Induction of Biofilm Formation in Klebsiella pneumoniae ATCC 13884 by Several Drugs: The Possible Role of Quorum Sensing Modulation.

Bacterial resistance is caused by several biochemical factors, the formation of biofilm being one of the main causes. This process is triggered by Quorum Sensing (QS), through the production of endogenous molecules, although other substances such as natural products can also do this. In this work, we aimed to determine whether some drugs are involved in the induction of biofilm formation in Klebsiella pneumoniae ATCC 13884, and thus, increase bacterial resistance. For this, the effect of 22 drugs on K. pneumoniae ATCC 13884 growth was determined at sub-plasmatic concentrations; the production

Attenuation of quorum-sensing-dependent virulence factors and biofilm formation by medicinal plants against antibiotic resistant Pseudomonas aeruginosa.

Pseudomonas aeruginosa use small signaling molecules such as acyl homoserine lactones (AHLs), which play an important role in release virulence factors and toxin for further establishment of host infection. Thus, involving with the QS system would provide alternative ways of preventing the pathogenicity. In the present study, totally six medicinal plants (Terminalia bellerica, Celastrus paniculatus, Kingiodendron pinnatum, Schleichera oleosa, Melastoma malabathricum, Garcinia gummi-gutta) were screened for anti-QS activity using biomonitor strain of Chromobacterium violaceum CV12472. The prima

Essential roles of methionine and S-adenosylmethionine in the autarkic lifestyle of Mycobacterium tuberculosis.

Multidrug resistance, strong side effects, and compliance problems in TB chemotherapy mandate new ways to kill Mycobacterium tuberculosis (Mtb). Here we show that deletion of the gene encoding homoserine transacetylase (metA) inactivates methionine and S-adenosylmethionine (SAM) biosynthesis in Mtb and renders this pathogen exquisitely sensitive to killing in immunocompetent or immunocompromised mice, leading to rapid clearance from host tissues. Mtb ΔmetA is unable to proliferate in primary human macrophages, and in vitro starvation leads to extraordinarily rapid killing with no appearance of

Paraoxonase-2 modulates stress response of endothelial cells to oxidized phospholipids and a bacterial quorum-sensing molecule.

OBJECTIVE: Chronic infection has long been postulated as a stimulus for atherogenesis. Pseudomonas aeruginosa infection has been associated with increased atherosclerosis in rats, and these bacteria produce a quorum-sensing molecule 3-oxo-dodecynoyl-homoserine lactone (3OC12-HSL) that is critical for colonization and virulence. Paraoxonase 2 (PON2) hydrolyzes 3OC12-HSL and also protects against the effects of oxidized phospholipids thought to contribute to atherosclerosis. We now report the response of human aortic endothelial cells (HAECs) to 3OC12-HSL and oxidized 1-palmitoyl-2-arachidonoyl-

Pseudomonas aeruginosa AlgR represses the Rhl quorum-sensing system in a biofilm-specific manner.

AlgR controls numerous virulence factors in Pseudomonas aeruginosa, including alginate, hydrogen cyanide production, and type IV pilus-mediated twitching motility. In this study, the role of AlgR in biofilms was examined in continuous-flow and static biofilm assays. Strain PSL317 (DeltaalgR) produced one-third the biofilm biomass of wild-type strain PAO1. Complementation with algR, but not fimTU-pilVWXY1Y2E, restored PSL317 to the wild-type biofilm phenotype. Comparisons of the transcriptional profiles of biofilm-grown PAO1 and PSL317 revealed that a number of quorum-sensing genes were upregul

Trace Hydrogen Sulfide Enhances Plasmid-Mediated Antibiotic Resistance Gene Transfer under Anaerobic Sewer-Like Conditions: Associations with Contact and Energy-State Responses.

