Saliva plays important roles in facilitation of a bloodmeal, lubrication of mouthparts, and parasite transmission for some vector insects. Salivary composition changes during the lifetime of an insect, and differences in the salivary profile may influence its functions. In this report, the amount and profile of salivary gland protein of the American visceral leishmaniasis vector Lutzomyia longipalpis (Lutz & Neiva, 1912) were analyzed at different times of insect development and diet. Protein content from unfed female sand flies increased significantly with age, and a significant difference wa

Leishmaniasis is a vector-borne disease with a complex epidemiology and ecology. Visceral leishmaniasis (VL) is its most severe clinical form as it results in death if not treated. In Latin America VL is caused by the protist parasite Leishmania infantum (syn. chagasi) and transmitted by Lutzomyia longipalpis. This phlebotomine sand fly is only found in the New World, from Mexico to Argentina. However, due to deforestation, migration and urbanisation, among others, VL in Latin America is undergoing an evident geographic expansion as well as dramatic changes in its transmission patterns. As plants and associated insects are at the bottom of some terrestrial food webs, they are the primary contributors to mercury (Hg) fluxes in ecosystems. In addition to the trophic position of these organisms, factors related to their life traits have been hypothesized to influence their exposure to Hg. This study investigates the transfer of Hg in a soil-nettle-insect system and the insect-related factors affecting their Hg concentrations in a revegetated chlor-alkali landfill. Twenty-three insect species were identified and classified according to their life traits, their relationship with n

The dynamic process of pathogen transmission by the bite of an insect vector combines several biological processes that have undergone extensive co-evolution. Whereas the host response to an insect bite is only occasionally confronted with the parasitic pathogens that competent vectors might transmit, the transmitted parasites will always be confronted with the acute, wound-healing response that is initiated by the bite itself. Invariably, this response involves neutrophils. In the case of Leishmania, infection is initiated in the skin following the bite of an infected sand fly, suggesting tha

Insect cell culture has greatly increased in part due to the widespread use of insect virus-based vectors for efficient expression of foreign proteins. Insect cells such as Sf9 cells are susceptible to arboviruses which may pose a safety concern by adventitious introduction during the production process. The objective of this study was to establish techniques for viral clearance validation of insect cell-derived biotechnological products using Japanese encephalitis virus (JEV) as a model, since JEV is a member of arthropod-borne flaviviruses that are known to be infectious in insect cells. BACKGROUND: Mosquitoes and other vectors are often exposed to sublethal doses of insecticides. Larvae can be exposed to the run-off of agricultural use, and adults can be irritated by insecticides used against them and move away before they have picked up a lethal dose. This sublethal exposure may affect the success of control of insect-borne diseases, for it may affect the competence of insects to transmit parasites, in particular if the insects are undernourished.
METHODS: We assessed how exposure of larvae and adults to a sublethal dose of permethrin (a pyrethroid) and how larval competiti

African trypanosomiasis, caused by parasites of the genus Trypanosoma, is a complex of devastating vector-borne diseases of humans and livestock in sub-Saharan Africa. Central to the pathogenesis of African trypanosomes is their transmission by the arthropod vector, Glossina spp. (tsetse fly). Intriguingly, the efficiency of parasite transmission through the vector is reduced following depletion of Trypanosoma brucei Procyclic-Specific Surface Antigen-2 (TbPSSA-2). To investigate the underlying molecular mechanism of TbPSSA-2, we determined the crystal structures of its ectodomain and that of

Chagas disease is endemic to the Americas and is transmitted by blood-feeding kissing bugs. We evaluated the insecticidal potential of a fungus (Beauveria bassiana strain Pr-11) against Triatoma infestans, an important vector in South America. This fungal species was isolated from a locust (Schistocerca piceifrons) that inhabits the Central Andes region of Peru. Ten days post inoculation, this strain induced high insect mortality (97%) at low fungal concentrations (2 × 107 conidia/ml) at 70% relative humidity. The Pr-11 strain outperformed reference strain CCBLE-216 B. bassiana, provided by th

Control of the vector is usually a crucial factor in control programs for tropical diseases spread by insect vectors. Successful control programs aim at vulnerable points in the interactions between the vector, the reservoir host, the pathogen, the human host, and the environment. The objective is to prevent potential transmission, or interrupt actual transmission, by reducing the abundance, longevity, or host contact of the vector--whichever is most appropriate to the particular pathogen or disease and the local situation. The importance of individual assessment in the light of local conditio

Recent omics studies (cytogenomics, genomics, and transcriptomics) have provided extensive information on Chagas disease (CD) vectors. Here, we conduct a review of proteomic and metabolomic studies on these insect vectors. We address proteomic studies in triatomines, focusing on the identification and characterization of proteins in their saliva, midgut, and salivary glands, emphasizing their role in blood feeding by providing vasodilatory and anticoagulant substances, along with anti-inflammatory and antiplatelet activities. In addition, the review investigates protein diversity in different

