Mucosal adjuvant activity of a PAR-2-activating peptide enhances lymph node immune cell recruitment and promotes immune cell activation in a prime-boost influenza vaccination model.

Influenza A virus (IAV) remains a global health challenge, and current intramuscular vaccines often fail to induce strong mucosal immunity in the airways. We have developed a novel intranasal vaccination strategy in mice using a Protease-Activated Receptor 2 (PAR-2)-activating peptide (AP) as an adjuvant co-delivered with influenza virosomes. We have previously shown that this peptide enhances virosome-induced CD8+ T cell responses and protection against lethal IAV challenge. Here we studied the immune changes induced by this peptide in draining lymph nodes and the lung. We show that AP promot

Immune Response Activation and Hepatoprotective Activity of Randia echinocarpa Soluble Melanins in Murine Models.

This research demonstrates the in vivo immunomodulatory and hepatoprotective activities of the soluble melanins of the Randia echinocarpa fruit (PSM). The splenocyte cellular metabolic activity and lymphocyte T γδ expression in mesenteric lymph nodes (MLNs) and Peyer patches (PPs) were measured in the mice model. The PSM hepatoprotective activity was evaluated in the CCl4-induced acute hepatotoxic injury (AHTI) in the rat model. Compared with the controls, the PSM treatment induced higher splenocyte cellular metabolic activity (in vitro, 24.1%-57.25%; in vivo, 28.8%-47.7%), activation of lymph

Precise In Situ Delivery of a Photo-Enhanceable Inflammasome-Activating Nanovaccine Activates Anticancer Immunity.

A variety of state-of-the-art nanovaccines (NV) combined with immunotherapies have recently been developed to treat malignant tumors, showing promising results. However, immunosuppression in the tumor microenvironment (TME) restrains cytotoxic T-cell infiltration and limits the efficacy of immunotherapies in solid tumors. Therefore, tactics for enhancing antigen cross-presentation and reshaping the TME need to be explored to enhance the activity of NVs. Here, we developed photo-enhanceable inflammasome-activating NVs (PIN) to achieve precise in situ delivery of a tumor antigen and a hydrophobi

Viral activation and ecological restructuring characterize a microbiome axis of spaceflight-associated immune activation.

Maintenance of astronaut health during spaceflight will require monitoring and potentially modulating their microbiomes, which play a role in some space-derived health disorders. However, documenting the response of microbiota to spaceflight has been difficult thus far due to mission constraints that lead to limited sampling. Here, we executed a six-month longitudinal study centered on a three-day flight to quantify the high-resolution microbiome response to spaceflight. Via paired metagenomics and metatranscriptomics alongside single immune profiling, we resolved a microbiome "architecture" o

Basophils in Immunity: Activation Pathways, Roles in Allergy and AllergoOncology and Application of the Basophil Activation Test.

Basophils, the least abundant leukocytes, are increasingly recognised as potent immunomodulators. Upon activation, they rapidly release preformed granule-associated mediators including histamine and lipid mediators such as LTC4, while cytokine production occurs over a longer timescale, contributing to downstream immune responses. Basophil activation is driven by IgE-dependent FcεRI crosslinking or IL-3, complement and Toll-like receptor (TLR) pathways, enabling diverse effector responses, including type I hypersensitivity to allergens. Through IL-4 and IL-13 secretion, basophils bridge innate

Latent and Active Tuberculosis Infection Increase Immune Activation in Individuals Co-Infected with HIV.

In recent years, chronic immune activation and systemic inflammation have emerged as hallmarks of HIV disease progression and mortality. Several studies indicate that soluble inflammatory biomarkers (sCD14, IL-6, IL-8, CRP and hyaluronic acid), as well as surface markers of T-cell activation (CD38, HLA-DR) independently predict progression to AIDS and mortality in HIV-infected individuals. While co-infections have been shown to contribute to immune activation, the impact of latent tuberculosis infection (LTBI), which is widely endemic in the areas most affected by the global AIDS epidemic, has

Coincident helminth infection modulates systemic inflammation and immune activation in active pulmonary tuberculosis.