The dissemination of antibiotic resistance genes (ARGs) in sewer systems poses a potential threat to public health. However, it is currently unclear how hydrogen sulfide (H2S) in anaerobic sewers affects plasmid-mediated ARG transfer. Here, we evaluated the effects of environmentally relevant concentrations (1-100 μmol·L-1) of H2S on ARG transfer across different sewer microbial communities. The results demonstrated that H2S significantly enhanced the transfer frequency of plasmid pKJK5 in sediment-derived (2.06-3.06-fold), sewage-derived (1.50-2.81-fold), and bioaerosol-derived (2.42-4.68-fol

Quorum sensing molecule C8-HSL enhances Chlorella growth and lipid accumulation via ROS mediated metabolic reprogramming.

Quorum sensing molecules play important roles in microbial metabolic regulation; however, their involvement in microalgal carbon allocation remains poorly understood. In this study, the effects of N-octanoyl-l-homoserine lactone (C8-HSL) on Chlorella sp. growth and lipid accumulation were systematically investigated, with particular focus on the involvement of reactive oxygen species (ROS). C8-HSL significantly promoted cell proliferation, resulting in a 30.9% increase in cell density at the late cultivation stage relative to the untreated control. It also enhanced lipid synthesis while mainta

Genomic Analysis of Pseudomonas aeruginosa Recovered From Surgical Site Infections From a Referral Hospital in Western Kenya Reveals Dominance of High-Risk Subtypes.

Pseudomonas aeruginosa is an important cause of surgical site infections (SSIs) and is characterized by extensive antimicrobial resistance and genomic plasticity. We analyzed 13 whole-genome sequences of clinical P. aeruginosa isolates recovered from SSIs at a Level 5 referral hospital in Western Kenya and integrated genomic analyses with antimicrobial susceptibility tests and phenotyping of quorum-sensing (QS). Multidrug resistance was common, with resistance observed primarily against ciprofloxacin, piperacillin, ceftazidime, and amikacin, whereas meropenem and piperacillin/tazobactam retain

MrtR of Mesorhizobium tianshanense reveals both activation and inhibition mechanisms of a LuxR-type quorum sensing receptor.

Quorum sensing (QS) enables common gram-negative bacteria to coordinate collective behaviors through small molecule signals, yet how these signals tune receptor activity remains incompletely understood. Here, we define a mechanism by which ligand structure controls function in a LuxR-type QS receptor. Using structural and biochemical analyses, we investigate MrtR from Mesorhizobium tianshanense and show that ligand acyl-chain length governs receptor assembly and activity. We present full-length structures of MrtR bound to activating and inhibitory ligands, revealing a switch in oligomeric stat

The Quorum-Sensing Regulator SdiA Activates npsA Expression and Modulates Cytotoxicity in Klebsiella oxytoca.

Toxigenic Klebsiella oxytoca strains linked to antibiotic-associated hemorrhagic colitis produce the cytotoxins tilimycin and tilivalline, which contribute to intestinal epithelial damage during infection. Tilimycin and tilivalline are synthesized by enzymes encoded within the nonribosomal peptide synthetase (NRPS) operon, yet the regulatory mechanisms controlling operon expression remain poorly understood. SdiA, an orphan LuxR-type quorum-sensing regulator, detects exogenous N-acyl homoserine lactones (AHLs) produced by neighboring bacterial species and modulates gene expression in response t

Pseudomonas aeruginosa lasR Mutants Resist Phagocytosis and Alter Inflammatory Cytokine Production by Cystic Fibrosis Macrophages.

Cystic Fibrosis is characterized by chronic muco-obstructive lung disease and infection. People with CF (pwCF) are often colonized with Pseudomonas aeruginosa for years to decades, allowing for evolutionary adaptation. In chronic P. aeruginosa lung isolates from pwCF, the quorum sensing regulator LasR frequently is nonfunctional, however the factors enabling lasR loss-of-function (LOF) mutant selection are incompletely understood. We hypothesized that LOF mutations in lasR could allow P. aeruginosa to resist the selective pressure of phagocytosis. We found that in multiple strain backgrounds,

Combinatorial discovery of microtopographical landscapes that resist biofilm formation through quorum sensing mediated autolubrication.