Malaria remains a serious public health problem in the Amazon. The main vector, Anopheles darlingi, uses natural and artificial water bodies as breeding grounds, places that harbor various aquatic insects, which should be considered in control programs. Therefore, the objective of this research was to determine the composition of the aquatic entomofauna associated with Anopheles spp. breeding sites and to evaluate the efficacy of a new Bacillus thuringiensis var. israelensis biolarvicide against An. darlingi, as well as its impact on non-target insects under simulated field conditions. BACKGROUND: Triatomine recognition by the public is an important factor in Chagas disease (CD) prevention campaigns. Citizen surveillance has been demonstrated as an effective alternative strategy, increasing the possibility of monitoring vector populations on a broad geographic scale. Thus, our study aimed to explore a database built from pictorial/video records and specimens sent through citizen participation, identify the main groups confused with triatomines, and systematize the data on insect diversity.
METHODS: Due to demand from the local community, the "WhatsBarb" project was establis

Some parasitic diseases, such as malaria, require two hosts to complete their lifecycle: a human and an insect vector. Although most malaria research has focused on parasite development in the human host, the life cycle within the vector is critical for the propagation of the disease. The mosquito stage of the Plasmodium lifecycle represents a major demographic bottleneck, crucial for transmission blocking strategies. Furthermore, it is in the vector, where sexual recombination occurs generating "de novo" genetic diversity, which can favor the spread of drug resistance and hinder effective vac

Insects are the most diverse form of life, and as such, they interact closely with humans, impacting our health, economy, and agriculture. Beneficial insect species contribute to pollination, biological control of pests, decomposition, and nutrient cycling. Pest species can cause damage to agricultural crops and vector diseases to humans and livestock. Insects are often exposed to toxic xenobiotics in the environment, both naturally occurring toxins like plant secondary metabolites and synthetic chemicals like herbicides, fungicides, and insecticides. Because of this, insects have evolved seve

Leishmaniasis is an important disease mediated by the protozoan parasite Leishmania via the bite of the female sandfly insect vector. Leishmaniasis is endemic in the tropical and subtropical regions. The most common form of the disease is cutaneous leishmaniasis, which affects more than 10 million people worldwide and includes at least 1.5 million new cases every year. So far, treatment of the disease relies on unsatisfactory chemotherapy that can be complicated by the rising appearance of drug-resistant parasites. Furthermore, it is challenging to achieve solid control of the insect vector an

Densovirus is classified as invertebrate virus belonging to the subfamily Densovirinae of Parvoviridae family. This group of viruses infects only insects and several densoviruses have been isolated from indigenous mosquitoes and mosquito cell lines. A number of mosquitoes, especially Aedes aegypti and Ae. albopictus are important vectors of viruses, which are the major causes of dengue hemorrhagic fever and yellow fever in humans. As densoviruses do not cause any pathology in humans, these viruses have been proposed to be a potential vector for use in biological control of mosquitoes and insec

Leishmania chagasi, the agent of visceral leishmaniasis in dogs in the Americas has a tropism to the male genital system, particularly the epididymis, prepuce, and glans penis, resulting in shedding of Leishmania in the semen. The goal of this study was to verify the possibility of venereal transmission of L. chagasi. Twelve Leishmania-free bitches, housed in the absence of the insect vector, copulated with multiple naturally infected dogs that were shedding Leishmania in the semen. PCR analysis of serially collected ejaculates indicated that shedding of Leishmania in the semen is intermittent

Despite the importance of circadian rhythms in vector-borne disease transmission, very little is known about its molecular control in hematophagous insect vectors. In Drosophila melanogaster, a negative feedback loop of gene expression has been shown to contribute to the clock mechanism. Here, we describe some features of the circadian clock of the sandfly Lutzomyia longipalpis, a vector of visceral leishmaniasis. Compared to D. melanogaster, sandfly period and timeless, two negative elements of the feedback loop, show similar peaks of mRNA abundance. On the other hand, the expression of Clock

Entomopathogenic fungi and bacteria offer promising alternatives to chemical insecticides for controlling mosquitoes and ticks-key vectors of malaria and Lyme disease. However, their efficacy largely depends on immune interactions between pathogens and hosts. Here, we focus on mosquitoes and ticks as representative disease vectors and dissect conserved and divergent immune responses, from physical barriers to cellular defenses and key humoral pathways, with mechanistic insights informed by insect model systems. Furthermore, we elucidate diverse virulence and immune evasion strategies that enab

The seasonality of African swine fever (ASF) in the summers and outbreaks in farms with high biosecurity levels suggest that the ASF virus (ASFV) may be transmitted by arthropod vectors. Arthropods were collected in this study from 14 pig farms with ASF outbreaks in Korea in 2019 to explore the role of arthropods as potential ASFV vectors. A total of 28,729 arthropods, including 28,508 (99.2%) Diptera, were collected using blacklight traps, insect nets and yellow sticky strips. All arthropod samples were negative for ASFV genomic DNA according to laboratory tests using real-time polymerase cha
