TTX-resistant NMDA receptor-mediated membrane potential oscillations in neonatal mouse Hb9 interneurons.

Conditional neuronal membrane potential oscillations have been identified as a potential mechanism to help support or generate rhythmogenesis in neural circuits. A genetically identified population of ventromedial interneurons, called Hb9, in the mouse spinal cord has been shown to generate TTX-resistant membrane potential oscillations in the presence of NMDA, serotonin and dopamine, but these oscillatory properties are not well characterized. Hb9 interneurons are rhythmically active during fictive locomotor-like behavior. In this study, we report that exogenous N-Methyl-D-Aspartic acid (NMDA)

Female contact activates male-specific interneurons that trigger stereotypic courtship behavior in Drosophila.

We determined the cellular substrate for male courtship behavior by quasinatural and artificial stimulation of brain neurons. Activation of fruitless (fru)-expressing neurons via stimulation of thermosensitive dTrpA1 channels induced an entire series of courtship acts in male Drosophila placed alone without any courting target. By reducing the number of neurons expressing dTrpA1 by MARCM, we demonstrated that the initiation of courtship behavior is significantly correlated with the activation of the transmidline P1 interneurons, the descending P2b interneurons, or both, indicating that these i

Lesions of cortical GABAergic interneurons and acetylcholine neurons in xeroderma pigmentosum group A.

Xeroderma pigmentosum (XP) is a rare genetic disorder caused by inherited disturbances in the nucleotide excision repair system; patients with XP groups A (XP-A), B, D, and G were shown to have progressive neurological disturbances. Particularly, XP-A patients, which account for approximately half of Japanese XP patients, show severe neurological disorders, including mental retardation and epilepsy. Herein, we performed an immunohistochemical analysis of the number of GABAergic interneurons (GABAis), including calbindin-D28K, parvalbumin, and calretinin, in the cerebral cortex and acetylcholin

Uncoordinated firing rate changes of striatal fast-spiking interneurons during behavioral task performance.

Basal ganglia circuits make key contributions to decision making. Distributed, synchronous feedforward inhibition of striatal medium spiny neurons by fast-spiking GABAergic interneurons (FSIs) has been argued to be important for the suppression of unwanted actions, and a deficit in FSIs has been found in human patients with Tourette syndrome. However, no studies have yet examined how striatal FSIs change their activity during behavioral tasks. Here I describe 36 presumed striatal FSIs recorded in rats during well practiced performance of a radial maze win-stay task. Although most FSIs showed r

Cortical interneurons become activated by deafferentation and instruct the apoptosis of pyramidal neurons.

Unlike peripheral nervous system neurons and certain groups of nerve cells in the CNS, cortical projection neurons are tolerant of axonal lesions. This resistance is incongruent with the massive death of pyramidal neurons in age-associated neurodegenerative diseases that proceed along corticocortical connections. Some insights have emerged from our previous work showing that pyramidal cells in piriform cortex undergo classical apoptosis within 24 h after bulbectomy via transsynaptic, but not retrograde, signaling. These findings allow the investigation of cellular and molecular changes that ta

The misplaced mouse Pax6 interneuron subclass: A cross-species transcriptomic reassignment.

Subclass cell types are strongly conserved across mammalian brains, making the systemic absence of a caudal ganglionic eminence (CGE) Pax6 interneuron subclass in mouse transcriptional atlases unexpected. Through cross-species transcriptomic analysis, we identify a Pax6 subclass homolog in mouse and uncover primate-specific divergence in the Sncg subclass. Analyses of electrophysiological and spatial datasets reveal limited subclass-specific structure in either modality, emphasizing the superior resolving power of transcriptomics for subclass distinctions. Our results highlight the pitfalls of

Computational Optimization of Spinal Cord Stimulation for Dorsal Horn Interneuron Polarization.

OBJECTIVES: The proposed mechanisms of spinal cord stimulation (SCS) follow the polarization of dorsal column axons; however, the development of subparesthesia SCS has encouraged the consideration of different targets. Given their relative proximity to the stimulation electrodes and their role in pain processing (eg, synaptic processing and gate control theory), spinal cord dorsal horn interneurons may be attractive stimulation targets. MATERIALS AND METHODS: We developed a computational modeling pipeline termed "quasiuniform-mirror assumption" and applied it to predict polarization of dorsal

Cortical interneuron-mediated inhibition delays the onset of amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis is a fatal disease resulting from motor neuron degeneration in the cortex and spinal cord. Cortical hyperexcitability is a hallmark feature of amyotrophic lateral sclerosis and is accompanied by decreased intracortical inhibition. Using electrophysiological patch-clamp recordings, we revealed parvalbumin interneurons to be hypoactive in the late pre-symptomatic SOD1*G93A mouse model of amyotrophic lateral sclerosis. We discovered that using adeno-associated virus-mediated delivery of chemogenetic technology targeted to increase the activity of the interneurons wit

Calretinin interneuron density in the caudate nucleus is lower in autism spectrum disorder.

Autism spectrum disorder is a debilitating condition with possible neurodevelopmental origins but unknown neuroanatomical correlates. Whereas investigators have paid much attention to the cerebral cortex, few studies have detailed the basal ganglia in autism. The caudate nucleus may be involved in the repetitive movements and limbic changes of autism. We used immunohistochemistry for calretinin and neuropeptide Y in 24 age- and gender-matched patients with autism spectrum disorder and control subjects ranging in age from 13 to 69 years. Patients with autism had a 35% lower density of calretini

Organization and neurochemical properties of intersegmental interneurons in the lumbar enlargement of the adult rat.