Bio-instructive materials that intrinsically inhibit biofilm formation have significant anti-biofouling potential in industrial and healthcare settings. Since bacterial surface attachment is sensitive to surface topography, we experimentally surveyed 2176 combinatorially generated shapes embossed into polymers using an unbiased screen. This identified microtopographies that, in vitro, reduce colonization by pathogens associated with medical device-related infections by up to 15-fold compared to a flat polymer surface. Machine learning provided design rules, based on generalisable descriptors,

Interkingdom Detection of Bacterial Quorum-Sensing Molecules by Mammalian Taste Receptors.

Bitter and sweet taste G protein-coupled receptors (known as T2Rs and T1Rs, respectively) were originally identified in type II taste cells on the tongue, where they signal perception of bitter and sweet tastes, respectively. Over the past ~15 years, taste receptors have been identified in cells all over the body, demonstrating a more general chemosensory role beyond taste. Bitter and sweet taste receptors regulate gut epithelial function, pancreatic β cell secretion, thyroid hormone secretion, adipocyte function, and many other processes. Emerging data from a variety of tissues suggest that t

Diffusible signal factor signaling controls bioleaching activity and niche protection in the acidophilic, mineral-oxidizing leptospirilli.

Bioleaching of metal sulfide ores involves acidophilic microbes that catalyze the chemical dissolution of the metal sulfide bond that is enhanced by attached and planktonic cell mediated oxidation of iron(II)-ions and inorganic sulfur compounds. Leptospirillum spp. often predominate in sulfide mineral-containing environments, including bioheaps for copper recovery from chalcopyrite, as they are effective primary mineral colonizers and oxidize iron(II)-ions efficiently. In this study, we demonstrated a functional diffusible signal factor interspecies quorum sensing signaling mechanism in Leptos

Chemical priming of immunity without costs to plant growth.

β-Aminobutyric acid (BABA) induces broad-spectrum disease resistance, but also represses plant growth, which has limited its exploitation in crop protection. BABA perception relies on binding to the aspartyl-tRNA synthetase (AspRS) IBI1, which primes the enzyme for secondary defense activity. This study aimed to identify structural BABA analogues that induce resistance without stunting plant growth. Using site-directed mutagenesis, we demonstrate that the (l)-aspartic acid-binding domain of IBI1 is critical for BABA perception. Based on interaction models of this domain, we screened a small li

Enhanced rapid acclimation of nitrifying sludge to sulfamethoxazole stress via exogenous quorum-sensing signal molecules.

Nitrifying sludge is capable of simultaneous removal of high-concentration ammonia nitrogen and sulfamethoxazole (SMX). Quorum sensing mediated by acyl-homoserine lactones (AHLs) has emerged as a potential regulatory tool to modulate microbial community function and stress resistance. However, the effects of AHLs on nitrifying sludge under SMX stress remain unclear. This study aimed to investigate whether AHLs can facilitate the acclimation of nitrifying sludge under SMX exposure. Sequencing batch reactor experiment results demonstrated that under the shock loading of 500 μg/L SMX, the additio

Catalytic Mechanism of γ-Elimination and Michael Addition Catalyzed by Pyridoxal-5'-Phosphate-Dependent γ-Substitution Enzyme Fub7 Involved in the Biosynthesis of 5-Alkyl-pipecolic Acid.

Fub7 is a rare pyridoxal 5'-phosphate (PLP)-dependent enzyme that can subsequently catalyze the γ-elimination of O-acetyl-l-homoserine (OAH) and Michael addition with n-valeraldehyde (NVA) to synthesize 5-alkyl-pipecolic acid, which shows significant potential in synthetic chemistry. In this study, we constructed the computational models and performed MD and QM/MM calculations to illuminate the complete catalytic cycle of Fub7, which contains five reaction stages, including external aldimine formation, γ-elimination, Michael addition, reformation of the internal aldimine, and final cyclization
