Symbiotic partnerships have opened new ecological niches and contributed to the remarkable diversification of insects. The leafhopper Scaphoideus titanus, a phloem-feeding insect known to be the primary vector of Flavescence dorée phytoplasma, harbours two primary endosymbionts: the bacterium 'Candidatus Karelsulcia muelleri' and a yeast-like symbiont (YLS). While most studies on insect-associated microorganisms have focused on obligate bacterial symbionts, fungal endosymbionts, although documented for almost a century, are only now gaining renewed attention for their evolutionary and ecologic

Across the animal kingdom, a remarkable diversity of chemoreceptors has evolved, reflecting the ecological and evolutionary pressures that shape species-specific sensory demands. Insects, the most biodiverse class of animals, play crucial roles in ecosystems and have an extensive chemosensory repertoire. At the heart of insect gustation lie the gustatory receptors (GRs), a large and remarkably diverse family of proteins characterized by their seven-transmembrane domain structure and tetrameric stoichiometry. These receptors are phylogenetically distinct from the taste receptors present in most

Insect cuticle acts as a primary protective barrier against environment stresses that may directly impact the insect body. Here, we report the mechanical defense function of a structural cuticular protein, BmorCPH24, to environmental stresses using a silkworm Bamboo (Bo) mutant with this gene mutation. Ultraviolet (UV) irradiation and topical application of an acetone insecticide were used as environmental stresses to determine the differences in susceptibility between Bo and wild-type larvae. UV irradiation resulted in a sunburn phenotype in the Bo strains earlier than the wild-type indicatin

Fusing insect derived neurotoxic peptides with Galanthus nivalis agglutinin (GNA) has been shown to enhance the insecticidal activity of the neuropeptides, especially when administered orally. This study produced a recombinant scorpion insect specific neurotoxin BjαIT, GNA, and a fusion protein BjαIT/GNA using Pichia pastoris as an expression host. Recombinant rBjαIT/GNA was found to be easily degraded during expression in yeast which and produced a main protein product with a molecular weight of approximately 14 kDa. Cytotoxicity results showed that rBjαIT, rGNA, and rBjαIT/GNA had no toxicit

Phytoecdysteroids, structurally similar to insect molting hormones, produce a range of effects in mammals, including increasing growth and physical performance. In skeletal muscle cells, phytoecdysteroids increase protein synthesis. In this study we show that in a mouse skeletal muscle cell line, C(2)C(12), 20-hydroxyecdysone (20HE), a common phytoecdysteroid in both insects and plants, elicited a rapid elevation in intracellular calcium, followed by sustained Akt activation and increased protein synthesis. The effect was inhibited by a G-protein coupled receptor (GPCR) inhibitor, a phospholip

BACKGROUND: Considering their high content of protein, insects are a valuable alternative protein source. However, no evaluation of their purine content has so far been done. High content of purine derivates may lead to the exclusion of such food from the diet of people with specific diseases. The aim of this study was to analyse the content of selected purine derivates and amino acid profile in the three insect species most often used for entomophagy in Europe and compare them with the purine content in egg white and chicken breast.
RESULTS: The content of individual purine derivates and the

This study addressed the urgent need for sustainable protein sources in aquaculture due to the depletion of marine resources and rising costs. Animal protein sources, particularly poultry by-product meal (PBM) and insect exuviae meal, were investigated as viable alternatives to fishmeal (FM). The research study confirmed the successful replacement of FM with a combination of PBM and insect exuviae meal (up to 50%) in the diet of European seabass without compromising growth, feed conversion, gut health, and liver fat content. In particular, growth was robust with both PBM formulations, with the

The cultivation of edible insects to provide an alternative, sustainable protein source is growing, and assuring their nutritional quality and safety is critical for acceptance by consumers. The original risk profiling was undertaken around ten years ago since when the knowledge base about farmed insects has increased. The updated safety data gap analysis from the Giant Leaps project has shown that many of the original conclusions remain true and risks can be mitigated by managing the quality of feedstock and hygienic rearing practices. Despite advances, evidence is still lacking regarding the

Insect prophenoloxidases (PPOs) are a group of important innate immunity proteins. Although there have been numerous studies dealing with the PPO activation cascade, the detailed biochemical behaviors of the PPO family proteins remain to be clearly established. This is due primarily to the difficulty in obtaining adequate amounts of PPO proteins for comprehensive characterization. In this study, we expressed three Drosophila melanogaster PPO genes in Escherichia coli, and extensively evaluated expression conditions for obtaining soluble proteins. Through the manipulation of expression conditio

Human gastric lipase (HGL) cDNA was synthesized by RT-PCR amplification and cloned into the PVL 1392 baculovirus transfer vector. The recombinant transfer vector was cotransfected with a modified baculovirus DNA (Baculogold) which contains a lethal deletion. Cotransfection of baculovirus DNA with the recombinant transfer vector rescues the lethal deletion of this virus DNA and reconstitutes viable virus particles inside the transfected insect cells. BTI-TN-5B1-4 insect cells (also called High Five cells) were used to express recombinant HGL. The level of HGL secretion was approximately 32 mg/l

p53 protein, the central molecule of the apoptosis pathway, is mutated in 50% of the human cancers. Of late, p53 homologues have been identified from different invertebrates including Drosophila melanogaster, Caenorhabditis elegans, Squid, and Clams. We report the identification of a p53-like protein in Spodoptera frugiperda (Sf9) insect cells, which is activated during oxidative stress, caused by exposure to UV-B or H(2) O(2) , and binds to p53 consensus DNA binding motifs as well as other p53 cognate motifs. Sf9 p53 motif-binding protein is similar to murine and Drosophila p53 in terms of mo

