Fluorescent in situ hybridization analysis of chromosomally normal gametes and abnormal arrested embryos.

OBJECTIVE: The purpose of the present study was to investigate if the arrested embryos from a couple with several previously failed IVF treatments were chromosomally normal. Probes for chromosomes X, Y and 18 were used. RESULTS: A couple had undergone 7 in vitro fertilization treatments over a 2 1/2-year period without achieving a pregnancy. In each cycle, where fertilization was obtained, the development of the embryos was arrested. Fluorescent in situ hybridization probes for chromosomes X and Y (and 18) was carried out on gametes and on embryos in 2 separate cycles. Sperm and oocytes were

Interphase in situ hybridization to disaggregated and intact tissue specimens of prostatic adenocarcinomas.

A comparative study was performed of interphase in situ hybridization (ISH) to deparaffinized 4-microns tissue sections and nuclear suspensions from eight prostatic adenocarcinomas, as well as one normal prostatic control. Whole nuclear suspensions were derived from the same tumor areas to evaluate differences of ISH to truncated versus whole nuclei. DNA probes specific for the centromeres of chromosome 1, 7, 8, 10, and Y were used for detection of numerical chromosomal changes and aneuploidy. In six adenocarcinomas chromosome aberrations (+7, +8, -8, -10, -Y) were seen. However, ISH to sectio

Five cases of cytomegalovirus infection detected by in situ hybridization and antigenemia assay.

We report five cases of cytomegalovirus infection in immunocompromised patients which were detected by either cytomegalovirus antigenemia assay or in situ hybridization. Four cases had leukemia and the other had chronic renal failure. All the three BMT recipients suffered from GvHD. Interestingly, there was an unique case of CMV disease without a history of BMT, which reminded us that CMV could attack immunocompromised patients who had not undergone transplantation, too. Four out of five cases died. We think that cytomegalovirus infection or disease should not be regarded as a minor problem in

Molecular cytogenetic characterization of the DiGeorge syndrome region using fluorescence in situ hybridization.

DiGeorge syndrome (DGS) is a developmental defect characterized by cardiac defects, facial dysmorphism, and mental retardation. Several studies have described a critical region for DGS at 22q11, within which the majority of DGS patients have deletions. We have isolated nine cosmid and three YAC clones using previously described and newly isolated probes that have been shown to be deleted in many DGS patients. Using fluorescence in situ hybridization and digital imaging, we have mapped and ordered these clones relative to the breakpoints of two balanced translocations at 22q11 (one in a DGS pat

Rapid in situ hybridization for the diagnosis of latent Epstein-Barr virus infection.

Latent Epstein-Barr virus (EBV) infection is associated with a variety of malignancies. Rapid in situ hybridization techniques have been described for various lytic viral infections because of limited gene expression. However, EBERS (Epstein Barr early RNAs) are expressed in abundance in tumour cells which are latently infected with EBV. We have targeted these transcripts in a rapid (3 h) in situ hybridization assay fo the detection of latent EBV in clinical specimens, including formalin-fixed paraffin-embedded material. EBER RNA was detected in control cell lines which have two copies of the

Fluorescence in situ hybridization to determine engraftment status after murine bone marrow transplant.

The mouse Y-specific DNA sequence pY2 was used as a probe for fluorescence in situ hybridization (FISH) to evaluate murine hematopoietic tissues after sex-mismatched bone marrow transplant (BMT). The pY2 probe was localized to the long arm of the Y chromosome on BM metaphases. Hybridization of pY2 in FISH of interphase cells from BM, spleen, and thymus after BMT was compared with Southern blot analysis; both methods gave comparable results. Only FISH was able to analyze post-BMT peripheral blood (PB) samples successfully, and provides a useful method for following engraftment status in the mou

Expression of peptidylglycine alpha-amidating monooxygenase: an in situ hybridization and immunocytochemical study.

Peptide alpha-amidation, an essential posttranslational modification that confers bioactivity to many neuroendocrine peptides, is catalyzed by peptidylglycine alpha-amidating monooxygenase (PAM; EC 1.14.17.3). To complement our previous studies on the distribution of PAM in neuroendocrine organs, we have examined expression of the PAM gene in several endocrine tissues by in situ hybridization and immunocytochemistry. In all instances, the autoradiographic densities for PAM mRNA correlated with staining patterns for PAM immunoreactivity. Very high levels of PAM mRNA were found in all heart atri

[Detection of HTLV-ImRNA by in situ hybridization technique using biotinylated oligonucleotide probe].

The expressions of human T-lymphotropic virus type I (HTLV-I) mRNA in the HTLV-I-infected lymphocytes were studied in the peripheral blood of two HTLV-I carriers and of three patients with adult T-cell leukemia (ATL) (acute, chronic and lymphoma types) by in situ hybridization technique using two biotinylated single-stranded oligodeoxynucleotide probes complementary to different nucleotide sequences in the mRNA for the HTLV-IpX region. A low percentage of leukocytes (1.5-7.4%) reacted with the probes in ATL patients, while less than 1% of leukocytes was reactive in HTLV-I carriers. These resul

Robertsonian translocation and an extra microchromosome: independent origin identified by in situ hybridization.