BACKGROUND: Helminth infections are known to modulate innate and adaptive immune responses in active and latent tuberculosis (TB). However, the role of helminth infections in modulating responses associated with inflammation and immune activation (reflecting disease activity and/or severity) in TB is not known. METHODOLOGY: We measured markers of inflammation and immune activation in active pulmonary TB individuals (ATB) with co-incidental Strongyloides stercoralis (Ss) infection. These included systemic levels of acute phase proteins, matrix metalloproteinases and their endogenous inhibitors

Early reduction of immune activation in lymphoid tissue following highly active HIV therapy.

OBJECTIVE: To evaluate immune reconstitution within HIV-infected lymphoid tissue during highly active antiretroviral therapy (HAART). DESIGN AND METHODS: In situ cellular responses were studied in sequential tonsillar biopsies in three asymptomatic HIV-infected (CD4 cells greater than 400 x 10(6)/l) antiretroviral treatment-naive volunteers enrolled in a clinical trial to determine the early effect of HAART. Computerized image analysis was used to study immunohistochemically stained sequential tonsil sections for the patterns of local cytokine production, chemokine receptor expression and cel

Modulation of in vitro immune reactions by platelet activating factor and a platelet activating factor antagonist.

Platelet activating factor (PAF) was found to suppress primary and secondary mixed lymphocyte reactions and BN52021, a naturally occurring PAF antagonist, blocked PAF-mediated suppression and enhanced the mixed lymphocyte reactions. The effect of delayed addition of PAF or BN52021 24 h or later after the initiation of cultures reduced the suppressive and enhancing effects, respectively. The removal of the antagonist BN52021 from mixed lymphocyte cultures up to 72 h after their initiation also was found to eliminate the potentiating effect of this antagonist. The continuous presence of PAF in m

Spatiotemporal immune gradients in gout: immune response-driven activation of the NLRP3-IL-1β axis and its transition to trained immunity.

Gout is a crystal-associated autoinflammatory disease triggered by monosodium urate (MSU) crystals, clinically characterized by recurrent transitions between acute inflammatory flares dominated by innate immunity and a state of "trained immunity" during the remission phase. However, previous studies have mostly focused on single time points or local lesions. Such approaches fail to systematically explain the recurrent nature of acute gout flares and the mechanisms underlying multi-system involvement. By integrating evidence from single-cell and spatial transcriptomics as well as mechanistic in

Polyclonal activation of the murine immune system by an antibody to IgD. IV. In vivo activation of B lymphocytes from immune defective CBA/N mice.

Previous studies with BALB/c mice have established that the injection of a goat antibody to IgD (GaM delta) induces splenic B lymphocytes to undergo T-independent, polyclonal increases in cell surface (s) Ia expression, size, and DNA synthesis 1 day after injection, as well as T-dependent maintenance of this activation and differentiation into sIgG1+ cells and IgG1-secreting cells 6 to 7 days after injection. Because B lymphocytes from mice homozygous or hemizygous for the CBA/N X-linked immune defect fail to proliferate in vitro when cultured with soluble anti-Ig antibodies, yet can make in v

From immune activation to symptom burden: mechanisms and translational implications of immune-symptom uncoupling in Sjögren's disease.

Sjögren's disease (SjD) is a heterogeneous autoimmune disease in which systemic activity, local tissue pathology, objective glandular or ocular function, biomarkers, and patient-reported symptoms frequently fail to align or change in parallel. We define immune-symptom uncoupling as partial, non-linear, or temporally asynchronous discordance across these domains rather than complete biological separation. This focused narrative review was informed by targeted, structured searches of PubMed and the Web of Science Core Collection through July 9, 2026. We distinguish four recurrent forms: systemic

Breaking Apoptosis-Induced Immune Silence: Ultrasound-Activated Nano-Oncolytic Therapy Reinvigorates Antitumor Immunity.