Background: Childhood malnutrition can have devastating consequences on health, behavior, and cognition. Edible insects are sustainable low cost high protein and iron nutritious foods that can prevent malnutrition. However, it is unclear whether insect-based diets may help prevent changes to brain neurochemistry associated with malnutrition.Materials and Methods: Weanling male Sprague-Dawley rats were malnourished by feeding a low protein-iron diet (LPI, 5% protein and ∼2 ppm Fe) for 3 weeks or nourished by feeding a sufficient protein-iron diet (SPI, 15% protein 20 ppm FeSO4) for the duration

N-glycosylation is an important regulator of insect development, tissue organization, neural function, and other physiological processes, yet the extent to which cultured insect cells reproduce organismal glycosylation remains poorly understood. Here, we characterized the whole-body N-glycoproteome of adult Drosophila melanogaster and compared it with our previously generated site-specific glycoproteome of Drosophila S2 cells. Intact glycopeptide analysis by LC-MS/MS identified 2144 glycopeptides representing 526 glycoproteins, 875 N-glycosylation sites, and 38 glycan compositions in adult fli

Clinical immunity to malaria is associated with the acquisition of IgG specific for members of the Plasmodium falciparum erythrocyte membrane protein 1 (PfEMP1) family of clonally variant antigens on the surface of infected erythrocytes (IEs). The VAR2CSA subtype of PfEMP1 mediates IE binding in the placenta. VAR2CSA-specific IgG is normally acquired only after exposure to placental parasites. However, it was recently reported that men and children from Colombia often have high levels of functional VAR2CSA-specific IgG. This potentially undermines the current understanding of malaria immunity

BACKGROUND: Although technical advances in genomics and proteomics research have yielded a better understanding of the coding capacity of a genome, one major challenge remaining is the identification of all expressed proteins, especially those less than 100 amino acids in length. Such information can be particularly relevant to human pathogens, such as Trypanosoma brucei, the causative agent of African trypanosomiasis, since it will provide further insight into the parasite biology and life cycle.
RESULTS: Starting with 993 T. brucei transcripts, previously shown by RNA-Sequencing not to coin

Saliva plays important roles in facilitation of a bloodmeal, lubrication of mouthparts, and parasite transmission for some vector insects. Salivary composition changes during the lifetime of an insect, and differences in the salivary profile may influence its functions. In this report, the amount and profile of salivary gland protein of the American visceral leishmaniasis vector Lutzomyia longipalpis (Lutz & Neiva, 1912) were analyzed at different times of insect development and diet. Protein content from unfed female sand flies increased significantly with age, and a significant difference wa

Sexual signaling is a fundamental component of sexual behavior of Ceratitis capitata that highly determines males' mating success. Nutritional status and age are dominant factors known to affect males' signaling performance and define the female decision to accept a male as a sexual partner. Wolbachia pipientis, a widespread endosymbiotic bacterium of insects and other arthropods, exerts several biological effects on its hosts. However, the effects of Wolbachia infection on the sexual behavior of medfly and the interaction between Wolbachia infection and adult food remain unexplored. This stud

Many insects elicit development of unique organs in their host plants called galls while feeding or ovipositing. Galls are organ-like entities that protect against biotic and abiotic stresses and provide a specialized nutrition supply. The signals used by gall-forming insects to elicit and shape galls are unknown, but phytohormones, peptides, small RNAs and arabinogalactan proteins are leading candidates. These four signal categories may work together to elicit galls and guide their development, but evidence of their transfer from insect to plant is incomplete. Galls vary in complexity from si

Insect pests like Plutella xylostella cause significant crop losses, and the overuse of synthetic pesticides has led to resistance and ecological imbalance. This study evaluated the insecticidal properties of 1-isoleucine, N-allyloxycarbonyl-, dodecyl ester, isolated from Actinokineospora fastidiosa. The compound showed strong antifeedant (89.19%), larvicidal (83.53%), and pupicidal (90.00%) activity at 25 µg/mL, with LC50 and LC90 values of 9.82 and 47.72 µg/mL, respectively. In addition, treated larvae exhibited delayed development, elevated antioxidant enzyme levels (SOD, CAT), DNA damage,

BACKGROUND: The life cycle of the parasitic protozoan Trypanosoma cruzi, the etiological agent of Chagas disease (CD), includes two well-recognised insect-dwelling stages: the replicative non-infective epimastigotes and the non-replicative infective metacyclic trypomastigotes. Nonetheless, the existence of multiple intermediate forms has been reported. Since nutrient restriction is considered one of the main factors driving metacyclogenesis and is very frequent due to the long-term starvation periods that the insect vectors commonly undergo, we have studied the transcriptomic effects of nutrie