Robertsonian translocation may result in either a monocentric or a dicentric product involving the long arms of the participating chromosomes. In the former case a microchromosome involving the short arms of both acrocentrics would be expected to arise, but in practice is rarely observed. Where such small bisatellited markers are seen in association with Robertsonian translocations, they are assumed to represent such reciprocal products. We present a case, involving a t(14q21q) and a DA/DAPI positive microchromosome, where using in situ hybridization we show that this assumption is incorrect a

Endothelin: visualization of mRNAs by in situ hybridization provides evidence for local action.

Endothelin (ET) is a recently identified vasoactive peptide with three isoforms for which three genes have been cloned. The cellular sites of synthesis of this peptide have not yet been identified in vivo. Using Northern blot analysis, we have detected two forms of ET mRNA in rat tissues: a 3.7-kilobase form in the kidney, eye, and brain, a 2.5-kilobase form in the intestine, and both forms in the lung. We have localized these forms of ET mRNA in several rat tissues using in situ hybridization. In the 19-day rat fetus, ET mRNA is highest in the lung, intestine, and meninges. At high resolution

Intrahepatic markers of hepatitis delta virus infection: a study by in situ hybridization.

The intrahepatic distribution of hepatitis delta virus RNA was studied by in situ hybridization in 33 formalin-fixed, paraffin-embedded biopsies from 17 chronic hepatitis B virus carriers superinfected with hepatitis delta virus. The findings were correlated with the expression of the hepatitis delta antigen, the duration of the hepatitis delta virus infection and the eosinophilic degeneration of the hepatocytes. Intranuclear hepatitis delta virus RNA and antigen were found in 28 specimens, whereas the remaining five were negative for both markers. Hepatitis delta virus RNA and antigen were mo

Progressive multifocal leukoencephalopathy: JC virus detection by in situ hybridization compared with immunohistochemistry.

In four cases of progressive multifocal leukoencephalopathy (PML), we compared biotin-labeled DNA:DNA in situ hybridization with peroxidase immunohistochemistry for the detection of JC virus (JCV). The localization of JCV DNA and JCV capsid protein was compared in formalin-fixed, paraffin-embedded brain tissues. Infected oligodendrocytes showed both JCV DNA and JCV protein. However, bizarre astrocytes demonstrated JCV capsid protein less often than JCV DNA. In situ hybridization with a biotinylated probe was as sensitive and specific as immunohistochemistry for diagnosis on formalin-fixed tiss

Putative Malignant Pleural Mesothelioma in situ (MPMIS) with Sequential Acquisition of Genomic Alterations on Fluorescence in situ Hybridization (FISH) Examination.

Malignant pleural mesothelioma (MPM) is a rare and deadly disease. A precursor in situ lesion, malignant pleural mesothelioma in situ (MPMIS), has recently been proposed. On cytological examination, the distinction between reactive and malignant mesothelial cells is often challenging, and sometimes even impossible without ancillary methods. Fluorescence in situ hybridization (FISH) for detection of 9p21 deletion is a powerful diagnostic tool in this context, both in histological and in cytological specimens. Here, we present a case of MPM with initial presentation as a putative MPMIS with diso

The surgical and cytopathology of viral infections: utility of immunohistochemistry, in situ hybridization, and in situ polymerase chain reaction amplification.

The diagnosis of viral infections is an important part of the daily work of a surgical and cytopathologist. Some viral infections, such as human papillomavirus infection of the lower genital tract, are seen commonly, whereas others, such as fatal enteroviral infection, cannot be diagnosed on routine histological examination and need to be addressed within the clinical context. In general, viral infections are best categorized for the surgical pathologist as low copy/RNA viruses and high copy/DNA viruses. In the latter, viral DNA enters the nucleus, undergoes rapid proliferation, and causes cer

Comparative Analysis of Gene Expression Analysis Methods for RNA in Situ Hybridization Images.

Gene expression analysis is pivotal in cancer research and clinical practice. Although traditional methods lack spatial context, RNA in situ hybridization (RNA-ISH) is a powerful technique that retains spatial tissue information. Here, RNAscope score, RT-droplet digital PCR, and automated QuantISH and QuPath were used for quantifying RNA-ISH expression values from formalin-fixed, paraffin-embedded samples. The methods were compared using high-grade serous ovarian carcinoma samples, focusing on CCNE1, WFDC2, and PPIB genes. The findings demonstrate good concordance between automated methods and

A novel approach for microRNA in situ hybridization using locked nucleic acid probes.

Identification of target tissue microRNAs (miR) using in situ hybridization (ISH), with digoxigenin-labeled locked nucleic acid (LNA) probes, is influenced by preanalytic parameters. To determine the best retrieval method for common microRNAs, a multiblock composed of paraffin-embedded tonsil, cervix, placenta, and hyperplastic prostate tissue were included. Tissue were fixed in 10% formalin in a range of 5-144 hours (h). Cut sections (5 μm) from the multiblock were subjected to combinations of pretreatment procedures: variable periods of proteinase K (PK) digestion or Heat-induced microRNA Re

ESHRE PGD consortium best practice guidelines for fluorescence in situ hybridization-based PGD.

In 2005, the European Society for Human Reproduction and Embryology (ESHRE) PGD Consortium published a set of Guidelines for Best Practice PGD to give information, support and guidance to potential, existing and fledgling PGD programmes. The subsequent years have seen the introduction of new technologies as well as evolution of current techniques. Additionally, in light of recent advice from ESHRE on how practice guidelines should be written and formulated, the Consortium believed it was timely to revise and update the PGD guidelines. Rather than one document that covers all of PGD, the new gu
