Designing of a multiepitope-based vaccine against echinococcosis utilizing the potent Ag5 antigen: Immunoinformatics and simulation approaches.

Echinococcosis is a significant parasitic zoonotic disease with severe implications for human and animal health. To date, there has been no effective vaccine candidate available for echinococcosis. Therefore, we employed computational approaches to develop a multiepitope-based vaccine using the most potent epitopes of MHC-I, MHC-II, and B-cell derived from the Ag5 protein of Echinococcus spp. The final vaccine construct containing the epitopes, linkers, and adjuvant exhibited potent antigenicity (score > 0.1) with no evidence of allergenicity (score < 0) and toxicity (score < 0) in several com

Development of a Multiple Epitopes-Based Dengue Vaccine: An Immunoinformatics Approach and Insights From Pakistani Population Genetics.

Dengue fever poses a grave threat to public health worldwide, claiming numerous fatalities each year with tropical regions being particularly hard hit by dengue outbreaks. The multiple-epitope construct is tailored to the dengue virus's geographical prevalence and the genetics of the Pakistani population. 14 experimentally validated MHC-I and MHC-II epitope sequences, were employed to generate the variants by taking into account the conservancy in all serotypes. Subsequently, the binding affinities of each derived variant against the human leukocyte antigen alleles most common among the Pakist

Immunoinformatics approach for designing a multiepitope subunit vaccine against porcine epidemic diarrhea virus genotype IIA spike protein.

BACKGROUND: Porcine epidemic diarrhea (PED), caused by the porcine epidemic diarrhea virus (PEDV), is associated with high mortality and morbidity rates, especially in neonatal pigs. This has resulted in significant economic losses for the pig industry. PEDV genotype II-based vaccines were found to confer better immunity against both heterologous and homologous challenges; specifically, spike (S) proteins, which are known to play a significant role during infection, are ideal for vaccine development. AIM: This study aims to design a multi-epitope subunit vaccine targeting the S protein of the

Developing a chimeric multiepitope vaccine against Nipah virus (NiV) through immunoinformatics, molecular docking and dynamic simulation approaches.

Nipah virus (NiV) is a highly lethal zoonotic pathogen that poses a significant threat to human and animal health. Unfortunately, no effective treatments have been developed for this deadly zoonotic disease. Therefore, we designed a chimeric multiepitope vaccine targeting the Nipah virus (NiV) glycoprotein and fusion protein through immunoinformatic approaches. Therefore, the vaccine was developed by combining promising and potential antigenic MHC-I, MHC-II, and B-cell epitopes obtained from the selected proteins. When combined, the MHC-I and MHC-II epitopes offered 100 % global population cov

A Vaccine Construction against COVID-19-Associated Mucormycosis Contrived with Immunoinformatics-Based Scavenging of Potential Mucoralean Epitopes.

Mucormycosis is a group of infections, caused by multiple fungal species, which affect many human organs and is lethal in immunocompromised patients. During the COVID-19 pandemic, the current wave of mucormycosis is a challenge to medical professionals as its effects are multiplied because of the severity of COVID-19 infection. The variant of concern, Omicron, has been linked to fatal mucormycosis infections in the US and Asia. Consequently, current postdiagnostic treatments of mucormycosis have been rendered unsatisfactory. In this hour of need, a preinfection cure is needed that may prevent

Immunoinformatics Design of Multi-Epitope Peptide-Based Vaccine Against Schistosoma mansoni Using Transmembrane Proteins as a Target.

Schistosomiasis remains a serious health issue nowadays for an estimated one billion people in 79 countries around the world. Great efforts have been made to identify good vaccine candidates during the last decades, but only three molecules reached clinical trials so far. The reverse vaccinology approach has become an attractive option for vaccine design, especially regarding parasites like Schistosoma spp. that present limitations for culture maintenance. This strategy also has prompted the construction of multi-epitope based vaccines, with great immunological foreseen properties as well as b

Heterologous Antigen Regimen and Bicistronic DNA Vaccination Targeting HPV16 E5 and E7 Oncoproteins: Immunoinformatics and Preclinical Antitumor Efficacy.