Conventional tumor therapies typically depend on the apoptotic pathway, which often leads to inadequate immunogenicity. This limitation underscores the urgent need for innovative treatments that enhance immunogenicity. In this study, an ultrasound (US)-activated nano-oncolytic system (designated as cRGD-Lip@PFP), is presented and consists of nanoliposomes (Lip) modified with the cRGD peptide for targeted tumor delivery. This system features a perfluoropentane (PFP) core that undergoes US-triggered acoustomechanical effects, enabling controlled expansion from nanoscale to microscale, ultimately

Bridging innate and adaptive tumor immunity: cGAS-STING pathway activation to potentiate immune checkpoint blockade.

The cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway is critical for innate immunity, as it detects cytoplasmic DNA and drives type I interferon signaling. Pharmacological stimulation of this pathway has been recognized as a valuable approach for cancer immunotherapy, especially when used together with immune checkpoint inhibitors (ICIs). Preclinical studies have demonstrated synergistic antitumor effects of cGAS-STING agonists and ICIs across various tumor models, while early-phase clinical trials are exploring their safety and efficacy in patients. Nonetheless, int

Neuroimmune and behavioral changes elicited by maternal immune activation in mice are ameliorated by early postnatal immune stimulation.

Though the etiology of autism spectrum disorder (ASD) is complex and not fully understood, it is believed that genetic risk factors, coupled with early life inflammation may predispose individuals to develop ASD. Maternal immune activation (MIA) is associated with increased incidence of ASD in offspring; however, not all mothers who experience inflammation during pregnancy have children with autism, suggesting that MIA may act as a disease primer that results in ASD pathology when paired with additional inflammatory insults. Here, we tested the hypothesis that MIA is a disease primer by using

Modulation of lipid nanoparticle-formulated plasmid DNA drives innate immune activation promoting adaptive immunity.

Nucleic acid vaccines have grown in importance over the past several years, with the development of new approaches remaining a focus. We describe a lipid nanoparticle-formulated DNA (DNA-LNP) formulation which induces robust innate and adaptive immunity with similar serological potency to mRNA-LNPs and adjuvanted protein. Using an influenza hemagglutinin (HA)-encoding construct, we show that priming with our HA DNA-LNP demonstrated stimulator of interferon genes (STING)-dependent upregulation and activation of migratory dendritic cell (DC) subpopulations. Complement activation profiles in patients with immune checkpoint inhibitor-associated neuromuscular immune-related adverse events.

BACKGROUND: Immune-related neuropathy (irNeuropathy) and myositis (irMyositis) are the most common neurologic adverse events (irAE-n) associated with immune checkpoint inhibitors. Although case reports suggest benefits of complement inhibitors, the role of complement activation in irAE-n is understudied. METHODS: In a retrospective multicenter study, we enrolled patients with irNeuropathy or irMyositis, cancer controls (CCs), and healthy controls (HCs). Serum levels of 11 complement components were measured using multiplex enzyme-linked immunosorbent assays. Associations with irAE-n severity

Peripheral immune reactions following human traumatic spinal cord injury: the interplay of immune activation and suppression.

Traumatic spinal cord injury (SCI) damages the nerve tissue of the spinal cord, resulting in loss of motor and/or sensory functions at and below the injury level. SCI provokes a long-lasting immune response that extends beyond the spinal cord and induces changes in the composition and function of the peripheral immune system. Seemingly contradictory findings have been observed, as both systemic immune activation, including inflammation and autoimmunity, and immune suppression have been reported. Differences in the levels and functions of various cell types and components of both the innate and

Role of PARP Inhibitors in Cancer Immunotherapy: Potential Friends to Immune Activating Molecules and Foes to Immune Checkpoints.

Poly (ADP-ribose) polymerase (PARP) inhibitors (PARPi) induce cytotoxic effects as single agents in tumors characterized by defective repair of DNA double-strand breaks deriving from BRCA1/2 mutations or other abnormalities in genes associated with homologous recombination. Preclinical studies have shown that PARPi-induced DNA damage may affect the tumor immune microenvironment and immune-mediated anti-tumor response through several mechanisms. In particular, increased DNA damage has been shown to induce the activation of type I interferon pathway and up-regulation of PD-L1 expression in cance
