of catecholamine release from adrenal chromaffin cells was studied in canine adrenal glands in situ,

On the two subdivisions and intrinsic synaptic connexions in the submandibular ganglion of the rat.

Parasympathetic neurones in the submandibular ganglion of the rat innervate the submandibular and sublingual salivary glands. Neurones which innervate the submandibular gland (s.m. neurones) are usually located along the salivary ducts which drain both glands. Neurones which innervate the sublingual gland (s.l. neurones) are located in the thin sheet of tissue which lies between the salivary ducts and the lingual nerve. The existence and characteristics of intrinsic synaptic connexions were studied electrophysiologically in these two divisions of the submandibular ganglion. Three days or more