Persistent infection with high-risk human papillomavirus, particularly HPV16, is a major cause of cervical cancer and orogenital malignancies. Therapeutic DNA vaccines targeting viral oncoproteins represent a promising strategy to induce antitumor immune responses. However, the impact of antigen combinations in bicistronic vectors and the immunization sequence on therapeutic efficacy remains poorly understood. Thus, this study aimed to characterize in silico previously described E5CP and E7CP vaccines, develop a new bicistronic construct, and evaluate its antitumor efficacy under different imm

Designing a novel vaccine against COVID-19 based on spike SARS-Cov-2 notable mutations using immunoinformatics approaches.

The rapid emergence of SARS-CoV-2 variants with spike protein mutations undermines the effectiveness of current vaccines, necessitating innovative strategies to ensure broad and lasting immunity. This study leverages an immunoinformatics approach to design two multi-epitope vaccine constructs Cov19-B (649 amino acids, 74 kDa) and Cov19-T (465 amino acids, 48 kDa) specifically targeting mutations in the spike protein observed in the Alpha, Beta, Gamma, and Omicron variants. Using sequence data retrieved from NCBI, GISAID, and UniProt, we predicted a range of epitopes, including linear B-cell, c

Immunoinformatics-Guided Computational Design and In Silico Validation of Multi-Epitope Vaccine Candidates Targeting Canine and Feline Parvoviruses.

Parvovirus infection causes severe diseases in both feline and canine species. It primarily affects adult cats and dogs but poses a higher risk to kittens and puppies. This virus is highly contagious and is easily transmitted through contaminated food, shared shelter environments, as well as the hands and clothing of people. The recovered species may continue to shed parvovirus in their feces for an extended period, leading to severe environmental contamination. There is no universal vaccine available that protects dogs and cats against parvovirus infections. The main goal of this study is to

In silico design of novel CTL based multi epitope vaccine for esophageal cancer using immunoinformatics and molecular docking.

Esophageal cancer is an aggressive malignancy with high morbidity, mortality, and limited durable treatment options due to tumor heterogeneity, immune evasion, and recurrence. This study addresses these challenges by computationally designing a novel CTL-based multi-epitope vaccine using experimentally validated epitopes from cancer-testis antigens (NY-ESO-1 and MAGE-A family), which are overexpressed in esophageal squamous cell carcinoma. To the best of our knowledge, this represents one of the most comprehensive in silico investigations for esophageal cancer, uniquely integrating experimenta

Immunoinformatics-Driven Identification of Conserved Multi-Epitope Vaccine Candidates from PDI Protein of Leishmania Donovani for Visceral Leishmaniasis.

Visceral leishmaniasis (VL), caused by Leishmania donovani, continues to pose a public health challenge, particularly in the Indian subcontinent. This warrants the need for a prophylactic vaccine; hence, a comprehensive immunoinformatics approach was employed to identify B-cell and T-cell epitopes from the parasite’s Protein Disulfide Isomerase (PDI), a known virulence factor. Strong CD8 + epitopes (RTAAGIASY, FLATAVLDY, SVAAFVEKY) and a CD4 + epitope (VKRFLATVA) were selected based on MHC-binding affinity, immunogenicity, and conservation across Leishmania species. In addition, four linear B-

A novel (-)-(2S)-7,4'-dihydroxyflavanone compound for treating age-related diabetes mellitus through immunoinformatics-guided activation of CISD3.

The iron-sulfur domain (CISD) proteins of CDGSH are classified into three classes: CISD1, CISD2, and CISD3. During premature ageing, mutations that affect these proteins, namely their binding sites, could result in reduced protein production and an inability to preserve cellular integrity. Consequently, this leads to the development of conditions such as diabetes. Notably, CISD3 plays a crucial role in the management of age-related disorders such as Wolfram syndrome, which is often referred to as DIDMOAD (diabetes insipidus, diabetes mellitus, optic atrophy, and deafness). Computational analys