Intersegmental interneurons with relatively short axons perform an important role in the coordination of limb movement but surprisingly little is known about their organization and how they contribute to neuronal networks in the adult rat. We undertook a series of anatomical tract-tracing studies to label cell bodies and axons of intersegmental neurons in the lumbar cord and characterized their neurochemical properties by using immunocytochemistry. The b-subunit of cholera toxin was injected into L1 or L3 segments of seven rats in the vicinity of lateral or medial motor nuclei. In L5 lumbar se

The contribution of the pleural type 12 interneuron to swim acceleration in Clione limacina.

The pteropod mollusc, Clione limacina, swims by alternate dorsal-ventral flapping movements of its wing-like parapodia. The basic swim rhythm is produced by a network of pedal swim interneurons that comprise a swim central pattern generator (CPG). Serotonergic modulation of both intrinsic cellular properties of the swim interneurons and network properties contribute to swim acceleration, the latter including recruitment of type 12 interneurons into the CPG. Here we address the role of the type 12 interneurons in swim acceleration. A single type 12 interneuron is found in each of the pleural ga

Regeneration after Spinal Cord Injury: A Review on the Crucial Aspects of V2a Interneurons.

Spinal cord injuries (SCI) are extremely difficult to treat due to the limited capacity for neural regeneration across the injury site. However, V2a interneurons have been a point of interest in SCI research over the last decade, as they have been shown to contribute to the promotion of neuroplasticity after injury. These excitatory interneurons contain either long or short projections that are effective at driving rhythmic motor firing. By possessing ipsilateral, contralateral, or propriospinal projections, subtypes of V2a interneurons expressing the visual system homeobox-2 (Vsx-2) gene have

Hyaluronic Acid-Alginate Hydrazone Crosslinked Hydrogels Support the Generation and Maturation of V2a Interneurons.

Injury to the central nervous system (CNS) causes inflammation, cell death, and glial scar formation that inhibits tissue repair. Injectable hydrogels modified with extracellular matrix (ECM)-derived peptides can provide biochemical cues to promote neural tissue repair and serve as a vehicle to deliver therapeutics across the blood-spinal/blood-brain barrier in a minimally invasive manner. We developed an injectable hydrazone crosslinked hyaluronic acid-alginate (HA-Alg) hydrogel for neural tissue repair. Autism-like behavior induced by conditional ablation of the Bassoon gene in GABAergic interneurons.

Synaptic dysfunction and resulting imbalance of excitatory vs. inhibitory transmission are fundamental aspects of autism spectrum disorders (ASD). Here we addressed the role of inhibitory synapse disturbances in ASD employing mice with a conditional ablation in GABAergic forebrain interneurons of the presynaptic active zone scaffolding protein Bassoon (Bsn), a key factor in synaptic development, activity and maintenance. The conditional Bsn gene knockout resulted in a reduction of synaptic vesicles and reduced synaptic efficacy of affected inhibitory synapses as well as diminished GABAergic in

4E-BP2-dependent translational control in GABAergic interneurons is required for long-term memory.

mRNA translational repression by eukaryotic initiation factor 4E-binding proteins (4E-BPs), plays a critical role in synaptic plasticity and the formation of long-term memory (LTM). Among the three 4E-BP paralogs, 4E-BP2 is the predominant form expressed in neurons, and its full-body deletion in mice causes memory deficits. Mice lacking 4E-BP2 in GABAergic inhibitory interneurons, but not excitatory neurons, display autistic-like behaviors and deficits in object location and recognition. The specific mRNAs translationally regulated by 4E-BP2 in GABAergic interneurons, and how they contribute t

Alteration of hippocampal parvalbumin interneurons underlies memory impairment in rat model of Parkinson's disease.

Cognitive decline is a major non-motor symptom in patients with Parkinson's disease (PD) that can be present as early as the prodromal stage. As a multisystem neurodegenerative syndrome, PD is associated with disturbances in various neurotransmitters, including dopamine, acetylcholine, serotonin, noradrenaline, glutamate, and gamma-aminobutyric acid (GABA). While the roles of dopaminergic and cholinergic deficiencies in cognitive impairment in PD are well documented, the contribution of the GABAergic system is less clear. We investigated spatial and recognition memory, along with changes in hi

Alcohol attenuates CRF-induced excitatory effects from the extended amygdala to dorsostriatal cholinergic interneurons.

Alcohol relapse is associated with corticotropin-releasing factor (CRF) signaling and altered reward pathway function, though the precise mechanisms remain unclear. Here, using both mice and rats, we investigated how CRF modulates cholinergic interneurons (CINs) in the dorsal striatum, a region critical in mediating cognitive flexibility and action selection. Using monosynaptic and retrograde circuit tracing, we identified direct inputs from CRF-expressing (CRF+) neurons in the central amygdala (CeA) and bed nucleus of the stria terminalis (BNST) to dorsal striatal CINs. We showed that CINs ex

Activity and Functional Connectivity of Lumbar Spinal Interneurons at Rest: Irregular, Sparse Yet Structured.

Spinal interneurons are central to sensorimotor integration. Even in the absence of overt sensory inputs or motor output, spinal interneurons remain active, yet their functional organization across spatial and temporal scales remains poorly understood. We characterized the discharge properties and functional connectivity of 2,847 lumbar spinal interneurons recorded in vivo from quiescent decerebrate cats (3 males, 5 females) using microelectrode arrays inserted into rostral (L3-4) and caudal (L5-7) spinal segments at superficial (0-1500 µm) and deep (1500-3000 µm) depths.
